Effect of urea on blood-luminol chemiluminescence reaction (Urea가 혈액-luminol 화학발광에 미치는 영향)

  • This study aimed to verify the hypothesis that urea denatures hemoglobin in the blood, thereby exposing active sites of enzymes and enhancing the chemiluminescence of the blood-luminol reaction. When blood was pretreated with urea, higher concentrations of pretreatment urea or longer pretreatment times resulted in enhanced chemiluminescence in the blood-luminol reaction, supporting the above hypothesis. However, the chemiluminescence was enhanced when blood was treated with luminol mixed with an 8 M urea solution, although the fact that the time for urea to denature hemoglobin was shorter compared to when blood was pretreated with urea and followed by luminol. In addition, the chemiluminescence was enhanced when a transition metal without hemoglobin was reacted with urea-containing luminol. Based on these results, it is anticipated that urea not only denatures hemoglobin but also plays a role in the luminol-hydrogen peroxide reaction.

  • In two separate experiments with crossbred bulls (Sahiwal $\times$ indigenous) the effect of access to a urea-molasses lickblock (MOL-U-MIN) on straw diets was studied. The animals were given either untreated (US) or urea treated (TS) rice straw with or without lickblock supplementation. In experiment 1, individual dry matter intake (DMI) and dry matter digestibility (DMD) were measured, while in experiment 2 in addition to the above rumen (pH, ammonia, minerals) and blood (protein, minerals and haemotological) parameters were also measured. With both experiments urea treatment did not effect DMI, but lickblock supplementation significantly (p < 0.05) increased DMI. The DMD values obtained in both experiments for TS were significantly (p < 0.05) higher than for US, but lickblock supplementation did not effect the DMD of either US or TS fed animals. Both urea treatment (6.97 vs 6.93) and lickblock supplementation (6.98 vs 6.92) significantly (p < 0.001) reduced the rumen pH. Urea treatment and lickblock supplementation increased the rumcn $NH_3-N$ concentration (mg/100 ml) from 8.7 to 11.9 and 9.2 to 11.4, respectively. Both US and TS diets fed with or without lickblock increased the molar ratio of Na : K in saliva. Phosphorus content in blood plasma was significantly (p < 0.01) increased due to lickblock supplementation, whereas the Fc content in blood was significantly increased (p < 0.01) by urea treatment. Haemoglobin content in blood ranged from 11.3 to 11.7 g/dl, and was not influenced by urea treatment or lickblock supplementation. Lickblock significantly reduced the number of red blood cells, but increased the mean corpuscular volume. It is concluded that feeding urea treated straw with proper mineral supplementation could be a more economical alternative to lickblock supplementation.

BLOOD BIOCHEMICAL PROFILE AND HISTOPATHOLOGY OF VITAL ORGANS IN RABBITS FED ON PROCESSED NEEM (Azadirachta indica) KERNEL MEAL INCORPORATED DIETS

  • Blood biochemical and histopathological changes in vital organs of rabbits were studied after 19 wk of feeding composite diets (75 concentrate : 25 roughage) incorporating either urea (2%, wt/wt) ammoniated or alkali (1.5%, wt/wt) treated neem kernel meal (NKM) replacing peanut meal protein of control diet by either 50 or 100%. The blood biochemical constituents (Haemoglobin, Alanine amino transferase, Aspartate amino transferase, Total protein, Blood urea nitrogen &Cholesterol) in rabbits fed on processed NKM diet at either levels, were comparable to the values of thos on control diet except a lowered (p < 0.05) blood glucose concentration in processed NKM fed rabbits as compared to that in control diet fed ones. Histological examination revealed increased goblet cell activity, stunting of jejunal villi, mild tubular degeneration in kidney and hepatic fibro-cellular reaction in rabbits fed on urea ammoniated and alkali treated NKM diets with less marked changes in the latter. Testicular changes with variable degree of disorganization and vacuolation of spermatogonial cells were noticed in rabbits fed higher levels of urea-ammoniated and alkali treated NKM. Thus, alkali treatment and urea-ammoniation were effective in detoxification of meal, but the processing technology is to be further perfected to prevent cumulative effect of residual neem bitters in long term feeding.

  • Renal dysfunction was experimentally induced with reductions in renal mass in dogs and then the remaining kidney function was estimated by blood urea nitrogen and serum creatinine concentrations. During experimental period, blood urea nitrogen and serum creatinine concentrations showed no variance in dogs with 50% reductions in renal mass, but these values were remarkably increased in dogs with 75% reductions in renal mass. It was considered that 75% reductions in renal mass was applicable as experimental models for renal dysfunction.

