Isolation and Characterization of 3,4-Dichloroaniline Degrading Bacteria from a Sandbank (갯벌에서 분리한 3,4-Dichloroaniline 분해 미생물의 특성)

  • The compound 3,4-dichloroaniline (DCA) is an aromatic amine used as an intermediate product in the synthesis of herbicides, azo-dyes and harmaceuticals. It is also a degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as active agent in the cosmetic industry. 3,4-DCA, however, is considered potential pollutants due to their toxic and recalcitrant properties to humans and other species. A bacterium capable of growth on 3,4-DCA was isolated by dilution method from 3,4-DCA-containing enrichment culture. Finally, a strain, YM-14, capable of degrading efficiently 3,4-DCA was isolated from a sandbank. The isolated strain, YM-14 was identified to be Arthrobacter sp.. Fifty ppm 3,4-DCA in 1/10 LB media was completely degraded by the growth of Arthrobacter sp. YM-14 for 12 h at $30^{\circ}C$. The isolated strain is capable of growth on 3,4-DCA as sole carbon source and also able to degrade other chloroaniline compounds. Also, the isolated strain showed high level of catechol 1,2-dioxygenase activity by 3,4-DCA exposure. The catechol 1,2-dioxygenase was supposed to be ones of the important factors for 3,4-DCA degradation.

Performance Analysis of the Localization Compensation Algorithm based on Measured Error Patterns of Distance in WPAN (WPAN에서 거리별 측정오차 패턴을 적용한 위치인식 보정 알고리즘의 성능 분석)

  • The performance characteristics and the disadvantages in the compensation algorithm based on the Measured Error Patterns of Distance that is already developed are analyzed, and the localization compensation algorithm(DCA2) based on measured error patterns of distance in WPAN that is the enhanced version of DCA1 is supposed in this paper. From the experimental results, it is confirmed that the localization performance of DCA1 and DCA2 is superior than SDS-TWR as each average above 60% and 75% of the total localizing measurement points in 2 experimental regions, and the localization performance of DCA2 is especially better than SDS-TWR as 91% of the points in $15m{\times}15m$ experimental region. In addition to this, it is confirmed that DCA2 is better than DCA1 as each 16% and 22% of the total localizing measurement points in $10m{\times}10m$ and $15m{\times}15m$ scaled experimental regions, and the average localization errors of DCA1 and DCA2 are lower than SDS-TWR to each 7~12% and 20%. Thus, it can be inferred that DCA2 is the best localization algorithm among 3 localization algorithms SDS-TWR, and DCA2.

Oxidative Coupling of Herbicide Propanil and Its Metabolite, DCA(3,4-dichloroaniline) to Humic Monomers (제초제 Propanil 및 그 분해산물인 DCA(3,4-dichloroaniline)와 Humic Monomer들과의 산화적 짝지움반응)

  • The herbicide propanil and its metabolite, DCA were incubated with oxidative catalysts in the presence or absence of humic monomers to evaluate the incorporation of them into humic substances. Propanil and DCA underwent little or no transformation by oxidatve catalysts in the absence of humic monomers. In the presence of humic monomers, the most effective co-substrate for transformation of propanil was syringic acid by laccase and HRP, that of DCA was catechol by laccase and HRP, and protocatechuic acid by birnessite. The transformation of DCA was the highest when it was incubated with catechol at pH 8.0 during 24 hrs by laccase, and with catechol at pH 3.0 during 2 hrs by HRP, and with protocatechuic acid at pH 5.0 during 2 hrs by birnessite. The DCA transformation increased with increasing concentration of humic monomers. The transformation of DCA was increased with about 5 times when it was incubated with lactase and birnessite together than lactase alone, but that of it was not effected when it was incubated with HRP and birnessite together. When DCA was incubated with dissolved organic carbon in the presence of oxidative catalysts, the transformation of it was not increased by laccase and birnessite but increased by HRP.

Removal Characteristics of Cadmium in Micellar Enhanced Ultrafiltration Using DCA (DCA 미셀을 이용한 한외여과에서 카드뮴의 제거특성)

  • The removal characteristics of cadmium in micellar enhanced ultrafiltration (MEUF) were investigated by using an anionic biosurfactant, deoxycholic acid (DCA). The ultrafiltration experiments were performed in a batch-type stirred cell. The ultrafiltration membranes used were Millipore YM1, YM3, YM10, and YM30 which had a difference in molecular weight cut-off. The presence of cadmium ion in DCA solution tends to reduce the critical micell concentration considerably and increase the micell size. The molecular weight cut-off of ultrafiltration membrane had a minimal effect on cadmium removal whereas DCA/Cd ratio had a substantial influence. When DCA/Cd ratio was 3, the removal efficiency of cadmium was more than 99.6%. A relative flux, which is defined as ratio of the flux for a separation trial to that of deionized water run using the same piece of membrane, decreased in the order of YM3 > YM1 > YM10 > YM30.

  • Chloroanilines are widely used in the production of dyes, drugs and herbicides. Chloroanilines, however, are considered potential pollutants due to their toxic and recalcitrant properties to humans and other species. With the increase of necessity of bioremediation, this study was conducted to isolate the chloroanilines-degrading bacteria. A bacterium capable of growth on 3,4-dichloroaniline (DCA) was isolated by the 3,4-DCA-containing enrichment culture. The strain KB35B was identified as Pseudomonas sp. and also able to degrade several chloroanilines. The isolated strain showed high level of catechol 2,3-dioxygenase activity in the presence of 3,4-DCA. The activity of catecho1 2,3-dioxygenase was supposed to be ones of the important factors for 3,4-DCA degradation. The activity toward 4-methykatechol was 60.6% of that of catechol, while the activity toward 3-methylcatechol and 4-chlorocatechol were 27.0 and 13.5%, respectively.

