Manure recycle is an emerging issue in agriculture in Korea these days. Farmers are keeping eye on legume mixture with grasses for nitrogen fixation and transfer to companion crops by legumes. We had a trial to investigate the effect of different legume mixtures on nitrogen fixation and transfer in spring soil. The treatment was arranged in a randomized complete block design with three replications. Three different mixtures were used(rye+hairy vetch, Italian ryegrass+crimson clover, oat+pea) and sowed in pots with paddy soil from western part of Korea(Seo Chon County). Pots with rye+vetch were transplanted from field of RDA(rural development administration) in Suwon on 2 February 2007 and other mixture treatments were sowed on early March with different sowing rate(7:3=Italian:crimson, 6:4=oat:pea). $(^{15}NH_4)_2SO_4$ solution at. $99.8\;atom%^{15}N$ was applied to the each pot at the rate of $2kg\;N\;ha^{-1}$. Application was done on 6 April at rye+vetch pots and remainder were applied on April 16. Forage were harvested from each pot at ground level in heading stage and separated into legume and grass. Total N content and $^{15}N$ value were determined using a continuous flow stable isotope ratio mass spectrometry. DM yield of rye+vetch, Italian+crimson and oat+pea were 6,607, 3,213 and 4,312 kg/ha, respectively. Proportion of N from fixation was 0.73(rye+vetch), 0.42(Italian+crimson) and 0.93(oat+pea). The percentages of N transfer from legume to grass were from 61% to 24% in different method by treatment and -35% to 21% in isotope dilution method.

By combining in situ hybridization (ISH) and immunocytochemistry (IC), microscopic topological localization of mRNAs and proteins can be determined. Although this technique can be applied to a variety of tissues, it is particularly important for use on neuronal cells which are morphologically complex and in which specific mRNAs and proteins are located in distinct subcellular domains such as dendrites and dendritic spines. One common technical problem for combined ISH and IC is that the signal for immunocytochemical localization of proteins often becomes much weaker after conducting ISH. In this manuscript, we report a simplified but robust protocol that allows immunocytochemical localization of proteins after ISH. In this protocol, we fix cultured cortical or hippocampal neurons with 4% paraformaldehyde (PFA), rinse briefly in PBS, and then further fix the cells with $-20^{\circ}C$ methanol. Our method has several major advantages over previously described ones in that (1) it is simple, as it is just consecutive routine fixation procedures, (2) it does not require any special alteration to the fixation procedures such as changes in salt concentration, and (3) it can be used with antibodies that are compatible with either methanol (MeOH-) or PFA-fixed target proteins. To our best knowledge, we are the first to employ this fixation method for fluorescence ISH + IC.
The objective of the present study was to assess the prevalence, occurrence pattern, and clinical outcomes of transfixation pinning and casting (TPC) for fractures in Korean native calves, and to identify the advantages of the procedure. The study investigated 52 cases of bone fractures documented from January 2017 to December 2020. Hindlimb fractures (72%) were the most common in bone fractures (n = 50). The distribution of fractures was highest in the tibia (n = 15), followed by the metatarsal (n = 12), femur (n = 9), radius/ulna (n = 8), metacarpal (n = 3), humerus (n = 3), mandible (n = 1), and caudal vertebrae (n = 1). All cases were diagnosed via radiographic evaluation. Closed fractures (86%) were the most common, followed by open fractures (14%). Surgery was performed on 34 calves using intramedullary pin (IM pin), TPC, cross pin, flexible wire (F-wire), and/or plate fixation. The rest of the calves were treated with external coaptation (n = 12) or were not treated (n = 6). Subsequently, 24 calves with follow-up records were evaluated in the bone fracture cases (n = 50). Bone union was observed in 15 (62%) calves; 3 (12%) calves showed non-union; 4 (16%) calves were dead after surgery; and 2 (8%) calves were euthanized at the owner's request. Among the 24 follow-up surgery cases, the most successful cases were tibia fractures (75%) surgically treated using TPC and/or IM pin. These findings suggest that TPC surgery is effective in the management of fractures in Korean native calves.
The aerobic heterotrophic bacteria in the histosphere associated with grasses (Gramineae, Caryphyllaceae, Crucifereae) and rice cultivars in saline and reclaimed saline paddy soils were varied with species and rice cultivars. The fraction of aerobic heterotrophic $N_2$-fixing bacteria to the total aerobic heterotrophic bacteria were averaged to eighteen percent in the histosphere of grasses and rice. Acetylene reducing activity of these bacteria were ranged from 1 to 24 n mole/tube/hr. Most of the bacteria strains were predominated of hydrogen utilizing bacteria. The majority of these bacteria were closed to Pseudomonas, Azospirillum, Klebsiella and Agrobacter.
