Drought Stress Influences Photosynthesis and Water Relations Parameters of Synurus deltoides (건조스트레스가 수리취의 광합성 및 수분관련 특성에 미치는 영향)

  • This study was conducted to find out the influence of drought stress on physiological responses of Synurus deltoides. Drought stress was induced by withholding water for 25 days. Leaf water potentials were decreased of both predawn (${\Psi}_{pd}$) and mid-day (${\Psi}_{mid}$) with increasing drought stress, but water saturation dificit (WSD) was 7 times increased. ${\Psi}_{pd}-{\Psi}_{mid}$ showed the significant difference of 0.22~0.18 MPa in stressed before 10 days, and nonsignificant as treatment time became longer. A strong reduction of stomatal conductance ($gH_2O$) and stomatal transpiration rate (E) were observed after 15 days of drought stress Significant reductions of net apparent quantum yield (${\Phi}$) and maximum photosynthesis rate ($Pn_{max}$) were observed after 20 days of drought stress; However, water use efficiency (WUE) was shown the opposite trend. This implies that decrease of photosynthesis rate may be due to an inability to regulate water and $CO_2$ exchanged through the stomata. From JIP analysis, flux ratios (${\Psi}_O$ and ${\Phi}_{EO}$) and performance index on absorption basis ($PI_{ABS}$) were dramatically decreased withholding water after 15 days, which reflects the relative reduction of photosystem II activity. The leaf of S. deltoides showed osmotic adjustment of -0.35 MPa at full turgor and -0.40 MPa at zero turgor, and also cell-wall elastic adjustment of 9.4 MPa, indicating that S. deltoides tolerate drought stress through osmotic adjustment and cell-wall elastic adjustment. The degree of change in water relations parameters such as Vo/DW, Vt/DW decreased with increasing drought stress. This result showed that S. deltoides was exhibited a strong reduction of photosynthetic activity to approximately -0.93 MPa of predawn leaf water potential, and both of osmotic adjustment and cell-wall elastic adjustment in drought stress condition appears to be an important adaptation for restoration in this species.

Chemical and Spectroscopic Characterization of Soil Humic and Fulvic Acids and Sorption Coefficient of Phenanthrene: A Correlation Study (토양 휴믹물질의 화학적.분광학적 특성에 따른 페난트린 흡착상수와의 상관성 규명에 대한 연구)

  • In this study, the organic carbon normalized-sorption coefficients (Koc) for the binding affinity of phenanthrene (PHE) to 16 different soil humic and fulvic acids of various origins were determined by fluorescence quenching. The humic and fulvic acids used in this study were isolated from 6 different domestic soils including Mt. Hanla soil, IHSS standard soil and peat as well as Aldrich humic acid and characterized by elemental composition, ultraviolet absorption at 254 nm, composition of main structural fragments determined by CPMAS $^{13}$C NMR. The Koc values($\times$10$^4$, L/kg C) for each of HA and FA samples were in the range of 1.48$\sim$8.65 and higher in HA compared to that of FA(3.13$\sim$8.65 vs 1.48$\sim$2.48) in the experimental condition([PHE]/[HS] = 0.02$\sim$0.2(mg/L)/(mg-OC/L), pH 6). The correlation study between the structural descriptors of humic and fulvic acids and log Koc values of phenanthrene, show that the magnitude of Koc values positively correlated with the UV$_{254}$ absorptivity([ABS]$_{254}$) and two $^{13}$C NMR descriptors (C$_{Ar-H,C}$, $\sum$C$_{Ar}$/$\sum$C$_{Alk}$), while negatively correlated with the independent descriptors of the(N+O)/C atomic ratios and $^{13}$C NMR descriptors (I$_{C-O}$/I$_{C-H,C}$). These results confirmed that the binding affinity for the hydrophobic organic compound, phenanthrene are significantly influenced by the polarity and aromaticity of soil humc and fulvic acids.

