Effect of Milling Time on the Microstructure and Mechanical Properties of Ta20Nb20V20W20Ti20 High Entropy Alloy (Ta20Nb20V20W20Ti20 하이엔트로피 합금의 미세조직 및 기계적 특성에 미치는 밀링 시간의 영향)

  • In this study, we report the microstructure and characterization of Ta20Nb20V20W20Ti20 high-entropy alloy powders and sintered samples. The effects of milling time on the microstructure and mechanical properties were investigated in detail. Microstructure and structural characterization were performed by scanning electron microscopy and X-ray diffraction. The mechanical properties of the sintered samples were analyzed through a compressive test at room temperature with a strain rate of 1 × 10-4 s-1. The microstructure of sintered Ta20Nb20V20W20Ti20 high-entropy alloy is composed of a BCC phase and a TiO phase. A better combination of compressive strength and strain was achieved by using prealloyed Ta20Nb20V20W20Ti20 powder with low oxygen content. The results suggest that the oxide formed during the sintering process affects the mechanical properties of Ta20Nb20V20W20Ti20 high-entropy alloys, which are related to the interfacial stability between the BCC matrix and TiO phase.

  • A mixture of 20(R)- and 20(S)-ginsenoside Rg$_{3}$ was obtained under mild acidic hydrolysis from protopanaxadiol saponins, ginsenosides Rb$_{1}$, Rb$_{2}$, Rc and Rd. The product was acetylated to give the peracetates, which were further converted into 20(R)-ginsenoside Rg$_{3}$, 20(S)-ginsenoside Rg$_{3}$, 20(R)-ginsenoside Rh$_{2}$ and 20(S)-ginsenoside Rh$_{2}$ by the direct alkaline treatment depending upon two kinds of temperature conditions respectively. The structure and physicochemical properties of a prosapogenin, 20(R)-ginsenoside Rh$_{2}$, were investigated.

Structural Characterization of the Anti-Complementary and Macrophage Activating Polysaccharides Isolated from Agaricus bisporus (양송이에서 분리한 보체계와 대식세포 활성화 다당류의 구조적 특성)

  • Three kinds of anti-complementary system and macrophage activating polysaccharides, AB-20-Ia, AB-20-IIa-2a and AB-20-IVa-2 were isolated from the fruit body of Agaricus bisporus and their structures were characterized. The proteoglycan, AB-20-IVa-2 showing the most potent anti-complementary and macrophage activity was composed of glucose, galactose, mannose, xylose, fucose and arabinose in a molar ratio of 3.48:1.83:1.00:0.79:0.74:0.11 and its main component amino acids were phenylalanine (34.72%) and valine (27.84%). The neutral polysaccharides, AB-20-Ia and AB-20-IIa-2a showing lower activity than AB-20-IVa-2, consisted of xylose, glucose, mannose, fucose and arabinose in molar ratios of <0.05:<0.05:2.07:1.00:2.72 and 2.16:1.58:1.00:0.20:0.14, respectively. The molecular weights of AB-20-Ia, AB-20-IIa-2a and AB-20-IVa-2 were 840,000, 750,000 and 650,000 respectively. In the $^1H-\;and\;^{13}C-NMR$ spectra of AB-20-Ia and AB-20-IIa-2a, AB-20-Ia showed only ${\beta}-configuration\;(^1H:\;4.8\;ppm,\;^{13}C:\;107.0\;ppm)$ in the anomerization of the glycosidic linkages, while AB-20-IIa-2a had both ${\alpha}-anomer\;(^1H:\;5.4\;ppm,\;^{13}C:\;102.0\;ppm)\;and\;{\beta}-anomer$. Especially, AB-20-Ia and AB-20-IIa-2a showed acetyl signals $(^1H:\;2.5\;ppm,\;^{13}C:\;21.0\;ppm)$. In the methylation analysis of the three polysaccharides, high proportion of 1,6-linked glucofuranosyl residues were detected in AB-20-Ia, whereas 1,6-linked glucopyranosyl residues and branches linked at position 4 of those mainly contained in AB-20-IIa-2a. AB-20-IVa-2 consisted mainly of 1,2-linked xylofuranosyl residues and 1,6-linked glucopyranosyl residues and branches linked at position 3 of those.

