A Study on Effects of Cyperus rotundus L. Essential Oil Inhalation on Stress Relaxation with HRV, EEG (향부자 정유 흡입이 스트레스 이완에 미치는 영향)

  • Objective : The purpose of this study was to assess the effects of Cyperus rotundus L. essential oil on relaxation in highly stressed volunteers with heart rate variability(HRV) and electroencephalography(EEG). Methods : 11 highly stressed volunteers participated in this study. The volunteers were examined with HRV and EEG before and after inhalation of Cyperus rotundus L. essential oil. Results : After smelling Cyperus rotundus L. essential oil, mean RR(mean of RR intervals) was incresed significantly(p<0.01), mean HRV(mean of heart rate), HF(high frequency) were decreased significantly(p<0.01). norm LF(low frequency), LF/HF ratio were decreased significantly(p<0.05), norm HF(normalized high frequency) was increased significantly(p<0.05) on HRV. After smelling Cyperus rotundus L. essential oil, relative ${\theta}$ power was decreased significantly(p<0.05) at P3(left parietal) and relative ${\alpha}$ power was increased significantly(p<0.05) at Fp1(left prefrontal), Fp2(right prefrontal) and relative ${\beta}$ power was decreased significantly(p<0.05) at Fp1(left prefrontal) and relative ${\gamma}$ power was decreased significantly(p<0.05) at Fp1(left prefrontal) on EEG. Conclusions : This results show that inhalation of Cyperus rotundus L. essential oil effects on relaxation and decreasing stress.

Agonistic Activities to the Benzodiazepine Receptor by Extracts of Medicinal Plants(I) -Screening of Some Sedative Plant Extracts- (생약의 Benzodiazepine 수용체 효현활성 검색(I) -수종의 신경안정 생약 추출물에 대한 활성 검색-)

  • This study was aimed to evaluate an agonistic activity to benzodiazepine receptor of several medicinal plants, which have been used as sedatives in oriental medicine. Methanol extracts of medicinal plants which were used in this study inhibited the binding of $[^3H]Ro15-1788$, a selective benzodiazepine receptor antagonist to benzodiazepine receptor of rat cortices. Inhibitory activity of Cyperus rotundus was observed to be the highest among the tested medicinal plants. Methanol extracts of Cyperus rotundus and Zizypus jujuba inhibited a $[^3H]flunitrazepam$, a selective benzodiazepine receptor agonist, binding to benzodiazepine receptor. GABA significantly enhanced the inhibition of $[3H]flunitrazepam$ binding by Cyperus rotundus and Zizypus jujuba, and these positive GABA shifts supported the strong possibility of agonistic activity to benzodiazepine receptor. From these results, it may be concluded that the substance or substances with neurochemical properties characteristic of a benzodiazepine receptor agonist may be important components and contribute to the sedative property of these medicinal plants.

  • This study was attempted to evaluate an agonistic activity to benzodiazepine receptor of several medicinal pants, which have been used as sedatives in oriental medicine. The activities of the methanol extracts of composite preparation of oriental drugs were compared with those of the simple drugs, furthermore, the active fraction was found out from the simple preparation. Inhibitory effects of composite preparations, Cyperus rotundus/Acorus gramineus, Thuja orientalis/Euphoria longan, Thuja orientalis/Albizzia julibrissin, on the binding of ${[^3H]}$Ro15-1788, a selective benszodiazepine receptor antagonist to benzodiazepine receptor of rat cortices, were observed to be lower than those of corresponding simple preparations. These unexpected results suggest that some components of the composite druge may rather act as an obstacle, not to show the sinergistic effect. The methanol extracts of Cyperus rotundus having the highest activity were fractionated using polar and nonpolar solvents to give ethylacetate and hexane fractions, respectively. The ethylacetate fraction containing relatively polar components exhibited much higher activity than the hexane fraction, which consiste of nonpolar agonist, binding to benzodiazepine receptor. However, in the presence of GABA, this fraction inhibited ${[^3H]}$flunitrazepan binding, and these positive GABA shift supported the strong possibility of agonistic activity to benzodiazepine receptro. As a result, it may be concluded that the substance or substances with neurochemical properties as a benzodiazepine receptor agonist may contribute to the sedative property of Cyperus rotundus.