The study on cytotoxicity of cytokines produced by the activated human NKT cells on neuroblastoma (활성화된 자연살상 T 세포(NKT)에서 생성된 사이토카인에 의한 신경모세포종의 세포독성에 관한 연구)

  • Purpose : ${\alpha}$-Galactosylceramide (${\alpha}$-GalCer)-stimulated human $V{\alpha}24$ natural killer T (NKT) cells exert antitumor activity against some leukemia in a CD1d dependent and TCR-mediated manner, but could not kill CD1d - negative neuroblastoma (NB) cells. There are few reports about the direct antitumor effect of highly secreted cytokines by these cells on activation. In this study, using a cell-free supernatant (SPN) collected from plate bound hCD1d/${\alpha}$ GalCer tetramers-stimulated NKT cells, we examined whether they could be helpful in the immunotherapeutic treatment of NB. Methods : Cells were cultured in IMDM. The cytokines produced by NKT cells were measured with Cytometric Bead Array (CBA) analysis. Cell viability was evaluated by calcein-AM fluorescence with digital image microscopy scanning (DIMSCAN). The percentage of specific apoptosis was calculated by flow cytometric detection of apoptosis using annexin V and 7-AAD. Results : The activated NKT cells secreted high levels of IL-2, INF-${\gamma}$, TNF-${\alpha}$. The SPN was significantly cytotoxic against four out of eight tested NB cell lines, through mainly apoptosis as evidenced by annexin-V staining and inhibition with the pretreatment of pancaspase blocker. This apoptosis was significantly inhibited when anti-TNF-${\alpha}$ and anti-IFN-${\gamma}$ neutralizing mAbs were used separately and it was completely abolished when the two mAbs were combined. Conclusion : IFN-${\gamma}$ and TNF-${\alpha}$ produced by NKT cells could exert synergistically direct antitumor activity through apoptosis on some NB cell lines.

Study on verification of various national standards regarding phthalate testing in industrial products (공산품 중 프탈레이트류 함유량 분석법에 관한 국내외 표준의 검증 연구)

  • As phthalates classified as toxic to reproduction category 2 and endocrine disrupting chemicals were more strictly regulated as Substances of Very High Concern (SVHC) for authorization in under EU REACH and considered as priority substances in RoHS II, standardization of phthalate testing method is now being proposed in IEC 62321 of IEC TC 111 and the 2nd revision of KS M 1991 is also finished. In order to assist standardization activities related to phthalating testing, solvent extraction part of existing national standards were compared and verified. Recovery of DEHP (diethylhexyl phthalate) from PVC (polyvinyl chloride) by Soxhlet extraction increased in the order of methanol, toluene, dichloromethane and hexane from 46.9% to 95.3% as measured by GC-MS. Optimum extraction time was verified to be 6 hours using hexane. Recovery of DBP (dibutyl phthalate), BBP (butylbenzyl phthalate), and DEHP from different matrixes such as PVC, nitro cellulose, ABS (acrylonitrile butadiene styrene). and EPDM(ethylene propylene diene monomer) rubber were evaluated to be more than 90% up to 99%. The detection limits of phthalates in solvent extraction followed by GC-MS analysis were 0.08~0.3 ${\mu}g/mL$ in solution and 8~30 mg/Kg in polymeric samples. GC-MS analyses of phthalates were carried out using different solvent extraction based on the EN 14372, ASTM D 7083, Japanese test method (MHLW 0906-4) and KS M 1991, proving that equivalent recoveries ranging from 98%~99% were obtained. DBP and DEHP were detected in three consumer products such as a child toy, a power cable and manicure with the amount of 22~1,910 mg/kg.

