Changes of Plant Cell Size Index by Culture Conditions (배양 조건에 따른 식물세포 크기 지수의 변화)

  • Effects of various environmental factors on cell size index(FCW/DCW) in Thalictrum rugosum. Lithospermum erythrorhizon and Taxus cuspidata plant cell suspension cultures were investigated. Time course change of cell size index were also observed. In batch cultures, FCW/DCW increased according to the decrease of sugar concentration. For short-term experiment within 24 hr, FCW/DCW value could be reduced significantly by increasing sugar concentration. When an osmoticum such as mannitol was added, FCW/DCW converged to a low value. Therefore, it was confirmed that osmolality of the medium was important in determining cell size or water content of the cells. Inorganic salts or treatment with organic solvent also exhibited some effect on the cell size index. However, pH and centrifugal force did not show any influences. On the other hand, it was found that the addition of Pluronic F-68 reduced FCW/DCW. By combining these results effectively, it may be possible to increase the cell concentration in high density culture to a higher extent.

  • The first step to design a cellular manufacturing system is to make part-families and machine-cells. This process is called the machine-part grouping. This paper considers a machine-cell size when grouping machine-cells. By considering a machine-cell size, an unrealistically big size of machine-cell which may be caused by the chaining effect can be avoid. A heuristic algorithm which minimizes the number of inter-cell movement of parts considering a machine-cell size is presented. The effectiveness and performance of the proposed heuristic algorithm are compared with those of several heuristic algorithms previously reported. The comparison shows that the proposed heuristic algorithm is efficient and reliable in minimizing the number of inter-cell movement of parts and also prevents the chaining effect.

  • Hybrid optical-electronic procedures are introduced for the automated estimation of cell parameters (e.g., size, size variation, and shape). Two different optical Fourier analysis procedures are applied to high contrast cell boundary patterns obtained from specular micrographs of the endothelial layer. In one case, a large number of cell patterns are illuminated to extract average cell size information. Once the average cell size information has been obtained, individual cells are illuminated to extract shape information.

  • When parental Chinese hamster ovary (CHO) cell clones that are capable of producing thrombopoietin (TPO) were subjected to high methotrexate (MTX) concentrations, clonal variations in cell growth were apparent. In the clones that had no significant enhancement in specific TPO productivity (q$\_$Tpo/)when a higher level of MTX was administered, their growth was not depressed significantly nor their cell size changed significantly. On the other hand, those clones that showed a significant-enhancement in q$\_$Tpo/ at higher a MTX dosage, cell growth was depressed initially but recovered during successive sub-cultures. Furthermore, their cell size increased, which suggested that changes in cell size may be indicative of an enhanced q$\_$Tpo/. When the enhancement of the q$\_$Tpo/ of 9 clones after a high MTX dosage was plotted against the extent of the increase of their size, there was a linear correlation (γ$^2$=0.80, p<0.001, ANOVA), which suggested that an enhancement of q$\_$Tpo/ after high MTX administration can be measured by the increase in their cell size. Taken together, our data demonstrate that the selection of amplified CHO cell clones with enhanced q$\_$Tpo/ can be done upon their increased size and growth pattern. This facilitates the development of highly productive recombinant CHO cell lines.

  • In this paper a new cell sizing method is proposed. The new method calculates cell size at a point using given size control elements directly without the aid of background grid as other cell sizing algorithms do. The calculation method and related definitions are described in detail, and typical cell sizing results are given.

  • Open microcellular foams having small-sized cell and good mechanical properties are desirable for many practical applications. As an effort to reduce the cell size, the microcellular foams combining viscosity improvers into the conventional formulation of styrene and water system were prepared via high internal phase emulsion polymerization. Since the material properties of foam are closely related to the solution properties of emulsion state before polymerization, the flow behavior of emulsions was investigated using a controlled stress rheometer. The yield stress and the storage modulus increased as viscosity improver concentration and agitation speed increased, due to the reduced cell size reflecting both a competition between the continuous phase viscosity and the viscosity ratio and an increase of shear force. Appreciable tendency was found between the rheological data of emulsions and the cell sizes of polymerized foams. Cell size reduction with the concentration of viscosity improver could be explained by the relation between capillary number and viscosity ratio. A correlative study for the cell size reduction with agitation speed was also attempted and the result was in a good accordance with the hydrodynamic theory.

  • Micro Cellular Plastics create a sensation at polymer industrial for lowering product cost & overcoming a lowering of mechanical intensity. Reduction of MCPs cell size increases the intensity of MCPs. This research based on the experiment about cell size reduction method. At this study, Quenching & Pressure foaming process are introduced to one of methods. Conclusion of study is that Quenching process is the simplest process for nano cell formation.

  • We investigate the field-dependence of energy barrier for various cell diameters and two type of geometry through the NEB method. We find that the energy barrier can depend strongly on the cell size when the switching is governed by the domain wall motion. Moreover we also examine the cell size dependence of energy barrier for two type of cell geometry. In the presentation, we will discuss the effect of domain wall formation and more various cell size on the energy barrier in detail.

  • Pseudomonas diminuta (ATCC 19146) has been typically used in the bacterial challenge test for validation of the sterilizing filtration process. Cell size is critical for determining the retention characteristics of membrane filters with pore-size of $0.2{\mu}m$. The changes of cell sizes after osmotic shocks at 150, 260, 500, and 700 mosM were measured by a particle size analyzer and the changes of their buoyant densities were analyzed with a Percoll gradient. The results indicated that there were no significant differences when cells were cultured in 260 mosM medium and osmotically shocked at 500 and 700 mosM. However, the osmotically shocked cells at 150 mosM showed a 38% increase of the cell size compared to the cells at 260 mosM. From these study, we concluded that the worst case condition for validation of a sterilizing filter would be 500 mosM, not because of changes in the cell size, but due to decrease in cell viability under those conditions.

Effects of Wax Guard on Weight, Triglyceride, Leptin and Fat Cell Size in Rats Fed on a High Fat Diet (고지방식이를 섭취시킨 흰쥐에서 동과가 체중, 중성지방, Leptin과 지방세포의 크기에 미치는 영향)

  • The purpose of this study was to investigate the effects of wax gourd on weight, triglyceride, leptin and fat cell size in rats fed a high-fat diet. Male Sprague Dawley rats were fed an experimental diet containing total dietary fat at 40% of calories with wax gourd 0%, 5%, 10% and 15% (w/w) for 4 weeks. Weight gain and triglyceride level fell significantly in the 15% wax gourd group compared to the control group. Epididymal fat pad, abdominal fat and perirenal fat tended to decrease in the 15% wax gourd group. Leptin and free fatty acid level were not significantly different among the groups. Fat cell size significantly decreased in the 10% and 15% wax gourd groups compared to the control group. Weight gain correlated positively with visceral fat masses and the levels of leptin and triglyceride. Fat cell size significantly correlated with visceral fat and leptin level. Therefore, the 15% wax gourd diet substantially reduced weight, triglyceride and fat cell size. (Korean J Nutrition 36(5): 446∼451, 2003)
