Extraction of the Skin-Whitening Ingredients from Hippophae rhamnoides L. Fruits: Optimization Using Solubility Parameters

산자나무 열매로부터 피부미백 성분의 추출: 용해도 파라미터를 이용한 최적화

Abstract

In this study, the optimal solvent for extracting L-ascorbic acid-a skin-whitening agent-from the fruit of Hippophae rhamnoides L. was determined using solubility parameters. The optimization process employed a central composite design (CCD) model of response surface methodology (RSM), with Hansen solubility parameters and extraction temperature set as independent variables. The optimal extraction conditions were found to be a solubility parameter of 36.7 MPa1/2 and an extraction temperature of 39.2 ℃. Under these conditions, the predicted responses were whitening activity of 49.4%, antioxidant activity of 87.4%, and yield of 322.5 mg/g. Experimental validation yielded results of 52.4% whitening activity, 87.5% antioxidant activity, and 327.8 mg/g yield, with an error margin of ± 2.5%.

본 연구에서는 용해도 파라미터를 이용하여 산자나무(Hippophae rhamnoides L.) 열매로부터 피부미백 성분인 L-ascorbic acid을 추출할 수 있는 최적 용매를 최적화하였다. 최적화 과정은 반응표면분석법 중 중심합성계획모델을 이용하였으며, 독립변수로는 Hansen 용해도 파라미터와 추출 온도를 설정하였다. 최적 추출 조건은 용해도 파라미터 𝛿t = 36.7 MPa1/2, 추출 온도 = 39.2 ℃로 산출되었으며, 예측된 반응치는 미백능 = 49.4%, 항산화능 = 87.4%, 수율 = 322.5 mg/g로 나타났다. 실제 실험을 통한 결과는 미백능 = 52.4%, 항산화능 = 87.5%, 수율 = 327.8 mg/g으로 측정되어 오차율은 ± 2.5%이다.
