Enhancement of Cyclo-His-Pro (CHP) Content from Soybean Fermented with Bacillus amyloliquefaciens CHP-12 and Its Anti-diabetic Effect (Bacillus amyloliquefaciens CHP-12에 의한 대두 발효물의 Cyclo-His-Pro (CHP) 함량 증진 및 항당뇨 효과)

  • To enhance cyclo-His-Pro (CHP) content, soybean hydrolysate was obtained using the strains isolated from Chungkukjang and further purified by various purification steps. First, twenty two strains were screened from Chungkukjang containing high level of CHP. Among them, the strain No. 12, which showed higher productivity of CHP from soybean ferment and have homologous sequence with 16S rDNA of Bacillus amyloliquefaciens, was named B. amyloliquefaciens CHP-12. Through various purification processes, CHP was concentrated from soybean ferment using ultrafiltration, which showed the best efficiency of CHP production, with the yield (71.3%) and CHP content (2.14 mg/g). Moreover, when glucose tolerance test was performed in Type I Sprague-Dawley rat induced by streptozotocin using the soybean ferments [0.5 g soybean ferment/kg body weight (CHP-0.5 group)] and 1.0 g soybean ferment/kg body weight (CHP-1.0 group), there were significant differences in glucose levels between diabetes-control group (265.3 mg/dL) and soybean ferment-treated groups (CHP-0.5 group: 84.3 mg/dL and CHP-1.0 group: 85.3 mg/dL) 120 min after glucose injection (2 g/kg body weight) (p < 0.05). Accordingly, it is suggested that the soybean ferment containing high level of CHP might be a candidate material as an anti-diabetic supplement for manufacturing functional healthy foods.

Optimization Process Models of Gas Combined Cycle CHP Using Renewable Energy Hybrid System in Industrial Complex (산업단지 내 CHP Hybrid System 최적화 모델에 관한 연구)

  • The study attempted to estimate the optimal facility capacity by combining renewable energy sources that can be connected with gas CHP in industrial complexes. In particular, we reviewed industrial complexes subject to energy use plan from 2013 to 2016. Although the regional designation was excluded, Sejong industrial complex, which has a fuel usage of 38 thousand TOE annually and a high heat density of $92.6Gcal/km^2{\cdot}h$, was selected for research. And we analyzed the optimal operation model of CHP Hybrid System linking fuel cell and photovoltaic power generation using HOMER Pro, a renewable energy hybrid system economic analysis program. In addition, in order to improve the reliability of the research by analyzing not only the heat demand but also the heat demand patterns for the dominant sectors in the thermal energy, the main supply energy source of CHP, the economic benefits were added to compare the relative benefits. As a result, the total indirect heat demand of Sejong industrial complex under construction was 378,282 Gcal per year, of which paper industry accounted for 77.7%, which is 293,754 Gcal per year. For the entire industrial complex indirect heat demand, a single CHP has an optimal capacity of 30,000 kW. In this case, CHP shares 275,707 Gcal and 72.8% of heat production, while peak load boiler PLB shares 103,240 Gcal and 27.2%. In the CHP, fuel cell, and photovoltaic combinations, the optimum capacity is 30,000 kW, 5,000 kW, and 1,980 kW, respectively. At this time, CHP shared 275,940 Gcal, 72.8%, fuel cell 12,390 Gcal, 3.3%, and PLB 90,620 Gcal, 23.9%. The CHP capacity was not reduced because an uneconomical alternative was found that required excessive operation of the PLB for insufficient heat production resulting from the CHP capacity reduction. On the other hand, in terms of indirect heat demand for the paper industry, which is the dominant industry, the optimal capacity of CHP, fuel cell, and photovoltaic combination is 25,000 kW, 5,000 kW, and 2,000 kW. The heat production was analyzed to be CHP 225,053 Gcal, 76.5%, fuel cell 11,215 Gcal, 3.8%, PLB 58,012 Gcal, 19.7%. However, the economic analysis results of the current electricity market and gas market confirm that the return on investment is impossible. However, we confirmed that the CHP Hybrid System, which combines CHP, fuel cell, and solar power, can improve management conditions of about KRW 9.3 billion annually for a single CHP system.

Preparation of Yeast Hydrolysate Enriched in Cyclo-His-Pro (CHP) by Enzymatic Hydrolysis and Evaluation of Its Functionality

  • In this study, we attempted to enrich cyclo-His-Pro (CHP) using enzymatic hydrolysis of yeast and to evaluate the functionality of yeast hydrolysate (YH)-enriched CHP. Flavourzyme offered a better performance in enhancing CHP content than other proteases. The CHP enrichment conditions were optimized as follows: addition of 1% Flavourzyme, 48-h incubation at 60oC, and pH 6.0. The CHP content significantly increased by 20-fold after ultra-filtration (UF). Maximal CHP translation was obtained after heating for 8 h at 50oC and pH 7.0. YH showed poor foaming capacity between pH 3.0 to 9.0. The emulsifying activities of YHs were slightly higher at near acidic pH. Increase in heating temperature and time resulted in decreased CHP content. The results indicate that YH is more heat stable after UF. Therefore, the CHP in YH after UF can be used as a food additive with physiological CHP activity and high heat stability.

  • Various crosslinked poly(4-vinylpyridines) (CHP4VP) having different degrees of crosslinking were synthesized by radical copolymerization of 4-vinylpyridine with if N,N' -1, 6-hexamethylenebisacrylamide, and CHP4VP- Cu(II) complexes were prepared by the method of adsorption equilibrium. The catalytic activity of the complexes for the oxidation of ascorbic acid (AA) was investigated. The oxidation of AA by these complexes showed a kinetic behavior of the Michaelis-Menten type. The catalytic activity of CHP4VP-Cu(I ) catalytic system was increased with increasing the degree of crosslinking of CHP4VP, and its activity was scarcely decreased even after repeated use. However, the tendency of the catalytic activity of CHP4VP-Cu(II) complexes was decreased for the oxidation of AA when compared with that of the previously reported catalytic system containing crosslinked poly(4-vinylpyridine) prepared using N,N'-methylenebisacrylamide as a crosslinker. These results indicate that the degree of crosslinking of CHP4VP and the hydrophobicity of the crosslinker play an important role in the catalytic system of the oxidation of AA.

