Effects of Temperature and Food on the Nymphal Development and Adult Longevity of the Green Mirid Bug, Cyrtorhinus lividipennis Router (Hemiptera: Miridae) and Its Egg Predation (온도 및 먹이가 등검은황록장님노린재의 약충발육과 성충수명 및 난 포식량에 미치는 영향)

  • This study was conducted to determine the effects of temperature and food onthe nymphal development and adult longevity of the green virid bug, Cyrtorhinus lividipennis Router. Nymphal period was 13.2 to 13.7 days at 24$^{\circ}C$, 10.4 to 10.9 days at 28$^{\circ}C$, and 9.0 to 9.3 days at 32$^{\circ}C$. Number of eggs predated by nymph was 10.6-14.3 at 24$^{\circ}C$, 7.5-9.2 at 28$^{\circ}C$, and 5.8-7.5 at 32$^{\circ}C$. Amount of predation was highest on BPH, followed by SBPH, WBPH, and GLH. Number of nympal moulting was mostly four times (55-75%) followed by three times (25-45%), regardless of temperature and food. Percent emergence rate was 52-70%, 48-66% and 22-38% at 24$^{\circ}C$, 28$^{\circ}C$ and 32$^{\circ}C$, respectively, showing a tendency to decrease with increasing temperature, and was affected by foods. Adult longevity, when supplied with one food, was 19.8 to 22.5 days at 24$^{\circ}C$, 15.6 to 17.0 days at 28$^{\circ}C$, and 10.3 to 11.7 days at 32$^{\circ}C$. There was no significant difference among foods in terms of adult longevity, but it tended to be slightly longer when provided with four foods than with one food. Number of eggs predated by an adult, when supplied with one food, was 35.7-54.2, 31.6-44.6 and 18.1-28.2 at 24$^{\circ}C$, 28$^{\circ}C$ and 32$^{\circ}C$, respectively. It was highest on BPH, followed by SBPH, WBPH and GLH regardless of temperature. Number of egg Predated by an adult, when supplied with four foods, was 4.0-28.9, 2.9-29.0 and 17-13.6 at 24$^{\circ}C$, 28$^{\circ}C$ and 32$^{\circ}C$, respectively, and was significantly different among foods supplied. The results suggest that the significantly different predation amount by adult was due to food preference amoung the different foods.

Effects of Tempearture and Food Source on Pupal Development, Adult Longevity and Oviposition of the Tobacco Cutworm, Spodoptera litura Fabricius (온도 및 기주조건이 담배거세나방의 용발육, 성충수명 및 산란에 미치는 영향)

  • This study was conducted to determine the effect of temperatures, 24"C, 28$^{\circ}$C and 32"C,and food sources on pupal development, adult longevity and oviposition of tobacco cutworm, Spodopteralitura Fabricius. Percent pupation of S. litura was became higher at higher temperature from 23% to 88%.And its percent pupation was highest on soybean leaf (SL), followed by perilla leaf (PL), sweet potato leaf(SPL) and artificial diet (AD). Pupal weight ranged from 0.28g on SPL to 0.40 g on PL and was tended toheavier with decreasing temperature. The mean pupal duration was 14, 10 and 7 days at 24"C, 28$^{\circ}$C and32"C, respectively. Percent emergence was in range of 21% to 89% with higher percent emergence as thetemperature increased and both 28$^{\circ}$C and 24$^{\circ}$C was highest on SL, followed by PL, SPL and AD, but at24$^{\circ}$C the order was SPL, PL, SL and then AD. Preoviposition duration was 3.2 days at 24"C, 2.8 days at28$^{\circ}$C and 2.5 days at 32$^{\circ}$C. Adult longevity became shorter as the temperature increased from 6.4 to 3.9days. Male longevity was longer than that of female. Adults lived longest when they were reared on PL,followed by SL, SPL and AD. Total number of eggs laid per female varied from 803 to 1,441 regardlessof the treatments, but those were significantly more on natural foods than on AD. Number of eggs per eggmass was 97.4 at 24$^{\circ}$C 155.8 at 28$^{\circ}$C and 104.7 at 32$^{\circ}$C. Number of egg mass was 12.0, 6.7 and 11.3 at24"C, 28$^{\circ}$C and 32"C, respectively.4"C, 28$^{\circ}$C and 32"C, respectively.

