Proline is known as an osmotrotectant to enhance tolerance against both salt and dehydration stresses. A P5CS ($\Delta^1$-pyrroline-5-carboxylate synthetase) plays a major role in regulation of synthesis of proline. An overexpression of the mothbean P5CS gene in transgenic tobacco plant increased the levels of proline and osmotolerance. In an attempt to look for the possibility to use content of proline as well as a level of P5CS gene expression as molecular markers for salt tolerance, the amounts of proline and transcript levels of P5CS were measured as functions of either concentration of NaCl or length of treatment period among different species of zoysiagrass. Hybridzoysia showed the highest level of proline ($329\mu\textrm{g}$/g.f.w.) among five different species of zoysiagrass at 250 mM NaCl in 24 hours. The level of P5CS transcript was also the highest in the hybridzoysia at 250 mM NaCl in 24 hours. The transcriptions of P5CS gene were induced at the rates of 1.2, 1.2, 1.8, and 1.8, upon treatment of 250 mM NaCl in Z. japonica, Z. matrella, Z. sinica and hybridzoysia respectively. Based on a correlation between the level of P5CS transcript and the proline content among different species of zoysiagrass, a comparative structural analysis of the gene for P5CS from either Z. sinica or hybridzoysia may lead to an understanding of mechanism for salt tolerance shown differently among zoysiagrasses.
Since $CO_2$ concentrations in industrial flue gases are usually 10%-20%, one of the prerequisites for efficient $CO_2$ removal by algae is the level of tolerance of microalgal species to exposure to high concentrations of $CO_2$. A newly isolated microalgal strain, Chlorella sp. MRA-1, could retain growth with high concentrations of $CO_2$ up to 15%. The highest lipid productivity for Chlorella sp. MRA-1 was 0.118 g/l/day with a 5% $CO_2$ concentration. Octadecenoic acid and hexadecanoic acid, the main components of biodiesel, accounted for 70% of the total fatty acids. A lipid content of 52% of dry cell weight was achieved with limited amounts of nitrogen. Chlorella sp. MRA-1 seems to be an ideal candidate for biodiesel production when cultured with high concentrations of $CO_2$.

BACKGROUND: Pre-harvest interval and decline pattern of thiamethoxam were determined in kiwifruit using liquid chromatography-tandem mass spectrometry (LCMS/MS). The study was carried out to propose import tolerance using OECD maximum residue limit (MRL) calculator for the export promotion of kiwifruit to Taiwan. METHODS AND RESULTS: The thiamethoxam residue in kiwifruit was determined by using the LC-TriQ-MS/MS with the analytical process to set up the import tolerance under greenhouse conditions for Taiwan. Excellent linearity was observed for all of the analytes with a determination coefficient (R2)≥0.99. The limit of quantification was determined to be 0.01 mg/kg for both thiamethoxam and clothianidin in kiwifruit. Linearity was determined from the co-efficient of determinants (R2) obtained from the seven-point calibration curve. The standard calibration curve showed as follows; 1) Site 1 (Gimje): y = 944,406X + 1,583 (R2=0.9995), 2) Site 2 (Goheung): y = 1,356,205X + 934 (R2=0.9983), and 3) Site 3 (Jangheung): y = 1,239,937X - 3,090 (R2=0.9908). The residue of thiamethoxam in the kiwifruit for three decline trials showed the range of 0.35 to 0.56 mg/kg in site 1 (Gimje), 0.24 to 0.55 mg/kg in site 2 (Goheung), and 0.28 to 0.42 mg/kg in site 3 (Jangheung), respectively. However, clothianidin was not detected in all of the treatments. The maximum residual amounts (decline) in the samples, sprayed according to the safe-use standard for thiamethoxam 10% WG in kiwifruit (30 days before harvest, 3 sprays every 7 days) were 0.56 mg/kg in site 1, 0.55 mg/kg in site 2, and 0.42 mg/kg in site 3, respectively. CONCLUSION(S): The import tolerance (IT) of thiamethoxam for kiwifruit may be proposed to be 0.9 mg/kg by using the OECD MRL calculator.
