The integrated fixed-film activated sludge (IFAS) system is a variation of the activated sludge wastewater treatment process, in which hybrid suspended and attached biomass is used to treat wastewater. Although the function and performance of the IFAS system are well studied, little is known about its microbial community structure. In this study, the composition and diversity of the bacterial community of suspended and attached biomass samples were investigated in a full-scale IFAS system using a high-throughput pyrosequencing technology. Distinct bacterial community compositions were examined for each sample and appeared to be important for its features different from conventional activated sludge processes. The abundant bacterial groups were Betaproteobacteria (59.3%), Gammaproteobacteria (8.1%), Bacteroidetes (5.2%), Alphaproteobacteria (3.9%), and Actinobacteria (3.2%) in the suspended sample, whereas Actinobacteria (14.6%), Firmicutes (13.6%), Bacteroidetes (11.6%), Betaproteobacteria (9.9%), Gammaproteobacteria (9.25%), and Alphaproteobacteria (7.4%) were major bacterial groups in the attached sample. Regarding the diversity, totals of 3,034 and 1,451 operational taxonomic units were identified at the 3% cutoff for the suspended and attached samples, respectively. Rank abundance and community analyses demonstrated that most of the diversity was originated from rare species in the samples. Taken together, the information obtained in this study will be a base for further studies relating to the microbial community structure and function of the IFAS system.
The authors investigated the bacterial community attached to granular activated carbon(GAC) particles and the susceptibility of the community to chlorine disinfection. The study was carried out at the G Water Treatment Plant in Seoul, which was in full-scale operation. Bacteria attached to the surface of GAC increased gradually with treatment from $0.4{\times}106{\sim}8.5{\times}106 CFU/g$. TOC removal was under 1.0 mg/L due to increased bacterial community on the surface of GAC. It was found that TOC removal was closely related with physical and biological parameters such as pore volume and the number of attached bacteria. When the washed and the attached cells were disinfected with 1.0mg/L of chlorine for 1 hour, the washed cells with chlorination could be controlled, but the number of the attached cells increased gradually. The results suggest a possibility that the treatment and disinfection barriers can be penetrated and pathogenic bacteria may break into the drinking water supplying system.

Nitrifying bacterial community structures of suspended and attached biomasses in a full-scale integrated fixed-film activated sludge process were investigated by analyzing 16S rRNA gene sequences obtained from pyrosequencing. The suspended biomass had a higher number of ammonia-oxidizing bacterial sequences (0.8% of total sequences) than the attached biomass (0.07%), although most of the sequences were within the Nitrosomonas oligotropha lineage in both biomasses. Nitrospira-like nitrite-oxidizing bacterial sequences were retrieved in the suspended biomass (0.06%), not in the attached biomass, whereas the existence of Nitrobacter-like sequences was not evident. The suspended biomass had higher nitrification activity (1.13 mg N/TSS/h) than the attached biomass (0.07 mg N/TSS/h). Overall, the results made it possible to conclude the importance of the suspended biomass, rather than the attached biomass, in nitrification in the wastewater treatment process studied.
This investigation was performed in eutrophic lake within the framework of a series of studies to evaluate the significance of gram reaction for both bacterioplankton and attached bacteria in the dynamics of organic materials at various aquatic ecosystems. In Lake Kasumigaura as a representative of the highly eutrophic freshwater environments, the gram-stain characteristics of the bacterial community changed with the influx of pulses of phytoplankton, as those in the meso trophic environments. The predominency of the gram-negative forms in the bacterial community was about 57% for bacterioplankton and about 53% for attached bacteria. The statistical analysis of the difference of these two distributions showed that these communites were different. Both gram-negative and gram-positive bacteria attached to particles were shown to effect the formation and degradation of particulate organic matter. Gram-negative bacteria plankton participate exclusively in the dynamics of dissolved organic matter.
The concentration of organic compounds was analyzed at each step of BAC process though $BDOC_{total/rapid/slow}$. Further, bacteria communities and biomass concentrations measured FISH and ATP methods were analyzed. The bed volume (BV) of steady state is different from that of based on assessment of organic compounds removal. Bed volumes in DOC, $BDOC_{rapid}$ and $BDOC_{total/slow}$ removal at steady state were around 27,500 (185.8 day), 15,000 (101.4 day) and 32,000 (216.2 day), respectively. A biomass didn't change after the bed volume reached 22,500 (152.0 day) according to analyzing ATP concentration of bacteria. The concentration of ATP was 2.14 ${\mu}g/g$ in BV 22,500 (152.0 day). The total bacterial number was $4.01{\pm}0.4{\times}10^7$ cells/g at the bed volume 1,150 (7.8 day) (the initial operation) and the number of bacteria was $9.27{\pm}0.2{\times}10^9$ at the bed volume 58,560 395.7 day) that increased more than 200 times. Bacterial uptrend was reduced and bacterial communities were stabilized since BV 18,720 (126.5 day). When BV were 1,150 (7.8 day), 8,916 (60.2 day), 18,720 (126.5 day), 31,005 (209.5 day), 49,632 (335.3 day), 58,560 (395.7 day), a proportion of total bacteria for the Eubacteria were 60.1%, 66.0%, 78.4%, 82.0%, 81.3% respectively. ${\gamma}$-Proteobacteria group was the most population throughout the entire range. The correlation coefficient ($r^2$) between Eubacteria biomass and ATP concentration was 0.9448.
A novel hydrogen fermentation technique by using polyvinyl alcohol (PVA) gel beads as a biomass carrier was investigated. The hydrogen gas was stably produced throughout the experimental period in a continuous reactor. Even though the hydrogen productivity was suddenly decrease by experimental troubles, the bacteria attached to the PVA gel beads played as an inoculum, it was promptly recovered. The hydrogen yield per glucose was not very high ($1.0-1.2mol-H_2/mol-glucose$), thus the optimization of the experimental conditions such as ORP and HRT should be considered to improve the hydrogen productivity. Bacterial community was stable during experimental period after the PVA gel beads applying, which indicated that applying of biomass carrier was specific to keep not only the biomass but also the bacteria commonly. Clostridium species were phylogenetically detected, which suggested that these bacteria contributed to the hydrogen production in the biofilm attached to the PVA gel beads.
Biolgical activated carbon (BAC) processes are known to effectively remove organic pollutants in raw water, and biomass and attached bacterial species play an important role in removing process. In the present study, changes of bacterial biomass in granular activated carbon (GAC) process according to the depth and operating period were investigated. In addition, changes of bacterial biomass were also confirmed after UV exposure prior to the GAC process. Results from this this study showed that the bacterial biomass was decreased dependently according to the depth of GAC process. In case of UV pre-treatment, the bacterial biomass was declined significantly over the period of operation. However, changes in bacterial community were not shown during operation period without UV pre-treatment process. In conclusion, findings from this study may provide the useful information about the management of BAC process.