Chemical Components of Cyperus rotundus L. and Inhibitory Effects on Nitric Oxide Production (향부자의 성분 및 Nitric Oxide 생합성 저해효과)

  • Five compounds were isolated from the methanol extract of the tubers of Cyperus rotundus (Cyperaceae). They were identified as cyperol(1), ${\alpha}-cyperone(2)$, cyperotundone(3), sugetriol triacetate(4) and ${\beta}-sitosterol(5)$ on the basis of spectroscopic and physicochemical analysis. Among them, cyperol, ${\alpha}-cyperone$ and cyperotundone were shown inhibitroy effects on LPS-induced nitric oxide production in RAW 264.7 cells in a dose dependent manner.

Polyphasic Analysis of the Bacterial Community in the Rhizosphere and Roots of Cyperus rotundus L. Grown in a Petroleum-Contaminated Soil

  • Cyperus rotundus L. is a perennial herb that was found to be dominating an area in northeast Brazil previously contaminated with petroleum. In order to increase our knowledge of microorganism-plant interactions in phytoremediation, the bacterial community present in the rhizosphere and roots of C. rotundus was evaluated by culture-dependent and molecular approaches. PCR-DGGE analysis based on the 16S rRNA gene showed that the bacterial community in bulk soil, rhizosphere, and root samples had a high degree of similarity. A complex population of alkane-utilizing bacteria and a variable nitrogen-fixing population were observed via PCR-DGGE analysis of alkB and nifH genes, respectively. In addition, two clone libraries were generated from alkB fragments obtained by PCR of bulk and rhizosphere soil DNA samples. Statistical analyses of these libraries showed that the compositions of their respective populations were different in terms of alkB gene sequences. Using culturedependent techniques, 209 bacterial strains were isolated from the rhizosphere and rhizoplane/roots of C. rotundus. Dot-blot analysis showed that 17 strains contained both alkB and nifH gene sequences. Partial 16S rRNA gene sequencing revealed that these strains are affiliated with the genera Bosea, Cupriavidus, Enterobacter, Gordonia, Mycoplana, Pandoraea, Pseudomonas, Rhizobium, and Rhodococcus. These isolates can be considered to have great potential for the phytoremediation of soil with C. rotundus in this tropical soil area.

Effects of methanol extract of Cyperus rotundus on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans (향부자 메탄올 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향)

  • Streptococcus mutans (S. mutans) is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of Cyperus rotundus (Cyperaceae) methanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of C. rotundus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 0.5 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of C. rotundus showed more than 10% inhibition over the concentration of 2 mg/ml. Thus, the application of C. rotundus can be considered a useful and a practical method for the prevention of dental caries.

Production Status and Economic Analysis in Chief Producing Area of Cyperus rotundus (향부자(香附子) 주산지(主産地)의 생산실태(生産實態)와 경제성(經濟性) 분석(分析))

  • Cyperus rotundus has been grown for a long time in korea to be used as medicinal crop. This study was conducted to survey the production status and economic analysis in chief producing area of Cyperus rotundus in Koryong, Korea. The Koryong area where was the major production area of Cyperus rotundus was composed of 0.6hectare cultivation size per farm household and the processes like cultivation, processing, shipping were formed unitedly. The most general management type was labor-extensive one which use the Nakdong river basin or idle land effectively. The cultivation area have been decreased gradually because the price was very low so that the income per hectare was just 3,880 thousand won and productivity per hectare was just 6,280kg. Thus, production condition was very inferior, it needs some self-help to encourage the production because it has some possibility as only regional special product.