Effect of Temperature, Salinity and Preservation Method on Hatching Rate of Resting Egg of Korean Rotifer, Brachionus plicatilis (S-type) (수온, 염분 및 보관방법에 따른 한국산 Rotifer, Brachionus plicatilis (S-type) 내구란의 부화율)

  • The effect of temperature ($24\~36^{\circ}C$ and salinities ($5\~30$ ppt) on hatching rate of the resting egg of the Korean rotifer, Brachionus plicatilis was investigated. The highest hatching rate of the resting egg was $85.7\%$ at $28^{\circ}C$ and 15 ppt. The hatching rates of the resting eggs with salinities were not significant at $326{\circ}C$ and $36^{\circ}C$. Hatching was faster at higher temperature. The resting eggs stored at $5^{\circ}C$ in dark hatched simultaneously within two days, however, those at $28^{\circ}C$ in dark hatched intermittently within 14 days. But, final hatching rate of the resting egg at $28^{\circ}C$ was higher at $28^{\circ}C$ than that at $5^{\circ}C$. With regard to drying temperature and time of resting egg, the resting egg dried at $30^{\circ}C$ for 1 hour showed the highest hatching rate ($46.4\%$).

Development of the Tea Red Spidermite, Tetranychus kanzawai (간자와응애의 발육(發育)에 관한 연구(硏究))

  • The tea red spider mite has recently outbroken as the major pest of tea at tea plantation area in southern part of Korea. This study was carried out to investigate some development of the tea red spider mite. The results were summarized as follows: 1. The body length and width of adult female and male were 0.40mm and 0.22mm, and 0.31mm and 0.20mm, respectively. 2. Durations of immature stage of females were 24.8 days at $16^{\circ}C$, 17.0 days at $20^{\circ}C$, 11.0 days at $24^{\circ}C$, 8.2 days at $28^{\circ}C$, and 6.7 days at $32^{\circ}C$. 3. The average longevities of female of the tea red spider mite were 31.5 days at $16^{\circ}C$, 13.4 days at $20^{\circ}C$, 8.4 days at $24^{\circ}C$, 7.8 days at $28^{\circ}C$, and 5.8 days at $32^{\circ}C$.

탈삽처리온도 및 처리시간에 따른 탈삽감의 품질변화

  • $CO_2$탈삽처리($CO_2$농도 90%이상, 0.08mmPE밀봉)한 탈삽감(도근조생)의 탈삽정도에 따른 유통중 품질변화를 비교하고자 2$0^{\circ}C$ 24시간, 2$0^{\circ}C$ 36시간, $25^{\circ}C$ 24시간, $25^{\circ}C$ 28시간, 38$^{\circ}C$ 10시간, 38$^{\circ}C$ 18시간 탈삽처리한후 18~2$0^{\circ}C$상온에서 출고후 시간경과에 따른 과육경도, 삽미, 가용성고형물, 과실색도(a값) 변화를 조사하였다. 과육경도는 각처리 공히 탈삽고로부터 출고직후의 경도는 1.9~2.0kg/$\Phi$5mm였으며, 시장판매시인 출고 48시간후와 소비자의 소비시기인 출고 72시간후의 경도는 1.6~l.9kg/$\Phi$5mm정도로 모든 처리에서 상품성이 있었으나, $25^{\circ}C$ 28시간 처리는 다른 처리에 비해 경도저하가 빠른 경향을 보였다. 삽미변화는 2$0^{\circ}C$ 36시간, 38$^{\circ}C$ 18시간 탈삽처리에서는 출하시인 출고 24시간후에 완전탈삽되었으며, $25^{\circ}C$ 28시간, 38$^{\circ}C$ 10시간 처리는 시장판매시인 출고 48시간후에 완전탈삽되었고 $25^{\circ}C$ 24시간처리는 출고 72 시간후에 완전탈삽되었다. 그러나 2$0^{\circ}C$ 24시간 처리는 출고후 72시간까지도 떫은맛이 남아있어 소비지에서 상품성이 크게 떨어졌다. 가용성고형물은 탈삽 3일후에 12.6~13.0 。Brix를 나타내어 탈삽전에 비해 3.9~3.5 。Brix정도 떨어졌으며 처리간에는 유의적인 차이는 없었다. 출고 48시간후 과실표면의 a값은 19.1~23.9로 탈삽후 시간이 경과할수록 증가하는 경향을 보였다. 이상의 결과를 요약하면 도근조생 탈삽감의 품질은 출하시에는 2$0^{\circ}C$ 36시간, 38$^{\circ}C$ 18시간 탈삽처리에서 과실경도가 2.0kg/$\Phi$5mm으로 높고 완전탈삽되어 품질이 가장 우수하였고, 시장판매시에는 2$0^{\circ}C$ 36시간, 38$^{\circ}C$ 18시간탈삽처리 및 $25^{\circ}C$ 28시간, 38$^{\circ}C$ 10시간 탈삽처리에서 상품성이 우수하였다. 그러나 25'E 28시간 탈삽처리는 탈삽의 균일도가 다른 처리에 비해 떨어지는 경향이었다.

