Quantitative analysis of Cynanchum wilfordii Hemsley (Cynanchum wilfordii의 성분함량 분석)

  • Objectives : Cynanchum wilfordii(Asclepiadaceae) has been traditionally used as a tonic, the prevention and treatment of various geriatric diseases in Korea. Acetophenone derivatives from C. wilfordii showed neuroprotective activity. In this study, two acetophenones were isolated and quantitative determination of acetophenones from C. wilfordii has been developed for quality stand. Methods : Three acetophenone derivatives were isolated from methanol extract of C. willfordii by the chromatographic separation. Their structures were identified as cynandione A, 2,5-dihydroxy acetophenone and cynanchone A on the basis of spectral data(MS, $^1H-NMR$, $^{13}C-NMR$) and chemical analysis. HPLC analysis was performed to determine the contents of cynandione A and 2,5-dihydroxyacetophenone in C. wilfordii. Results : According to the results, the contents of cynandione A and 2,5- dihydroxyacetophenone were 0.274%, 0.035% by HPLC, respectively. Conclusions : In these results, we have determined the contents of cynandione A and 2,5-dihydroxyacetophenone in Cynanchum wilfordii, respectively. We hope that this study will contribute to the standardization and quality control of herbal medicine.

Effects of Cynanchum wilfordii Extract on Serum Lipid Components and Enzyme Activities in Hyperlipidemic and Streptozotocin-Induced Diabetic Rats (백하수오 추출액이 고지혈증 및 Streptozotocin 유발 당뇨성 흰쥐의 혈청 지질성분 및 효소활성에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Cynanchum wilfordii extract on the improvement of the blood glucose, lipid components in the serum of dietary hyperlipidemic and streptozotocin(STZ) -induced diabetic rats(S.D. strain, ♂) fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-Cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose in serum were significantly higher in the cholesterol administration groups((group 2(cholesterol+water), 4(cholesterol+Cynanchum WIlfordii 3.5g% extract)) than those in the control group (group1 , basal diet+water). But the concentrations of total cholesterol. atherosclerotic index, LDL, LDL- cholesterol. free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum were remakably lower in the group 4 than those in the group 2. In the STZ(55mg/kg B.W.)-induced diabetic groups((group 3(STZ, IP.)+water), 5(STZ(IP.)+Cynanchum WIlfordii 3.5g% extract? the serum total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose concentrations actions were rather lower in the group 5 than those in the group 3. In the ratio of HDL -cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Cynanchum wilfordii extract administration groups were higher percentage than III the groups 2 and 3. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase (ALP) and lactate dehydrogenase(LDH) in serum were rather lower in the Cynanchum wllfordii extract administration groups (group 4,5) than in the cholesterol diet group(group 2) and STZ-induced diabetic group (group 3). From the above research, the physiological activity substances in Cynanchum wllfordii were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic and STZ-induced diabetic rats. And particularly, physiological activity substance in Cynanchum wilfordii was more effective therapeutic regimen for the control of metabolic derangements in adult disease.

  • Definitive identification of original plant species is important for standardizing herbal medicine. The herbal medicines Cynanchi Wilfordii Radix (Baekshuoh in Korean and Beishuwu in Chinese) and Polygoni Multiflori Radix (Hashuoh in Korean and Heshuwu in Chinese) are often misidentified in the Korean herbal market due to morphological similarities and similar names. Therefore, we developed a reliable molecular marker for the identification of Cynanchi Wilfordii Radix and Polygoni Multiflori Radix. We used random amplified polymorphic DNA (RAPD) analysis of three plant species, Polygoni multiflorum, Cynanchum wilfordii, and Cynanchum auriculatum, to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed six sequence characterized amplification region (SCAR) markers for distinguishing Polygoni Multiflori Radix and Cynanchi Wilfordii Radix. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. These SCAR markers can be used for efficient and rapid authentication of these closely related species, and will be useful for preventing the distribution of adulterants.