Preparation and Characterization of Polyurethane/Organoclay Nanocomposites by UV Curing (UV경화에 의한 폴리우레탄/유기화클레이 나노복합재료 제조와 물성 연구)

  • Polyurethane (PU)/organoclay nanocomposites were prepared by mixing UV curable urethane acrylate oligomer with organoclay, and a subsequent curing by UV irradiation. As organoclays, commercially available Cloisite 20A (C20A) and acrylsilane modified C20A were used. XRD and TEM analyses revealed that the UV cured PU/clay nanocomposites formed intercalated nanocomposites, and acrylsilane modified C20A are dispersed more finely than unmodified C20A in PU matrix. DMTA, pencil hardness and adhesion test onto PET substrate showed that the clay nanolayers induced an increase in the properties, and the enhancement in the properties was more pronounced in the PU/acrylsilane modified C20A nanocomposites than in the PU/unmodified C20A nanocomposites. It was also observed that the PU/surface modified clay nanocomposites showed remarkably lower shrinkage upon UV curing than the unfilled PU. The nanocomposites showed excellent optical transparency but lower gloss as compared to unfilled PU.

Ginsenoside 20(S)-Rg3 reduces KIF20A expression and promotes CDC25A proteasomal degradation in epithelial ovarian cancer

  • Background: Ginsenoside 20(S)-Rg3 shows promising tumor-suppressive effects in ovarian cancer via inhibiting NF-kB signaling. This study aimed to explore the downstream tumor suppressive mechanisms of ginsenoside Rg3 via this signaling pathway. Materials and methods: A systematical screening was applied to examine the expression profile of 41 kinesin family member genes in ovarian cancer. The regulatory effect of ginsenoside Rg3 on KIF20A expression was studied. In addition, we explored interacting proteins of KIF20A and their molecular regulations in ovarian cancer. RNA-seq data from The Cancer Genome Atlas (TCGA) was used for bioinformatic analysis. Epithelial ovarian cancer cell lines SKOV3 and A2780 were used as in vitro and in vivo cell models. Commercial human ovarian cancer tissue arrays were used for immunohistochemistry staining. Results: KIF20A is a biomarker of poor prognosis among the kinesin genes. It promotes ovarian cancer cell growth in vitro and in vivo. Ginsenoside Rg3 can suppress the transcription of KIF20A. GST pull-down and co-immunoprecipitation (IP) assays confirmed that KIF20A physically interacts with BTRC (β-TrCP1), a substrate recognition subunit for SCFβ-TrCP E3 ubiquitin ligase. In vitro ubiquitination and cycloheximide (CHX) chase assays showed that via interacting with BTRC, KIF20A reduces BTRC-mediated CDC25A poly-ubiquitination and enhances its stability. Ginsenoside Rg3 treatment partly abrogates KIF20A overexpression-induced CDC25A upregulation. Conclusion: This study revealed a novel anti-tumor mechanism of ginsenoside Rg3. It can inhibit KIF20A transcription and promote CDC25A proteasomal degradation in epithelial ovarian cancer.

TOMM20 as a potential therapeutic target of colorectal cancer

  • Translocase of outer mitochondrial membrane 20 (TOMM20) plays an essential role as a receptor for proteins targeted to mitochondria. TOMM20 was shown to be overexpressed in various cancers. However, the oncological function and therapeutic potential for TOMM20 in cancer remains largely unexplored. The purpose of this study was to elucidate the underlying molecular mechanism of TOMM20's contribution to tumorigenesis and to explore the possibility of its therapeutic potential using colorectal cancer as a model. The results show that TOMM20 overexpression resulted in an increase in cell proliferation, migration, and invasion of colorectal cancer (CRC) cells, while siRNA-mediated inhibition of TOMM20 resulted in significant decreases in cell proliferation, migration, and invasion. TOMM20 expression directly impacted the mitochondrial function including ATP production and maintenance of membrane potential, which contributed to tumorigenic cellular activities including regulation of S phase cell cycle and apoptosis. TOMM20 was overexpressed in CRC compared to the normal tissues and increased expression of TOMM20 to be associated with malignant characteristics including a higher number of lymph nodes and perineural invasion in CRC. Notably, knockdown of TOMM20 in the xenograft mouse model resulted in a significant reduction of tumor growth. This is the first report demonstrating a relationship between TOMM20 and tumorigenesis in colorectal cancer and providing promising evidence for the potential for TOMM20 to serve as a new therapeutic target of colorectal cancer.