The maximum nitrogen fixation activity of the associative, microaerobic and acid tolerant bacteria, Azospirillum amazonense Kp1 was obtained with 0.2Kpa of $O_{2}$ and showed a reversible inhibition by the higher concentrations. Ammonium treatment caused a gradual inhibition of the activity up to 350mM. The nitrogenase systems were purified by gradient chromatography on DEAE-52 cellulose, heat treatment and preparative PAGE. The MoFe protein showed molecular weight of 210,000 including two nonidentical subunits with apparent molecular weights of 55,000 and 50,000 and an isoelectricpoint of 5.2 and contained 2, 24 and 28 atoms of Mo, Fe and acid labile S per molecule. The Fe protein revealed molecular weight of 66,000 including two types of subunits with molecular weights of 35,000 and 31,000 and an isoelectric point of 4.6, and contained 4 atoms of Fe and 6 atoms of S per molecule. The maximum specific nitrogenase activity attained 2,200 and 1,700nM $C_2H_4mg^{-1} min^{-1}$, respectively for MoFe and Fe proteins at pH7 and $35^{\circ}C$. The activity was lost after 10 and 30 days under the cold room ($4^{\circ}C$) condition for Fe and MoFe proteins, respectively.
The effects of two-step fixation of steaming and baking on the dischargeability of cotton fabrics dyed with C.I. Reactive Black 5(Bl-5) were investigated when the concentrations of $K_2CO_3$ and benzaldehyde sodium bisulfite(BASB) were increased over 120/kg. Remarkably increased dischargeability resulted from baking for 3 min or more at 160t after steaming for 8 min or more at $102^\circ{C}$, but 120g/kg or more amounts of $K_2CO_3$ and BASB(50%) had little influence on dischargeability. Therefore the discharge mechanism can be suggested that covalent bonds between cellulose and Bl-5 undergo $S_N2$ attack by hydroxide ion formed by the reaction of $K_2CO_3$ and water in steaming at $102^\circ{C}$ first and then, through transition states they are cleavaged in baking at 160t to yield hydrolyzed Bl-S and compounds of BASB and Bl-5 isolated from fiber, which are undyeable and removed by washing. The effect of urea, one of the hydrotrope agents, on discharge printing was also studied. The result which dischargeability was greatly improved by increasing the steaming time from 8 min to 15 min at $102^\circ{C}$ or by increasing the amount of urea obviously shows that water in steaming and urea in print paste play an important role in discharge printing. And as an increase of the baking time from 5 min to 7 min at $160^\circ{C}$ makes it possible to improve dischargeability, it is once more confirmed that high temperature of about 160t is exactly required to discharge the dyed Bl-5. The colored discharge printing demands a more amount of urea because urea contributes to the putting color fixation as well as the discharge reaction.
Purpose: This study aimed to compare the effects of three interventions on pain, blood pressure, and pulse rate during infiltration anesthesia in patients about to undergo gamma knife surgeries. Methods: The three interventions employed in a university-affiliated Hospital in J City, South Korea were as follows: EMLA cream plus Vapocoolant spray (Vapocoolant, n=30), EMLA cream plus 10.0% Lidocaine spray (Lidocaine, n=30), and EMLA cream only (EMLA, n=30). The equivalent control-group pre test - post test study design was used. Pain was assessed subjectively using the numeric rating scale (NRS) and objectively using a Galvanic Skin Response (GSR) tester. NRS scores were assessed after infiltration anesthesia and the GSR was assessed during infiltration anesthesia. Blood pressure and pulse rate were assessed twice: before and after infiltration anesthesia. Data were collected between August 3, 2016 and March 24, 2017. Results: NRS scores after infiltration anesthesia and the GSR during infiltration anesthesia were significantly lower in the Vapocoolant group than in the Lidocaine and EMLA groups (F=13.56, p<.001 and F=14.43, p<.001, respectively). The increase in systolic blood pressure (F=4.77, p=.011) and in pulse rates (F=4.78, p=.011) before and after infiltration anesthesia were significantly smaller in the Vapocoolant group than in the Lidocaine and EMLA groups; however, no significant differences were observed in diastolic blood pressures (F=1.51, p=.227). Conclusion: EMLA cream plus Vapocoolant spray was the most effective intervention to relieve pain and to lower increase in systolic blood pressure and pulse rate caused by infiltration anesthesia for stereotactic frame fixation. Thus, application of Vapocoolant spray in addition to EMLA cream is highly recommended as a nursing intervention for patients undergoing gamma knife surgeries.