Chlorophyll Fluorescence and $CO_2$ Fixation Capacity in Leaves of Camellia sinensis, Camellia japonica, and Citrus unshiu (차나무, 동백나무, 귤나무 잎에서 엽록소 형광 및 $CO_2$ 흡수능의 비교 분석)

  • The chlorophyll fluorescence and photosynthetic $CO_2$ fixation capacity of leaves from three major crop trees found on Jeju Island, Camellia sinensis L., Camellia japonica L., and Citrus unshiu M., were analyzed. The photosynthetic $CO_2$ fixation rate of C. sinensis was similar to that of C. unshiu, and much higher than that of C. japonica which belongs to the same genus. Stomatal conductance in the three species was high at dawn and low during daytime. The intercellular $CO_2$ concentration of the three species was also high at dawn and decreased at midday. The transpiration rate showed an opposite trend from the intercellular $CO_2$ concentration. The photochemical efficiencies of PSII (Fv/Fm) in C. sinensis were slightly lower at midday compared to the level at dawn and/or dusk. The decline in Fv/Fm of C. sinensis at midday was much smaller than that of C. japonica. These results indicate that C. sinensis is better acclimated to high levels of radiation under natural conditions in late summer, although its PSII reaction center was inhibited by strong radiation. Of the chlorophyll fluorescence parameters in the species, the RC/CS decreased significantly while the ABS/RC, TRo/RC, ETo/RC, and DIo/RC increased significantly at midday in late summer. However, C. unshiu did not show significant changes in these values depending on the time of day. Among the three species, the daily $CO_2$ fixation rate in C. sinensis ($320.1mmol\;m^{-2}d^{-1}$) was the highest, followed by that of C. unshiu ($292.5mmol\;m^{-2}d^{-1}$) and C. japonica ($244.8mmol\;m^{-2}d^{-1}$). Thus, C. sinensis may be a valuable crop tree in terms of the uptake of $CO_2$ under natural field conditions.

Quality Characteristics and Antioxidant Activity of Puffed Rice Vinegar Added with Lemon Balm Extracts (레몬밤 추출물을 함유한 팽화미 식초의 품질 특성 및 항산화 활성)

  • To enhance the usability of puffed rice, we evaluated the quality characteristics and antioxidant activity of vinegar made with puffed rice and lemon balm. First, puffed rice containing lemon balm extracts was fermented for alcohol production. The alcoholic fermented liquid was adjusted to an alcohol content of 6% and an initial acidity of 2% for vinegar fermentation. The pH and alcohol content of the lemon balm vinegar decreased overall as fermentation progressed. The total acidity was 5.20±0.01% (2.0% lemon balm) to 5.80±0.01% (0.0% lemon balm) on the 15th day of fermentation, and the total acidity was slightly lower as the amount of added lemon balm was increased. Citric, succinic, and acetic organic acids were detected in the vinegar and the acetic acid content was highest at 4.81% to 5.37% on the 15th day of fermentation. We found that the total polyphenol and total flavonoid content of the lemon balm vinegar increased as we added lemon balm, and the content on the 15th day of fermentation was 179.4±4.5 (0.0% lemon balm) - 340.8±2.6 mg/kg (2.0% lemon balm) and 1.5±0.3 (0.0% lemon balm) - 31.7±0.3 mg/kg (2.0% lemon balm), respectively. The higher the amount of lemon balm added, the higher the DPPH radical scavenging activity, the FRAP (reducing power), and the ABTS radical scavenging activity. Moreover, those activities after 2.0% lemon balm treatment were 82.99±0.61%, 1.08±0.01 abs., and 87.67±0.68%, respectively, on the 15th day of fermentation. These results suggest that vinegar with added lemon balm may be used as an antioxidant agent.

