Electrochemical Behaviors of ABTS2- on the Thiol-modified Gold Electrodes

  • The electrochemical properties of the redox mediator, 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonate) ($ABTS^{2-}$) were studied using cyclic voltammetry. The measured potentials (${E^o}'$ vs SCE) of the two redox couples of ABTS are 0.45 V for $ABTS^{2-}/ABTS^{\cdot-}$ and 0.87 V for $ABTS^{\cdot-}/ABTS^0$. The rate constant for heterogeneous electron transfer and the diffusion coefficients for $ABTS^{2-}$ are $5x10^{-3}cm\;s^{-1}$ and $3.1x10^{-6}cm^2\;s^{-1}$, respectively. Our interest in $ABTS^{2-}$ stems from the fact that this molecule functions as a substrate to the copper oxidase, laccase, by providing the reducing equivalents necessary for the biocatalyzed reduction of dioxygen to water. Consequently, when laccase is tethered to an electrode surface or dissolved in solution, $ABTS^{2-}$ can be used to quantify enzyme activity electrochemically.

  • Free radical scavengers in the bisdemethoxycurcumin (BDMC), demethoxycurcumin (DMC) and curcumin of turmeric (Curcuma longa) were screened, identified, quantified and isolation using coupled off-line-2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and on-line screening high-performance liquid chromatography (HPLC)-ABTS assay. There was a very small margin of error between the off-line-ABTS method and the on-line screening HPLC-ABTS method.

Screening of New Mediators for Lignin Degradation Based on Their Electrochemical Properties and Interactions with Fungal Laccase

  • This study was performed to evaluate extensive electrochemical characteristics of 23 commercially available mediators for laccase. Electrochemical properties, interactions with laccases, and ability to degrade lignin were compared for selected mediators. Among them, NNDS has very similar electrochemical properties in terms of reversibility and redox potential (about 470 mV vs. Ag/AgCl at pH=7) compared to ABTS which is a well-known mediator. Specific activity of purified laccase from Cerrena unicolor was determined by both 2,2'-azino-bis-(3-ethylbenz-thiazoline-6-sulfonic acid) (ABTS) and 1-nitroso-2-naphthol -3,6-disulfonic acid (NNDS). The specific activity of the laccase was 23.2 units/mg with ABTS and 21.2 units/mg with NNDS. The electron exchange rate for NNDS with laccase was very similar to that for ABTS, which meant that NNDS had similar mediating capability to ABTS. Determining methanol concentration after reacting with laccase compared to lignin degradation capabilities of both ARTS and NNDS. ARTS or NNDS alone cannot degrade lignin, but in the presence of laccase enhanced the rate of lignin degradation. ABTS showed better activity in the beginning, and the reaction rate of NNDS with lignin was about a half of that of ABTS at 10 minute, but the final concentration of methanol produced in 1 hour was very similar each other. The reason for similar methanol concentration for both ABTS and NNDS can be interpreted as the initial activity of ABTS was better than that of NNDS, but ABTS would be inhibited laccase activity more during the incubation.

ABTS+ Radical, Hydroxy Radical (OH), Nitric Oxide (NO), and Ferric Ion Reducing Antioxidant Power (FRAP) Effects of Ethanol Extracts from Four Seaweed Species for Noodles (국수에 대한 4종 해조류 에탄올 추출물에 의한 ABTS+, OH 라디칼, NO 라디칼, 철 이온 환원력)

