Influence of Bornyl Acetate on Blood Pressure and Aortic Strips Contractility of the Rat

  • The present study was conducted to investigate the effects of bornyl acetate on arterial blood pressure and vascular contractile responses in the normotensive rats and to establish the mechanism of action. Both phenylephrine (an adrenergi$\alpha$-receptor agonist) and high potassium (a membrane-depolarizing agent) caused greatly contractile responses in the isolated aortic strips. These phenylephrine (10$^{-5}$ M)-induced contractile responses were depressed in the presence of high concentrations of bornyl acetate (10∼20 $\mu\textrm{g}$/ml), but not affected in low concentrations of bornyl acetate (2.5∼5$\mu\textrm{g}$/ml). High potassium (5.6 ${\times}$ 10$^{-2}$ M)-induced contractile responses were also greatly inhibited in the presence of bornyl acetate (2.5∼20 $\mu\textrm{g}$/ml) in a dose-dependent fashion. Bornyl acetate (1∼10 mg/kg) given into a femoral vein of the normotensive rat produced a dose-dependent depressor response, which is transient (data not shown). Interestingly, the infusion of a moderate dose of bornyl acetate (3mg/kg/30 min) made a significant reduction in pressor responses induced by intravenous norepinephrine. Collectively, these results obtained from the present study demonstrate that intravenous bornyl acetate causes a dose-dependent depressor action in the anesthetized rat at least partly through the blockade of adrenergic $\alpha$$_1$-receptors. bornyl acetate also causes vascular relaxation in the isolated aortic strips of the rat via the blockade of adrenergic $\alpha$$_1$-receptors, in addition to the unknown mechanism of direct vasorelaxation.

  • The objective of this study was to investigate the effects of 11 active compounds of essential oils (ACEO) on rumen fermentation characteristics and methane production. Two trials were conducted. In trial 1, ACEO (eugenol, carvacrol, citral, limonene, 1,4-cineole, p-cymene, linalool, bornyl acetate, ${\alpha}$-pinene, and ${\beta}$-pinene) at a dose of $1,000{\mu}L/L$ were incubated for 24 h in diluted rumen fluid with a 70:30 forage:concentrate substrate (16.2% crude protein; 36.6% neutral detergent fiber). Three fistulated Holstein cows were used as donors of rumen fluid. The reduction in methane production was observed with nine ACEO (up to 86% reduction) compared with the control (p<0.05). Among these, only limonene, 1,4-cineole, bornyl acetate, and ${\alpha}$-pinene did not inhibit volatile fatty acid (VFA) production, and only bornyl acetate produced less methane per mol of VFA compared with the control (p<0.05). In a subsequent trial, the effects on rumen fermentation and methane production of two concentrations (500 and $2,000{\mu}L/L$) of bornyl acetate, the most promising ACEO from the first trial, were evaluated using the same in vitro incubation method that was used in the first trial. In trial 2, monensin was used as a positive control. Both doses of bornyl acetate decreased (p<0.05) methane production and did not inhibit VFA production. Positive effects of bornyl acetate on methane and VFA production were more pronounced than the effects of monensin. These results confirm the ability of bornyl acetate to decrease methane production, which may help to improve the efficiency of energy use in the rumen.

Allelopathic Effect of Volatile Extracts from Eupatorium rugosum (서양등골나물 휘발성 추출물의 알레로파시 효과)

  • This study was carried out to find the allelopathic effect of volatile materials released from Eupatorium rugosum. The GC method was employed for analysis of volatile materials from E. rugosum and 49 chemical substances were identified such as $\beta$-caryophyllene, $\alpha$-terpinenol, chamazulene, bornyl acetate, $\alpha$-pinene, etc. including unidentified three chemicals. Germination test in Phaseolus radiatus was done to find the inhibition effect of volatile materials using some chemicals which were proved to be important component or much amounts ones in E. rugosum. It was strongly inhibited by linalool and terpinen-4-ol. Seedling elongation and radicle growth of that were proportionally inhibited by the concentration of the essential oil, especially $\alpha$-pinene and bornyl acetate. Biomass of receptor plant was slightly decreased more than 58 ${\mu}l$ of the extract in case of $\alpha$-pinene, while it was decreased more than 19 ${\mu}l$ of that in bornyl acetate but it was shown non-significant. From the above results, it was found that volatile materials from E. rugosum showed a allelopathic effect and also $\alpha$-pinene, bornyl acetate, linalool and terpinen-4-ol used in bioassay were some of major allelochemicals in germination inhibition and especially linalool and terpinen-4-ol are prominent effect on growth inhibition of other plant.

  • The needle oils of Pious rigida Mill and Pious densiflora Sieb & Zucc were analyzed by gas chromatography/mass spectrometry. The major components of Pinus rigida were ${\alpha}-pinene$, 1-hexen-3-ol formate, sabinene, ${\beta}-pinene,\;{\alpha}-terpineol$ and ${\beta}-caryophyllene,\;{\alpha}-pinene$, bornyl acetate, ${\beta}-pinene$ and ${\beta}-hpellandrene$ were the major components fo Pious densiflora. Pious densiflora had sweeter and more greenish note than Pines rigida because the bornyl acetate content of Pious densiflora was about three times more than that of Pious rigida.

