꾸지뽕나무 (Cudrania tricuspidata)의 폴리페놀 화합물 함량과 항산화 활성 (Antioxidative Activities and Contents of Polyphenolic Compound of Cudrania tricuspidata)

  • Polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess strong antioxidant activity. The antioxidative substances of water soluble extract from leaves, stem bark, root bark and fruit powder of Cudrania tricuspidata were tested in three different in vitro experimental models. In oxidation models using DPPH( , ' diphenyl picrylhydrazyl) method, Fe2+ induced linoleic acid peroxidation, and autooxidation of hepatic microsomal membranes, the antioxidative activities of water soluble extract from stem bark were strong than that from leaves and root bark. Fruits of Cudrania tricuspidata contained the highest amounts of polyphenolic compounds among the parts of this plant. The changes in polyphenolic compound contents of fruit powder caused by heat treatment (20oC, 40oC, and 60oC) were also monitored. After water blanching, contents of phenolic compounds was increased slightly in the following order; 20oC(1454mg), 40oC(1487mg), and 60oC(1511mg). These results supports that water soluble extracts from Cudrania tricuspidata contain antioxidative compounds.

꾸지뽕 열매 추출물이 인간 유래 각질 형성 세포의 밀착연접에 미치는 영향 (Investigation of the effect of water extract from Cudrania tricuspidata fruit on tight junction in human keratinocyte)

  • Objectives : Cudrania tricuspidata (C. tricuspidata) is well-known traditional herbal remedy and its root, leaf and fruit were used for treatment of inflammation, tumor and painkilling. However, effect of C. tricuspidata fruit on tight junction is still unknown. The aim of this research was to determine the effect of C. tricuspidata fruit extract on human keratinocyte HaCaT cells. Methods : The antioxidant effects of water extract of C. tricuspidata (WECT) and ethanol extract of C. tricuspidata (EECT) were analyzed by using an ABTS assay. To confirm the cytotoxicity of WECT and EECT, MTS assay was performed. The mRNA expression levels of tight junction related genes were analyzed using quantitative RT-PCR analysis. Furthermore, dispase assay was used to investigate the alteration of cell-cell adhesion strength of EECT treated HaCaT cells. Results : WECT and EECT showed strong antioxidant activity. No obvious cytotoxicity was observed in both WECT and EECT until $2.0mg/m{\ell}$ concentration. The mRNA expression level of Claudin 6 were significantly increased by EECT treatment, whereas the WECT did not affect the expression of Claudin 6. Furthermore, EECT treatment enhances cell-cell adhesion strength. Conclusions : In this study, we investigated the physiological activities of the extracts of Cudrania tricuspidata fruit extracts on human keratinocytes by two different extraction methods. EECT might have an anti-aging activity on the skin by reducing oxidative stress. Moreover, it may be a useful ingredient in atopic dermatitis and skin-moisturizing, given its effects of altering Claudin 6 gene expression and enhancing cell-cell adhesion strength.

국내 자생 꾸지뽕나무 잎과 열매의 형태적 특성에 관한 연구 (A Study on the Morphological Characteristics of Leaves and Fruit of Cudrania tricuspidata in Korea)

  • 꾸지뽕나무의 수요 증가에 따라 자생지 파괴가 심각한 반면 꾸지뽕나무에 대한 연구는 미비하기 때문에 자생지 복원과 새로운 농가 소득원으로 이용하기 위하여 국내에 자생하는 꾸지뽕나무 16개 집단을 대상으로 엽과 열매의 형태적 특성을 분석한 결과는 다음과 같다. 조사된 꾸지뽕나무 집단에서 엽의 형태적 특성에 대한 유집분석 결과 4개의 그룹으로 구분되었다. 열매의 형태적 특성을 분석하기 위해 꾸지뽕나무 15개 집단의 열매길이, 폭, 무게 등 3가지 형질을 분산 분석한 결과, 유의확률이 p=0.05로 각각의 형질에 따라 집단 간에 차이가 있는 것으로 나타났으며 열매의 3가지 형질별 특성을 이용하여 군집분석을 실시한 결과 세 집단으로 구분되었다. 본 연구결과에 따르면 꾸지뽕나무 엽과 열매의 형태학적 특징은 각 집단 간 차이가 있었으며, 엽의 형태적 특징과 열매의 특징이 일치하지는 않았다.

