Crown and Foot Rot of Grafted Cucumber Caused by Fusarium solani f. sp. cucurbitae (Fusarium solani f. sp cucurbitae에 의한 오이 근경썩음병)

  • In March 2010, grafted cucumber cultivated in the greenhouse showed a severe rot on crown resulting yellowing and wilting of the leaves. The symptoms of naturally infected plants showed dark brown, watersoaked lesions at the base of the stem. The fungus produced mass of white mycelium and yellow to orange spores in necrotic lesions on dead and dying plants. Fungus was isolated from rotted tissues of the crown and root. On the basis of morphological characteristics, ITS sequence and pathogenicity tests, the isolate was identified as Fusarium solani f. sp. cucurbitae. This is the first report of the crown and foot rot of grafted cucumber caused by F. solani f. sp. cucurbitae in Korea.

  • Out of 60 quarantine insect pests in Korea, 42 species are classified into tephritid fruit flies. Most of these fruit flies are attracted to two natural products, methyl eugenol (ME) or raspberry ketone. Paraffin wax has been devised to formulate these lures. The formulated lures were applied to field test in Taiwan to attract quarantine fruit flies. Wax-ME formulation was installed in Delta trap and could attract 60-80 males of B. dorsalis per day during late August, while a wax formulation of Cue-lure (a methylated compound of raspberry ketone) attracted both B. cucurbitae and B. tau. These wax formulations can be applied to monitor these three quarantine species in Korea.

In Vitro Inhibitory Activity of Cow Urine and Dung to Fusarium solani f. sp. cucurbitae

  • This paper deals with the study on comparative efficacy and in vitro activity of cow urine and cow dung for controlling root rot disease of cucumber caused by Fusarium solani f. sp. cucurbitae Snyder & Hansen following slide germination and mycelial growth inhibition tests. Results showed that both germination of conidia and the percentage inhibition of mycelial growth decreased or suppressed and varied greatly with respect to different hour and days of incubation and kind of bio-matters. In between two bio-matters cow urine was found more effective than that of cow dung in conidial germination. No germination of conidia was recorded after one hour of incubation in any medium whereas in cow urine germination of conidia was not also observed even after 2 hours of incubation. After 7 hours of incubation out of 200 conidia of F. solani f. sp. cucurbitae, 28 in cow urine and 64 in cow dung were germinated while in control a total germinated conidia was 185. In case of percentage inhibition of conidial germination the highest percentage(100%) was recorded in cow urine after 2 hours of incubation followed by 3 hours(96.0%), 4 hours(91.0%) and 6 hours(89.4%). During the test on inhibition of mycelial growth, the highest percentage(62.8%) was recorded in cow urine potato dextrose agar(CUPDA) medium tested after 4 days of incubation, followed by 3 days(60.5%), 5 days(56.5%) and 2 days(55.0%). In this test cow dung potato dextrose agar(CDPDA) had less efficacy in suppression of the percentage inhibition of mycelial growth.

  • Monitoring exotic fruit flies is essential for quarantine procedure. Wax formulation containing fruit fly lures is relatively long-lasting in field conditions and has been applicable to monitor the fruit flies. This study was performed to extend the application of wax formulation against different fruit flies. The wax formulation containing lures was tested in Thailand, at which various exotic fruit flies inhabited. Captured flies were identified to be Bactrocera dorsalis, Zeugodacus cucurbitae, and Zeugodacus caudata by molecular diagnosis technique.

  • Fusarium species, a large group of plant pathogens, potentially pose quarantine concerns worldwide. Here, we focus on the development of a method for detecting four Fusarium species in quarantined plants in Korea: F. solani f. sp. cucurbitae, F. stilboides, F. redolens, and F. semitectum var. majus. Species-specific primers were designed from the nucleotide sequences of either the translation elongation factor-1 alpha (TEF1) gene or RNA polymerase II subunit (RPB2) gene. Two different primer sets derived from TEF1, all specific to F. solani f. sp. cucurbitae, were able to differentiate the two races (1 and 2) of this species. A set of nested primers for each race was designed to confirm the PCR results. Similarly, two primer sets derived from RPB2 successfully amplified specific fragments from five F. stilboides isolates grouped within a single phylogenetic clade. A specific TEF1 primer set amplified a DNA fragment from only four of the 12 F. redolens strains examined, which were grouped within a single phylogenetic clade. All of the F. semitectum var. majus isolates could be specifically detected with a single RPB2 primer set. The specificity of the primer sets developed here was confirmed using a total of 130 Fusarium isolates.

Isolation and identification of a phenol-degrading bacterium from the sewage sludge (하수슬러지로부터 페놀분해세균의 분리 및 동정에 관한 연구)

  • A bacterium which grow on phenol as an only carbon and energy source was isolated from the sewage sludge at Nangi municipal wastewater treatment plant in Seoul. This bacterium was found to be a Gram negative rod with high motility, and well grew on 0.05%, 0.1%, and 0.15% of phenol. No matching strain was found from the result of the BBL test. Phylogenetic analysis of the strain by comparison of the 16s-rDNA has revealed that this bacterium has 99% of similarity with Stenotrophomonas maltophilia strain of Xanthomonas group, which belongs t the Gamma (${\gamma}$) subdivision of Proteobacteria. This strain has also shown 98% of similarity with nitrogen fixing bacterium MAGDE3 and Pseudomonas cissicola strain, and 97% of similarity with Stenotrophomonas sp. LMG198 and Xanthomonas cucurbitae.

