  • Effects of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on sheep red blood cells (sRBC)-induced arthus reaction (AR) were studied in ICR male mice and determined the plaque forming cells (PFC) numbers and hemagglutinin (HA) titer. Two weeks after sensitization of i.p. injection of sRBC (4x10$^{8}$ cells), ICR male mice were challenged by i.p. injection of sRBC (2$\times{10}^8$ cells). Five days after the challenge of antigen, paw edema induced three hours after the last challenge by Arthus reaction. (omitted)

  • Effect of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on heterologous passive cutaneous anaphylaxis (HPCA), the release of histamine, and Phospholipase $A_2$ (PLA2) and phosphadiesterase (PDE) activities were studied by the method of Levine and Vaz. Anti-serum was prepared from ovalbumin (OA)-sensitized male Balb/c mouse at two weeks after the last challenge of OA and alumina gel. Heterologous PCA test in rats were carried out to determine the contents of leakaged pigment in the dorsal skin 30 minutes after i.v. injection of 0.2 ml of 1 % OA and 1 % Evans blue mixture (1:1).   • Effect of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on the early- (EAR) and late-phase asthmatic responses (LAR) of guinea pigs were studied in vivo. Guinea pigs were sensitized by injection of 100 mg of ovalbumin (OA). Twenty-one days after sensitization, animals were challenged with exposure to aerosolized 1 % OA for five minutes in double-chambered plethysmograph box with jet nubulizer. Immediately and twenty-four hours after challenge, EAR and LAR ashmatic responses were determined the tidal volume (TV), respiration rate (RR) and specific airway resistance (sRaw), and then animals anethetized and taken the bronchoalveolar lavage fluid (BALF) by lavage the lung with HEPES buffer through cannulation into trachea.   • 새로운 혈소판 활성화인자 (PAF) 수용체 결합 저해활성 유효성분인 pinusolide ($IC_{50}$/=2.5$\times$$10^{-7}$M)와 arctigenin($IC_{50}$/=5.2$\times$$10^{-6}$M)을 측백엽(Biota orientalis)과 우방자(Arctium lappa)로부터 분리하여 이미 보고한 바 있다. PAF 수용체에 대한 구조활성 상관관계를 규명하고자 또한 용해도가 우수한 강력한 PAF 길항제를 개발하고자 이 길항제들의 유도체들을 다양한 유기화학반응을 이용하여 합성하였고 in vitro PAF receptor binding assay로 그 활성의 강도를 비교 검토하였다. 그 결과, lactone ring 부분은 unsaturated lactone이 saturated lactone보다 활성이 강하였고 ring이 개열되면 활성이 현저히 감소하였다. pinusolide 경우 exocylclic double bond 부분은 sp$^2$구조의 hydrophobic한 유도체가 활성이 강하였으며, methyl ester 부분은 hydrophobic한 유도체가 활성이 강하였다. Arctigenin의 경우 aromatic unit가 hydrophobic할수록 활성이 강하였다.

Analysis of Nutritional Components and Antioxidant Activity of Roasting Wooung (Burdock, Arctium lappa L.) and Jerusalem Artichoke (Helianthus tuberosus L.) (볶음 처리한 우엉과 돼지감자의 영양성분 및 항산화 활성)

