Histometrical evaluation of biphasic calcium phosphate in surgically created 1-wall periodontal intrabony defects in dogs (성견 일벽성 치주 결손부에 이식한 biphasic calcium phosphate의 조직계측학적 평가)

  • Purpose: The aim of this study was to evaluated biphasic calcium phosphate applied in surgically created 1-wall periodontal intrabony defects in dogs by histometrical analysis. Material and Method: Critical sized($4\;mm\;{\times}\;4\;mm$), one wall periodontal intrabony defects were surgically produced at the proximal aspect of mandibular premolars in either right and left jaw quadrants in four canines. The control group was treated with debridement alone, and experimental group was treated with debridement and biphasic calcium phosphate application. The healing processes were histologically and histometrically observed after 8 weeks. Results: In biphasic calcium phosphate group, more new bone and cementum formation, less epithelium and connective tissue attachment were observed compared to other groups. But there was no statistical significance. Conclusion: Though the statistically significant difference could not be found, it seemed that there was more new bone and cementum formation with applying biphasic calcium phosphate in 1 wall intrabony defects in dogs by preventing junctional epithelium migration.

  • The co-precipitation technique has been applied to synthesize biphasic calcium phosphate (BCP). $Ca(NO_3)_2{\cdot}4H_2O$ and $(NH_4)_2HPO_4$ as the starting materials was used. X-ray diffraction (XRD) and Fourier transformed infrared (FT-IR) spectroscopy were used to characterize the structure of as-synthesized and calcined BCP powders. After immersion in Hanks' Balanced Salt Solution (HBSS), for 1 week a precipitation started to be formed with individual small granules on the specimen surface. An MTT assay indicated that BCP powders have no cytotoxic effects on MG-63 cells, and that they have good biocompatibility.

  • The present study was to determine the influence of micro-macro biphasic calcium phosphate(MBCP) on proliferation and differentiation of human marrow-derived mesenchymal stem cells. Primary stem cells were cultured from bone marrow and 3-4 passaged cells were used. This study tested the proliferative effects by cell counting. Collagen sythensis, alkaline phosphatase activity, expression of osteocalcin and bone sialoprotein by Western blot analysis were evaluated. The cellular proliferation of ASC was not influenced by MBCP. Collagen synthesis of ASC cultured on MBCP significantly increased at 5th and 7th days(p<0.05). The ALP activity in ASC cultured on MBCP significantly increased at 5th and 7th days(p<0.05). The expression of OC and BSP incresaed in ASC cultured on MBCP. These results suggest that MBCP may stimulates the osteoblastic activity of ASC.

  • Si-substituted biphasic calcium phosphates (Si-BCP) were prepared by co-precipitation method. X-ray diffraction and fourier transform infrared spectroscopy were used to characterize the structure of Si-BCP powders. The Si-BCP powders with various Ca/(P+Si) molar ratio were carried out on structural change of hydroxyapatite (HAp) and ${\beta}$-tricalcium phosphate ($\ss$-TCP). The in-vitro bioactivity of the Si-BCP powders was determined by immersing the powders in SBF solution, after that observing the chemical composition and morphology change by X-ray diffraction, scanning electron microscope and energy dispersive spectroscopy.

Simple surface biofunctionalization of biphasic calcium phosphates for improving osteogenic activity and bone tissue regeneration

  • In this study, we found a simple surface biofunctionalization of biphasic calcium phosphate (BCP) based on the high affinity between alendronate and the calcium ions of BCP, and the strong interaction between heparin and bone morphogenic protein-2 (BMP-2). The biofunctionalized BCP did not be precipitated well and display a remarkable enhancement of osteogenic activity of human adipose-derived stem cells by showing increased alkaline phosphatase (ALP), calcium deposition and osteogenic-related genes (i.e., Runx-2, ALP, osteocalcin, and osteopontin), and bone regeneration in the calvarial defect model. Therefore, this simple surface technique can be used to easily functionalize various calcium phosphates.