Potentiation of decursinol angelate on pentobarbital-induced sleeping behaviors via the activation of GABAA-ergic systems in rodents

  • Angelicae Gigantis Radix (AGR, Angelica gigas) has been used for a long time as a traditional folk medicine in Korea and oriental countries. Decursinol angelate (DCA) is structurally isomeric decursin, one of the major components of AGR. This study was performed to confirm whether DCA augments pentobarbital-induced sleeping behaviors via the activation of $GABA_A$-ergic systems in animals. Oral administration of DCA (10, 25 and 50 mg/kg) markedly suppressed spontaneous locomotor activity. DCA also prolonged sleeping time, and decreased the sleep latency by pentobarbital (42 mg/kg), in a dose-dependent manner, similar to muscimol, both at the hypnotic (42 mg/kg) and sub-hypnotic (28 mg/kg) dosages. Especially, DCA increased the number of sleeping animals in the sub-hypnotic dosage. DCA (50 mg/kg, p.o.) itself modulated sleep architectures; DCA reduced the counts of sleep/wake cycles. At the same time, DCA increased total sleep time, but not non-rapid eye movement (NREM) and rapid eye movement (REM) sleep. In the molecular experiments. DCA (0.001, 0.01 and $0.1{\mu}g/ml$) increased intracellular Cl- influx level in hypothalamic primary cultured neuronal cells of rats. In addition, DCA increased the protein expression of glutamic acid decarboxylase ($GAD_{65/67}$) and $GABA_A$ receptors subtypes. Taken together, these results suggest that DCA potentiates pentobarbital-induced sleeping behaviors through the activation of $GABA_A$-ergic systems, and can be useful in the treatment of insomnia.

Differential Display Analysis of Gene Expression Induced under DCA Treatment in Rat Liver

  • The expression of genes induced by Dichloroacetate (DCA) treatment was analyzed by mRNA differential display. Purified total RNAs from rat liver treated with saline or DCA (100 mg/100 g b.w.) were reverse transcribed by using a set of oligonucleotide primers. The PCR products were resolved on a denaturing sequencing gel. PCR band representing mRNA expressed specifically in DCA-treated liver was excised and reamplified by PCR. A 120-bp c-DNA clone named IC1 was isolated and the DNA sequence of IC1 was analyzed. IC1 revealed 50% homology with 3' end of a mouse fibroblast growth factor mRNA This result indicates that DCA induces the expression of a gene which has a 50% homology with a Mouse fibroblast growth factor, and expression of this gene might be involved in non genotoxic process caused by DCA.

  • In this paper, we proposed a Dynamic Channel Assignment (DCA) scheme called Graph Coloring based DCA (GC-DCA) to improve system performance for downlink femtocell networks with high density femto Access Point (AP) deployments. The proposed scheme consists of two steps: one is a femto AP grouping step considering interference and the other is a DCA step considering Signal to Interference plus Noise Ratio (SINR) for femto User Equipments (UEs). Simulation results show that the proposed GC-DCA outperforms other schemes in terms of the mean femto UE capacity and probability of femto UEs which have capacities less than a given transmit rate.

  • Purpose : Patient immobilization is crucial factor for radiation therapy. Generally, we have been used vacuum bag immobilization device(Vac-Lock) for whole body immobilization. In order to easily set up of vacuum bag(Vac-Lock), we made a 3DCA(3-Dimensional Conformal Accessary). The purpose of this study is evaluation of effectiveness of 3DCA using Vack-Lock for patient immobilization. Materials and Methods : We made 3DCA(3-Dimensional Conformal Accessary) tool of wooden boards. The reasons to choice of wooden boards are its easily handling nature and cheap expenses. Results : (1) We reduced man power from $5{\sim}6$persons to 1person to make immobilizations, (2) Shortened work time from 1hour to within 10minutes. (3) Avoid a collision to treatment gantry head. (4) Its shapes are smart and clean. Conclusion : We have made and used 3DCA(3-Dimensional Conformal Accessary) tool was very effective and convenience for the patients and users.

Dichloroacetate Inhibits the Proliferation of a Human Anaplastic Thyroid Cancer Cell Line via a p53-independent Pathway (Dichloroacetate의 p53 비의존적 경로를 통한 인간 역분화 갑상선 암세포주의 성장억제 효과)

  • Occurrence of the Warburg effect in solid tumors causes resistance to cancer chemotherapy, and targeting energy metabolisms such as aerobic glycolysis is a potential strategy for alternative treatment. Dichloroacetate (DCA), an inhibitor of pyruvate dehydrogenase kinase (PDK), shifts glucose metabolism from aerobic glycolysis to oxidative phosphorylation (OxPhos) in many cancers. In this study, we investigated the anticancer effect of DCA on a human anaplastic thyroid cancer (ATC) cell line, 8505C. We found that DCA selectively inhibits cell proliferation of the 8505C line but not of a normal thyroid line. In 8505C, the cell cycle was arrested at the G1/S phase with DCA treatment as a result of decreased antiapoptotic proteins such as $HIF1{\alpha}$, PDK1, and Bcl-2 and increased proapoptotic proteins such as Bax and p21. DCA treatment enhanced the production of reactive oxygen species which consequently induced cell cycle arrest and apoptosis. Interestingly, DCA treatment not only reduced lactate production but also increased the expression of sodium-iodine symporter, indicating that it restores the OxPhos of glucose metabolism and the iodine metabolism of the ATC. Taken together, our findings suggest that PDK inhibitors such as DCA could be useful anticancer drugs for the treatment of ATC and may also be helpful in combination with chemotherapy and radiotherapy.
