The Inhibitory Effects of Quercetin from Houttuynia cordata against Cadmium-Induced Cytotoxicity (어성초로부터 분리된 Quercetin의 카드뮴에 대한 독성억제효과)

  • The aim of this study was to develop antitoxic compound for cadmium-induced cytotoxic Vero cells. These cells were divided into five groups; control group (medium only), cadmium group (cadmium only), and two experimental groups. SRB (sulphorodamine B) assay was performed to evaluate the cytotoxicity of cell organelles. After cadmium was treated on Vero cells, we determined IC50 values to examine the detoxification effects of Houttuynia cordata methanol extract and quercetin under cadmium-induced cytotoxicity. Furthermore, morphological changes were observed by the light microscope. In Vero cells, methanol extract of Houttuynia cordata, and quercetin showed inhibitory effects on cadmium-induced cytotoxicity and these detoxification effects were increased in a concentration-dependent manner. These results suggest that methanol extract and quercetin from Houttuynia cordata retain a potential antitoxic activity.

  • Polysaccharide hydrogels constitute a structurally diverse class of biological macromolecules with a wide range of physicochemical properties. They also constitute important members of the family of industrial water-soluble polymers. They find application in Pharmacy as binders, disintegrants, suspending, emulsifying and sustaining agents. According to the International Pharmaceutical Excipients Council (IPEC), an excipient must have an established safety profile. Hence, in the present study, in vitro cytotoxicity on Vero and HEp-2 cell lines, and in vivo acute toxicity in rats were carried out to establish the safety of polysaccharide hydrogels from the seeds of Plantago ovata and Ocimum basilicum. The in vitro cytotoxicity was determined by MTT and SRB assays. In the in vivo acute toxicity, the effects of three different doses of hydrogels (100, 200 and 400 mg/kg body weight) on food and water intake, body weight, biochemical and hematological parameters were studied. The results of in vitro did not show any cytotoxicity on both the cell lines used. In the in vivo acute toxicity, the hydrogels did not show any toxic symptoms in all three dose levels. This establishes the safety of the selected hydrogels. Hence, they can be used as excipients in pharmaceutical dosage forms.

A Study on Cytotoxicity of Dangkwi-Yonghoe-Hwan (당귀용회환의 세포독성(細胞毒性)에 관(關)한 연구(硏究))

  • The purpose of this research was to investigate cytotoxicity of DangkwiYonghoe-Hwan(DYH) and the constitutive crude drugs on several cancer cell-lines, thymocytes, splenocytes and 3T3 cells. The DYH consists of Coptidis Rhizoma, Scutellariae Radix, Phellodendri Cortex, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus, Saussureae Radix and Angelicae Gigantis Radix. The cytotoxicity was determined by MTT method. The DYH inhibited the proliferation of MOLT-4, K562, HL-60, Jurkat, L1210, P815, S180 and Yac-1, thymocyte, splenocyte and 3T3 cells. The cytotoxicity of Coptidis Rhizoma on the cancer cell-lines was the most potent in the constitutive crude drugs. The proliferation of cancer cell-lines was partly inhibition and partly increase by the treatment of Scutellariae Radix, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus and Angelicae Gigantis Radix. Phellodendri Cortex and Saussureae Radix had a poor cytotoxicity on cancer cell-lines. Coptidis Rhizoma and Phellodendri Cortex inhibited the proliferation of thymocyte, splenocyte and 3T3 cells.