  • In this study, we evaluated the nutritional components and functional activities of Wooung (burdock, Arctium lappa L.) and Jerusalem artichoke (Helianthus tuberosus L.) tea. Roasting burdock' contained 75.87% carbohydrates; in addition, the moisture content, crude fat, crude protein, and crude fiber were 10.43%, 1.77%, 8.50%, and 3.43%, respectively. Roasting Jerusalem artichoke showed 77.477% carbohydrate content, with moisture content, crude fat, crude protein, and crude fiber of 10.67%, 1.23%, 7.83%, and 2.80%, respectively. Roasting burdock's water-soluble dietary fiber content was 4.8 g/100 g and insoluble dietary fiber content was 1.5 g/100 g; whereas, roasting Jerusalem artichoke' water soluble dietary fiber content was 2.4 g/100 g and insoluble dietary fiber content was 1.6 g/100 g. The highest mineral contents in roasting burdock and Jerusalem artichoke were potassium and magnesium, in order. The results of amino acid analyses s indicated a total of 25 types in roasting burdock, with total amino acid content of 1,382.112 mg/100 g, and essential amino acid content of 766.031 mg/100 g. In total, 24 types of amino acids were separated and identified in roasting Jerusalem artichoke, with total amino acid content of 2,678.018 mg/100 g, and total essential amino acid content of 157.294 mg/100 g. Roasting burdock and Jerusalem artichoke' polyphenol contents were 32.56 and 29.56 mg GAE/g each, and their flavonoid contents were 16.54 and 16.71 CE/g each. $IC_{50}$ values of DPPH radical-scavenging activity of roasting burdock and Jerusalem artichoke were 12.99 and 19.74, respectively; and $IC_{50}$ values of hydroxyl radical-scavenging activity were 25.96 and 22.93, respectively.

  • This study evaluated the qualitative properties of burdock (Arctium lappa L) peels as food material. Proximate composition, sweetness, pH, total acidity, mineral, fatty acid, free sugar, and organic acid contents of preprocessed burdock were measured. Crude ash, protein, fat, and total dietary fiber contents of non-peeled treated lotus roots were higher than those of peeled treated lotus roots (p<0.05). $^{\circ}Bx$, pH, and total acidity were not different in the peeling process. Redness and yellowness of lotus roots were reduced by the peeling process, whereas lightness increased (p<0.05). The mineral contents of Ca, K, Fe, and P were higher in non-peeled lotus roots, whereas non-peeled treated lotus root showed higher K contents (p<0.05). Maltose content was not significantly different, whereas fructose, sucrose and maltose contents were significantly different. Total organic acid contents were higher in non-peeled lotus roots (p<0.01). However total free amino acid contents were higher in peeled lotus roots (p<0.01).

Proton Beam Sensitivity of Radical Scavenging Activity from Plant Resources (식물자원의 양성자 빔 조사에 대한 라디칼 소거능활성의 민감도)

  • 20 species of plants were extracted with Methanol and were investigated for DPPH radical scavenging activity and ABTs free radical scavenging activity to quest anti-oxidation ability depending on the proton beam irradiation quantity. In the proton beam irradiation, 15 species's activities increased but among them, Pharbities nil Choisy decreased at 10 KGray and 4 species' activity didn't change at all. In hydrogen ion radical elimination activity, Ulmus macrocarpa (84 %) showed the highest and Pharbitis nil Choisy showed 6 % decreasing at more than 1 KGray. By comparison with untreated $IC_{50}$ value, the beam-treated $IC_{50}$ value increased 6.3 times for Dioscorea batatas Decne. at 1 KGray, 2.1 times for Trichosanthes kirilowii Max., and 2.8 times for Dioscorea batatas Decne. at 5 KGray. In ABTs free radical elimination activity, the activity increased 60 % for Terminalia chebula Retzius compared with untreated one. Besides, the beam-treated $IC_{50}$ value increased 2 times for Gray Ephedra sinica Stapf, 2.5 times for Terminalia chebula Retz. and 2.4 times for Arctium lappa Linne at 1 KGray.

A Study on the Frequencies of Medicinal Herb Mombinations in the Prescriptions of "Wenbingtiaobian(溫病條辨)" ("온병조변(溫病條辨)" 수록(收錄) 처방(處方) 내(內)의 약물(藥物) 조합 빈도(頻度) 연구)

