Synergistic Effect of Citric Acid on Antioxidant Property of Red Pepper (고추의 산화성(酸化性)에 대한 Citric Acid의 상승효과)

  • The antioxidant activity of ground red pepper, ethyl alcohol extracts of ground red pepper peel and alcohol extracts plus organic acid was studied by using edible soybean oil as a substrate. All the substrates were stored in a dark place at $45.0{\pm}0.5^{\circ}C$ respectively. The results of the study were as follow: 1. Ground red pepper, black pepper and cassia exhibited slight antioxidant activity and there was no significant difference in antioxidant activity among them. 2. Although both ground red pepper peel and red pepper seed showed antioxidant activity, ground red pepper peel exhibited greater antioxidant activity than ground red pepper peel did. 3. Ethyl alcohol extracts of red pepper peel showed strong antioxidant activity, but the residue did not show the antioxidant activity. 4. The antioxidant activity of the ethyl alcohol extracts of red pepper peel increased in direct proportion to the concentration of the extracts. 5. In connection with the synergistic effect of organic acid to the ethyl alcohol extracts of red pepper peel, citric acid showed a very strong synergistic effect on the antioxidant activity of red pepper, while ascorbic acid showed a weak effect. On the other hand, phosphoric acid showed an accelerating effect on the oxidation of oils. 6. The maximum antioxidant activity of the ethyl alcohol of ground red pepper peel was observed at the citric acid concentration of 0.04%, when tested at various concentration of citric acid.

Antioxidant and Pro-oxidant Activities of Hamcho (Salicornia herbacea L.) (함초의 항산화 및 산화 촉진 효과)

  • Hamcho (Salicornia herbacea) extracts were evaluated for total polyphenol content, antioxidant and pro-oxidant activities. The total polyphenol content was 1.81 g and 0.72 g per 100 g of dried sample in water and ethanol extracts respectively. Both water and ethanol extracts of Hamcho significantly exhibited antioxidant activity. The scavenging activity of hydroxyl radical was 13.8~26.2% and 14.2~16.0% in water and ethanol extracts respectively. The inhibitory effect of conjugated diene formation was 24.6~39.1% and 28.4~39.6% in water and ethanol extracts respectively. However, pro-oxidative effect was also observed in the Hamcho extracts. The Hamcho water extract showed the pro-oxidant effect by enhancing the formation of hydroxyl radical and conjugated diene. The ethanol extract of Hamcho induced conjugated diene formation at 0.5 mg/mL but not at 1 mg/mL. The hydroxyl radical formation was not induced by the Hamcho ethanol extract. Taken together, these results show that Hamcho extracts can act as pro-oxidants by generating hydroxyl radical or conjugated diene in addition to their antioxidant properties. Therefore, this study suggests that the physiological properties of Hamcho and its use as food materials should be considered with caution because antioxidant dietary plants such as Hamcho possess possible adverse effects induced by pro-oxidant activity.

Antioxidant Activity of Flavonoids Isolated from Vitex rotundifolia (순비기나무(Vitex rotundifolia)로부터 분리한 플라보노이드 성분의 항산화 활성)

  • The aim of this investigation was to evaluate antioxidant activity of crude extracts from the halophyte Vitex rotundifolia, their solvent fractions, and isolated compounds (1-3). Antioxidant capacity was determined by measuring DPPH radical, and authentic $ONOO^-$ and $ONOO^-$ generated from 3- morpholinsydnonimine (SIN-1) in vitro as well as degree of occurrence of intracellular ROS, NO and GSH in mouse macrophage Raw 264.7 cells. From comparative analysis, MeOH extract, n-BuOH, and 85% aq. MeOH solvent fractions showed significant antioxidant effect in DPPH radical and $ONOO^-$ assay systems. Activity-guided purification of n-BuOH and 85% aq. MeOH fractions led to the isolation of flavonoids 1-3. Among them, compound 1 exhibited excellent antioxidant effect in all bioassay systems tested. On the other hand, compounds 2 and 3 revealed potent inhibitory effect against $ONOO^-$ generated from SIN-1, comparable with the positive control penicillamine.