Constituents Released from Streptococcus mutans Attenuate Arecoline-mediated Cytotoxicity in HGF Cells by Altering Intracellular $Ca^{2+}$ Signaling

  • Streptococcus mutans (S. mutans) is a facultative anaerobic bacterium mainly found in the oral cavity and is known to contribute to tooth decay and gingivitis. Recent studies on intestinal microbiota have revealed that microorganisms forming a biofilm play important roles in maintaining tissue homeostasis through their own metabolism. However, the physiological roles of oral microorganisms such as S. mutans are still unclear. In our current study, we identified that constituents released from S. mutans (CR) reduce arecoline-mediated cytotoxicity without producing toxic effects themselves. Arecoline, as a major alkaloid of areca nut, is known to mediate cytotoxicity on oral epithelial cells and induces a sustained intracellular $Ca^{2+}$ ($[Ca^{2+}]_i$) increase that is cytotoxic. The exposure of human gingival fibroblast (HGF) cells to CR not only inhibited the sustained $[Ca^{2+}]_i$ increase but also the initial $[Ca^{2+}]_i$ elevation. In contrast, CR had no effects on the gene regulation mediated by arecoline. These results demonstrate that S. mutans has physiological role in reducing cytotoxicity in HGF cells and may be considered a novel pharmaceutical candidate.

Effect of Antioxidant of Citri Reticulatae Pericarpium on Cytotoxicity of Oxygen Free Radicals in Cultured NIH3T3 Fibroblast (배양섬유모세포에서 산소유리기의 세포독성에 대한 진피의 항산화효과에 관한 연구)

  • Objectives : It is demonstrated that oxygen free radicals have cytotoxic effect on NIH3T3 fibroblast cells. Recently, many of herb extracts have an effect of antioxidant in oxygen free radical-induced cytotoxicity. But, the toxic mechanism of oxygen free radical is left unknown. The purpose of this study was to examine the cytotoxicity of hydrogen peroxide ($H_2O_2$) and antioxidant effect of Citri reticulatae pericarpium (CRP) on NIH3T3 fibroblasts. Methods : The cytotoxicy was measured by cell viability by XTT assay in NIH3T3 fibroblasts. XTT assay is regarded as a very sensitive screening method for the determination of the cell viability on various chemicals. Results : In this study, H2O2 decreased cell viability according to the dose- and time dependent manners after NIH3T3 fibroblasts were treated with various concentrations of H2O2 for 4 hours. And also, CRP showed the effect of antioxidant on $H_2O_2-induced $ cytotoxicity in cultured NIH3T3 fibroblasts. Conclusion : These results suggest that $H_2O_2$ has highly cytotoxic effect on cultured NIH3T3 fibroblasts by the decrease of cell viavility, and the herb extract such as CRP was showed the effect of antioxidant on $H_2O_2-induced$ cytotoxicity in these cultures.

Inhibitory effect of Se-Yeon-Eum on nicotine- and cigarette smoke extract induced cytotoxicity in human lung fibroblast

  • There are over 4,000 different chemicals in cigarette smoke, including nicotine and tar. These compounds influence on lung tissue directly or indirectly. In this study, we have examined whether an aqueous extract of Se-Yeon-Eum (SYE), composed of Oriental medicine that has been known to be effective to symptom by smoking, inhibits nicotine- or cigarette smoke extract (CSE)-induced cytotoxicity in human embryonic lung fibroblast, MRC-9. Assessment of cell viability using 3-(4,5-dimethythiazol-2-yl)-2,5-diphenyltetrazolium bromide colorimetric assay indicated that SYE inhibited not only nicotine-induced cytotoxicity but also CSE-induced cytotoxicity. These results suggest the possibility that the use of SYE may be useful for improvement of many symptoms by smoking.

Protective Effect of Omega-3 of Polyunsaturated Fatty Acid Docosahexaenoic Acid on the Organic Mercury-Induced Cytotoxicity in Cultured NIH3T3 Fibroblasts