Pathogenicity of Spodoptera litura Nucleopolyhedrovirus with Different Temperatures (온도조건에 따른 담배거세미나방핵다각체병바이러스의 병원성 검정)

  • This experiment was conducted to investigate pathogenicity of Spodoptera litura nucleopolyhedrovirus (SINPV) with different temperatures to mass production. In laboratory, LC$_{50}$ values of SINPV were 5.534$\times$10$^3$ PIBs/ml and 4.l0$\times$10$^2$PIBs/ml in 1st instar larvae at 2$0^{\circ}C$ and 28$^{\circ}C$, respectively, but those were increased at 32$^{\circ}C$. LC$_{50}$ at 3rd and 5th instar larvae were showed the lowest value at 28$^{\circ}C$. LT$_{50}$ values of SINPV in 1.0$\times$10$^{5.7}$ PIBs/ml were determined at various temperature conditions (20-32$^{\circ}C$). The result showed that treatments at 28$^{\circ}C$ and 32$^{\circ}C$ shortened LT$_{50}$ values, but treatments at 2$0^{\circ}C$ and 24$^{\circ}C$ was relatively longer. Also LT$_{50}$ values was shortened in high concentration and young larva. LT$_{50}$ values are dependent on the temperature, viral concentration and larval instar.

The Development and Oviposition of Bean Bug, Riptortus clavatus Thunberg (Hemiptera: Alydidae) at Temperature Conditions (온도조건에 따른 톱다리개미허리노린재의 발육 및 산란)

  • This study was conducted to determine the effect of temperatures on the egg and nymphal development, adult longevity and oviposition of bean bug, Riptortus clavatus Thunberg, using Saealkong seed as food sources in fibrous nylon-tube at four different temperatures (20, 24, 28 and $32^{\circ}C$). Hatchability showed the highest value of 100% at $28^{\circ}C$ and decreased with increasing temperature. Egg duration ranged from 7 days at $32^{\circ}$ to 16.7 days at $20^{\circ}C$. Instar duration was longer with increasing instar stage. Nymphal duration was 38 days at $20^{\circ}C$, 30 days at $24^{\circ}C$, 23 days at $28^{\circ}C$, and 18 days at $32^{\circ}C$ Emergence rates to adult were 16, 41, 72 and 68% at 20, 24, 28 and $32^{\circ}C$, respectively. Female adult longelity ranged from a minimum 20 days at $20^{\circ}C$ to a maximum 63 days at $28^{\circ}C$, while the longevity of male ranged from 19 days at $20^{\circ}C$ to 60 days at $28^{\circ}C$. Preoviposition duration was shorter with increasing temperature and ranged from 11 days at $20^{\circ}C$ to 5 days at $32^{\circ}C$. Total number of eggs laid per female ranged from a minimum 21 eggs at $20^{\circ}C$ to a maximum 67 eggs at $28^{\circ}C$. Consequently, the estimated lower threshold tempeatures of each developmental stage were $10.3^{\circ}C$ for egg, and 9.3, 12.7, 10.0, 11.0 and 8.7 for the 1st, 2nd, 3rd, 4th and 5th instar, respectively.

Studies on Genetics and Breeding in Rainbow Trout(Oncorhynchus mykiss) VII. Fertilization of Fresh Egg with Co-Preserved Sperm and Ultrastructural Changes (무지개 송어의 유전 육종학적 연구 VII. 동결보존시킨 정자와 신선한 난모세포의 수정 및 미세구조적 변화)