Effects of a Rubus coreanus Miquel supplement on plasma antioxidant capacity in healthy Korean men

  • Korean raspberry, Rubus coreanus Miquel (RCM), contains high concentrations of phenolic compounds, which prevent oxidative stress. To determine the effect of RCM on antioxidant capacity in humans, we assessed in vivo lipid oxidation and antioxidant enzyme activities from plasma in 15 healthy men. The subjects ingested 30 g of freeze-dried RCM daily for 4 weeks. Blood was taken at baseline and at the end of the study to determine blood lipid profiles, fasting plasma glucose, liver function, lipid peroxidation, and antioxidant enzyme activities. RCM supplementation had no effect on blood lipid or fasting plasma glucose concentrations but decreased alkaline phosphatase activity. RCM supplementation increased glutathione peroxidase activities (P<0.05) but had no effect on lipid peroxidation. These results suggest that short-term RCM supplementation may offer health benefits by enhancing antioxidant capacity in a healthy population.

Antioxidant Effects of Noni (Morinda citrifolia) Extracts Treated with Hel and Trypsin (염산과 트립신으로 처리한 노니(Morinda citrifolia) 추출물의 항산화 효과)

  • To investigate biological activity of noni extracts treated with HCl and trypsin, we measured the antioxidant activity through vitro assay and cellular system. Both water and lipid soluble fraction of noni extracts dose-dependently scav­enged DPPH radical. Superoxide scavenging activity of lipid soluble fraction after treating HCl and trypsin was significantly more potent than those of other fractions in NBT/xanthine oxidase assay, which suggests that antioxidant activity of noni extracts was increased by the treatment with HCl and trypsin. In antioxidant assay using RBL 2H3 cells, water soluble frac­tion of noni extracts had little effect on silica-induced reactive oxygen species generation, whereas lipid soluble fraction inhibited in a dose dependent manner. In non-treated noni extracts, effect of water soluble fraction on silica/$CuSO_4$-induced lipid peroxidation was more potent than that of lipid soluble fraction. However, the effects of noni extracts were reversed in noni extracts treated with HCl and trypsin. These data suggest that water soluble substances may be converted into lipid soluble substances by the treatment with HCl and trypsin. From the above results, it is suggested that lipid soluble fraction of noni extracts contain antioxidant used in vitro assay and RBL 2H3 cellular system. Such an effect of noni extracts may be increased by the treatment with HCl and trypsin.

Antioxidant Activity and Differentiation Effect of Taraxacum mongolicum Extracts against Hydrogen Peroxide-induced Oxidative Damage of MC3T3-E1 Osteoblast Cells (민들레 추출물의 항산화 활성 및 H2O2로 산화적 스트레스를 유도한 조골세포의 활성과 분화에 미치는 영향)

  • The correlation between osteoporosis and reactive oxygen species (ROS)-induced oxidative stress was investigated. Thus, interest in food and plants with antioxidant effects that can reduce damage caused by ROS during bone metabolism is heightening. In this study, the antioxidant effect of Taraxacum mongolicum on proliferation and differentiation of MC3T3-E1 cells under H2O2-induced oxidative stress was studied to investigate its protective effect against oxidative stress and its availability as an antioxidant material related to bone diseases. As a result, total polyphenol and total flavonoid contents of T. mongolicum were 33.65 mg/g and 4.45 mg/g, respectively. The T. mongolicum extract increased proliferation of both MC3T3-E1 cells and differentiated osteoblasts under $H_2O_2$-induced oxidative stress conditions. In addition, two differentiation markers, alkaline phosphatase activity and mineralization level in the T. mongolicum extract, tended to increase. These results indicate that T. mongolicum extract suppressed the damage to osteoblasts under oxidative stress and that it is potential antioxidant materials for preventing bone diseases.