  • To clerify the protective effect of omega-3 of polyunsaturated fatty acid docosahexaenoic acid (DHA) on the cytotoxicity induced by organic mercury in cultured NIH3T3 fibroblasts. The measurement of cell viability on ogranic mercury wad done by XTT assay after NIH3T3 fibroblasts were cultured with various concentrations of methyl mercuric chloride (MMC). And also, the effect of DHA on the MMC-mediated cytotoxicity was examined by cell viability, and antioxidant effect of DHA was also assessed by superoxide dismutase (SOD)-like activity and the lipid peroxidation activity in cultured NIH3T3 fibroblasts. In this study, MMC decreased cell viability and $XTT_{50}$ value was determined at $50{\mu}M$ of MMC in these culture. In the effect of DHA against the cytotoxicity induced by MMC, DHA significantly increased the cell viability damaged by MMC in cultured NIH3T3 fibroblasts. And also, DHA showed the antioxidant effect by showing the increase of SOD-like activity and the decrease of lipid peroxidation activity. From these results, it is suggested that organic mercury such as MMC has highly toxic effect on cultured NIH3T3 fibroblasts, and also, omega-3 of polyunsaturated fatty acid, DHA showed the protection on MMC-induced cytotoxicity and antioxidant effect.

Effect of Kaempferol on the Cytotoxicity Induced Oxygen Free Radicals in Skin Fibroblast Derived from Human In Vitro

  • In order to evaluate on the effect of kaempferol on the cytotoxicity of oxygen tree radicals, XTT assay was performed to determine the cell viability after skin fibroblasts derived from human (Detroit 51) that were treated with various concentrations of hydrogen peroxide $(H_2O_2)$. And also, the effect of kaempferol on the cytotoxicity induced by H202 that was examined by cell viability, lactate dehydrogenase (LDH) activity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. $H_2O_2$ decreased cell viability in dose-dependent manner in these cultures and the $XTT_{90}\;and\;XTT_{50}$ values were determined at concentration of $35{\mu}M\;and\;90{\mu}M$ of $H_2O_2$ after skin fibroblasts derived from human were treated with $15{\sim}90{\mu}M$ of $H_2O_2$ for 6 hours, respectively. $H_2O_2$ was highly toxic on cultured skin fibroblasts derived from human by toxic criteria of Brenfreund and Puerner (1984). In the protective effect of kaempferol on $H_2O_2$-induced cytotoxicity, kaempferol increased DPPH radical scavenging activity and significantly decreased LDH activity. From these results, it is suggested that oxygen tree radical, $H_2O_2$, was highly toxic on cultured skin fibroblasts derived from human, and also kaempferol of flavonoid showed the protection on $H_2O_2$-induced cytotoxicity.

Synthesis and In Vitro Cytotoxicity of Cinnamaldehydes to Hyman Solid Tumor Cells

  • Cinnamaldehydes and related compounds were synthesized from various cinnamic acids based on the $2^{I}$-hydroxycinnamaidehyde isolated from the bark of Cinnamomum cassia Blume. The cytotoxicity to human solid tumor cells such as A549, SK-OV-3, SK-MEL-2, XF498 and HCT15 were measured. Cinnamic acid, cinnamates and cinnamyl alcohols did not show any cytotoxicity against the human tumor cells. Cinnamaldehydes and realted compounds were resistant to A549 cell line up to 15 .mu.g/ml. In contrast, HCT15 and SK-MEL-2 cells were much sensitive to these cinnamaidehyde analogues which showed $ED{50} values 0.63-8.1{\mu}g/ml.$Cytotoxicity of the saturated aldehydes was much weak compared to their unsaturated aldehydes. From these studies, it was found that the key functional group of the cinnamaldehyde-related compounds in the antitumor activity is the propenal group.p.

  • Water-soluble phosphate glasses containing Ag and Cu ion were prepared through melting process. Dis-solution characteristics, bactericidal effect and cytotoxicity were investigated with composition and time in D. I. water using the powdered sample. Surface change were observed with increasing dissolution time us-ing the bulk specimen. Dissolution amount was maximum at the molar Ag:Cu ratio of 1 and increased with increasing time. The behavior of dissolution was total dissolution from the early stage. But no new layer was observed at the glass surface during dissolution. The bactericidal effect for pseudomonas sp, e. coli, sta-phylococcus aureus, and salmonella increased with increasing dissolution amount and therefore great bac-tericidal effect appeared. The result of cytotoxicity experiment to L929 showed that solution with more than 10 ppm of Ag ion had strong cytotoxicity.