Background: In dental intravenous sedation, continuous intravenous infusion of a low-dose drug requires an infusion pump such as a syringe pump. To develop a new syringe pump for clinical use, the functions of the pump must meet certain international standards. Various safety and efficacy tests must be performed on the syringe pump, as stipulated by these standards, and an approval must be received from the approving agency based on such test results. Methods: The authors of the present study developed a novel syringe pump and performed efficacy evaluation by testing its infusion speed at 1 and 25 ml/h, and infusion performance testing at 2 and 24 h. Moreover, performance evaluation was conducted by comparing the novel pump to an existing pump with the infusion speed varied from 1 to 5 ml/h. Results: In the efficacy testing on the newly developed syringe pump, infusion with the infusion speed initially set to 1 ml/h resulted in infusion speeds of 1.00 and 0.99 ml/h in the 2- and 24-h assessment, respectively. Changing the infusion speed setting to 25 ml/h resulted in an infusion speed of 25.09 and 23.92 ml/h in the 2- and 24-h assessment, respectively. These results show no significant differences when compared with other commercially available pumps. Conclusions: The efficacy testing of the newly developed syringe pump showed the accuracy to be within tolerance. Based on these findings, we believe that the newly developed syringe pump is suitable for clinical use.
The laser welding and its analysis of thin-sheet carbon steels were carried out with high power $CO_{2}$ laser. The main factor of weld quality of laser welding is gap and edge quality. This work was preformed to focus on the gap tolerance problem during laser welding. First, bead on plate welding of thin sheet was examined to investigate the effect of laser welding variables, and to obtain optimum welding condition. Butt welding was also carried out to show the effect of gap on the laser weldability of thin sheet. In order to investigate the effect of gap on formability of welded thin sheet, LDH test was caried out. At high welding speed, the partial penetration was obtained by low heat input. Otherwise, porosity was formed in the bead at low weld speed because of too much heat input. The optimum welding condition of welding was derived from bead width, penetration and hardness property. The maximum gap tolerance on laser welding was observed to be about 0.2mm. This gap size has good relationship with beam size of laser spot(about 0.3mm). The formability of welded sheet was about $80{\%}$ value of base metal and the gap size has not affected on the formability, although weld quality is dependent on the gap size.
This study was conducted to compare probiotic activities and physiological functions of Bifidobacterium longum Mk-G7 with weveral commercial and type strains of bifidobacteria. bif. longum MK-G7 showed the highest acid tolerance against HCl and acetic acid, whereas bif. infantis Y-1 showed the lowest acid tolerance and more than 4 log cycles of viable cell count decreased due to acid injuty. Viable cell counts of bifidobacteria strains decreased more than 1.5 log cycles owing to oxygen toxicity, with the exception of Bif. longum MK-G7, Bif. infantis Y-2, Bif. longum Y-3, Bif. longum Y-6, and Bif. longum RD-13 showed the highest bile tolerance, whereas Bif. longum MK-G7 showed a medium level of bile tolerance. Only Bif. longum MK-G7 howed much higher antibiotic resistance against both tetracycline and penicillin-G in the MIC(minimum inhibitory concentration) level of 24.8 mg/I and 0.52mg/I, respectively. Bif longum Y-6, and Bif. bifidum ATCC 29539 showed more than 80% of anti-mutagenicity against NQO(4-nitroquinolinel-oxide). Since the production of cytokines such as $TNF(tumor necrosis factor)-{\alpha}$ and IL (interleukin)-6, and NO(nitric oxide) in the macrophage cell line Raw 264.7 cells increased as Bif. longum MK-G7 cell concentration increased, ti was suggested that Bif. longum MK-G7 is able to enhance immunopotentiating activity in vitro. When freeze-dred Bif. longum MK-G7 was administered to mice at the dose of 1,2,4, and 6 g/kg of body weight, all of the mice survived in all feeding groups, proving the GRAS(generally recognized as safe) status of Bif. longum MK-G7. When fermented milk containing Bif. longum MK-G7 was administered to human volunteers, viable cell count of total bifidobacteria and anaerobes in the feces increased up to 0.5 log cycles more than before the administration. In particular, Bif. logum MK-G7 ingibited the growth of Bacteroides at the level of 1.0-1.5 log cycles.
