Discovery of Cyclin-dependent Kinase Inhibitor, CR229, Using Structure-based Drug Screening

  • To generate new scaffold candidates as highly selective and potent cyelin-dependent kinase (CDK) inhibitors, structure-based drug screening was performed utilizing 3D pharmacophore conformations of known potent inhibitors. As a result, CR229 (6-bromo-2,3,4,9-tetrahydro-carbolin-1-one) was generated as the hit-compound. A computational docking study using the X-ray crystallographic structure of CDK2 in complex with CR229 was evaluated. This predicted binding mode study of CR229 with CDK2 demonstrated that CR229 interacted effectively with the Leu83 and Glu81 residues in the ATP-binding pocket of CDK2 for the possible hydrogen bond formation. Furthermore, biochemical studies on inhibitory effects of CR229 on various kinases in the human cervical cancer HeLa cells demonstrated that CR229 was a potent inhibitor of CDK2 ($IC_{50}:\;3\;{\mu}M$), CDKI ($IC_{50}:\;4.9\;{\mu}M$), and CDK4 ($IC_{50}:\;3\;{\mu}M$), yet had much less inhibitory effect ($IC_{50}:>20\;{\mu}M$) on other kinases, such as casein kinase 2-${\alpha}1$ (CK2-${\alpha}1$), protein kinase A (PKA), and protein kinase C (PKC). Accordingly, these data demonstrate that CR229 is a potent CDK inhibitor with anticancer efficacy.

Influence of Crystal Structure on the Chemical Bonding Nature and Photocatalytic Activity of Hexagonal and Cubic Perovskite Compounds

  • We have investigated the influence of the crystal structure on the chemical bonding nature and photocatalytic activity of cubic and hexagonal perovskite A[$Cr_{1/2}Ta_{1/2}$]O3 (A = Sr, Ba) compounds. According to neutron diffraction and field emission-scanning electron microscopy, the crystal structure and particle size of these compounds are strongly dependent on the nature of A-site cations. Also, it was found that the face-shared octahedra in the hexagonal phase are exclusively occupied by chromium ions, suggesting the presence of metallic (Cr-Cr) bonds. X-ray absorption and diffuse UV-vis spectroscopic analyses clearly demonstrated that, in comparison with cubic Sr[$Cr_{1/2}Ta_{1/2}$]$O_3$ phase, hexagonal Ba[$Cr_{1/2}Ta_{1/2}$]$O_3$ phase shows a decrease of Cr oxidation state as well as remarkable changes in interband Cr d-d transitions, which can be interpreted as a result of metallic (Cr-Cr) interactions. According to the test of photocatalytic activity, the present semiconducting materials have a distinct activity against the photodegradation of 4-chlorophenol. Also the Srbased compound was found to show a higher photocatalytic activity than the Ba-based one, which is attributable to its smaller particle size and its stronger absorption in visible light region.

Hardness and Oxidation Resistance of Ti0.33Al0.67N/CrN Nano-multilayered Superlattice Coatings

  • $Ti_{0.33}Al_{0.67}N/CrN$ nano-multilayers, which are known to have excellent wear resistance, were prepared using an unbalanced magnetron sputter to have various periods of 2-5 nm. $Ti_{0.33}Al_{0.67}N$ had a hexagonal structure in a single layer, but converted to a cubic structure by forming a multilayer with CrN, which has a cubic structure. Thus, $Ti_{0.33}Al_{0.67}N$ formed a superlattice in the multilayer. The $Ti_{0.33}Al_{0.67}/CrN$ multilayer with a period of 2.5 nm greatly exceeded the hardness of the $Ti_{0.33}Al_{0.67}N$ and the CrN single layer, reaching 39 GPa. According to the low angle X-ray diffraction results, the $Ti_{0.33}Al_{0.67}N/CrN$ multilayer maintained its as-coated structure to a temperature as high as $700^{\circ}C$ and exhibited hardness of 30 GPa. The thickness of the oxide layer of the $Ti_{0.33}Al_{0.67}N/CrN$ multilayered coating was less than one-tenth of those of the single layers. Thus, $Ti_{0.33}Al_{0.67}N/CrN$ multilayered coating had hardness and oxidation resistance far superior to those of its constituent single layers.

  • Let M be an 0-minimal structure on the standard structure :=( , +, ,<) of the field of real numbers. We study Cr -G manifolds (0$\leq$r$\leq$w) which are generalizations of Nash manifolds and Nash G manifolds. We prove that if M is polynomially bounded, then every Cr -G (0$\leq$r<$\infty$) manifold is Cr -G imbeddable into some n, and that if M is exponential and G is a compact affine Cw -G group, then each compact $C\infty$ -G imbeddable into some representation of G.