β-Carotene, Cucumisin Content and Fruit Morphology of Melon (Cucumis melo L.) Germplasm Collections

  • Melon fruits are a year-old plant in the family Cucurbitae and are the most cultivated fruit in tropical countries. Melon flesh is an important source of anti-cancer, antioxidant effects, attracting attention as a functional food. We investigated the morphological properties, β-carotene content, and cucumisin activity of seeds of 58 melon genetic resources. Melon resources have shown various morphological properties. Melons grouped by morphological properties were the five groups. β-carotene content varied between 82.34 mg/kg, 86.75 mg/kg, 25.56 mg/kg, 86.25 mg/kg, and 54.65 mg/kg. Between β-carotene, cucumisin activity and other quantitative fruit morphological properties, the color of the pulp and the firmness of the pulp had a significant amount of correlation between the β-carotene content. However, cucumisin activity and β-carotene content had significant negative correlation, and the color of the fruit and shape of the fruit had significant negative correlation with β-carotene content and activity of cucumisin. Our study vindicated that high diversity in melon morphological characters of genetic sources that provide beneficial baseline data for the future and in the activity of β-carotene and cucumisin, and these results could predict indicators of β-carotene content by the length of leaf, the width of fruit, the length of joint, and the length of seed, and collectively the morphological properties of melons could help predict indicators of β-carotene content and help develop functional sarcoma and farmhouse cultivation.

Geographical Variation of the Oriental Fruit Fly, Bactrocera dorsalis, Occurring in Taiwan (오리엔탈과실파리 유전변이 - 대만 지역 집단변이)

  • This study analyzed genetic variation of the Oriental fruit fly (OFF), Bactrocera dorsalis, which is designated to be a quarantine insect pest in Korea. OFF samples endemic to Taiwan were collected at three different locations (Taipei, Taichung, and Kaohsiung) for three days from July 30 to August 1 in 2018 and assessed in their age and mitochondrial DNA sequence variations. In these places, 1,085 OFF males were collected using methyl eugenol lure while 30 males of Zeugodacus cucurbitae and one male of Bactrocera tau were collected using Cuelure. A protein diet lure attracted 6 flies including one OFF and 5 flies of Z. cucurbitae. Male heads of OFF contained pterin, which increased in contents with age from 32 to $59{\mu}g/head$. There was a local variation in pterin amounts in OFF heads, in which Kaohsiung population had lower amounts of pterin than Taipei and Taichung populations. Genetic distance among these three populations were measured by random amplified polymorphic DNA and showed that Taipei population was separated from Taichung/Kaohsiung cluster. Genetic variation was also analyzed in sequence variations in cytochrome oxidase I (CO-I) and NADH dehydrogenase I (ND-I). There was 7.8% variation in CO-I sequence (360 residues) and 6.6% variation in ND-I sequence (213 residues). These polymorphic sites are proposed to be used to develop SNP (single nucleotide polymorphism) markers characteristic to Taiwan OFF populations.

A Tuber Lectin from Arisaema jacquemontii Blume with Anti-insect and Anti-proliferative Properties

  • A tuber lectin from Arisaema jacquemontii Blume belonging to family Araceae was purified by employing a single step affinity chromatography using column of asialofetuin-linked amino activated silica beads and the bound lectin was eluted with 100 mM glycine-HCl buffer pH 2.5. The purified A. jacquemontii lectin (AJL) showed a single protein band with an apparent molecular mass of 13.4 kDa when submitted to SDS-polyacrylamide gel electrophoresis under reducing as well as non-reducing conditions. The native molecular mass of AJL determined by gel filtration on a Biogel P-200 column was 52 kDa and its carbohydrate content was estimated to be 3.40%. Thus AJL is a tetrameric glycoprotein. The purified lectin agglutinated erythrocytes from rabbit but not from human. Its activity was not inhibited by any of the mono- and disaccharides tested except N-acetyl-D-lactosamine having minimal inhibitory sugar concentration (MIC) 25 mM. Among the glycoproteins tested only asialofetuin was found to be inhibitory (MIC $125\;{\mu}g/mL$). A single band was obtained in native PAGE at pH 4.5 while PAGE at pH 8.3 showed two bands. Isoelectric focusing of AJL gave multiple bands in the pI range of 4.6-5.5. When incorporated in artificial diet AJL significantly affected the development of Bactrocera cucurbitae (Coquillett) larvae indicating the possibility of using this lectin in a biotechnological strategy for insect management of cucurbits. Larvae fed on artificial diet containing sub-lethal dose of AJL showed a significant decrease in acid phosphatase and alkaline phosphatase activity while esterase activity markedly increased as compared to larvae fed on diet without lectin. Out of various human cancer cell lines employed in sulphorhodamine B (SRB) assay, this lectin was found to have appreciable inhibitory effect on the in vitro proliferation of HCT-15, HOP-62, SW-620, HT-29, IMR-32, SKOV-3, Colo-205, PC-3, HEP-2 and A-549 cancer cell lines by 82, 77, 73, 70, 41, 41, 37, 29, 21 and 21% respectively.