꾸지뽕나무 뿌리 열수추출물로부터 미백 및 주름개선 화장품 소재의 개발 (Cudrania tricuspidata Root Extract as Whitening and Antiwrinkle Cosmetic Agent)

  • 본 연구에서는 꾸지뽕나무의 뿌리, 줄기, 열매 및 잎으로부터 유효성분을 열수 추출하고 다양한 화장품 소재 시험을 실시하여 화장품 소재로서의 개발 가능성을 검토하였다. 세포독성시험에서 꾸지뽕나무의 뿌리, 줄기 그리고 열매 추출물은 독성을 나타내지 않았지만 잎 추출물은 $200{\mu}g/ml$ 이상의 농도에서 다소의 독성을 나타내었다. 꾸지뽕나무 뿌리추출물은 $500{\mu}g/ml$의 농도에서 81%의 tyrosinase 억제효과(미백효과)와 58%의 elastase 억제효과(주름개선효과)를 나타내었고, UVB 자외선 흡수능도 다소 보여주었다. 21일간의 온도 안정성 시험 결과, 꾸지뽕 뿌리 추출물을 포함한 액제와 로션제형은 $25^{\circ}C$에서는 안정하였으나 $47^{\circ}C$에서 점도와 입자크기가 불안정하였다. 본 연구 결과 꾸지뽕 뿌리추출물은 높은 폴리페놀 및 플라보노이드의 함량, 우수한 미백 및 주름개선 효과를 나타내어 천연 기능성 화장품 소재로서 높은 가능성을 보여주었다.

  • Polysaccharides produced in microorganisms and plants are known to increase the immune response in the body. We proposed analysis of beta-glucan contents of phellinus linteus fruit body (FB) and mycelium (MC) cultured in cudrania tricuspidata. Also, we examined whether fruit body and mycelium can increase the immune response in cyclophosphamide-induced immunosuppression animal models. We injected cyclophosphamide (50 mg/kg) twice to produce immunosuppression mice. Then, FB (200 mg/kg) and MC (200 mg/kg) were oral administered for 14 days. In order to confirm the immune-enhancing effect of FB and MC, we analyzed spleen weight, the number of immune cells, cytokines, and immunoglobulins levels. Cyclophosphamide decreased the weight of spleen, the number of immune cells. However, FB and MC have significantly increased the weight of spleen, the number of white blood cell, lymphocyte and monocyte. In addition, they have significantly increased immune-related cytokines (IL-2 and IFN-${\gamma}$) and immunoglobulins (IgA, IgG, IgM) levels. As a results, phellinus linteus fruit body (FB) and mycelium (MC) cultured in cudrania tricuspidata can be used as effective natural materials for immune-enhancing.

전통발효식품으로부터 분리한 초산균을 이용한 꾸지뽕 열매 발효식초 제조 및 발효특성 (Fermentation characteristics of mulberry (Cudrania tricuspidata) fruit vinegar produced by acetic acid bacteria isolated from traditional fermented foods)

  • 꾸지뽕 열매의 부가가치를 증대하기 위해 꾸지뽕 발효식초를 제조하고 그의 발효조건을 확립하였다. 전통발효식품으로부터 초산내성, 초산 고생산능, ethanol 내성 및 아황산 내성이 우수한 49종의 초산 균주를 분리하였고, 16S rRNA 유전자 염기서열의 해독 결과, Acetobacter indonesiensis, A. cerevisiae, A. orientalis, A. tropicalis, A. fabarum, A. pasteurianus 및 A. syzygii으로 동정되었다. 이들 중 GRAS 균주인 A. pasteurianus SCMA5와 SCMA6를 발효 균주로 최종 선정하였다. 최적 발효는 꾸지뽕 열매 함량이 40%(v/v)인 즙액과 5%(v/v) ethanol을 첨가하여 $25^{\circ}C$에서 72시간 발효가 가장 적절하였다. 관능평가 결과, SCMA06 균주를 적용한 발효액의 선호도가 SCMA05 균주를 적용한 발효액보다 높았다. SCMA06 균주를 사용한 발효식초에서 항산화 능력을 측정하는 DPPH 라디컬 소거활성의 경우 대조구에 비해 $53.02{\pm}0.78%$이상 높게 나타났고, 항당뇨 능력을 측정하는 AGI활성은 발효 72시간에 $91.40{\pm}2.43%$ 저해능을 보여 시판중인 acarbose보다 활성이 높았다. 이번 연구는 꾸지뽕 열매를 활용한 발효식초 제조를 위한 산업화 연구에 기여할 수 있을 것이다.