Der Verlust der Amtsfähigkeit bzw. des Wahlrechts und das Gebot der Individualisierung der Strafen (선거범에 대한 자격제한과 형벌개별화원칙)

  • Wer wegen eines Wahldelikts zu Geldstrafe von mehr als 1 Million Won verurteilt wird, verliert nach ${\S}$ 18 Abs. 1 Satz 3, ${\S}$ 19 Satz 1, ${\S}$ 266 des Koreanischen Wahlgesetzes $f{\ddot{u}}r$ die Dauer von $f{\ddot{u}}nf$ Jahren die $F{\ddot{a}}higkeit$, ${\ddot{o}}ffentliche$ ${\ddot{A}}mter$ zu bekleiden und Rechte aus ${\ddot{o}}ffentlichen$ Wahlen. Bei Verurteilung zu Freiheitsstrafe wegen eines Wahldelikts ${\ddot{A}}mter$ sich die Dauer des Verlusts der $Amtsf{\ddot{a}}higkeit$ und des aktiven bzw. passiven Walhrechts auf 10 Jahre. Dies erfolgt kraft Gesetzes. Das $hei{\ss}t$, dass die Entscheidung ${\ddot{u}}ber$ das Ob und die Dauer des Verlusts nicht im Ermessen des Gerichts steht. Allerdings sollte $diesbez{\ddot{u}}glich$ nicht verkannt werden, dass ein deratriger Entzug von $Amtsf{\ddot{a}}higkeit$, $W{\ddot{a}}hlbarkeit$ u.s.w., mit dem eine Straftat geahndet werden soll, selber von Natur aus eine Art Strafen darstellt. Der im ${\S}$ 41 des Koreanischen StGB geregelte Strafen-Katalog $enth{\ddot{a}}lt$ $n{\ddot{a}}mlich$ eine zeitlich begrenzte Aberkennung des oben genannten ${\ddot{o}}ffentliche$n Rechtsstatus als eine Art Ehrenstrafen. Nicht einleuchtend ist, warum das Wesen der Sanktion $gem{\ddot{a}}{\ss}$ ${\S}$ 18 Abs. 1 Satz 3, ${\S}$ 19 Satz 1, ${\S}$ 266 des Koreanischen Wahlgesetzes, die den gleichen Zweck und die gleiche Rechtsfolge wie die im ${\S}$ 41 des Koreanischen StGB geregelte Ehrenstrafe hat, nicht als Strafe aufgefasst werden sollte. Handelt es sich bei der oben genannten Sanktion um eine Art Ehrenstrafen, so stellt sich die Anforderung, sie je nach der Eigenart der begangenen Tat bzw. des $T{\ddot{a}}ters$ zu individualisieren. Das Gebot der Individualisierung der Strafen, welches $haupts{\ddot{a}}chlich$ vom materiellen Rechtsstaatsprinzip ableitbar ist, kann im Grunde nur verwirklicht werden, wenn das Gericht dazu befugt ist, unter $Ber{\ddot{u}}cksichtigung$ der konkreten $Umst{\ddot{a}}nde$ jedes Einzelfalls ${\ddot{u}}ber$ eine angemessene Strafe zu befinden. Somit ist der kraft Gesetzes eintretenden Verlust der $Amtsf{\ddot{a}}higkeit$ und der $W{\ddot{a}}hlbarkeit$ nur schwer mit dem Gebot der Individualisierung der Strafen vereinbar. Es $w{\ddot{a}}re$ deshalb $w{\ddot{u}}nschenswert$, wenn der Gesetzgeber eine Reform in Betracht ziehen $w{\ddot{u}}rde$, welche den Ersatz des kraft Gesetzes automatisch eintretenden Entzugs der $Amtsf{\ddot{a}}higkeit$ bzw. des Wahlrechts durch die gerichtliche fakultative Aberkennung von diesen Statusrechten beinhaltet.

Photosynthesis, Growth and Yield Characteristics of Peucedanum japonicum T. Grown under Aquaponics in a Plant Factory (식물공장형 아쿠아포닉스에서 산채 갯기름의 광합성, 생육 및 수량 특성)