For laser welding in shipbuilding industry, gap bridging capability is one of the most important characteristics to achieve the high productivity and good weld quality. Recently, laser-GMA hybrid welding process is regarded as a distinctive method to overcome the tight gap tolerance with improving the productivity. In this study, the influence of process parameters on the bead formation was experimentally analyzed and the relationship between the process parameters and geometric imperfections was investigated. It was revealed that undercut, excessive weld metal, excessive penetration and incompletely filled groove were the major geometric imperfections. The optimized wire feeding and arc pressure were necessary to ensure the gap bridging ability. The approach to select the process parameters was conducted for butt welding with up to 2mm joint gap, in which the sound weld beads were generated without changing the welding speed.
BACKGROUND: Recent studies have described the importance of microbes and enzymes that can compost food waste. This study was carried out to improve production of protease of isolated microbes from food waste. METHODS AND RESULTS: Seven bacteria isolated from various sources were screened for protease production by adding skim milk into the agar medium. About 7 microbes producing protease were tested, and strain JH-35 showed the highest protease activity among them. The strain was identified as Bacillus amyloliquefaciens JH-35 based on morphological, cultural, physiological characteristics and 16S rRNA. In the fermentation experiment, the assay B. amyloliquefaciens JH-35 showed the highest protease activity in the condition of 1% glucose, 1.5% yeast extract and 0.2%$ K_2HPO_4$. The optimal condition of culture with temperature $35^{\circ}C$, initial pH of 7 and shaking speed of 200 rpm and 24 hr. CONCLUSION: The protease of the B. amyloliquefaciens JH-35 had its activity at pH 7 and the optimal culture time was 24 hr. Also, B. amyloliquefaciens JH-35 was high salt tolerance. Our results suggest that B. amyloliquefaciens JH-35 from food waste may have the potential to degrade protein and carbohydrate in food waste.
For three cultivars of chilling-sensitive cucumber plants, chilling sensitivity was evaluated in terms of photosynthetic activity using Chl fluorescence techniques. Low-temperature treatment caused a decrease in photosynthetic activities of cucumber leaves, measured as CO$_2$ exchange, as well as the decrease in the stomatal conductance. FR of the three cultivars decreased after chilling for 24 h in light and the extent of decline of F$_R$ was the greatest in 'Chosaeng' cultivar. When these plants were recovered from light-chilling, 'Chosaeng' and 'Samchuk' cultivars did not fully restore the original value of F$_R$ after 24 h of recovery, in contrast to 'Ilmi' cultivar which showed a rather efficient recovery. The results of FR study showed that 'Chosaeng' was most susceptible, whereas Ilmi was most resistant, to chilling among the three cultivars of cucumber plants. When quenching coefficients for chlorophyll fluorescence was analyzed after chilling the cucumber plants for 24 h in light, 'Chosaeng' elicited more rapid declines in the coefficients for photochemical quenching (qQ), non-photochemical quenching (qNP) and energy-dependent quenching (qE) than 'Ilmi' and 'Samchuk'. The implications of these observations are discussed in relation to the growth habits of the respective cultivars in the field. The results showed that measurement of chlorophyll fluorescence was an effective means of screening chilling tolerance of cucumber plants. Furthermore, the study on the chlorophyll fluorescence induction and fluorescence quenching charactersitics showed that low temperature could accelerate inhibition of photosynthesis in chilling-sensitive plants, by limiting Calvin cycle activity and disrupting, in part, the energy dissipation mechanims of the photosystem II.
