Genetic Diversity among Dioscorea, spp. Using Molecular Markers (분자표지를 이용한 마(Dioscorea spp.)의 다양성 분석)

  • A lot of clones of the genus Dioscorea species have been introduced from some tropical and subtropical regions since 1997. The aim of this study was to determine characteristics of the production process in yams(Dioscorea spp). By utilizing 51clones of water yams(Dioscorea spp), some morphological characteristics were investigated at the field. Intraspecific genetic relationship of 51 variation types of the Yam classified by their external morphological characteristics such as leaf and tuber shape were assessed by DNA using random and specific primers. D. Alata and D. opposita were showed different relationship between yield and the growth of their aboveground parts. This suggests that even in the same species there were differences in yield volume and translocation of assimilation products, depending on the types. D. alata were distinguished from others Dioscorea species at 62% level in AFLP analysis. Also in principal component analysis, D. alata were showed the class from II to V.

Physicochemical Composition and Antioxidant Activities of Korean Dioscorea Species (국내산 마 품종별 이화학적 성분 및 항산화 활성)

  • The objective of this study was to determine the proximate composition and antioxidant activities of Dioscorea spp. Samples were divided into Dunggeunma (D. bulbifra L), Danma (D. polystachya Turcz), Jangma (D. batatas Decaisne) and Danpungma (D. quinqueloba Thunb). The samples were extracted with 70% ethanol and analyzed for total flavonoid contents, polyphenol contents, DPPH radical scavenging activity, and ABTS radical scavenging activity. The moisture contents ranged from 11.96% to 15.01%. The crude protein contents were in the range of 7.86% to 11.55% (Dunggeunma 11.55%, Danpungma 10.48%, Danma 9.43%, Jangma 7.86%). The crude fat contents were between 0.37% and 0.43% (Danpungma 0.43%, Jangma 0.37%, Dunggeunma 0.37%, Danma 0.37%). In the antioxidant activities assay, total polyphenol content of Danma was significantly higher than those of other species at 115.47 mg/100 g gallic acid equivalent (GAE) (p<0.05), whereas it was significantly lowest in Jangma at 66.31 mg/100 g gallic acid equivalent (GAE) (p<0.05). ABTS and DPPH radical scavenging activities of Danma were significantly highest at 104.43 and 56.72 mg trolox eq/100 g (p<0.05). Therefore, antioxidant activities of Dioscorea spp. provide a possible scientific basis for assessing the performance of functional material of Dioscorea spp.

Cryopreservation of zygotic embryos of wild yams(Dioscorea spp.) in Korea

  • A simplified technique that cryoprotects zygotic embryos by desiccation was developed for germplasm conservation of wild yam species(Dioscorea spp.) in Korea. Comparative studies with three other cryogenic techniques were conducted. The maximum survival of zygotic embryos were achieved at a frequency of 96.6% when embryos were cryopreserved by the desiccation method. For the successful cryopreservation of yam zygotic embryos, those that were excised from immature/mature seeds were dried in the air stream of a laminar flow cabinet for 30 min at room temperature and then directly immersed in liquid nitrogen.

  • Detailed morphological descriptions and illustrations of the last instar larva of $Lilioceris$ ($Lilioceris$) ruficollis (Baly, 1865) with the host species, $Dioscorea$ spp., are presented for the first time. Taxonomical and ecological remarks on their biology are also given herein.

The Quality and Potential of DHEA Formation after the Addition of Diosgenin of Yam(Dioscorea spp.) during the Fermentation of Soybean Paste (Diosgenin을 함유한 마(Dioscorea spp.) 첨가 전통된장의 발효과정 중 품질변화 및 DHEA의 생성)

  • The principal objective of this study was to determine the quality and potential of DHEA formation after the addition of diosgenin of yam(Dioscorea spp.) during the fermentation of soybean paste. All samples were divided into three groups based on the variety of yam. In sample A, the soy source was not separated from soybean paste and in sample B, the soybean source was separated from soybean paste. The range in salt concentration in the samples was as follows : sample A: 10.5~11.8%, samples B: 7.9~8.8%. However, no substantial differences between the two samples, in terms of salt content, were observed. The crude protein and crude lipid content were found to decrease with the fermentation time of soybean paste(passage of 30 days: 26.4~32.3% and 4.45~5.90%, passage of 60 days: 24.5~28.5% and 3.95~4.90%, respectively). As the fermentation time of soybean paste increased, the nitrogen from amino compounds increased(passage of 30 days: 531.9~687.7 mg%, passage of 60 days: 551.3~706.8 mg%). In regards to color and preference, sample B, where the soy source was separated from soybean paste, was better than sample A, where the two components were not separated. After a soybean paste fermentation time of 60 days, the diosgenin of yam, which was added to the soybean paste, was not detected in tested samples, and the contents of DHEA formation in soybean paste were as follows: sample A: $488.0\pm42.5{\sim}719.3\pm44.8{\mu}g/g$, sample B: $287.3\pm20.6{\sim}482.7{\pm}36.5{\mu}g/g$.
