Continuous rapid Production of Soy Sauce by Coimmobilized Mixed Culture system of Zygosaccharomyces rouxii and Candida versatilis using Air Bubble Column Reactor (Zygosaccharomyces rouxii와 Candida versatilis의 동시 고정화에 의한 Air Bubble Column Reactor에서 간장의 연속적 속성 생산)

  • This study was designed to find out the rapid fermentation of soy sauce from koji hydrolyzates using air bubble column reactor packed with coimmobilized mixed culture system. Continuous ripid production was performed by coimmobilized Z. rouxiii BH-90 and C. versatilis BH-91. Coimmobilized cells of Zygosaccharomyces rouxii BH-90 and Candida versatilis BH-91 mixture cells in the column reactor produced 2.8% ethyl alcohol and 18mg/L 4-ethylguaiacol over 96 hours under the optimal conditions. Coimmobilized cells produced 2.30∼2.4% ethyl alcohol during 30 days, and decreased gradually from 40 days to 70 days. Also coimmobilized cells produced 4-ethylguaiacol at the constant rate of 16∼18mg/L and decreased gradually after 40 days. Final product of soy sauce contained 2.4% ethyl alcohol and 18mg/L 4-ethylguaiacol. However, amino acid compositions of soy sauce were consisted of predominantly glutamic acid, leucin, arginine, aspartic acid, Iysine and valine, which were more than 50% of total amino acid.

  • This present study was carried out particulary focusing on rapid fermentation of soy sauce by using column type reactor $(30\;cm{\times}5\;cm)$ packed each immobilized cells of Pediococcus halophilus R-22, Saccharomyces rouxii R-60 and Candida etchellsii H-50. When immobilized P. halophilus R-22 by column type reactor was performed continuously fermentation, lactic acid was produced $0.62{\sim}0.64%$ during 25 days and then decreased gradually after 30 days. S. rouxii R-60 was Produced the $2.1{\sim}2.5%$ ethylalcohol constantly for 35 days and also C. etchellsii H-50 was produced $14{\sim}16\;mg/l$ 4-ethylguaiacol for 35 days and then this products were decreased gradually after fermentation of 40 days. Final Products of fish sauce contained 1,721.6 mg% total nitrogen, 1,584.1 mg% amino-nitrogen, 0.75% lactic acid, 2.7% ethylalcohol and 18.2 mg/l 4-ethylguaiacol.

  • This study was performed to rapid fermentation from sardine hydrolyzate by using column reactor. The column reactor was constructed from three glass columns $(30cm{\times}5cm)$ and each column was packed with colloidal silica and sodium alginate (1:5) on which Pediococcus halophilus R-22, Saccharomyces rouxii R-60 and Candida etchellsii H-50, respectively, was previously fixed. At that time, optimal conditions for rapid fermentation were found the pH of 5.2, temperature of $30^{\circ}C$ and 10% NaCl. For rapid fermentation, immobilized whole cells of P. halophilus R-22, S. ruoxii R-60 and C. etchellsii H-50 packed the each column reactor were produced 0.75% lactic acid, 2.5% ethylalcohol and 18 mg/l 4-ethylguaiacol under the optimal conditions.

  • The aim of this study was to find out the rapid fermentation of soy sauce from koji hydrolyzate using column type reactor packed with immobilized yeast cells. Each immobilized cell of Zygosaccharomyces rouxii BH-90 and Candida versatilis BH-91 in the packed column type reactor produced 2.8% ethyl alcohol and 18mg/l 4-ethylguaiacol over 96 hours under the optimal condition. Continuous fermentation was performed by immobilized Z. rouxii BH-90 packed in column type reactor. Immobilized Z. rouxii BH-90 produced 2.30~2.85% ethyl alcohol during 30 days, and decreased gradually from 40 days to 80 days. Also C. versatilis BH-91 produced 4-ethylguaiacol at the constant rate of 16~18mg/l and decreased gradually after 40 days. Final product of soy sauce contained 2.8% ethyl alcohol and 18mg/l 4-ethylguaiacol. However, amino acid compositions of final products were consisted of predominantly glutamic acid, leucine, arginine, aspartic acid, lysine and valine, which were more than 50% of total amino acid.

Analysis of Off-flavor Compounds from Over-extracted Coffee (과추출에 의한 커피의 이취성분 분석)

  • To verify the volatile compounds contributing to coffee off-flavor, medium roasted Colombian, Sumatra, Ethiopian and light and dark roasted Colombian bean coffees were sequentially and studiously over extracted. Eight espresso samples and eight filter-dripped samples were analyzed by gas chromatography/mass spectrometry with the solidphase microextraction method and evaluated by sensory tests. In total, 67 compounds were detected, and desirable aroma decreased sharply, while off-flavors increased rapidly after 30 mL of espresso and 200 mL of drip coffee. Percent peak area of 4-ethylguaiacol revealed a linear increase during extraction and was highly correlated with increasing off-flavor, suggesting that it could be an indicator of over extraction. Considering the odor activity value, guaiacol and 4- vinylguaiacol were also contributory compounds to off-flavors.