  • The essential oil of Korean valerian root ( Valeriana fauriei roar. dasycarpa Hara) was isolated by simultaneous distillation Sl extraction. The oil content of fresh root was 0.7% (wb) and that of dried root was 2.1 5 (db) and sensory analysis of the oil indicated sweet-balsamic, woody and floral characteristic aroma notes. The oil was fractionated into one hydrocarbon fraction and three oxygenated hydrocarbon fractions by using silica gel column chromatograpy. Each fraction was analyzed by capillary GC and GC-MS. Out of 81 characterized compounds, the major compounds were a-pinene, camphene, $\beta$-pinene, bornyl acetate, borneol , bornyl iso-valerate and sesquiphellandrene and the characteristic floral and woody aroma of neutral fraction of Korean valerian root could be due to be the presence of oxygenated compounds such as borneol, bornyl acetate, bornyl iso-valerate, p-ionone ana $\beta$-ionone epoxide. Comparing the yield of Korean valerian root with those from other origins reported, oil content of Korean valerian root was higher than those of European and Indian origins.

  • The plastochron index (PI) according to leaf development of Ambrosia artemisiifolia var. elatior were estimated. The amount and composition of monoterpenes in the leaves of A. artemisiifolia var. elatior were determined, and the seasonal monoterpene variations in representative of plastochron index were investigated. The total amount of monoterpenes in A. artemisiifolia var. elatior was increased continuously from PI 2.0, the highest amount was in PI 11.89. The major monoterpenes in A. artemisiifolia var. elatior consisted of 29 compounds. The leaf monoterpene fraction of A. artemisiifolia var. elatior were dominated by No.2, octene, α-pinene, β-myrcene, 1,3,6-octatrine, endo-borneol and (-)-bornyl acetate.

  • This study was conducted to know the effect of cultivation area with different sea levels on essential oil content and its composition and content of major active compounds in Valerian fauriei var. dasycarpa HARA and Valerian officinalis L. The total contents of essential oil were not different according to the locality in both valerian, however, its compositions were varied. The major components in V. fauriei were detected bornyl acetate and sesquiterpene alcohol in three cultivation areas. The major compounds were borneol, borney acetate, ${\beta}\;-sesquiohellandrene$ in V. officinalis other. Among the valepotrate components known as major pharaceutically active components in Europe and Nepal valerian, valtrate was detected in a small amount in V. fauriei, but its quantity was not changed according to the cultivation area.

  • The compositions of essential oils isolated from the leaves and fruits of Chamaecyparis obtusa (Sieb. et Zucc). Endl. and Chamaecyparis pisifera (Sieb. et Zucc.) Endl. were analyzed through GC and GC-MS. The oil yields were 0.83% (as fresh weight) and 1.36% in the leaves and the fruits of C. obtusa, and were 0.92% and 1.28% in those of C. pisifera, respectively. More than 90 components were identified, including high contents of monoterpenoids and sesquiterpenoids. Contents of monoteipenoids in the leaf and fruit oils of C. pisifera were higher than in those of C. obutsa. The major constituents in the leaf oil of C. obtusa were sabinene (11.81% as determined through GC peak area), limonene (7.73%), bornyl acetate (6.92%), $borneol+{\alpha}-teirineol$ (15.67%), and elemol (12.82%), and those in the fruit oil were myrcene (8.12%), ${\gamma}-terpinene$(5.91%), p-cymene(7.62%), $borneol+{\alpha}-terpineol$(6.53%) and ${\beta}-caryophyllene$ (23.74%). The major constituents in the leaf oil of C. pisifera were ${\alpha}-pinene$(32.34%), ${\delta}-3-carene$(25.28%), myrcene(11.72%), and bornyl acetate (8.77%), and those in the fruit oil were ${\alpha}-pinene$ (29.38%), ${\delta}-3-carene$(30.27%), myrcene(15.05%), and limonene(8.10%).

  • We examined the effect of 12 main monoterpenes in Pinus plants on growth inhibition of Escherichia coli and Aspergillus nidulans. We tested four concentrations of each compound by comparing the clear zone with controls. (R)-(-)carvone, (S)(+)carvone, (1R)(-)fenchone, (-)menthone, α-pinene, (1S) (-)verbenone and (+)β - pinene had a inhibition effect on E. coli. (R)-(-)carvone, (S)(+)carvone, (+) β-pinene, geranyl-acetate, α-pinene, and (1S)(-)verbenone had inhibitory effects on the growth of A. nidulans. Geranyl-acetate inhibit growth of A. nidulans, however not to E. coli. And (1R)(-)fenchone and (-)menthone inhibit growth of E. coli, but not to A. nidulans. Myrcene, sabinene, bornyl acetate, and limonene had no inhibitory effects on E. coli and A. nidulans, eventhough at the highest concentration. All these results suggested that some selected monoterpenes had antifungal activities depend on the species of microorganism.

Changes in Esterase Isozyme Activity After Pesticides Treatment in Digestive Juice of Monochamus saltuarius (Gebler) Adult (북방수염하늘소(Monochamus saltuarius) 성충의 살충제 처리에 따른 소화 효소의 활성 변화)

  • Esterase isozymes were investigated from digestive juice of M. saltuarius adults after pesticide treatment. Twelve esterase isozymes were separated on 12% native-PAGE gel and stained with three different substrates(${\alpha}$-naphthyl acetate, ${\beta}$-naphthyl acetate, and ${\alpha}$-naphthyl butyrate). Interestingly, the isozyme of Est1(${\alpha}$-naphthyl acetate) was strongly inhibited by the carbofuran and methomyl. The Est1 activity was completely inhibited by the chlorpyrifos and partially inhibited by methidation about 70 %. In addition, eserine suppressed esterase isozyme activities of Est1 about 70% and isozyme activities of Est2, Est3, and Est4 were weakly inhibited. ${\alpha}$-pinene did not suppressed esterase isozyme activities but activities of esterases were very weakly inhibited in camphor and bornyl acetate.