Comparison of Cynanchum wilfordii, C. auriculatum, Metaplexis japonica and Polygonum multiflorum by Morphological Characters (백수오, 이엽우피소, 박주가리 및 하수오의 형태적 형질 비교)

  • These studies were conducted to discriminate Cynanchum wilfordii, C. auriculatum, Metaplexis japonica, and Polygonum multiflorum by morphological characters. Cynanchum wilfordii, C. auriculatum, and Metaplexis japonica(Asclepiadaceae) were easily distinguished from Polygonum multiflorum by twining stem to the left, producing opposite leaves, having laticiferous stems and leaves at cutting cross-sectionally, and lacking lignification of the stem. Four species had different morphological characters of flower. Comparing four species to fruit type, three species of Asclepiadaceae had follicle, while Polygonum multiflorum had achene with three wings. Follicle shape was lanceolate, narrowly lanceolate, and widely lanceolate in C. wilfordii, C. auriculatum, and Metaplexis japonica, respectively. Whereas seed color of four species was dark brown similarly, seed shape and 1,000 seed weight were useful characters to discrimate between three species of Asclepiadaceae and Polygonum multiflorum. Shape and color of root were thickened and yellowish brown in C. wilfordii and C. auriculatum. They were thin and long and white in Metaplexis japonica, and fusiform and reddish brown in Polygonum multiflorum. They would be useful characters to distinguish Cynanchum spp., Metaplexis japonica, and Polygonum multiflorum, however, impossible characters to discriminate between C. wilfordii and C. auriculatum.

Cytogenetical Study of Cynanchum wilfordii and Cynanchum auriculatum using Fluorescence In Situ Hybridization (FISH) (rDNA FISH를 이용한 큰조롱과 넓은잎큰조롱의 세포유전학적 연구)

  • Background: Dried tuberous roots of Cynanchum wilfordii are known to relieve menopause symptoms. However, the dried roots of C. wilfordii are morphologically similar to those of C. auriculatum, which makes it difficult to distinguish when used as a medicine. Various comparative studies have focused on chemical or molecular analysis of these roots. However, the differences between the two species at the cytogenetic level based on chromosome structure and composition remain to be elucidated. Methods and Results: For chromosome slides, the roots were fixed in 8-hydroxyquinoline, digested with enzyme mixture, and spread on slides. 5S and 45S rDNA were used as cytogenetic markers for the analysis of nuclear genomes by FISH. The chromosome number of the two species was 2n = 22, with a relatively short length, 1.13 ㎛ - 4.24 ㎛ and 1.00 ㎛ - 3.42 ㎛ with respect to each other. Both species represent one pair of 5S and 45S rDNA signal on chromosome 1, at the proximal region and peri-centromeric region, respectively. Conclusions: These preliminary cytogenetic data using FISH in C. wilfordii and C. auriculatum could be valuable for the comprehension of Cynanchum genome history.

Effect of Peeling Blades and Abrasives on Mechanical Peeling of Cynanchum wilfordii Hemsley (박피(剝皮)날 및 연마재(硏磨材)가 백하수오(白何首烏) 기계박피(機械剝皮)에 미치는 영향(影響))

  • A rapid increasing in cultivation area due to favorable price temporarily and decreasing in consumer's demands caused by economic depressions recently, lead the price of Cynanchum wilfordii Hemsley to decline sharply. Thus, it may give rises to weakening of cultivation bases in Cynanchum wilfordii Hemsley. To investigate optimal mechanical peeling conditions of Cynanchum wilfordii Hemsley, protruded rubber, plastic pad, diamond shape's steel and palstic brush were introduced as blades and artificial stone, sand and small pebble were done as abrasives. The main results obtained were summarized as follows; 1. It took 2 minutes per 1kg in mechanical peeling of Cynanchum wilfordii Hemsley's raw root whereas 36 minutes in manual peeling and values of lightness showed more higher in manual peeling than in mechanical peeling. 2. Yield in combination of diamond shape's steel blade and sand abrasive showed the lowest at 89.9% among treatments and peeling rates in combination of diamond shape's steel blade and small pebble showed the highest at 71.3% in mechanical peeling. 3. Lightness, one of the most important factors in determining quality of Cynanchum wilfordii Hemsley, showed the highest in combination of diamond shape's steel blade and small pebble brasive at 61.90 in mechanical peeling. 4. As the speed of rotation gets faster, yields tend to lower in mechanical peeling. Peeling rates and lightness showed the highest at 66.8%, 57.96 respectively among treatments at 30 r.p.m. in mechanical peeling.

Effects of Isoflavone and Cynanchum wilfordii Supplementation on Premenstrual Syndrome, Dysmenorrhea - Focus on 20's Women - (이소플라본과 백수오 보충 섭취가 월경전 증후군, 월경통에 미치는 영향 - 20대 여성을 중심으로-)

