Cardiovascular and neurodegenerative diseases are major health threats in many developed countries. Recently, target tissues derived from human embryonic stem (hES) cells and induced pluripotent stem cells (iPSCs), such as cardiomyocytes (CMs) or neurons, have been actively mobilized for drug screening. Knowledge of drug toxicity and efficacy obtained using stem cell-derived tissues could parallel that obtained from human trials. Furthermore, iPSC disease models could be advantageous in the development of personalized medicine in various parts of disease sectors. To obtain the maximum benefit from iPSCs in disease modeling, researchers are now focusing on aging, maturation, and metabolism to recapitulate the pathological features seen in patients. Compared to pediatric disease modeling, adult-onset disease modeling with iPSCs requires proper maturation for full manifestation of pathological features. Herein, the success of iPSC technology, focusing on patient-specific drug treatment, maturation-based disease modeling, and alternative approaches to compensate for the current limitations of patient iPSC modeling, will be further discussed. Heart disease is one of the major life-threatening diseases with high mortality and incidence worldwide. Several model systems, such as primary cells and animals, have been used to understand heart diseases and establish appropriate treatments. However, they have limitations in accuracy and reproducibility in recapitulating disease pathophysiology and evaluating drug responses. In recent years, three-dimensional (3D) cardiac tissue models produced using tissue engineering technology and human cells have outperformed conventional models. In particular, the integration of cell reprogramming techniques with bioengineering platforms (e.g., microfluidics, scaffolds, bioprinting, and biophysical stimuli) has facilitated the development of heart-on-a-chip, cardiac spheroid/organoid, and engineered heart tissue (EHT) to recapitulate the structural and functional features of the native human heart. These cardiac models have improved heart disease modeling and toxicological evaluation. In this review, we summarize the cell types for the fabrication of cardiac tissue models, introduce diverse 3D human cardiac tissue models, and discuss the strategies to enhance their complexity and maturity. Finally, recent studies in the modeling of various heart diseases are reviewed.

As the aging society continues in recent years, interest in dementia is increasing. Among them, Alzheimer's disease is a degenerative brain disease that accounts for the largest percentage of all dementia patients, with the medical community currently not offering clear prevention and treatment for Alzheimer's disease, and the importance of early treatment and early prevention is emphasized. In this paper, we intend to find the most efficient activation function by combining various activation functions centering on convolutional neural networks using MRI datasets of normal people and patients with Alzheimer's disease. In addition, it is intended to be used as a dementia classification modeling suitable for the medical field in the future through Alzheimer's dementia classification modeling.
Kidney disease affects a significant portion of the global population, yet effective therapies are lacking despite advancements in identifying genetic causes. This limitation can be attributed to the absence of adequate in vitro models that accurately mimic human kidney disease, hindering targeted therapeutic development. However, the emergence of human induced pluripotent stem cells (PSCs) and the development of organoids using them have opened up a way to model kidney development and disease in humans, as well as validate the effects of new drugs. To fully leverage their capabilities in these fields, it is crucial for kidney organoids to closely resemble the structure and functionality of adult human kidneys. In this review, we aim to discuss the potential of using human PSCs or adult kidney stem cell-derived kidney organoids to model genetic kidney disease and renal cancer.
Objectives: The purpose of this study was to analyze research trends in dental hygiene using topic modeling and semantic network analysis. Methods: A total of 261 published studies were collected 686 key words from the Research Information Sharing Service (RISS) by 2019-2021. Topic modeling and semantic network analysis were performed using Textom. Results: The most frequently and frequency-inverse document frequently key words were 'dental hygienist', 'oral health', 'elderly', 'periodontal disease', 'dental hygiene'. N-gram of key words show that 'dental hygienist-emotional labor', 'dental hygienist-elderly', 'dental hygienist-job performance', 'oral health-quality of life', 'oral health-periodontal disease' etc. were frequently. Key words with high degree centrality were 'dental hygienist (0.317)', 'oral health (0.239)', 'elderly (0.127)', 'job satisfaction (0.057)', 'dental care (0.049)'. Extracted topics were 5 by topic modeling. Conclusions: Results from the current study could be available to know research trends in dental hygiene and it is necessary to improve more detailed and qualitative analysis in follow-up study.
C-C chemokine receptor type 4 (CCR4) is a chemokine receptor with seven transmembrane helices and it belongs to the GPCR family. It plays an important role in asthma, lung disease, atopic dermatitis, allergic bronchopulmonary aspergillosis, cancer, inflammatory bowel disease, the mosquito-borne tropical diseases, such as dengue fever and allergic rhinitis. Because of its role in wide spectrum of disease processes, CCR4 is considered to be an important drug target. Three dimensional structure of the protein is essential to determine the functions. In the present study homology modeling of human CCR4 was performed based on crystal structure of CCR5 chemokine receptor. The generated models were validated using various parameters. Among the generated homology models the best one is selected based on validation result. The model can be used for performing further docking studies to identifying the critical interacting residues.
The CRISPR system has revolutionized gene editing field. Cas9-mediated gene editing such as Indel induction or HDR enable targeted gene disruption or precise correction of mutation. Moreover, CRISPR-based new editing tools have been developed such as base editors. In this review, we focus on gene editing in human pluripotent stem cells, which is principal technique for gene correction therapy and disease modeling. Pluripotent stem cell-specific drug YM155 enabled selection of target gene-edited pluripotent stem cells. Also, we discussed base editing for treatment of congenital retina disease. Adenine base editor delivery as RNP form provide an approach for genetic disease treatment with safe and precise in vivo gene correction.
The concentration effect relationship (pharmacokinetic pharmacodynamic model, PKPD) of drugs used for Parkinson's disease is complex. The benefits and adverse effects of drug treatment have to be considered in terms of short term and long term effects. Acute effects, observed over hours and days, reflect symptomatic benefit while chronic effects, observed over months and years, also reveal influences on the progress of the disease. (omitted)
We develop a mathematical model for the obesity dynamics to investigate the long term obesity trend with the consideration of psychological and social factors due to the increasing prevalence of obesity around the world. Many mathematical models for obesity dynamics adopted the modeling idea of infectious disease and treated overweight and obese people infectious and spreading obesity to normal weight. However, this modeling idea is not proper in obesity modeling because obesity is not an infectious disease. In fact, weight gain and loss are related to social interactions among different weight groups not only in the direction from overweight/obese to normal weight but also the other way around. Thus, we consider these aspects in our model and implement personal weight gain feature, a psychological factor such as body image dissatisfaction, and social interactions such as positive support on weight loss and negative criticism on weight status from various weight groups. We show that the equilibrium point with no normal weight population will be unstable and that an equilibrium point with positive normal weight population should have all other components positive. We conduct computer simulations on Korean demography data with our model and demonstrate the long term obesity trend of Korean male as an example of the use of our model.
Objectives : The purpose of this study was to investigate the depressive change and predictors of chronic illnesses and to examine whether depressive change predictors are different according to chronic disease types. Methods : Data from the 1st to the 10th year of the Korean Welfare Panel of 5,023 adults over 20 years of age who were treated for chronic diseases for more than three months were examined. We assessed demographic, sociological, and socioeconomic factors including depression predictors of chronically ill patients, and analyzed the data using latent growth modeling and multiple group analysis. Results : Depression symptoms of chronically ill patients gradually decreased, showing a linear relationship across time. In addition, multiple group analysis was performed for each type of chronic disease. Conclusions : We described the phenomenon of depression in chronically ill patients, and discussed the practical and policy interventions required to improve depression in chronically ill patients.