  • Calcium phosphates such as HA (hydroxyapatite), β-TCP (tricalcium phosphate) and biphasic HA/β-TCP, were synthesized by wet chemical precipitation in aqueous solution combined with ball milling process. Nanosize powders of the calcium phosphates were synthesized using Ca(OH)2 and H3PO4. The effects of initial precursor Ca/P ratio (1.30, 1.50 and 1.67), ball milling process and post heat-treatment on the phase evolution behavior of the powders were investigated. The phase of resulting powder was controllable by adjusting the initial Ca/P ratio. HA was the only phase for as-prepared powders in both cases of Ca/P ratios of 1.50 and 1.67. The single HA phase without any noticeable second phase was obtained for the initial Ca/P ratio of 1.67 in the overall heat-treatment range. Pure β-TCP and biphasic calcium phosphate (HA/β-TCP) were synthesized from precursor solutions having Ca/P molar ratios of 1.30 and 1.50, respectively, after having been heat-treated above 700 ℃. The β-TCP phase has appeared on the pre-existing DCPD (dicalcium phosphate dihydrate) and/or HA phase. Dense ceramics having translucency were obtained at a considerably lower sintering temperature. The modified process offered a fast, convenient and economical route for the synthesis of calcium phosphates.

  • Nanoscale-biodegradable behaviors of synthesized biphasic calcium phosphates (BCP) powders were characterized using X-ray diffraction (XRD), lattice parameter, and field emission microscopy (FE-SEM). The calcined BCP powders in vitro tested in Hank's balanced salt solution (pH = 7.4, $36.5^{\circ}C$) for 3 weeks. The calculated unit cell parameters for BCP have shown lattice distortion and expansion as irregular changes in the a and c-axis after in vitro.

  • The calcium to phosphate ratio (Ca/P) in biphasic calcium phosphates powders using X-ray diffraction analysis (XRD) was characterized. The BCP powders with various stoichiometric Ca/P molar ratio were synthesized with coprecipitation process and calcination. Compositions of the powders with Ca/P molar ratio between 1.5 and 1.67 were subjected to starting Ca/P molar ratio, pH = 10, and thermal treatment up to $900^{\circ}C$. The structural, morphological and chemical characterizations for BCP powders with stoichiometric Ca/P ratio were carried out with scanning electron microscope (SEM) and inductively coupled plasma atomic emission spectroscopy (ICP-AES) and a phase quantification was investigated by XRD. The solubility of HAp, $\beta$-TCP, and BCP powders was tested in the phosphate buffer solution (PBS) at $36.5^{\circ}C$ and pH = 7.4.

  • 목적 : 이 연구의 목적은 치과 영역에서 골 재생을 촉진하기 위해, 현재 많이 사용하고 있는 BDX(bovinederived xenograft)와 비교하여 BCP(biphasic calcium phosphate)의 효과를 알아보기 위함이다. 실험 재료 및 방법: 본 연구는 태아골모세포주(hFOB 1.19)를 사용하였으며, 사용된 골 이식재에 따라 2개의 실험군으로 구분하였고, 각 실험에 적절한 농도의 BDX와 BCP를 첨가하였다. 그리고, 세포 증식도 검사, 교원질 합성량 분석, 염기성 인산분해효소 활성도 측정, Western blot 분석을 통한 OC과 BSP의 발현 정도등의 실험을 진행하였다. 결과 : BDX와 BCP는 대조군과 비교하여 세포 증식에서 유의한 차이가 없었지만, 교원질 합성량, 염기성 인산분해효소의 활성, 그리고 OC과 BSP의 발현에 있어 대조군과 비교하여 유의하게 증가를 보였다. 그러나, 두 이식재간의 유의한 차이는 보이지 않았다. 결론 : 본 실험실적 연구에서 BCP는 골모세포분화에 긍정적인 영향을 미침으로써 효과적인 이식재로 사용할 수 있음을 가늠할수 있었다.