발효 꾸지뽕(Cudrania tricuspidata) 열매 추출물이 마우스 비장세포의 cytokine 생성에 미치는 영향 (Effect of Fermented Cudrania tricuspidata Fruit Extracts on the Generation of the Cytokines in Mouse Spleen Cells)

  • 꾸지뽕(Cudrania tricuspidata) 열매의 면역증강 작용을 검토하기 위하여 유산균을 이용하여 발효한 후 70% 에탄올 추출하여 그 추출물을 마우스 비장세포에 작용한 후 분비되는 cytokine으로 면역활성을 조사하였다. 그 결과 발효에 의해 성분의 변화로 총 페놀함량을 측정한 결과 발효에 의해 폴리페놀 함량이 약 47% 증가하였으며, 면역활성에서는 발효 꾸지뽕 열매에서 cytokine IL-2와IL-4의 분비량이 증가하였다. 그리고 LPS 자극제와 함께 처리하였을 때, Th1 cell 유도 cytokine인 IL-2, IL-12, TNF-${\alpha}$의 분비는 억제되었으나, Th2 cell 유도 cytokine인 IL-4, IL-5, IL-6, IL-10의 분비량이 유의적으로 증가하였다. 이는 발효 꾸지뽕 나무 열매 추출물이 Th1 cell의 발현에 의해 유도되는 세포성 면역작용 반응은 억제하며, B 림프구에 의한 항체생성과 관련된 체액성 면역작용을 유도하는 Th2 cell의 발현을 활성화 하는 것으로 사료된다.

  • The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.

Evaluation of Antioxidant Activity of Cudrania tricuspidata (CT) Leaves, Fruit Powder and CT Fruit in Pork Patties during Storage

  • The objective of this study was to assess antioxidant activities of leaves and fruit powder of Cudrania tricuspidata (CT) with different particle sizes (crude, 500 ㎛, 150 ㎛), and determine the physicochemical properties and microbial counts of pork patties with various levels of CT fruit powder (CTFP) during refrigerated storage. Total phenolic content of crude leaves had the highest value of 3.54 g/100 g (p<0.05). Overall, CT leaves (CTLP) had higher total phenolic content than CTFP (p<0.05). 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of CTFP was higher than that of CTLP (p<0.05), which showed higher iron chelating ability and reducing power than CTFP (p<0.05). After pork patties were manufactured with 0.5% and 1.0% of CTFP at 500 ㎛, pH, color values, thiobarbituric acid reactive substance (TBARS), and peroxide values (POV) were then measured. The addition of CTFP into pork patties significantly (p<0.05) increased redness and yellowness values of patties. TBARS values of pork patties containing CTFP were lower (p<0.05) than those of CTFP-0 patties after 10 days of storage. Pork patties added with CTFP showed no significant (p>0.05) difference TBARS values among different storage periods. POV values of pork patties containing CTFP were lower than those of the control from 3 days up to the end of refrigerated storage (p<0.05). These results suggest that CTFP could be used as a natural antioxidant to retard lipid oxidation in meat products during refrigerated storage.

Antiplatelet Effect of Cudraxanthone L Isolated from Cudrania tricuspidata via Inhibition of Phosphoproteins

  • Cudrania tricuspidata (C. tricuspidata) is a deciduous tree found in Japan, China and Korea. The root, stems, bark and fruit of C. tricuspidata has been used as traditional herbal remedies such as eczema, mumps, acute arthritis and tuberculosis. In this study, we investigated the potential efficacies of this natural compound by focusing on the inhibitory effect of cudraxanthone L (CXL) isolated from the roots of C. tricuspidata on human platelet aggregation. Our study focused on the action of CXL on collagen-stimulated human platelet aggregation, inhibition of platelet signaling molecules such as fibrinogen binding, intracellular calcium mobilization, fibronectin adhesion, dense granule secretion, and thromboxane A2 secretion. In addition, we investigated the inhibitory effect of CXL on thrombin-induced clot retraction. Our results showed that CXL inhibited collagen-induced human platelet aggregation, intracellular calcium mobilization, fibrinogen binding, fibronectin adhesion and clot retraction without cytotoxicity. Therefore, we confirmed that CXL has inhibitory effects on human platelet activities and has potential value as a natural substance for preventing thrombosis.
