형방지황양(荊防地黃揚)의 항Allergy 및 항염증 효과 (Anti-Allergic Effect of Hyeongbangjiwang-Tang)

  • The present study was conducted to investigate the anti-allergic activity of HBT. We investigated the anti-allergic effects of HBT in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator. HBT significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. The in vitro anti-inflammatory activities of HBT in LPS-stimulated RAW 264.7 cells were investigated. HBT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that HBT may be beneficial in the treatment of allergic inflammtory disease.

  • Besides (-) pimara-8(14), 15-dien-19-oic acid [I] which had already been isolated as an active anti-inflammtory principle of Aralia continentalis, (-) kaur-16-en-19-oic acid [II] was separated as another active component of the plant, by tracing albumin stabilizing activity. $IC_{50}$ of [II] for the protein stabilizing activity was 0.026mg/3ml, when those of [I] and phenylbutazone were 0.032 and 0.32 mg/3ml, respectively. Being investigated employing carrageenin-induced edema test in rat hind paw, the anti-inflammatory activity of [II] administered s. c. was slightly lower than that of phenylbutazone, whereas the activity of [II] administered p. p. was three times greater than that of phenylbutazone. These results of [II] were contrary to those of [I] in the aspect of administration routes.

  • Objectives : This study is to investigate the anti-inflammatory and anti-oxidant effects of Tongbi-san extract (TS) on RAW264.7 macrophages using by cell cytotoxicity, Nitric Oxide (NO) and Prostaglandin $E_2$ ($PGE_2$) production and 1,1-diphenyl-2-picryl ghdrazyl (DPPH) free radical scavenging capability. Methods : Cell cytotoxicity was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The production of NO was measured by Griess assay. The production of $PGE_2$ was measured by immunoassay. And the anti-oxidant activity was measured by the DPPH method. Results : TS did not increased significantly compared to the TS untreated group in the cell cytotoxicity. TS inhibited NO and $PGE_2$ production in lipopolysaccharide-stimulated RAW 264.7 cells. TS had the DPPH free radical scavenging capability. Conclusion : The anti-inflammtory and anti-oxidant effects of TS may be use for a treatment of anti-inflammatory diseases.

  • New prednisolone derivatives including methyl $20{\xi}-acetoxy-prednisolonate$ have been synthesized and evaluated for local anti-inflammatory agents with reduced systemic side effect. Methyl $20{\beta}-dihydroprednisolonate$ and methyl $20{\xi}-acetoxyprednisolonate$ showed comparable anti-inflammtory activity in vivo with significantly reduced systemic side effects.

  • Handayeolso-Tang(HDT) has been used as traditional medicine for the treatment of Taeumin TaeYang-Hanguel. The present study was conducted to investigate the anti-allergic activity of Handayeolso-Tang(HDT). We investigated the anti-allergic effects of HDT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. HDT significantly inhibited ${\beta}-hexosaminidase$ and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, HDT effectively inhibited anaphylactic shock in mice by 45% at a dose 120 mg/mouse versus PBS treated control after the I.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of HDT in LPS-stimulated RAW 264.7 cells were investigated. HDT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS -stimulated RAW 264.7 cells. These result provide evidences that HDT may be beneficial in the treatment of allergic inflammtory disease.

We investigated the anti-allergic effects of GHT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. GHT significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHT effectively inhibited anaphylactic shock in mice by 40% at a dose 100 mg/mouse versus PBS treated control after the l.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of GHT in LPS-stimulated RAW 264.7 cells were investigated. GHT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that GHT may be beneficial in the treatment of allergic inflammtory disease.

궁귀향소산(芎歸香蘇散)의 항Allergy 및 항염증 효과 (Inhibitory Effect of Allergic Reaction by Gunggwihyangso-San)

  • The present study was conducted to investigate the anti-allergic activity of Gunggwihyangso-San(GHS). We investigated the anti-allergic effects of GHS in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. Gunggwihyangso-San significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHS effectively inhibited anaphylactic shock in mice by 50% at a dose 80 mg/mouse versus PBS treated control after the I.p injection(8 mg/kg) of compound 48/80. GHS inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. The inhibition of inflammatory molecule expression on 3T3-L1 adipocytes by berberine is not mediated by leptin signaling

  • In our previous study, we have shown that berberine has both anti-adipogenic and anti-inflammatory effects on 3T3-L1 adipocytes, and the anti-adipogenic effect is due to the down-regulation of adipogenic enzymes and transcription factors. Here we focused more on anti-inflammatory effect of berberine using real time RT-PCR and found it changes expressions of adipokines. We hypothesized that anti-adipogenicity of berberine mediates anti-inflammtory effect and explored leptin as a candidate mediator of this signaling. We studied this hypothesis by western blot analysis, but our results showed that berberine has no effect on the phosphorylations of STAT-3 and ERK which have important roles on leptin signaling. These results led us to conclude that the anti-inflammatory effect of berberine is not mediated by the inhibition of leptin signal transduction. Moreover, we have found that berberine down-regulates NF-${\kappa}B$ signaling, one of the inflammation-related signaling pathway, through western blot analysis. Taken together, the anti-inflammatory effect of berberine is not mediated by leptin, and berberine induces anti-inflammatory effect independent of leptin signaling.

소음인(少陰人) 궁귀향소산(芎歸香蘇散)의 항(抗) 알레르기 및 항(抗) 염증에 미치는 실험적 연구 (Experimental Study on Anti-allergic and Anti-inflammatory Effects of Soeumin-Googwihyangso-san Methanol Extract in Vitro)

  • Soeumin-Googwihyangso-san(SGGHSS) has been used for the prevention or treatment of Soeumin-allergic rhinitis. This study was performed to demonstrate anti-allergic and anti-inflamatory effects of SGGHSS methanol extract in HMC cell lines and activated mouse B cells and CD4+ T cells. SGGHSS inhibited the production of TNF-$\alpha$ in PMA plus A23187 activated HMC-1 cells but not that of IL-6, as measured by ELISA. SGGHSS inhibited the expression of CD23 and surface IgE in B cells as determined by flowcytometry. It also inhibited secretion of IFN-$\gamma$ and IgG1, the Th1 related IgG type, but increased that of IL-4 in anti-CD40 and IL-4 treated B cells as measured by ELISA. As for Th cell differentiation, SGGHSS did not much affect IL-4 or IFN-$\gamma$. Taken together, our data showed that SGGHSS exerted an anti-inflammtory effect by inhibiting TNF-$\alpha$ in mast cells and has anti-allergic activity not via inhibition of CD4+ T cell, but via inhibition of B cells. These results suggest some evidence that SGGHSS can be applied to allergic disease.
