Comparative Performance of Broilers Fed Diets Containing Processed Meals of BT, Parental Non-BT Line or Commercial Cotton Seeds

  • An effort was made to assess comparative production performance in broiler chickens fed diets containing solvent extracted cottonseed meal (CSM) processed from BT and Parental Non-BT lines. Processed meal of national check and commercial produce cottonseeds were also used for comparison. The free gossypol contents were 0.02, 0.02, 0.44 and 0.03% in meals of BT, Parental Non-BT, national check and commercial produce cottonseeds, respectively. Day-old broiler chicks (n=243) were divided to 27 groups of 9 each. Nine dietary treatments (iso-nitrogenous, 23% CP and iso-caloric, 2,800 kcal ME/kg) were formulated viz., D1 (control, soybean meal-SBM based), D2 and D3 (BT CSM at 10% of diet with and without additional iron), D4 and D5 (non-BT CSM with and without additional iron), D6 and D7 (national check CSM with or without additional iron), and D8 and D9 (commercial produce CSM with or without additional iron at 2 ppm for every 1 ppm of free gossypol, respectively). Each dietary treatment was offered to three replicated groups up to 6 weeks of age. At the end of 6 weeks of age, 10 birds were taken out randomly from each treatment and were sacrificed to study carcass traits, organs' yield and histo-pathological changes in vital organs. The broiler chickens received CSM processed from BT (D2, 1,753 g and D3, 1,638 g) and Parental Non-BT (D4, 1,653 g and D5, 1,687 g) with or without additional Fe grew at same rate as observed in soybean meal (solvent ext.) based diet (D1, 1,676 g). The feed intake and feed conversion efficiency (feed: gain) in these dietary treatments (BT, non-BT line based diets) also did not differ significantly (p>0.05) from control diet. Similar observation was also observed in dietary treatments (D8 and D9) containing solvent extracted cottonseed meal of commercial produce origin. However, a decrease (p<0.05) in body weight gain and feed intake was observed in D6 containing national check CSM with high gossypol content. Addition of Fe in the diet (D7) improved (p<0.05) feed intake and weight gain but not to the extent as observed in diets containing BT, parental non-BT, and commercial produce CSM or control. However, any type of CSM did not affect feed conversion efficiency when fed with or without additional iron. The carcass characteristics in terms of dressing percentage, liver weight and heart weight was not significantly (p>0.05) different between the treatments. The eviscerated yields emanated from diets containing either BT, non-BT or commercial produce were statistically similar to control. However, eviscerated yield of broilers fed national check CSM with or without iron supplementation was lower (p<0.05) than BT cotton with Fe supplementation and commercial produce CSM. The study envisaged that BT, parental non-BT and commercial produce solvent extracted cottonseed meal can be included at 10% in soybean meal based broiler diet replacing soybean meal and rice bran without additional iron.

Comparison of 2D and 3D Brachytherapy Planning for Cervical Cancer (자궁경부암 근접방사선치료 시 2차원, 3차원 치료계획 비교평가)

  • To evaluate the usefulness of 3-dimensional brachytherapy(BT) planning technique based on CT in cervical cancer. Patients with cervical cancer underwent 2-D BT treatment planning and then CT scan with HDR intracavitary applicators in place with same positions. Dose was prescribed to Point A with 5Gy per fraction on 2-D BT planning. For 3-D BT planning, and dose was prescribed to the High risk CTV for BT (HR CTV) with 5Gy. The 3-D BT planning goal was to cover at least 90% of the HR CTV with target 5Gy isodose surface while limiting the dose to $2cm^3$ of bladder to less than 7.5 Gy, and $2cm^3$ of rectum to less than 5Gy. In one patient of 10 patients, $D_{2cm3}$ of rectal dose was over 5Gy and 6patients at $D_{2cm3}$ of bladder dose on 2-D BT planning. There was a tendency to underestimate ICRU bladder dose than ICRU rectal dose. CT based 3-D BT planning for cervical cancer will enable evaluation of dose distributions for tumor and critical organs at risk. So, rectal and bladder morbidity as well as geographic miss will be reduced in case of the bulky disease or uterine malposition.

