Degradation of Crystalline Cellulose by the Brown-rot Basidiomycete Fomitopsis palustris

  • This study demonstrated that the brown rot basidiomycete Fomitopsis palustris was able to degrade crystalline cellulose (Avicel). This fungus could also produce the three major cellulases (exoglucanases, endoglucanases, and $\beta-glucosidase$) when the cells were grown on $2.0\%$ Avicel. Avicel degraded by F. palustris showed a decrease in relative crystallinity from $83\%\;to\;78.5\%$ after 14 days of incubation. The characterization study indicated that optimum pH was 4.5 and optimum temperature was $70^{\circ}C$ for exoglucanase (cellobiohydrolase) activity. Hydrolysis of Avicel by the crude enzyme from F. palustris yielded 1.6 mg/ml of glucose after 43 h, which corresponded to a cellulose conversion degree of $3.2\%$. Therefore, this study revealed for the first time that the brown rot basidiomycete F. palustris produces cellulases capable of yielding soluble sugars from crystalline cellulose.

  • To investigate the antibiotic constituents of Korean basidiomycetes the carpophores of the wood-rotting basidiomycetes were collected from several locations of Korea, and from them 17 mycelial strains were isolated on potato-dextrose-agar plates supplemented with tetracycline ($20\;{\mu}g/m{\ell}$. The isolated strains were shake-cultured in glucosepeptone-yeast extract medium and then the antibacterial activities of the culture filtrates were assessed by disc-plate method. Among them, 12 strains (70.6%) were active, and basidiomycete strain LMCB-109 (Daedalea quercina) and LMCB-116 showed potent activities against all the six bacterial target organisms including Serratia marcescens.

Transformation of the Edible Basidiomycete, Pleurotus ostreatus to Phleomycin Resistance

  • For transformation of Pleurotus ostreatus, two novel vectors, pPhKM1 and pPhKM2, were constructed, using the regulatory sequences of the P. sajor-caju $\beta$-tubulin gene(TUB1) and the ble gene encoding phleomycin binding protein. pPhKM1 contains ble fused to the TUB1 promoter and the Schizophyllum commune GPD terminator. pPhKM2 contains ble fused to the promoter and terminator regions of P. sajor-caju TUB1. To confirm phleomycin-resistance activity, each vector was cotrans-formed with pTRura3-2 into the P. ostreatus homokaryotic $ura^-$ strain. The transforming DNA was stably integrated into the genomic DNA. Subsequently, phleomycin resistance was conferred on wild-type dikaryotic P. ostreatus by transformation with pPhKM1 or pPhKM2. This transformation system generated stable phleomycin-resistant transformants.

  • It has been known that MAO (monoamine oxidase) catalyses the oxidation of endogenous neurotransmitter amines. From its key role in the regulation of some physiological amines, it has been the target of inhibitors used as antidepressive agents. In our continuing search for MAO inhibitors from Basidiomycete. sp., strain 8082 was selected. Two metabolites (8082-1 and 8082-2) were isolated by adsorption chromatography and HPLC from the culture broth of strain 8082. The structure of 8082-1 and 8082-2 were elucidated by $^1$H-, $\^$13/C-NMR and HMBC spectral data, and these products were identified as 5-methylmellein and nectriapyrone, respectively, which have significant inhibitory effect against mouse brain MAO.

Cultivation of Phanerochaete chrysosporium and Lignin Peroxidase Activity

  • Effects of exogenous veratryl alcohol addition on the growth of basidiomycete Phanerochaete chrysosporium ME-446 and the induction of lignin peroxidase activity were investigated in this study. The organism was grown in ligninolytic (low-nitrogen) culture conditions in which extracellular enzymes are produced. Analyses showed that a statistically significant decrease of cell growth was associated with the veratryl alcohol addition. The effect of veratryl alcohol addition on LiP activity was nearly instantaneous and this effect diminished with culture aging. The extent of this effect was different depending on the time of addition, which led to a speculation that there might be some other effector species which played a role in regulation of lignin peroxidase activity.