The purpose of this research was to investigate the effect of P fertilization and cutting fkquency without N fertilizer on nutritive value and nitrogen fixation activity of alfalfa. A field experiment was conducted on the experimental farm of Kongju National University at Yesan from 1994 to 1996. The experimental design was a split plot design with two P levels as main plot and two cutting frequencies as the sub-plot. The results obtained are summarized as follows. Available phosphate, exchangeable Ca and Na in the soil were slightly increased with P fertilizer and basal treatment of lime. Average DM yield was 1.5 ton and 1.4 ton in second and third year. There was no significant difference within P level and cutting kquency. The first cut showed higest CP value as cutting times increased the value tend to be lowered. Crude protein content in 3 and 4 cutting regimes were from 16.3 to 16.6% 17.4 to 17.9% respectively, but no significant effect were found among the main and sub-plot. ADF and NDF in 1995 were ranged from 38.2 to 39.9, 45.1 to 58.1% respectively. ADF was 2% more higher in 1996 compared to previous year and NDF was slightly higher in 1996. There was significant difference of ADF between main plot in 1995. .P, Ca, K and Mg of 200kg P level with 3 times cutting treatment in 1995 and 1996 were 0.32 and 0.23, 0.95 and 0.84, 3.24 and 2.87, and 0.36 and 0.26. Significant difference of K and Mg between cutting frequency was observed in 1995. Nitrogen fixing activity of 400kg P fertilizer plots were higher than 200kg P level and 3 times cutting plot also showed higher acetylene reduction activity than that of 4 cutting per year. Phosphate level and cutting frequency treatment of did not significantly affect nitrogen fixation activity.

Photobiological hydrogen production by nitrogen-fixing unicellular cyanobacteria has long been considered to be an environmentally sound and very promising method for the future supply of renewable clean energy. Using six Korean nitrogen-fixing unicellular cyanobacterial strains and the Synechococcus sp. strain Miami BG043511 we performed cultivation experiments to find out the strain-specific optimal temperature for population growth and $H_2$ production. Under $20^{\circ}C$ the population growth of all the tested strains was significantly retarded in contrasts to the faster and higher growth under 25, 30 or $35^{\circ}C$. The highest growth rates in all the 7 strains were measured under $30^{\circ}C$ while the maximal biomass yields were under $30^{\circ}C$ (strains CB-MAL 026, 054, and 055) or $35^{\circ}C$ (strains 002, 031, 058, and Miami BG043511). The difference between the maximal biomass yields at $30^{\circ}C$ and $35^{\circ}C$ was not greater than 10%. The quantity of photobiologically produced $H_2$ was only slight larger under $35^{\circ}C$ than that under $20^{\circ}C$. Our result may suggest a two-step process of $H_2$ production which includes rapid and sizable production of biomass at $30^{\circ}C$ and the following high $H_2$ production at $20^{\circ}C$ by the test strains of marine nitrogen-fixing unicellular cyanobacteria.
A green microalga, Chlorella vulgaris UTX 259, was cultivated in a bench-scale raceway pond. During the culture, 15%(v/v) $CO_2$ was supplied and industrial wastewater discharged from a steel-making plant was used as a culture medium. In a small scale culture bottle, the microalga grew up to 1.8 g $dm^{-3}$ of cell concentration and ammonia was completely removed from the wastewater with an yield coefficient of 25.7 g dry cell weight $g^{-1}\;NH_3-N$. During the bottle-culture, microalga was dominant over heterotrophic microorganisms in the culture medium. Therefore, the amount of carbon dioxide fixation could be estimated from the change of dry cell weight. In a semi-continuous operation of raceway pond with intermittent lighting (12 h light and 12 h dark), increase of dilution rate resulted in increase of the ammonia removal rate as well as the $CO_2$ fixation rate but the ammonia removal efficiency decreased. Ammonia was not completely removed from the medium (wastewater) of raceway pond which was operated in a batch mode under a light intensity up to 20 klux. The incomplete removal of ammonia was believed due to insufficient light supply. A mathematical model, capable of predicting experimental data, was developed in order to simulate the performance of the raceway pond under the light intensity of sun during a bright daytime. Simulation results showed that the rates of $CO_2$ fixation and ammonia removal could be enhanced by increasing light intensity. According to the simulation, 80 mg $dm^{-3}$ of ammonia in the medium could be completely removed if the light intensity was over 60 klux with a continuous lighting. Under the optimal operating condition determined by the simulation, the rates of carbon dioxide fixation and ammonia removal in the outdoor operation of raceway pond were estimated as high as $24.7 g m^{-2} day^{-1}$ and $0.52 g NH_3-N m^{-2} day^{-1}$, respectively.