  • The main purpose of this study is to investigate the technology related facts which determine the difference of the output of companies in the fields of IT and BT industries in Korean. The determining facts consist of the technology development strategy, the characteristic of R&D groups, and the ability of managing knowledge assets. Three kinds of regression model are made by using 3 determining facts from total model to IT and BT model to investigate the research purpose of this study. The technology development strategy consists of 2 sub facts, as technology innovation and product upgrade. The characteristic of R&D group consists of 2 sub facts, as the characteristic of task and the intensity of R&D group members. The ability of managing knowledge assets consist of 3 sub elements, as the ability of creating knowledge assets, the ability of holding in common knowledge assets, and the ability of developing the value of knowledge assets.

Effects of Benzoic Acid and Thymol on Growth Performance and Gut Characteristics of Weaned Piglets

  • A total of 144 weaned crossed pigs were used in a 42-d trial to explore the effects of different concentrations/combinations of benzoic acid and thymol on growth performance and gut characteristics in weaned pigs. Pigs were randomly allotted to 4 dietary treatments: i) control (C), basal diet, ii) C+1,000 mg/kg benzoic acid+100 mg/kg thymol (BT1), iii) C+1,000 mg/kg benzoic acid+200 mg/kg thymol (BT2) and, iv) C+2,000 mg/kg benzoic acid+100 mg/kg thymol (BT3). Relative to the control, pigs fed diet BT3 had lower diarrhoea score during the overall period (p<0.10) and improved feed to gain ratio between days 1 to 14 (p<0.05), which was accompanied by improved apparent total tract digestibility of ether extract, Ca and crude ash (p<0.05), and larger lipase, lactase and sucrose activities in the jejunum (p<0.05) at d 14 and d 42. Similarly, relative to the control, pigs fed diet BT3 had higher counts for Lactobacillus spp in digesta of ileum at d 14 (p<0.05), and pigs fed diets BT1, BT2, or BT3 also had higher counts of Bacillus spp in digesta of caecum at d 14 (p<0.05), and lower concentration of ammonia nitrogen in digesta of caecum at d 14 and d 42 (p<0.05). Finally, pigs fed diet BT3 had higher concentration of butyric acid in digesta of caecum at d 42 (p<0.05), and a larger villus height:crypt depth ratio in jejunum and ileum at d 14 (p<0.05) than pigs fed the control diet. In conclusion, piglets fed diet supplementation with different concentrations/combinations of benzoic acid and thymol could improve feed efficiency and diarrhoea, and improve gut microfloral composition. The combination of 2,000 mg/kg benzoic acid+100 mg/kg thymol produced better effects than other treatments in most measurements.

Expression of Rat Hepatic Glutathione-S-Transferases by Benzoazoles (Benzoazole계 화합물이 glutathione-S-transferases의 유도발현에 미치는 영향)

  • Glutathione-S-transferases (GSTs) detoxify electrophilic xenobiotics and reactive metabolites. Recently benzene-fused heterocycles have been shown to increase the total amount of hepatic GSTs in rats. Primarily this study aimed to determine the induction of GSTs by benzoazoles (BAs) including benzoxazole (BX), 2-methylbenzoxazole (M-BX), 2,5-dimethyl benzoxazole (D-BX), benzothiazole (BT), aminobenzothiazole (A-BT) and 2-mercaptobenzothiazole (M-BT) in rats. Hepatic cytosol and poly(A)$^+$ mRNA were prepared from rats after oral administration of BX, BT, M-BX, D-BX, A-BT and M-BT for 5 consecutive days at doses of 1 mmol/kg. Western immunoblot and northern blot analysis were conducted with rabbit anti-GST Ya, Yb$_1$, Yb$_2$, Yc antibodies and cDNA probes containing = 500 bps in the specific coding regions of Ya, Yb$_1$, Yb$_2$, Yc$_1$, and Yc$_2$, respectively. All BAs increased the amount of enzymes and mRNA levels of GSTs. BT was the most effective inducer of GSTs among the compounds examined in this study. Although A-BT and M-BT, the derivatives of BT, induced GSTs, these chemicals had lesser effect on induction of GSTs than BT. The derivatives of BX also induced less GSTs than the parent compound and the addition of methyl group to the benzene ring of BX reduced the induction of GSTs. BAs had better inductive effects on the class $\alpha$(Ya, Yc) than class $\mu$ GSTs (Yb$_1$, Yb$_2$). BAs enhanced mRNA levels of GSTs in parallel with the protein levels. These results indicate that 1) most of BAs induced various isozymes of GSTs, 2) the induction of GSTs appears to be correlated with the chemical structure of the derivatives, and 3) the expression of GST by BAs is presumably under the transcriptional regulation.