  • The stream cipher Salsa20 and its reduced versions are among the fastest stream ciphers available today. However, Salsa20/7 is broken and Salsa20/12 is not as safe as before. Therefore, Salsa20 must completely perform all of the four rounds of encryption to achieve a good diffusion in order to resist the known attacks. In this paper, a new variant of Salsa20 that uses the chaos theory and that can achieve diffusion faster than the original Salsa20 is presented. The method has been tested and benchmarked with the original Salsa20 with a series of tests. Most of the tests show that the proposed chaotic Salsa of two rounds is faster than the original four rounds of Salsa20/4, but it offers the same diffusion level.

Bioinformatics Analysis of Hsp20 Sequences in Proteobacteria

  • Heat shock proteins are a class of molecular chaperones that can be found in nearly all organisms from Bacteria, Archaea and Eukarya domains. Heat shock proteins experience increased transcription during periods of heat induced osmotic stress and are involved in protein disaggregation and refolding as part of a cell's danger signaling cascade. Heat shock protein, Hsp20 is a small molecular chaperone that is approximately 20kDa in weight and is hypothesized to prevent aggregation and denaturation. Hsp20 can be found in several strains of Proteobacteria, which comprises the largest phyla of the Bacteria domain and also contains several medically significant bacterial strains. Genomic analyses were performed to determine a common evolutionary pattern among Hsp20 sequences in Proteobacteria. It was found that Hsp20 shared a common ancestor within and among the five subclasses of Proteobacteria. This is readily apparent from the amount of sequence similarities within and between Hsp20 protein sequences as well as phylogenetic analysis of sequences from proteobacterial and non-proteobacterial species.

A Study on Morphology and Mechanical Properties of Biodegradable Polymer Nanocomposites (생분해성 고분자 나노복합체의 형태학 및 기계적 특성 연구)

  • BBiodegradable polymers have attracted great attention because of the increased environmental pollution by waste plastics. In this study, PLA (polylactic acid)/Clay-20 (Cloisite 20) and PLA (polylactic acid)/PBS (poly(butylene succinate)/Clay-20 (Cloisite 20) nanocomposites were manufactured in a twin-screw extruder. Specimens for mechanical properties of PLA/Clay-20 and PLA/PBS (90/10)/Clay-20 nanocomposites were prepared by injection molding. Thermal, mechanical, morphological and raman spectral properties of two nanocomposites were investigated by differential scanning calorimetry (DSC), tensile tester, scanning electron microscopy (SEM) and raman-microscope spectrophotometer, respectively. In addition, hydrolytic degradation properties of two nanocomposites were investigated by hydrolytic degradation test. It was confirmed that the crystallinity of PLA/Clay-20 and PLA/PBS/Clay-20 nanocomposite was increased with increasing Clay-20 content and the Clay-20 is miscible with PLA and PLA/PBS resin from DSC and SEM results. Tensile strength of two nanocomposites was decreased, but thier elongation, impact strength, tensile modulus and flexural modulus were increased with an increase of Clay-20 content. The impact strength of PLA/Clay-20 and PLA/PBS/Clay-20 nanocomposites with 5 wt% of Clay-20 content was increased above twice than that of pure PLA and PLA/PBS (90/10). The hydrolytic degradation rate of PLA/Clay-20 nanocomposite with 3 wt% of Clay-20 content was accelerated about twice than that of pure PLA. The reason is that degradation may occur in the PLA and Clay-20 interface easily because of hydrophilic property of organic Clay-20. It was confirmed that a proper amount of Clay-20 can improve the mechanical properties of PLA and can control biodegradable property of PLA.

  • This study was carried out to determine the optimum planting time for in vitromultiplied tuber of Pinellia ternata(Thunb.) Breit. The tubers were planted on April 20, May 20, June 20, July 20, August 20 and September 20 in 1990. Emergence ratios were 68 to 87% in any planting time except planting on July 20. The number of tubers per $m^2$ at harvest in plantings on May 20 and June 20 were significantly higher with 1,110 and 1,021, respectively, while in plantings after July 20, those were drastically decreased. As compared with fresh yield of planting on April 20(352kg /10a), that of May 20 was 109% and June 20 was 103%, while those of after July 20 were from 41% to 19%. There was a highly positive correlation between dry tuber yield and the number of tubers per $m^2(r=0.991^{**})$. Tuber yields for commercial use(diameter over 7.1mm) were high in planting on May 20(322kg /10a) and on June 20(299kg /10a). It was suggested that optimum field planting time for in vitro multiplied tuber of Pinellia ternata(Thunb.) Breit was from May 20 to June May 20.