CheY-OmpR Hybrid Protein Acting on the Osmoregulatory System (CheY-OmpR 혼성 단백질의 삼투조절효과)

  • In the previous study(6), we constructed the CheY-OmpR hybrid, Chp, which affects the expressions of ompF and om pC genes. Here we further characterize these effects and present the regulatory mechanism based on in vivo and in vitro data. Although Chp retained the sequence-specific DNA-binding ability, it was not possible to enhance transcriptional activity, suggesting that it may act as a competitive inhibitor to OmpR. The DNA-binding affinity of Chp was not modulated by phosphorylation of its Che Y portion. Chp was able to increase ompR transcription. FurthemlOre, it was found that the wild-type OmpR also exerts the same effect, which is also eOlltrolled by changes in medium osmolarity and in EnvZ activity.

  • This paper presents and analyzes the effects of on-grid electricity cost, fuel price and initial capital cost of a CHP system, on the optimum DG and AC capacity and NPV, by using the ORNL CHP Capacity Optimizer, which was applied to a library in a university. By considering the current domestic energy cost and initial capital cost, it is shown that the installation and operation of the CHP system is not economical. However, with the current domestic CHP installation cost and fuel price, the NPV achieved by the installation of CHP system is greater when the on-grid electricity price is a factor of ${\times}1.5$ the present value. Regarding the initial capital cost of the CHP system, the reduction of the DG cost is much more economical than that of the AC cost, with respect to NPV. Electricity cost and fuel price have opposite effects on NPV, and NPV is more sensitive to an increase of the electricity cost than an increase of the fuel price.

Cost-Benefit Analysis on Community Health Practitioner (보건진료원 활동의 비용-편익 분석)

  • Purpose : The purpose of the study were to describe outcomes of CHP activities, and to evaluate the economic validity of CHP through a cost-benefit analysis. Method : The sample size was 272. Data were collected using a researcher developed questionnaire from November 1999 to March, 2000. Result : The mean age of CHPs was 39.6 (SD-36). In regard to marital status, 90.8% of the respondents were married. 72% of the CHPs had associate degree. Among CHP activities, providing medical services was 50%, followed by home care visits 20% and health promotion services 20%, preventive services 10%. Total costs per month incurred to CHP activities was \3,053,437($2,442.7). Total benefits per month was \6,711,525($5,369.2). Hence, net benefit was calculated as \3,658,089($2,926). Conclusion : Cost-benefit ratio was 2.20, which provides the evidence of the economic viability of CHP program. The result of cost-benefit analysis, however, would more strongly support the economic value of CHP if intangible benefits of CHP activities such as decreases in pain and suffering and increased quality of life, could be counted.

  • For the study of method for salt elimination aimed at reforming tidal land into normal paddy fields in a short period with reduction of periods requiring for elimination of saline, CHP (a kind of Ca-hum ate), a soil conditioner made of peat as a main material was tried. In the pot experiment, effect on elimination of salt, improvement of physical-chemical characteristics and rice cultivation test were studied. The results of these tests are as follows: 1, CHP treatment somewhat improves aggregation state with some effect on aggregation. 2. CHP treatment is remarkably effective in permeability which increases with 1.0 percent treatment by three times in percolation rate, and by 4.5 times in volume of leached water respectively. 3. With the increase of CHP amounts, salt was eliminated in short period. When 80% of the total Na was leached in 1.0% CHP-A treated pot, control pot begins permeable. 4. CEC and phosphorous absorption capacity are not influenced by CHP treatment. 5. Growing state of rice is greatly influenced by rainfalls. Growth of rice in tidal land however are almost similar to those in normal paddy fields with layer amounts of CHP treatment. With salt content in the soils, saline hazard and numbers of ineffective stems, amounts of unmatured grain are increased. 6. With the treatment of CHP yields of rough rice were increased. With 0.5% CHP treatment the yields were similar to those of the normal paddy fields. With 1.0% CHP-A treatment, the yields were increased by 15 times more than those of none treated soil and by 25 percent more than normal paddy soils.

Fuel cell based CHP technologies for residential sector (연료전지와 마이크로 열병합 발전기술)

  • This article reports current status of micro fuel cell-combined heat and power (${\mu}FC$-CHP) systems which utilize both power and heat generated by fuel cells. There are several options for constructing CHP systems and among them, fuel cells are the most useful and their total energy efficiency combining heat and power can reach up to about 90%. Fuel cells are classified as five types based on the electrolyte, but the most suitable fuel cell types for the ${\mu}FC$-CHP system are proton exchange membrane fuel cells (PEMFCs) and solid oxide fuel cells (SOFCs). ${\mu}FC$-CHP systems have several advantages such as decrease of the transmission-distribution loss, reduced costs of electricity due to distributed power generation, and environmental-friendliness owing to zero emission. The main drawback of the ${\mu}FC$-CHP systems is the high initial investment, however, it keeps decreasing as the technology development reduces production costs. Currently, Japan is the most leading country of the ${\mu}FC$-CHP market, however, Korea tries to expand the market by planning the deployment of 1 million units of ${\mu}FC$-CHP systems and governmental subsidiary supporting of half of the install price. In this report, integration technologies for connecting FC and CHP, and technology trends of leading countries are presented as well.