  • This study was carried out to develop new techniques useful for cryopreservation, thawing and artificial insemination, and ultrastructural changes of cryopreserved spermatozoa in rainbow trout(Oncorhynchus mykiss) . Two extenders, such as Tyrode solution and Whittingham's $T_6$ solution, were used to preserve rainbow trout sperm in refrigerator $(-20,\;-40\;and\;-70^{\circ}C)$ or liquid nitrogen $%(-196^{\circ})$. Hand-stripped semen was diluted to 1:16 with two extenders, an then the semen were frozen after mixing semen and each extender containing 1M or 1.5M DMSO solution to 1:1. After 60 days cryopreserved semen was thawed in a $13^{\circ}$ water bath, and subsequently centrifugated. After centrifugation at 1,000 rpm for 5 min thawed semen was washed with extenders, and then fertilized with fresh eggs. The results obtained in these experiments were summarized as follows: After cryopreservation, over 75% of spermatozoa were appeared motile and the survival rate was high. Following cryopreservation by the addition of cryoprotectant such as DMSO, methanol and glycerol, the fertilization rate of the thawed spermatozoa appeared over $99\%$ compared with the control having $99\%$ of fertilization rate. There was no difference between the control and experimental groups such as $(-20^{\circ}C\;-40^{\circ}C\;and\;-70^{\circ}C)$ and $-196^{\circ}$ in fertilization rate. Following cryopreservation at $-196^{\circ}$ by the addition of 1M DMSO of cryoprotectant, each fertilization rate following 24 hours and hatching rate following 24 days showed $96\%$ and $8\%$ by the addition of BSA, but showed $98\%\;and\;10%$ by no addition of BSA. Following 2 months of cryopreservation by the addition of 1M DMSO of cryoprotectant, there were $10%$ of hatching rate at $-196^{\circ}\;and\;10\%\;and\;35\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C$. Following 2 months of cryopreservation by the addition of 1M methanol of cryoprotectant, there were $22\%$ of fertilization rate at $-20^{\circ}C,\;and\;28\%,\;at\;-70^{\circ}C$ Following 2 months of cryopreservation by the addition of 1M glycerol of cryoprotectant, there were $22\%$ of fertilization rate at $-20^{\circ}C$, and $33\%,\;at\;-70^{\circ}C$. pollowing 2 months of cryopreservation by the addition of 1.5M DMSO of cryoprotectant, there were $27\%$ of fertilization rate at $-20^{\circ}C,\;an\;36\%\;and \;35\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C$. Following 2 months of cryopreservation by the addition of 1.5M glycerol of cryoprotectant, there were $34\% \;of\;fertilization\;rate\;at\;-20^{\circ}C, \;and\;31\%\;and\;31\%,\;respectively,\;at \;-40^{\circ}C\;and\;-70^{\circ}$. Following 2 months of cryopreservation by the addition of 1.5M methanol of cryoprotectant, there were $28\%$ of fertilization rate at $-20^{\circ}C,\;and\;29\%\;and\;28\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C.$ From 10 days and 15 days following fertilization at $13^{\circ}C\;and\;10^{\circ}C$, respectively, the mortality rate of fertilized ova was markedly increased. The middle piece of spermatozoa had two set of central doublets, nine set of outer coarse fibres, and mitochondrial sheath. Spermatozoa went through morphological changes during storage, e.g. winding of flagella, detachment of the nuclear envelope and the plasma membrane from the nucleus of the sperm head. There were $1\%$ abnormal spermatozoa in fresh sperm and about $15\%$ during storage.

The effect of four different temperatures on the growth of Aedes albopictus larva (네 가지 다른 온도가 흰줄숲모기(Aedes albopictus) 유충 생장에 미치는 영향)

  • We investigated to know the growth patterns of Aedes albopictus larva at the four different temperature conditions. Each of 120 individuals was placed into a $20m{\ell}$ vial and 12 sets (a set of 10 vials) were separated into 12 water tanks ($17{\times}24{\times}18cm^3$). Each water tank was composed of 3 the $1^{st}$ instar, 3 the $2^{nd}$ instar, 2 the $3^{rd}$ instar, and 2 the $4^{th}$ instar. Three sets of water tanks were placed under the four different incubator temperatures ($17^{\circ}C$, $21^{\circ}C$, $24^{\circ}C$, $28^{\circ}C$). We found that the eclosion rates were $20.00{\pm}5.77%$ at $21^{\circ}C$ and $3.33{\pm}3.33%$ at other temperatures. For the mosquito larva mortality rate, $1^{st}$ instar was $19.24{\pm}3.65%$, $2^{nd}$ instar was $16.48{\pm}3.25%$, $3^{rd}$ instar was $23.54{\pm}5.06%$, and $4^{th}$ instar was $40.74{\pm}7.08%$. The lowest mortality rate in growth stages according to temperature was $13.33{\pm}6.67%$ at $17^{\circ}C$ in $1^{st}$ instar larva, $7.41{\pm}7.41%$ at $21^{\circ}C$ at $2^{nd}$ instar larva, $10.74{\pm}6.43%$ at $24^{\circ}C$ in $3^{rd}$ instar larva, and $20.37{\pm}5.46%$ at $28^{\circ}C$ in $4^{th}$ instar larva. The survival period of mosquitoes in underwater were $26.33{\pm}0.67days$ at $17^{\circ}C$, $23.33{\pm}1.33days$ at $21^{\circ}C$, $20.00{\pm}2.52days$ at $24^{\circ}C$, and $11.67{\pm}1.20days$ at $28^{\circ}C$. From our results the most effective temperature to the normal growth of mosquito larva was $21^{\circ}C$, and the highest mortality rate was shown at the $4^{th}$ instar stage of larva growth. Our results would provide the basic data for the mosquito larva's growth pattern.