  • This study aimed to determine the photosynthesis and growth characteristics of Peucedanum japonicum T. grown under aquaponics in a plant factory (AP) by comparing those grown under hydroponic cultivation system (HP). The AP system raised 30 fishes at a density of 10.6 kg·m-3 in a 367.5 L tank, and at HP, nutrient solution was controlled with EC 1.3 dS·m-1 and pH 6.5. The pH level ranged from 4.0 to 7.1 for the AP system and 4.0 to 7.4 for the HP system. The pH level in the AP began to decrease with an increase in nitrate nitrogen (NO3-N) and lasted bellower than pH 5.5 for 15-67 DAT. It was found that ammonium nitrogen (NH4-N) continued to increase even under low pH conditions. EC was maintained at 1.3 to 1.5 dS·m-1 in both systems. The concentration of major mineral elements in the fish tank was higher than that of the hydroponics, except for K and Mg. There was no significant difference in the photosynthesis characteristics, but the PIABS parameters were 30.4% lower in the AP compared to the HP at the 34DAT and 12.0% lower at the 74DAT. There was no significant difference in the growth characteristics, but the petiole length was 56% longer in the leaf grown under the AP system. While there was no significant difference in the fresh and dry weights of leaf and root, the leaf area ratio was 36.43% higher in the AP system. All the integrated results suggest that aquaponics is a highly-sustainable farming to safely produce food by recycling agricultural by-products, and to produce Peucedanum japonicum as much as hydroponics under a proper fish density and pH level.

Comparison of Antioxidant Activity According to Silkworm Cultivars (품종에 따른 국내산 누에의 항산화 활성 비교)

  • Although varieties of silkworms, which have recently attracted attention as a health functional food, are being produced, studies on the differences in the functionality of different silkworm varieties are insufficient. Therefore, in this study, the antioxidant activities of different silkworm breeds bred in domestic farms were analyzed, and the potential for their cultivation as specialized varieties with excellent antioxidant function was investigated. To compare antioxidant activity, four varieties of silkworms, white Bakokjam, Golden silk, Yeonokjam, and Hanseongjam, were bred; water and ethanol extracts of these silkworms were prepared on the 3rd and 5th days of the 5th instar larval stage. The highest extraction yield was seen for the water extract from the Golden silk variety on the 3rd day of the 5th instar; the highest total phenolic compound and flavonoid contents were observed for the water extract (86.11±4.04 ㎍/mg GE) and 70% (v/v) ethanol extract (46.70±2.81 ㎍/mg QE). Bakokjam and Yeonokjam exhibited DPPH radical scavenging activity of up to 78% and showed the highest nitrite scavenging activity (85%) at pH 1.2. The maximum SOD-like activity of Yeonokjam was about 47%. Furthermore, 48 ㎍/ml of the Yeonokjam extract showed a reducing power of 0.7 abs, which was the best among the four varieties. Considering these results, the Yeonokjam (on the 3rd day of the 5th instar) had antioxidant activity and represents a silkworm cultivar that would be suitable for cultivation as a health food.

Physiological and Growth Responses of M. thunbergii to Different Levels of Fertilization (시비처리에 따른 후박나무의 생리 및 생장 반응)

  • In the current study, four groups; control, 500, 1000, and 2000 mg/L, were treated to investigate the effects of physiological and growth characteristics on Machilus thunbergii under various fertilization levels. As a result of the physiological response to the fertilization treatment, the fertilized group demonstrated relatively higher levels of A, ITE, WUEi, Vcamx, PIabs, and SFIabs in comparison to the control. The best photosynthetic mechanism was most clearly seen at 1000 mg/L, which involved gas exchange through active stomatal opening and closing. For a productive photosynthetic mechanism. As seen in the growth response of M. thunbergii to fertilization treatment, the fertilized group has significantly higher height, DRC, leaf dry weight, stem dry weight, total dry weight, LWR, and SWR than the control group. A healthy seedling quality index was particularly evident at 1000 mg/L, and other growth indicators were also at a decent level. 500 mg /L also demonstrated growth characteristics that were comparable to those at 1000 mg/L. As a result, M. thunbergii featured the best physiological and growth characteristics in response to the fertilization treatment at 1000 mg/L, and 500 mg/L also showed a similar trend, which is considered to be a good option from an economical perspective.
