Effect of Supplementation with Docosahexaenoic Acid from Gestation to Adulthood on Spatial Learning Performance in Rat (임신기부터 성장기 동안 Docosahexaenoic Acid 보충에 의한 흰쥐의 공간기억력 개선 효과)

  • The effect of supplementation with docosahexaenoic acid into n-3 fatty acid deficient diet on improvement of loaming related brain function was investigated. On the second day after conception, Sprague Dawley strain dams were subjected to a diet containing either n-3 fatty acid deficient (Def) or n-3 fatty acid deficient + docosahexaenoic acid (Def+DHA). After weaning, male pups were fed on the same diet of their respective dams until adulthood. Motor activity and Morris water maze tests were measured at 10 weeks old. In motor activity test, there were no statistically significant differences in moving time and moving distance between the Def and Def+DHA diet groups. The n-3 fatty acid deficient with DHA (Def+DHA) group exhibited a shorter escape latency, swimming time and swimming distance (P<0.05) compared to the n-3 fatty acid deficient group (Def) but there was no difference in resting time and swimming speed between the experimental diet groups. In memory retention trial, the number of crossing of the platform position (region A) was significantly greater than those of other regions for the Def+DHA group. However, the Def group swam randomly without preference for the provisions platform location, indicating poorer memory retention. From those results, supplementation with DHA into the n-3 fatty acid deficient diet improved the spatial loaming ability in rats as assessed by Morris water maze test.

  • RNA interference (RNAi)-mediated transcriptional knock-down of Crassostrea gigas big defensin 1 and 2 genes (Cg-BigDef1 and Cg-BigDef2) was investigated. The cDNA sequences of Cg-BigDef1 and Cg-BigDef2 were identical, excluding an additional fragment of 20 nucleotides in Cg-BigDef1; thus, a long double-stranded RNA (dsRNA) targeting the mRNA of Cg-BigDef2 effectively downregulated both Cg-BigDef2 and Cg-BigDef1. In addition, long dsRNA targeting green fluorescent protein (GFP) did not affect transcription of the two big defensin genes. These results suggest that the transcriptional downregulation of Cg-BigDef1 and Cg-BigDef2 was mediated by sequence-specific RNA interference (RNAi). Despite injection of long dsRNA targeting Cg-BigDef2 into only the adductor muscle, knock-down of Cg-BigDef1 and Cg-BigDef2 was observed in the adductor muscle, hemocytes, mantle, and gills, suggestive of systemic spread of RNAi in C. gigas. Furthermore, the inhibitory effect of dsRNA persisted until 72 h post-injection, indicative of a long-lasting RNAi-mediated knock-down of target genes.

Effect of Treatment with Docosahexaenoic Acid into N-3 Fatty Acid Deficient and Adequate Diets on Rat Brain and Liver Fatty Acid Composition (필수 지방산 조성이 다른 식이의 docosahexaenoic acid 투여가 흰쥐 뇌 및 간의 지방산 조성에 미치는 영향)

  • Previous studies have suggested that docosahexaenoic acid (DHA) supplementation into n-3 fatty acid deficient diet improved spatial learning performance, but there was no significant difference in brain related function when DHA was added into a n-3 fatty acid adequate diet. Here, we investigated the effect of adding DHA into an n-3 fatty acid deficient or adequate diet on brain and liver fatty acid composition. On the second day after conception, Sprague Dawley strain dams were divided into four groups as follows; n-3 fatty acid deficient (Def), n-3 fatty acid deficient plus DHA (Def+DHA, 10.2% DHA), n-3 fatty acid adequate (Adq, 3.4% linolenic acid), and n-3 fatty acid adequate plus DHA (Adq+DHA, 3.31% linolenic acid plus 9.65% DHA). After weaning, male pups were fed on the same diets of their respective dams until adulthood. In brain fatty acid composition, the Def group showed a lower brain DHA (64% decrease), which was largely compensated for by an increase in docosapentaenoic acid (22:5n-6). Brain DHA in the Def+DHA group was increased to almost the same extent as in the Adq and Adq+DHA groups and there were no significant differences among them. Liver fatty acid composition showed a similar pattern to that of the brain, but liver DHA in the Def+DHA showed the highest percentage among the diet groups. In conclusion, n-3 fatty acid deficiency from gestation to adulthood leads to decreased brain DHA, which has been shown to be highly associated with poor spatial leaning performance. Thus, adequate brain DHA levels are required for optimal nervous function.

  • The purpose of this study was to clarify the relationship between dental caries status, and the content of fluoride in drinking water, and the content of fluoride in enamel of deciduous teeth of Korean children. The specific areas selected in this study were Asan-Kun and Seouchon-Kun (Chung Cheoung Nam-Do) where the fluoride content in the deep well water were 0.9 ppm and 0.4 ppm respectively. The dental caries status of 720 children aged 6 to 11 who have been lived since birth was detected. The following results were obtained upon this study: 1. The def person rate, deft rate and def index of the Asan-Kun children were revealed 46.11%, 14.41% and 1. 69 respectively. The fluoride content in the outer layer of enamel was 430 ppm. 2. The def person rate, deft rate and def index of the Seouchon-Kun children were revealed 59.17%, 20.97% and 2.40 respectively. The fluoride content in the outer layer of enamel was 391 ppm. 3. The def person rate, deft rate and def index of the Asan-Kun children were showed lower value than the Seouchon-Kun children. The fluoride content of the outer layer of enamel of the Asna-Kun children were showed higher value than the Seouchon-Kun children.