The use of biological-activated carbon (BAC) processes in water treatment involves biofiltration, which maximizes the bacteria's capabilities to remove organic matter. In this study, the distribution of the bacterial community was assessed in response to different types of BAC processes applied downstream in the Nakdong River. The bacterial biomass and activity were $1.20{\sim}34.0{\times}10^7$ CFU/g and 0.61~1.10 mg-C/$m^3{\cdot}hr$ in coal-based BAC, respectively. The attachment of the bacterial biomass and the removal efficiency of the organic carbon were greatest with the coal-based activated carbon. The bacteria attached to each activated carbon material were detected in the order of Pseudomonas genus, Chryseomonas genus, Flavobacterium genus, Alcaligenes genus, Acinetobacter genus, and Spingomona genus. Pseudomonas cepacia was the dominant species in the coal-based materials, and Chryseomonas luteola was the dominant species in the wood-based material.

The Biological Activated Carbon (BAC) process in the water treatments represents a kind of biofiltration process which capabilities of bacteria to remove organic matters are maximized. It enables to eliminate organic matters and effectively reduce microbial regrowth potentials. As attached bacteria employ natural organic matter as a substrate, they are significantly dependent on indigenous microorganisms. In this study, characteristics of bacterial community by culturable and unculturable Methods have been conducted in a pilot plant using SAC in water treatment process at the downstream of the Nakdong River. Based on the results, HPC and bacterial- production for coal-based activated carbon material were $1.20{\sim}56.2{\times}l0^7$ cfu/g and $1.2{\sim}3.7\;mgC/m^{3}h$, respectively, in the SAC process. The highest level of attached bacteria biomass and organic carbon removal efficiency was found in the coal-based activated carbon. The genera Pseudomonas, Flavobacterium, Alcaligenes, Acilzetobacter, and Spingomonas were identified for each activated carbon material. Pseudomonas vesicularis was the dominant species in the coconut- and coal-based materials, where as Pseudomonas cepacia was the dominant species in the wood-based material. The Scanning Electron Microscope (SEM) observation of the activated carbon surface also found the widespread distribution of rod form and coccus. The community of attached bacteria was investigated by performing Fluorescent in situ hybridization (FISH) analysis. a group was dominant in coal, wood and coccunt-based materials, ${\alpha},\;{\beta}\;and\;{\gamma}$ group ranged from 27.0 ${\sim}$ 43.0%, 7.1 ${\sim}$ 22.0%, 11.3 ${\sim}$ 28.6%, respectively. These results suggest that a group bacterial community appears to be regulated removal efficiency of organic material in water treatment process.

The concentration of organic compounds was analyzed at each step of BAC (biological activated carbon) process though BDOC (biodegradable dissolved organic carbon) total/rapid/slow. Further, bacteria communities and biomass concentrations measured DGGE (denaturing gradirnt gel electrophoresis) and ATP (adenosine triphosphate) methods were analyzed. The bed volume of steady state is different based on assessment of organic compounds removal. Bed volumes at steady state in DOC, $BDOC_{rapid}$ and $BDOC_{total/slow}$ removal were around 27,500, 15,000 and 32,000, respectively. A biomass didn't change after the bed volume reached 22,500 according to analyzing HPC (heterotrophic plate count) and ATP concentration of bacteria. The concentration of HPC and ATP were $3.3{\times}10^8$ cells/g and $2.14{\mu}g/g$, respectively. The number of the DGGE band were only 5 at the bed volume 8,916, but increased up to 11 at the bed volume 49,632. As operation time increase, bacterial group were more diversity. Four bacteria species including Pseudomonas fluorescens, the uncultured bacterium similar to Acinetobacteria, uncultured Novosphingobium sp. and Flavobacterium frigidarium have detected from the early stages and Proteobacteria group were dominantly detected.