  • The objective of this study was to evaluate the composition, pH, titratable activity, microbial properties, and antioxidant effect of yogurt using ginseng extract powder (GEP), Lactobacillus plantarum NK181, and Streptococcus thermophilus as the starter culture. Different concentration of GEP (0%, 0.5%, 1%, 1.5%, and 2% (w/v)) were used in the yogurt. During yogurt fermentation, pH was decreased; however, titratable acidity and viable cell counts were increased. The addition of GEP to yogurt led to a decrease in moisture content and an increase in the fat, ash, and total solids content. The antioxidant effect using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, ${\beta}$-carotene bleaching, and ferric reducing antioxidant power (FRAP) assay gradually increased with added GEP. Overall, yogurt fermented with 1% GEP was acceptable in terms of cell viability and antioxidant effect. These results might provide information regarding development of ginseng dairy products with enhanced antioxidant activities and probiotic properties.

The Effect of Antioxidant in the Electrical Properties of Homemade Insulating Oil (국산절연유의 전기적 특성에 미치는 산화방지제의 효과)

  • The main purporse of this paper is to study on the influence of temperature effect on electrical-properties, i.e., dielectric breakdown voltage, dielectric constant and resitivity of homemade insulating oil containing antioxidant. Transformer oil manufactured in Korea is taken for a sample, and this sample is classified into seven samples in which antioxidant is contained and not contained, and antioxidant is divided into three kinds of amount: 0.3%, 0.5%, 0.8%. Changing the tempe5rature of the samples by oil bath, the electric properties measured from the samples are as follows: (1) the variation of either the dielectric breakdown voltage or the resistivity are considerably large, but the dielectric constants are almost unchanging. (2) In view of electrical characteristic, the deterioration rate of the samples containing antioxidant is somewhat lower than that of other samples. (3) Nearly the same value is shown in the dielectric-breakdown voltage of the samples, in which contained the different amount of antioxidant: 0.3%, 0.5%, 0.8%, but the decrease rate of resitivity is lowest, 0.3%, of all. As the result of the experiment, This study can suggest that 0.3% be the optimum amount of antioxidant which will be added transformer oil manufactured in Korea and is useful for the understanding about the electrical properties of transformer oil containing antioxidant.

Antioxidant Activities of Flavonoids from the Leaves of Smilax china Linne (청미래덩굴 잎의 항산화 활성 성분)

  • In this study, we have investigated the antioxidant activities of the leaves of Smilax china Linne in order to find the antioxidant substances which has an scavenging effect of reactive oxygen species (ROS) from natural products. As a result, EtOAc and n-BuOH extracts of the leaves of Smilax china Linne exhibited potent antioxidant effect on various antioxidant experiment. The major components of antioxidant activity were isolated from EtOAc and n-BuOH extracts of the leaves of Smilax china Linne. Their structure of compounds were identified as kaempferol-7-O-${\alpha}$-L-rhamnopyranoside and kaempferol-3,7-O-${\alpha}$-L-dirhamnopyranoside by spectroscopic evidence, respectively. Antioxidant activities were observed in kaempferrol-7-O-${\alpha}$-L-rhamnopyranoside at 23 ${\mu}g$ and in kaempferol-3,7-O-${\alpha}$-L-dirhamnopyranoside at 27 ${\mu}g$ as concentration of DPPH 50% reduction.

  • In order to isolate and clarify the antioxygenic substances from the browning reaction products, the antioxidant activity of various amino acids and their browning reaction products were measured when they were reacted with xylose. Among nonpolar amino acids Met and Trp appeared to have stronger antioxidant effect than others. Most of polar and basic amino acids, however, did not have antioxidant activity. Ser and Cys showed a rather slight prooxidant effect. The browning reaction products of Trp and His had a higher level of antioxidant activity than that they were reacted as free amino acids. But the browning product of Met did not show the antioxidant activity. When all amino acids were divided on their polar characteristics, the higher optical density of the browning reaction products showed, the stronger antioxidant activity revealed.