  • 본 연구에서는 분리대두단백(ISP: isolated soy protein)과 탈지대두박(DSF: defatted soy flake)을 Devolase Flavourzyme 효소로 가수분해한 각각의 HVP를 발효 배지로 사용하여 Candida utilis(KCCM 50342) 효모의 ethylalcohol, 4-ethylguaiacol의 생성과 향 생성의 주요 인자인 a -galactosidase의 활성을 측정하였다. Ethylalcohol은 발효 1일째에 탈지대두박을 원료로 하고 여과후 미생물 처리한 경우에서 가장 높게 생성되었고, 4-ethylguaiacol은 일부 여과하지 않고 미생물 처리한 경우에서 낮은 농도의 생성을 보였다. a-galactosidase 효소활성은 탈지대두박 보다 ISP를 원료로 한 HVP에서 높은 활성을 보였다. 관능검사결과 탈지대두박을 사용한 HVP의 관능적 특성이 ISP를 원료로 한 HVP보다 더 우수하였다. 결과적으로 탈지대두박을 원료로 한 HVP의 향기성분 및 관능적 특성이 우수함을 확인하였다.

Selective Production of Monomeric Phenols from Lignin via Two-step Catalytic Cracking Process (2단계 촉매 분해공정을 이용한 리그닌 유래 선택적 페놀화합물 생산)

  • In this study, lignin was efficiently degraded via two-step catalytic cracking process and lignin-oil, char, and gas was produced as lignin degraded products. Three kinds of catalysts (MgO, CaO, and Pt/C) were used in first catalytic cracking step and the highest lignin-oil yield (76.2 wt%) was obtained in Pt/C catalyst with the smallest char formation (4.1 wt%). GC-MS/FID analysis revealed that 18 kinds of monomeric phenols existed in lignin-oil and sum of them was the highest in Pt/C condition (97.8 mg/g lignin). Meanwhile, relatively lower yield of monomeric phenols was produced in MgO and CaO condition because of their absorption on catalysts. Lignin-oil produced over Pt/C was introduced to second catalytic cracking process with porous Pd/activated carbon aerogel catalyst. From this process, four kinds of monomeric phenols such as 4-ethylguaiacol, 4-propylguaiacol, 4-ethylsyringol, 4-propylsyringol were selectively produced at 0.89 - 1.82 wt% level.

Component Analysis of major Softwood and Hardwood Vinegars (주요 침ㆍ활엽수 목초액의 성분분석)

  • To collect the basic data for the use of major softwood and hardwood vinegar, the chemical compositions were analized. In softwood vinegar the phenolic compounds derieved from lignin and extractives are vanillin, guaiacol, 3-methylphenol, acetoguaiacone, 1,2-benzenediol, ethylguaiacol, pyrocatechol. The compound derived from carbohydrates is 2-hydroxy-3-methyl-2-cyclopenten-1-one. The isolated compounds are the most in the phenolic fraction, and in the vinegar of Pinus koraiensis. In hardwood vingar 2-methoxyphenol, 2,6-djmethoxyphenol, 1-(4-hydroxy-3-methoxyphenyl) -ethanone, 3-methoxy-4-hydroxybenzylalcohol, 1-(4-hydroxy-3,5-dimethoxy-phenyl)-ethanone are the compounds derived from lignin and extractives. 2-hydroxy-3-methyl-2,4-cyclopen-1-one, methyl-4-t-butyl-2-furoate are the major compounds derived from the decomposition of carbohydrates. Many different kinds of compounds are analized in the neutral, phenolic and acidic fraction of hardwood vinegar The amount and kinds of compounds in hardwood vinegar are more higher than those of softwood than in softwood.

Development of a Novel Yeast Strain Which Ferments Soy Sauce by Protoplast Fusion

  • In order to develop a novel yeast which produces the charateristic aroma of soy sauce, a protoplast fusion between Zygosaccharomyces rouxii WFS4 and Torulopsis versatilis IAM 4993 was carried out. Auxotrophic mutants as selective markers were obtained from Zygosaccharomyces rouxii and Torulopsis versatilis by treatment of N-methyl-N -nitro-N-nitrosoguanidine. The conditions of the protoplast formation and the regeneration for fusion were examined. The protoplast fusion using polyethylene glycol 4000 led to the fusion frequency of $4~5{\times}10^{-7}\;cells/ml$. Among fusants, a fusant ST723-F31 presented the best results in terms of the aromaticity of fragrance, the growth pattern, the resistance against salt and the degree of growth according to pH. It makes easy to control the production and the balance of aroma components so that it gives a good flavor, shortens the fermentation period and, simplifies the preparation process when using a bioreactor into which fusant is immobilized.

  • In order to identify of volatile flavor compounds and polycyclic aromatic hydrocarbons (PAHs) in commercial liquid smokes, this study was conducted to analyze volatile flavor compounds by solvent extraction and/or Purge & Trap method/GC/MSD. A total of 156 volatile flavor compounds were detected in 6 commercial liquid smokes, and these compounds were composed mainly of 12 aldehydes, 60 ketones, 7 alcohols, 14 acids, 20 esters, 24 aromatic compounds, 7 furans and 12 miscellaneous compounds. Ketones $(806.6-7,573.9\mu g/mL)$ and aromatic compounds $(282.6-7,896.3 \mu g/mL)$ were more abundant than others. The PAHs known as carcinogen have not been detected in this study. The acids $(422.9-4,903.1\mu g/mL)$ was identified in relatively high concentration compared to other groups. Phenol and its derivatives among aromatic compounds were in relatively high concentration. Especially, the phenol and its derivatives including o-cresol, guaiacol, 4-ethylguaiacol and syringol were in higher concentration.