  • Aim To evaluate blood pressure, blood glucose and serum lipid level in obese and nonobese type 2 diabetic patients. Methods 206 obese(76 male, 130 female) and 442 nonobese(208 male, 234 female) type 2 diabetic patients underwent fasting blood glucose, 2-hour postprandial blood glucose, $HbA_1c$ total cholesterol, triglyceride, high density lipoprotein, microalbuminuria, blood urea nitrogen, creatinine and C-peptide were measured. Diabetes was diagnosed according to the American Diabetes Association(ADA)criteria. Obesity was defined as body mass index(BMI, kilograms per meters squared)${\geq}25$. Results In male, systolic blood pressure, triglycerides, microalbuminuria and C-peptide were significant higher in obese than nonobese patients. Fasting blood glucose were significantly lower in obese than nonobese patients. Diastolic blood pressure, 2-hour postprandial blood glucose, $HbA_1c$, total cholesterol, high density lipoprotein, blood urea nitrogen, and creatinine were no difference between 2 groups. In female, triglycerides and C-peptide were significant higher in obese than nonobese patients, Blood pressure, fasting blood glucose, 2-hour postprandial blood glucose, $HbA_1c$, total cholesterol, high density lipoprotein, microalbuminuria, blood urea nitrogen, and creatinine were no difference between 2 groups. Conclusion Our present study supports that increased triglycerides play a major role in increasing the risk of coronary heart disease(CHD) in obese women type 2 diabetic patients.

Simulation Study on the Efficacy of Toxin Removal by Pulsatile Flow in Blood Purification Systems that use Semipermeable Membranes

  • Using numerical models, we investigated the efficiency of toxin removal using pulsatile flow in blood purification systems that use semipermeable membranes. The model consisted of a three-compartmental mass transfer model for the inside body and a solute kinetics model for the dialyzer. The model predicted the toxin concentration inside the body during blood purification therapy, and the toxin removal efficiencies at different flow configurations were compared quantitatively. According to the simulation results, the clearances of urea and ${\beta}_2$ microglobulin (B2M) using a pulsatile pump were improved by up to 30.9% for hemofiltration, with a 2.0% higher urea clearance and 4.6% higher B2M clearance for high flux dialysis, and a 3.9% higher urea clearance and 8.2% higher B2M clearance for hemodiafiltration. These results suggest that using a pulsatile blood pump in blood purification systems with a semipermeable membrane improves the efficacy of toxin removal, especially for large molecules and hemofiltration treatment.

  • The effects of casein diet supplemented with excessive levels of three basic amino acidslysine, histidine and arginine - were studied in male, weanling, I. C. R. strain mice. The relationship between weight gain and concentration of blood urea nitrogen was also discussed with the follows results : 1. The groups supplemented with basic amino acids were demonstrated depressed growth and food intake than the control ($10\%$ casein diet) group, $5\%$ L-histidine HCl supplemented group being most depressed. 2. The concentration of blood urea nitrogen in the basic amino acid supplemented groups were higher than the control group. The value of L-arginine supplemented group was highest. 3. The concentration of blood urea nitrogen was related to the amounts of nitrogen contained in the diet not related to the growth gain.

  • The development of a portable system to be used by diabetes patients and renal discase patients is needed to monitor their urea concentration and their glucose concentration in blood. This paper reports a compound sensor composed of a urea sensor, a glucose sensor and a micro-channel. This paper also reports the development of a portable measurement system to measure the concentrations, display the values, and save the values to be used by doctors.

A Study on the Change of Blood Constituents During Growth Period of Velvet Antler in Korean Spotted Deer (꽃사슴의 녹용 성장기간 중 혈액성분의 변화에 관한 연구)

  • This study was carried out to evaluate the change of blood constituents during the growth of antlers in Korean spotted deer (Cervus nippon). Samples of blood, obtained from the jugular vein of twenty five deer (3 to 6 year-old males) were taken in 10 days interval from casting to cutting of antler and were analyzed the blood parameters. Total-protein concentration in blood serum showed significantly differences during growth period of velvet antler (p<0.01), and it had similar tendency for total protein and albumin concentration, but albumin concentration was about 40-45% of total protein concentration. Urea and creatinine concentration was tended to inverse proportion, and it was almost 30:1 for ratio of urea and creatinine concentration. The concentration of total protein, creatinine, and uric acid concentration in blood during growth period of velvet antler was similarly increased and decreased but urea concentration was opposed. The concentration of total-bilirubin and direct-bilirubin at casting was higher than those at other period(P<0.05).

  • This study was aimed to investigate the effects of Ginseng ethanol extract on Diabetes induced by alloxan in comparison with that of insulin. The blood sugar, blood urea nitrogen, glutamic oxaloacetic transaminase and cholesterol were measured by spectrophotometry. The results were obtained as follows. 1. Ethanol extract of Ginseng increased the blood sugar level in normal rats. 2. Ethanol extract of Ginseng restrained the increase of blood sugar in diabetes induced by alloxan. 3. Ethanol extract of Ginseng acted to oppose a rapid decrease of blood sugar by insulin. 4. Blood urea nitrogen, glutamic oxaloacetic transaminase and cholesterol level in blood were not affected by Ginseng.
