  • From the stems of Solanum lyratum Thunb.(Solanaceae), two sesquiterpenoids together with two 5$\alpha$, 8$alpha-epidioxy sterols have been isolated and identified as atractylenolide I, dehydrocarissone, engosterol peroxide, 9, 11-dehydroergosterol peroxide. These compounds never previously isolated from this genus.

Purification and Characterization of Chitinase from a Marine Bacterium, Vibrio sp. 98CJ11027

  • Chitin-degrading marine bacterial strain 98CJ11027 was isolated from bryozoa from the coastal area of Cheju Island, Korea, and identified as a member of the genus Vibrio. The molecular mass of the main extracellular chitinase (chitinase I), purified from strain 98CJ11027, was estimated to be 98 kDa. The optimal condition for chitinase I activity is pH 6.0 and 45℃. The activity was inhibited by $Fe^+2$ and$Cu^+2$. Chitinase I displayed the hydrolysis type of chitobiosidase and catalyzed reversed hydrolysis leading to the synthesis of tetraacetylchitotetraose.

Pharmacolgocial Characterization of LB50016, N-(4-Amino)Butyl 3-Phenylpyrrolidine Derivative, as a New 5-HT_{1A}Receptor Agonist

  • LB50016 was characterized as a selective and potent$ 5-HT_{1A}$ receptor agonist and evaluate it anxiolytic and antidepressant activities. It shows high affinity for $ 5-HT_{1A}$receptor, moderate affinity for $\alpha$2 adrenergic and $ 5-HT_{2A}$receptors and no significant affinity for other receptors tested. Hypothermia and increased serum corticosterone level were observed in LB50016-treated rats, which are mediated mostly by post synaptic $ 5-HT_{1A}$ receptor activation. In the mouse forced swim model for depression, LB50016-elicited dose-dependent reductions in immobility time, showing $ED_{50}$ of approximately 3 mg/kg i.p., which was blocked by pretreatment of NAN-190, $ 5-HT_{1A}$antagonist. In face-to-face test for anxiolytic activity in mice, estimated $ED_{50}$ was 2 mg/kg, i.p.. In isolation-induced aggression test with mice, fifty-fold increases in latency to attack were observed at 30 min and last up to 4 h after LB50016 treatment (3 mg/kg, i.p.). Taken together, LB50016-induced pharmacological activities are mediated by activation of $ 5-HT_{1A}$receptors, offering an effective therapeutic candidate in the management of anxiety and depression in humans.

  • A coil pyrolyzer and processor-controlled gas chromatograph were used for analysis of rubber for compounding ratio of the single and blend rubber vlucanizates. Variables such as sample size, pyrolysis temperature, time allowed for pyrolysis, the column packing material, its length and programmable temperature for gas chromatography were examined to obtain optimum condition for application to NR, BR and SBR blends. By application fixed conditions, three kinds of standard curves were finally obtained from thirty samples of blend vulcanizates which were prepared in the pilot plant, NIRI. It is possible to determine rubber composition and their ratio in NR, BR and SBR products by pyrolysis.

A Study on the Chemical Powdering Process of Polypropylene Resin for Anti-slip Floor Paint (미끄럼 방지 바닥 도료용 폴리프로필렌 수지의 화학적 분쇄에 관한 연구)

  • In this study, some experimental results of the peroxide-degradation process of polypropylene(PP) in a co-rotating twin-screw extruder to produce controlled rheology polypropylene(CRPP) are presented. The peroxide was dicumyl peroxide(DCP) and the concentration of DCP was in the range 0-0.3 wt%. It was found that the rheological properties of PP change significantly during reactive extrusion. Melt flow index(MFI) increased with DCP concentration. Intrinsic viscosity decreases with increasing DCP concentration. From dynamic rheological data, number average molecular weight(Mn), weight average molecular weight(Mn) and molecular weight distribution(MWD) were calculated. Results indicated that Mw decreases and MWD becomes narrower with increasing peroxide concentration. Especially, particle size distribution of CRPP decreases with increasing DCP concentration by chemical powdering process, and anti-slip floor paint, CRPP(DCP 0.2 wt%) powder by 10phr was friction coefficient 2.15 ${\mu}$, abrasion resistance 511.18%.

  • One sesquiterpenoid was isolated from the methanol extract of Atractylodes rhizomes which has been used as traditional medicine for diuresis and the compound was established as selina-4(14),7(11)-dien-8-one by the spectroscopic methods. The compound$(IC_{50}<10\;ppm)$ has inhibitory effects on melanogenesis in B16 mouse melanoma.

  • Cytomegalovirus (CMV), a beta-herpesvirus with worldwide distribution, exhibits host persistence, a distinguishing characteristic of all herpesviruses. This persistence is dependent upon restricted gene expression in infected cells as well as the ability of productively infected cells to escape from normal cell-mediated anti-viral immunosurveillance. Type I (IFN-α/β) and type II (IFN-γ) interferons are major components of the innate defense system against viral infection. They are potent inducers of MHC class I and II antigens and of antigen processing proteins. Additionally, IFNS mediate direct antiviral effects through induction effector molecules that block viral infection and replications such as 2′, 5-oligoadenylate synthetase (2, 5-OAS). IFNS function through activation of well-defined signal transduction pathways that involve phosphorylation of constituent proteins and ultimate formation of active transcription factors. Recent studies have shown that a number of diverse viruses, including CMV, EBV, HPV mumps and Ebola, are capable of inhibiting IFN-mediated signal transduction through a variety of mechanisms. As an example, CMV infection inhibits the ability of infected cells Is transcribe HLA class I and II antigens as well as the antiviral effector molecules 2, 5-OAS and MxA I. EMSA studies have shown that IFN-α and IFN-γ are unable to induce complete signal transduction in the presence of CMV infection, phenomena that are associated with specific decreases in JAKl and p48. Viral inhibition of IFN signal transduction represents a new mechanistic paradigm for increased viral survival, a paradigm predicting widespread consequences in the case of signal transduction factors common to multiple cytokine pathways.