  • Menstrual problem (premenstrual syndrome (PMS) and dysmenorrhea) is a common gynecological complaints among women in reproductive age. The aim of this study was to compare the effects of isoflavone and Cynanchum wilfordii for improvement of premenstrual syndrome, dysmenorrhea. Subjects were 30 college women in Busan, divided into three groups (CG; control group, n=10; IS group; isoflavone supplement group, n=10; CS group; Cynanchum wilfordii supplement group, n=10). They showed 5.0 or higher for menstrual pain when tested by the VAS. IS group was allocated a isoflavone pill (500 mg/day) for 8 weeks, and CS group was allocated a Cynanchum wilfordii pill (6 g/day) for 8 weeks. We administered a menstrual distress questionnaire (MDQ), and visual analogue scale(VAS). There were no differences among the three groups in general characteristics, nutrients intake, MDQ, or VAS. In the CG group, there were no differences in MDQ, or VAS after 8 weeks. In the IS group, negative emotions, behavioral changes, concentration, pain, autonomic nervous system response, water retention, and skin changes significantly improved (P<0.05). In the CS group, negative emotions, pain, autonomic nervous system response, and water retention significantly decreased after 8 weeks (P<0.05). Changes in negative emotion, pain, autonomic nervous system response, water retention, skin changes significantly improved in the CS group compared to the control group (P<0.05). VAS significantly decreased in the IS and CS groups (P<0.05). The decrease in VAS differed among three groups, with the CS group showing a larger decrease than the control group and IS group (P<0.001).

Experiment Study on Antioxidant Activites of Aqua-acupuncture of Cynanchum Wilfordii Hemsley (백화수오약침의 항산화작용에 관한 실험적 연구)

  • This study was performed to prove the antioxidant activities of Cynanchum Wilfordii Hemsley(白何首烏) by way of aqua- acupuncture methods. After 10% & 20% concentrations of liquid extract of Cynanchum Wilfordii Hemsley were acupunctured on Joksamri(足三里) of rats with acute and chronic experimental oxidation by AAPH(2, 2'-azobis(aminoidinopropane), hydrochoride), various kinds of experiments were measured. The results were obtained as follows: 1. In acute antioxidant experiment, the amount of TBARS indicated irregular increase and decrease to over 68% in experiment group in comparison with normal group. 2. In the serum test of acute antioxident experiment, uric acid and total protein and albumin were significantly increased in experiment group in comparison with control group. 3. In chronic antioxident experiment, the change of body weight was significantly decreased in 20% experimental groups in comparison with control group, and only liver significantly increased in 10% & 20% experiment group. 4. In chronic antioxident experiment, total protein significantly increased in 10%, 20% experimental groups and creatinine and BUN were significantly decreased in 10% experiment group. 5. In the chronic antioxident experiment, serum GOT, GPT activity didn't show no significantly change, LDH activity was significantly increase in 10% experiment group in comparison with control group. 6. In the chronic antioxident experiment, the tolerance test with enzyme activities of erythrocyte membrane was significantly decreased in 10% & 20% experimental groups. 7. In the chronic antioxident experiment, the change of TBA was significantly decreased in 10% & 20% experimental group. 8. In the chronic antioxident experiment, the amount of cytochrome p-450 was significantly increased in 10% & 20% experimental groups in comparison with control group. 9. In the chronic antioxident experiment, the amount of cytochrome b5 was significantly increase in 20% experimental group in comparison with control group. 10. In the chronic antioxident experiment, the change of activity of superoxide dismutase, and catalase were significantly increased in 20% experiment group in comparison with control group. From the above result, it was concluded considers the liquid extract of Cynanchum Wilfordii Hemsley(白何首烏) could be used as antiaging drugs, because it delayed the aging process and retarded senility.

Effects of Cynanchum spp. on the hyperlipidemia in rats induced by Triton WR-1339 (국산 및 중국산 백수오(白首烏)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구)

  • Objective : In order to compare of hyperlipidemia effect of C. wilfordii, C. auriculatum in korea and C. auriculatum in China, it was performed on the hyperlipidemia in rats induced by Triton WR-1339. Method : The 70% MeOH extracts from dried root of C. wilfordii and C. auriculatum in korea and C. auriculatum in china were fractionated into H2O layer and CHCl3 layer. The level of total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, AST and ALT in serum were used to observe the effectiveness of C. wilfordii korean C. auriculatum and chinese C. auriculatum. Results : In the hyperlipidemic rats induced by Triton WR-1339, C. wilfordii and korean C. auriculatum significantly decreased total cholesterol, triglyceride, otherwise significantly increased HDL. And C. wilfordii and chinese C. auriculatum significantly decreased AST and ALT. Conclusion : This study indicate that C. wilfordii korean C. auriculatum and chinese C. auriculatum were effective in the hyperlipidemic rats induced by Triton WR-1339. According to the results, C. wilfordii korean C. auriculatum and chinese C. auriculatum need to considered for the application in hyperlipidemia.