Effects of Conjugated Linoleic Acid Supplement on the Composition of Fatty Acids, Expressions of delta-5 Desaturase (D5D) and Fatty Acid Desaturase2 (FADS2) Genes in Mice (식이지방에 첨가한 Conjugated Linoleic Acid가 지방산 조성, delta-5 Desaturase(D5D)와 Fatty Acid Desaturase2(FADS2) 유전자 발현에 미치는 영향)

  • We investigated the effects of conjugated linoleic acid (CLA) on the fatty acid composition in the plasma and liver, and the expressions of delta-5 desaturase (D5D) and fatty acid desaturase2 (FADS2) genes in ICR male mice using two different sources of fats in the diets. The experimental groups were divided into four groups: beef tallow (BT) and fish oil (FO), BT with CLA supplementation (BTC), and FO with CLA supplementation (FOC) groups. Ten mice in each group were fed with the experimental diets for 4 weeks. All mice were fed experimental diets containing 12% of total dietary fat (w/w) either with or without 0.5% CLA (w/w). Fatty acid compositions were analyzed in the plasma and liver using gas chromatography. The levels of D5D and FADS2 expression were analyzed by RT-PCR in the liver The results showed that CLA participates competitively with C18:2 in the elongation and desaturation processes, leading to significant increase in the levels of C20:4 and C22:6 in BTC group (p<0.05). The expression levels of D5D and FADS2 were higher in BT and BTC group than those of FO and FOC group. In particular, the expression of D5D gene was greatly upregulated in BTC group. Furthermore, the conversion ratios from C18:2 to C20:4 in the liver were higher in BTC group than those in other groups. Thus our results suggest that increased expressions of DSD and FADS2 genes may be responsible for the enhanced CLA effects on the desaturation in the BT containing saturated fatty acids rather than the FO rich in n-3 PUFA.

Piezoelectric and Dielectric Properties of NKN-(1-x)BNT-xBT Ceramics (NKN-(1-x)BNT-xBT 세라믹스의 압전 및 유전특성)

  • In this study, piezoelectric and dielectric properties of the $(Na_{0.5}K_{0.5})NbO_3-(1-x)(Bi_{0.5}Na_{0.5})TiO_3-xBaTiO_3$ [NKN-(1-x)BNT-xBT] ceramics were investigated. The lead-free NKN-(1-x)BNT-xBT ceramics were fabricated by a conventional mixed oxide method. The results indicate that the addition of $BaTiO_3$ significantly influences the sintering, microstructure, phase transition and electrical properties of NKN-BNT ceramics. A gradual change in the piezoelectric and dielectric properties was observed with the increase of BT contents. The dielectric constant, piezoelectric constant ($d_{33}$) and electromechanical coupling factor ($k_p$) increased at the morphotropic phase boundary (MPB). The $d_{33}$=184 pC/N, $k_p$=0.38, dielectric constant=1455 with dielectric loss value of less than 1% were obtained for the NKN-0.95BNT-0.05BT ceramics sintered at $1150^{\circ}C$ for 2h. These results demonstrate that the NKN-(1-x)BNT-xBT ceramics is an attractive candidate for lead-free piezoelectric materials.