Signal Detection Based on a Decreasing Exponential Function in Alpha-Stable Distributed Noise

  • Signal detection in symmetric alpha-stable ($S{\alpha}S$) distributed noise is a challenging problem. This paper proposes a detector based on a decreasing exponential function (DEF). The DEF detector can effectively suppress the impulsive noise and achieve good performance in the presence of $S{\alpha}S$ noise. The analytical expressions of the detection and false alarm probabilities of the DEF detector are derived, and the parameter optimization for the detector is discussed. A performance analysis shows that the DEF detector has much lower computational complexity than the Gaussian kernelized energy detector (GKED), and it performs better than the latter in $S{\alpha}S$ noise with small characteristic exponent values. In addition, the DEF detector outperforms the fractional lower order moment (FLOM)-based detector in $S{\alpha}S$ noise for most characteristic exponent values with the same order of magnitude of computational complexity.

Fused Polypeptide with DEF Induces Apoptosis of Lung Adenocarcinoma Cells

  • To analyze the effects of a new unknown peptide DEF on the growth of tumor cells, a fused polypeptide TAT-DV1-DEF was designed and synthesized. The lung adenocarcinoma cell line GLC-82 treated with TAT-DV1-DEF was analyzed with a cell counting kit 8, and the location of polypeptides in cells was observed under laser confocal microscopy. The efficiency of polypeptide transfection and changes in nuclear morphology were analyzed by flow cytometry and fluorescence microscopy, respectively. Finally, the mechanism of tumor cell growth inhibition was evaluated by Western blotting. We found that TAT-DV1-DEF could significantly inhibit the growth of the lung adenocarcinoma cell line GLC-82, but not the normal human embryonic kidney cell line HEK-293. Polypeptides were found to be mostly localized in the cytoplasm and some mitochondria. The efficiency of polypeptide transfection in the two cell types was approximately 99%. Apoptotic nuclei were observed under fluorescence microscopy upon treatment with polypeptides and DAPI staining. Western blot analyses indicated that the polypeptide inhibition of tumor cell growth was apoptosis dependent. In the present study, we demonstrated that fused polypeptides could induce apoptosis of the lung adenocarcinoma cell line GLC-82, indicating that the new unknown peptide DEF has antitumor effects.

  • The Spray-ICP technique uses the ICP(Inductively Coupled Plasma) of ultra-high temperature which is produced by r.f power. The ICP is well-kwown as a clean heat source for the preparation of pure ceramic particles because the ICP is a electrodeless-thermal plasma without contamination. In this study,{{{{ { SnO}_{2 } }}}} particles were sythesized from metal salt solution by Spray-ICP technique. The effects of concentration of solution, collecting location of powders were investicated. The prepared {{{{ { SnO}_{2 } }}}} particles from each concentration of solution had same crystalline phase(tetragonal {{{{ { SnO}_{2 } }}}}) a nd the mean size decreased in proportion to the increase of solution concentration. Each {{{{ { SnO}_{2 } }}}} p owders collector in reactor and electrostatic collector had same crystalline phase and morphologies. The mean size of {{{{ { SnO}_{2 } }}}} p articles prepared by Spray-ICP technique was below 30nm.

  • In the first paper of this series, [2], we investigated how the conformal change enforces the connections and gave the complete relations between connections in Einstein's $^*g^{{\lambda}{\nu}}$-unified field theory. In the current paper we wish to investigate how the vector def $$S_{{\lambda}{{\mu}}{{^\mu}}{=^{def}}S_{\lambda}$$ is transformed by the conformal change. This topics will be studied for all classes and all possible indices of inertia.

  • This paper presents the new algorithm for fault analysis and the fault analysis package for executing this algorithm. This algorithm obtains requisite term for fault analysis by the two-port network technique. Therefore, the fault calculation time is minimized because ${Y_{BUS}}^{-1}$ calculation time is removed. And, the graphic user environment for fault analysis is implemented in mouse-oriented user interface with window and pull-down menu. Therefore, this package can be a useful tool for fault analysis.

Synthesis and Properties of PCPP-Based Conjugated Polymers Containing Pendant Carbazole Units for LEDs

  • New poly(cyclopenta[def]phenanthrene) (PCPP)-based conjugated copolymers, containing carbazole units as pendants, were prepared as the electroluminescent (EL) layer in light-emitting diodes (LEDs) to show that most of them have higher maximum brightness and EL efficiency. The prepared polymers, Poly(2,6-(4-(6-(Ncarbazolyl)- hexyl)-4-octyl-4H-cyclopenta[def]phenanthrene)) (CzPCPP10) and Poly(2,6-(4-(6-(N-carbazolyl)- hexyl)-4-octyl-4H-cyclopenta[def]phenanthrene))-co-(2,6-(4,4-dioctyl-4H-cyclopenta[def]phenanthrene)) (CzPCPP7 and CzPCPP5), were soluble in common organic solvents and used as the EL layer in light-emitting diodes (LEDs) of configuration with ITO/PEDOT/polymer/Ca/Al device. The polymers are thermally stable with glass transition temperature (Tg) at 77-100 °C and decomposition temperature (Td) at 423-457 °C. The studies of cyclic voltammetry indicated same HOME levels in all polymers, although the ratios of carbazole units are different. In case of PLEDs with configuration of ITO/PEDOT/CzPCPPs/Ca/Al device, The EL maximum peaks were around 450 nm, which the turn-on voltages were about 6.0-6.5 V. The maximum luminescence of PLEDs using CzPCPP10 was over 4400 cd/m2 at 6.5 V, which all of the maximum EL efficiency were 0.12 cd/A. The CIE coordinates of the EL spectrum of PLEDs using CzPCPP10 was (0.18, 0.08), which are quite close to that of the standard blue (0.14, 0.08) of NTSC.