  • The structural changes of Al-Cr alloys due to the difference in the growth rates were investigated in the study using the water cooled copper chill apparatus, the levitation apparatus, and the melt spinner. Growth rate was evaluated by means of thermal analysis could measured the cooling rate up to 10$^{5}$ K/sec. The transformation from the cell structure to the massive transformed structure was obtained the Al-3.43wt%Cr alloy in the melt spinner method.

Control of Thin Film Media Microstructure by Using Very Thin Seedlayer Material with Different Affinity for Oxygen

  • To reduce the grain size and the media noise in a typical CrMo/CoCrPtB longitudinal media, a sputtering process which includes the exposure of oxygen onto the surface of CrW$_x$ (x=0, 25, 50, 75, 100 at.%) and CrTi$_{15}$ seedlayers with the thickness of 0.5 nm have been utilized. The main results are: (1) the media grain size and the media noise are reduced when using CrW$_x$ (x=0, 25, 50 at.%) seedlayers, and not reduced when using CrTils or CrW$_x$ (x=75, 100 at.%) seedlayers, (2) AES and RHEED results suggest that W seedlayer, which has the highest melting point, forms layer-like film with very small and dense island grain, due to its high free surface energy and low mobility. On the other hand, CrW$_{50}$ and Cr seedlayers, which have lower melting point than W seedlayer, form island film, (3) to effectively reduce the media grain size and improve the media signal to noise ratio, it is essential to utilize a very thin Cr-based seedlayer with high affinity for oxygen and which forms island-like structure, such as CrW$_{50}$ seedlayer.

Effect of Oxygen on the Microstructure and Mechanical Properties of Cr-O-N Coatings (Oxygen 함량에 따른 Cr-O-N 코팅막의 미세구조 및 기계적 특성에 관한 연구)

  • Cr-O-N coatings having different oxygen contents were deposited on Si wafer and SUS 304 substrates by an arc ion plating technique using Cr target in $Ar/O_2/N_2$ gaseous atmosphere. As increasing oxygen content in the coating, the microstructure of Cr-O-N coating changed from polycrystalline having NaCl structure to amorphous structure. Further increase of oxygen content resulted in phase transformation from amorphous to rhombohedral structure. From the variations of d value and average grain size, it was revealed that the maximum solubility of oxygen in Cr-O-N coating was about 21 at.%. And the maximum micro-hardness of 2751HK was obtained in this composition. The lowest friction coefficient was measured in the coating having 34.8 at.% of oxygen. However, more narrow width of wear track was found in the coating having 30.1 at.% of oxygen.

  • The structure and mechanical properties of the extruded specimens were investigated in rapid solidified Al-(1, 3, 5) Cr alloys after mechanical alloying. Finer lamellar microstructure could no longer be resolved in the bars obtained by extrusion of the spherical particles after 200 min. of processing time. The structure of extruded bars are highly depended on the processing time of splats. The isothermal annealing of the extruded bars showed that all the alloys had good thermal stability up to $400^{\circ}C$ and did not show the recrystallization phenomena. Severe working of Al-(1, 3, 5) Cr splats produced a very fine grain size and substructural strengthening (high dislocation density and fine grain size). Effects of mechanical alloying on the thermal stability of the extruded bars Al-(1, 3, 5) Cr alloys decreases, with increasing Cr content. But the ultimate tensile Strength in the extruded bars increases with increasing Cr content.

  • We study an (n+3)($n\;{\geq}\;7-dimensional$ real submanifold of a (4m+3)-unit sphere $S^{4m+3}$ with Sasakian 3-structure induced from the canonical quaternionic $K\"{a}hler$ structure of quaternionic (m+1)-number space $Q^{m+1}$, and especially determine contact three CR-submanifolds with (p-1) contact three CR-dimension under the equality conditions given in (4.1), where p = 4m - n denotes the codimension of the submanifold. Also we provide necessary conditions concerning sectional curvature in order that a compact contact three CR-submanifold of (p-1) contact three CR-dimension in $S^{4m+3}$ is the model space $S^{4n_1+3}(r_1){\times}S^{4n_2+3}(r_2)$ for some portion $(n_1,\;n_2)$ of (n-3)/4 and some $r_1,\;r_2$ with $r^{2}_{1}+r^{2}_{2}=1$.

MAGNETORESISTANCE OF EPITAXIALLY GROWN METALLIC MULTILAYERS

  • The epitaxial TM/bcc-Cr(001) (TM=Fe, Co, Ni) multilayers have been prepared using MBE. The crystal structure, interlayer exchange coupling and magnetoresistance of those multilayers have been discussed. The structure of Fe, Co and Ni grown on bcc-Cr(001) exhibited bcc(001), distorted hcp(1120) and fcc(110), respectively. In Fe/Cr multilayes, an oscillatory exchange coupling has been observed, but not observed in Ni/Cr system, which may come from the large mixing at interfaces. Large MR ratio (116%, 4.2K) has been obtained in Fe/Cr system, but only 2% in Co/Cr system. This difference can be understood from the view point of the relative potential geight for down spin electrons between TM and Cr.