An investigation on the improvement of neutron radiography system of the Tehran research reactor by using MCNPX simulations

  • Applying the available neutron flux for medical and industrial purposes is the most important application of research reactors. The neutron radiography system is used for non-destructive testing (NDT) of materials so that it is one of the main applications of nuclear research reactors. One of these research reactors is the 5 MW pool-type light water research reactor of Tehran (TRR). This work aims to investigate on materials and location of the beam tube (BT) of the TRR radiography system to improve the index parameters of BT. Our results showed that a through-type BT with 20 cm thick carbon neutron filter, 1.2 cm and 9.4 cm of the diameter of inlet (D1) and output (D2) BT, respectively gives thermal neutron flux almost 25.7, 5.6 and 1.1 times greater than the former design of the TRR (with D1 = 1.8 cm and D1 = 9.4 cm), previous design of the TRR with D1 = 3 cm and D1 = 9.4 cm, and another design with D1 = 5 cm and D1 = 9.4 cm, respectively. Therefore, the design proposed in this paper could be a better alternative to the current BT of the TRR.

Evaluation of Bt-cotton Genotypes for Resistance to Cotton Leaf Curl Disease under High Inoculum Pressure in the Field and Using Graft Inoculation in Glasshouse

  • Bt-cotton germplasm, consisting of 75 genotypes was evaluated against cotton leaf curl disease (CLCuD) under high inoculum pressure in the field and using graft inoculation in glasshouse by visual symptom scoring assessments. None of the tested genotype was found disease free under both evaluation tests. Under field conditions in 2011, 3 genotypes were found resistant, 4 moderately resistant, 3 tolerant, 2 moderately susceptible and one susceptible; in 2012, 3 genotypes were tolerant, 7 moderately susceptible, 5 susceptible and 38 highly susceptible; in 2013, one was moderately susceptible and 51 were highly susceptible with varying degree of percent disease index (PDI) and severity index (SI). However, through graft evaluation in glasshouse, none of the graft inoculated plant was symptomless. All tested genotypes showed disease symptoms with SI values ranging between 5.0 and 6.0, and latent period between 12 and 14 days. Of the 75 genotypes evaluated using graft inoculation, 11 were found susceptible with SI values of 5.0 to 5.4 while remaining 64 were highly susceptible with SI values of 5.5 to 6.0. Inoculated plants of all tested genotypes exhibited severe disease symptoms within 10 days after the appearance of initial symptoms. No reduction in SI value was observed until the end of the experiment i.e., 90 days after grafting. Information generated under the present study clearly demonstrates that no sources of resistance to CLCuD are available among the tested Bt-cotton genotypes. So, a breeding programme is needed to introgress the CLCuD-resistance from other resistant sources to agronomically suitable Bt-cotton genotypes.

Purification of xylose reductase from Candida sp. BT001 and characterization of its properties (Candida sp. BT001의 xylose reductase의 정제 및 성질)

  • Xylose reductase (alditol: $NADP^+$ 1-oxidoreductase, EC 1.1.1.21) from the xylose-fermenting yeast, Candida sp. BT001, was purified via salt fractionation, ion-exchange, gel filtration and affinity chromatography, and its properties were characterized. The enzyme from the yeast was active with both NADPH and NADH as coenzyme. The xylose reductase activity with NADH was approximately 51% of that with NADPH and the specific activities of purified enzyme with NADPH and NADH were 11.78 U/mg and 6.01 U/mg, respectively. Molecular weight of the purified enzyme was 31,000 on SDS-PAGE and 61,000 on gel filtration. The Km for D-xylose, NADPH, and NADH was $94.2{\times}10^{-3}M,\;0.011{\times}10^{-3}M\;and \;0.032{\times}10^{-3}M$, respectively. The purified xylose reductase had relatively higher substrate affinity for L-arabinose than other aldoses tested. The optimal pH was 6.2 and the optimal reaction temperature was $45^{\circ}C$. The thermal stability of the enzyme was for 20 minutes at $30^{\circ}C$.