  • Through analyzing the frequencies of medicinal herb combinations in the prescriptions of "Wenbingtiaobian(溫病條辨)", we could understand the characteristics of prescription composition and author's medical thoughts. So we conducted this study that analyze combination of 258 medicinal herbs in 206 prescriptions, and found meaningful combinations of 700. Among these, the most meaningful combinations are the two of 8 herbs. One consists of Glycyrrhiza uralensis Fischer, Platycodon grandiflorum, Lonicera japonica Thunberg, Sojae Semen Praeparatum, Mentha piperascens (Malinv.) Holmes, Forsythia suspensa, Phyllostachys nigra var henonis, Schizonepeta tenuifolia var. japonic, the other consists of Glycyrrhiza uralensis Fischer, Platycodon grandiflorum, Lonicera japonica Thunberg, Mentha piperascens (Malinv.) Holmes, Forsythia suspensa, Arctium lappa, Phyllostachys nigra var henonis, Schizonepeta tenuifolia var. japonica. The former is Yeonkyo-san without Arctium lapp, the letter is Yeonkyo-san without Sojae Semen Praeparatum. It means these combinations are kind of modified Yeonkyo-san which is effective in outthrustting through the exterior with pungent-cool and clearing heat and detoxify. In conclusion, prescriptions of "Wenbingtiaobian(溫病條辨)" involve herbs mostly used to outthrust through the exterior with pungent-cool and clear heat and detoxify. Through this study, we know the author' s one of medical view points on outthrustting through the exterior with pungent-cool and clearing heat and detoxify.

Antimicrobial Activity of Medicinal Plants Against Bacillus subtilis Spore

  • Bacterial endospores, especially those of Bacillus and Clostridium genera, are the target of sterilization in various foods. We used Bacillus subtilis ATCC 6633 spores to screen novel antimicrobial substances against spores from medicinal plants. We collected 79 types of plant samples, comprising 42 types of herbs and spices and 37 types of medicinal plants used in traditional medicine in Korea and China. At a concentration of 1%(w/v), only 14 of the ethanol extracts exhibited antimicrobial activity against B. subtilis spores of at least 90%. Crude extracts of Torilis japonica, Gardenia jasminoides, Plantago asiatica, Fritllaria, and Arctium lappa showed particularly high sporicidal activities, reducing the spore count by about 99%. Consideration of several factors, including antimicrobial activity, extraction yields, and costs of raw materials, resulted in the selection of T. japonica, G. jasminoides, A. lappa, and Coriandrum sativum for the final screening of novel antimicrobial substances. Verification tests repeated 10 times over a 4-month period showed that the ethanol extract of T. japonica fruit reduced aerobic plate counts of B. subtilis spores the most, from $10^7$ to $10^4\;CFU/mL$ (99.9%) and with a standard deviation of 0.21%, indicating that this fruit is the most suitable for developing a novel antimicrobial substance for inactivating B. subtilis spores.

Further Screening for Antioxidant Activity of Vegetable Plants and Its Active Principles from Zanthoxylum schinifolum (식용식물의 항산화 효과 검색과 산초의 항산화 성분)

  • The antioxidant activity of methanol extracts of thirty plants was tested using the methol of 1, 1-diphenyl-2-pi-cryl hydrazyl (DPPH) reactivity. Four methanol extracts from Zingiber officinale, Piper nigrum , Zanthoxylum schinifolium and Capsocum annuum were found to be the most effective on DPPH radical scavenging activity. The next effective ones were Perilla frutescens , Sedium sarmentosum , Raphnus sativas, aArctium lappa, Beta vulgaris. Brassica oleracea var. Acephala, bBrassica juncea inorder, and the others did not show a considerable activity. The methanol extract obtained from the seed coats of Zanthoxylum schinifolium was fractinated with several sovlents. The interphase materials exhibited the strongest antioxidant activity and was further purified by silica gel and Sephadex LH-20 column chormatography. Two active principles were isolated and identified as quercetin -3-O-$\alpha$-L-rhamonopyranoiside(quercitrin) and quercetin 3-O-$\alpha$-D-galactopyranoside (hyperoside) by ultraviolet(UV), proton nuclear magetic resonance (1H-NMR) and carbon nuclear magnetic resonance (13C-NMR). Its antioxidative activity was a little higher that that of L-ascorbic acid.