Comparison of Naphthalene Degradation Efficiency and OH Radical Production by the Change of Frequency and Reaction Conditions of Ultrasound (초음파 주파수 및 반응조건 변화에 따른 나프탈렌 분해효율과 OH 라디칼의 발생량 비교)

  • Naphthalene is a volatile, hydrophobic, and possibly carcinogenic compound that is known to have a severe detrimental effect to aquatic ecosystem. Our research examined the effects of various operating conditions (temperature, pH, initial concentration, and frequency and type of ultrasound) on the sonochemical degradation of naphthalene and OH radical production. The MDL (Method detection limit) determined by LC/FLD (1200 series, Agilient) using C-18 reversed column is measured up to 0.01 ppm. Naphthalene vapor produced from ultrasound irradiation was detected under 0.05 ppm. Comparison of naphthalene sonodegradion efficiency tested under open and closed reactor cover fell within less than 1% of difference. Increasing the reaction temperature from $15^{\circ}C$ to $40^{\circ}C$ resulted in reduction of naphthalene degradation efficiency ($15^{\circ}C$: 95% ${\rightarrow}$ $40^{\circ}C$: 85%), and altering pH from 12 to 3 increased the effect (pH 12: 84% ${\rightarrow}$pH 3: 95.6%). Pseudo first-order constants ($k_1$) of sonodegradation of naphthalene decreased as initial concentration of naphthalene increased (2.5 ppm: $27.3{\times}10^{-3}\;min^{-3}\;{\rightarrow}$ 10 ppm : $19.3{\times}10^{-3}\;min^{-3}$). Degradation efficiency of 2.5 ppm of naphthalene subjected to 28 kHz of ultrasonic irradiation was found to be 1.46 times as much as when exposed under 132 kHz (132 kHz: 56%, 28 kHz: 82.7%). Additionally, its $k_1$ constant was increased by 2.3 times (132 kHz: $2.4{\times}10^{-3}\;min^{-1}$, 28 kHz: $5.0{\times}10^{-3}\;min^{-1}$). $H_2O_2$ concentration measured 10 minutes after the exposure to 132 kHz of ultrasound, when compared with the measurement under frequency of 28 kHz, was 7.2 times as much. The concentration measured after 90 minutes, however, showed the difference of only 10%. (concentration of $H_2O_2$ under 28 kHz being 1.1 times greater than that under 132 kHz.) The $H_2O_2$ concentration resulting from 2.5 ppm naphthalene after 90 minutes of sonication at 24 kHz and 132 kHz were lower by 0.05 and 0.1 ppm, respectively, than the concentration measured from the irradiated M.Q. water (no naphthalene added.) Degradation efficiency of horn type (24 kHz) and bath type (28 kHz) ultrasound was found to be 87% and 82.7%, respectively, and $k_1$ was calculated into $22.8{\times}10^{-3}\;min^{-1}$ and $18.7{\times}10^{-3}\;min^{-1}$ respectively. Using the multi- frequency and mixed type of ultrasound system (28 kHz bath type + 24 kHz horn type) simultaneously resulted in combined efficiency of 88.1%, while $H_2O_2$ concentration increased 3.5 times (28 kHz + 24 kHz: 2.37 ppm, 24 kHz: 0.7 ppm.) Therefore, the multi-frequency and mixed type of ultrasound system procedure might be most effectively used for removing the substances that are easily oxidized by the OH radical.