  • The authors evaluated the scavenging activities of ABTS+ radical, hydroxy radical (OH), nitric oxide (NO), and ferric ion reducing antioxidant power (FRAP) from ethanol extracts of four edible alga, Enteromorpha linza, Porphyra tenera, Sargassum fusiforme, and Undaria pinnatifida. ABTS+ scavenging activity was analyzed according to the method of Brand-Williams et al. ABTS+ scavenging activity of S. fusiforme was evaluated to 61.8% at 8.0 mg/ml. ABTS+ scavenging activity of P. tenera was evaluated to 35.7% at 8.0 mg/ml. P. tenera and U. pinnatifida showed similar inhibitions of ABTS+ scavenging activity. According to the results of the OH assay in seaweed, inhibitory activities were in the order of S. fusiforme > P. tenera > U. pinnatifida > E. linza. The results showed scavenging activity for NO in the following order of potency: S. fusiforme > P. tenera > U. pinnatifida > E. linza with concentration values of 8.0 mg/ml. The NO scavenging activities of dough, which was instant noodles mixed with S. fusiforme and 3.5% salt, were 27.2% at 8.0 mg/ml. After boiling for 5 minutes, FRAP scavenging activity of instant noodles mixed with extracts of U. pinnatifida was evaluated to 31.5% at 8.0 mg/ml. S. fusiforme showed the highest inhibition activity of ABTS+, OH, NO, and FRAP among the four algae. Thus, these findings provide evidence that P. tenera, U. Pinnatifida, S. fusiforme, and E. linza extracts could become sources of natural antioxidants.

Antioxidant Properties (ABTS, FRAP, Total Phenolic Content) of Alaska and Gochujang Pollock Roes and Fermented Pollock Roe Seasoning (고추장과 발효액이 첨가된 알래스카 산 프리미엄 명란의 ABTS, FRAP, total phenolic acid의 항산화 특성 분석)

  • The Alaska Pollock (Gadus chalcogrammus) is distributed in an arc across the North Pacific Ocean. Distilled water extracts (DWE) and ethanol extracts (ETE) of 1.0 mg/ml concentrations of raw Alaska Pollock roe, premium Gochujang Pollock roe, and premium fermented Pollock roe seasoning were evaluated for estimated 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS), ferric reducing antioxidant potential (FRAP), and total phenolic content. The ABTS scavenging activity of the raw Alaska roe DWE and ETE were evaluated at 50.1% and 53.1%, respectively. The ABTS scavenging activity of the Gochujang roe DWE was 68.7% and of the ETE was 70.4%; for the fermented seasoning it was 71.3% and 71.6% for the DWE and ETE, respectively. The ABTS $EC_{50}$ values of the raw roe DWE and ETE were 12.49 ug/ml and 12.21 ug/ml, respectively. The FRAP $EC_{50}$ values of the Gochujang roe DWE and ETE were 10.67 ug/ml and 10.56 ug/ml, respectively, and the $EC_{50}$ values for total phenolic content for the fermented seasoning DWE and ETE were 10.45 ug/ml and 10.31 ug/ml, respectively. When Gallic was acid used as a control, the relative total phenolic content scavenging activity in each ETE was 52.0% (raw Alaska roe), 61.1% (Gochujang roe), and 63.6% (fermented seasoning). In the present study, higher ABTS, FRAP, and total phenolic content were observed in the Gochujang Pollock roe and fermented roe seasoning than in the Alaska Pollock roe.

Synthesis, Structure, and Peroxidase Activity of an Octahedral Ru(III) Complex with a Tripodal Tetraamine Ligand

  • A new octahedral Ru(III) complex with a tripodal tetraamine ligand, tpea = tris[2-(1-pyrazoyl)ethyl]amine, has been prepared and characterized. The single crystal X-ray crystallographic study of the complex revealed that the complex has a near octahedral geometry with the tetradentate ligand and two chloride ions. Peroxidase activity was examined by observing the oxidation of 2,2'-azinobis(3-ethylbenzothiazoline)-6-sulfonic acid (ABTS) with hydrogen peroxide in the presence of the complex. Amount of $ABTS^{+{\bullet}}$ generated during the reaction was monitored by UV/VIS and EPR spectroscopies. After the initiation of the peroxidase reaction, $ABTS^{+{\bullet}}$ concentration increases and then decreases after certain time, indicating that both ABTS and $ABTS^{+{\bullet}}$ are the substrates of the peroxidase activity of the Ru(III) complex.

  • The Antioxidant activity screening identification of five kind compounds in Artemisiae annuae herba with the on-line screening high performance liquid chromatography (HPLC) $ABTS^+$ assay. The various experimental variables such as the extraction time (h) and extraction solvent composition (%) of dipping method were investigated efficiently extraction at the room temperature $25^{\circ}C$. The results, the highest yield of total extract amount (0.458 g, 15.250%) was obtained by dipping method with 100% water and extraction time to 3 h. And the on-line screening HPLC-$ABTS^+$ assay method was rapid and efficient to search for bioactivity from natural products.

  • In this work, we describes analysis of the antioxidant potential of Korean green tea phenolics using an high-performance liquid chromatography (HPLC) on-line $ABTS^{+}$ antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) radical scavenging ability, the extraction of catechine compounds from Korean green tea were performed by various temperature and time. The optimum operating conditions were experimentally determined to analyze the catechine compounds in the pretreatment extracts. From the results, the extraction temperature $60^{\circ}C$, time 3 min was selected as an optimal antioxidant activity condition. The analysis by $C_{18}$ column was performed, the flow rate of mobile phase and UV wavelength was fixed at 1.0 ml/min and 254 nm, respectively. the mobile phase was composed from acetonitrile and water, and the gradient elution mode were applied.

Antioxidant Activity and Functional Components of Corn Silk(Zea mays L.) (옥수수 수염의 항산화 활성과 기능성분 분석)

  • In order to find out superior corn line which has a strong antioxidant activity in the corn silk, antioxidant compounds and antioxidant activity were measured from various inbred and F1 lines. As a result, the contents of phenolic and flavonoid compounds range from 532 mg/100 g to 3,274 mg/100 g and from 980 mg/100 g to 2,420 mg/100 g respectively. Absorbance at 517 nm for contents of anthocyanins ranges from 0.05 to 0.76. Correlation coefficients between antioxidant compounds and various antioxidant assays such as DPPH, ABTS and FRAP were analyzed. Correlation coefficients between antioxidant activity and phenolic compounds and anthocyanin were significantly high in ABTS and FRAP assays. Considering acidic extraction condition and intervention between anthocyanins and DPPH solution, ABTS assay and FRAP assay are more suitable methods to evaluate antioxidant activity of corn silks. Especially, ABTS assay is thought to be the best method among three assays because the antioxidant activity in ABTS assay showed high correlation with phenol, flavonoid and anthocyanin compounds respectively. Among the samples, S15 which showed the highest contents of total phenolic compounds and the most potent antioxidant activity in ABTS and FRAP assay will be a good source for functional material.

Monitoring of antioxidant activities with dried Gugija (Lycium chinensis Mill) extraction (건조 구기자의 추출에 따른 항산화 효능 모니터링)

  • Thise study aimed to determine the optimum antioxidant extraction conditions of dried Gugija (Lycium chinensis Mill). To determine the operational parameters, including ethanol concentration ($X_1$, 0~80%) and extraction time ($X_2$, 1~5 hr), a response surface methodology was applied to monitor brown color intensity, total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity. Coefficients of determinations ($R^2$) of the models were 0.8486~0.9214 (p<0.05~0.1) in dependent parameters. Brown color intensity of Gugija extracts reached a maximum of 0.75 (OD in 420 nm) under extraction conditions of 2.88 hr in 78.10% ethanol. Total phenolic compounds reached a maximum of $2,355{\mu}g$ under extraction conditions of 4.94 hr in 30.17% ethanol. ABTS radical scavenging activity was 13.83% at 4.61 hr and 16.21% ethanol. $Fe^{2+}$ chelating activity showed a maximum of 58.54% under extraction conditions of 3.39 hr in 0.76% ethanol. Optimum extraction conditions (5 hr extraction in 15% ethanol) were obtained by superimposing the contour maps with regards to total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity of dried Gugija. Maximum values of total phenolic compounds, ABTS radical scavenging activity, and $Fe^{2+}$ chelating activity under optimum extraction condition were $2,397{\mu}g$, 15.62% and 54.78%, respectively.
