This test process on screening method for the detection of residual antibiotics in milk is simple, economic, sensitive to residual antibiotics and was given approval international organs. Thus, this study was carried out the comparison of Disk assay method and TTC-II method for sensitivity and minimum detectable range of antibiotics in raw milk. The results of this study was summarized as follows ; 1. The number of samples requested for treatment of mastitis was 198 samples. Comparison or analytical results among the methods of TTC-II, disk assay and Delve sp was that TTC-II 37 samples(18.6% ), Disk assay 125samples(63.1%), Delve SP 130 samples(65.7% ) reacted positively. Conformity rate of Delve SP and Disk assay was 70%. 2. Detectable limits of disk assay method in some antibiotics were more sensitive than those of official method(0.05-0.0025ppm in the $\beta$-lactams, 1ppm in two aminoglycoside, 0.2 ppm in one tetracycline, similar in one macrolide) 3. For sensitivity of residual sulfonamides TTC-II was much more sensitive than disk assay. Detectable limits of sulfamethazine and sulfadimethoxine were 30 to 50ppm levels. 4. The best medium preservation period is 1-2 days. 5. Concentration of brome cresol purple related to resistance for B stearothermophilus culture was 24ppm/ml. These results show that disk assay method for screening detection of antibiotics residuces in milk is worthy of use.
The resistance of 10 varieties of cucumber (Cucumis sativus L.) to powdery mildew, caused by Sphaerotheca fusca, was evaluated by a leaf disk assay. Leaf disks (10 mm in diameter) were removed from fully expanded leaves and then placed in petri dishes containing 0.16% water agar amended with benzimidazole. Leaf disks were inoculated by dropping a 10 $\mu$l of conidia suspension. Conidiophore formation of powdery mildew was the greatest at $25^{\circ}C$. The response of the host to powdery mildew, based on the inoculation onto disks of the first leaf, highly correlated with results obtained from harvesting stage of cucumber plants in greenhouse test (r = 0.99$^{**}$). It is indicating that a leaf disk assay may precisely predict the response of cucumber plant to S. fusca.a.
Safflower is called as the 'beneficial flower' because 'it helps human health', and it was introduced as red flower in Tonguibogam due to the red color of floral leaf. From old times, it has been used for the material of cloth and rouge. Recently, polyphenol compound, the main ingredient of safflower, known as anti-aging and anti-oxidizing material in the healthy food industry becomes the emerging hot topic. This study aims to confirm by DDT (Disk Diffusion Test) assay, MTT assay, and NF-${\kappa}$B Luciferase activity inhibition assay in vitro that polyphenol compound, which is the main ingredient of safflower, has the anti-microbial efficacy to inhibit the growth of acne germs that make troubles for the teenagers or middle aged. Also it aims to evaluate its clinical efficacy on the acne skin, utilizing the facial cleansing cosmetic form of soap sample. This study can contribute to take a major step forward to the development of cosmetic soap for acne in the cosmeceutical industry.
The possibility of identification of families of antibacterial agent residues in fish tissue was studied by disk assay using three test organisms, Bacillus subtilis BGA, Micrococcus luteus ATCC 9341, and Bacillus cereus var. mycoides ATCC 11778. In the present method, a simple clean-up procedure was performed to obtain the aqueous solution from homogenized flounder muscle sample(10g) in Mcilvaine buffer. Then, aqueous solution was fractionated into A and B to be used in disk assay by choloroform and Sep-Pak $C_{18}$ cartridge column after being defatted in hexane. The chloroform layer of fraction A was used for the analysis of macrolide antibiotics(ML), sulfa drugs(SA), chloramphenicol(CP), and quinolone antibiotics(QN). Adsorbed materials to Sep-Pak $C_{18}$ of fraction B were also employed for the analysis of penicillins(PC), tetracyclines(TC), and nitrofuran derivatives(NF) Minimun-detectable concentrations by the present method were, $0.1{\mu}g$/g for oxytetracycline, tetracycline, doxycycline, spiramycin and ciprofloxacin, $0.025{\mu}g$/g for erythromycin and ampicillin, $1.0{\mu}g$/g for sodium nifurstyrenate and florfenical, $0.25{\mu}g$/g for sulfamonomethoxie and sulfadimethoxine, $2.5{\mu}g$/g for oxolinic acid and flumequine, and $15{\mu}g$/g for piromidic acid, respectively. Three test organisms showed different sensitivity patterns for each family of antibacterial agent. Sensitivity patterns were B. cereus > B. subtilis > M. luteus for TC and NF, M. luteus > B, subtilis > B. cereus for ML and PC, B. cereus = B. subtilis > M. luteus for CP and QN, and B. subtilis > B. cereus=M. luteus for SA. The present method utilizing these characteristics could be useful as a routine screening test for the determination of family of antibacterial agent residues in fish tissue.

Propionibacterium acnes have been recognized as pus-forming bacteria triggering an inflammation in acne. The present study was conducted to evaluate antimicrobial activities of Dryopteris crassirhizoma Nakai against these etiologic agents of acne vulgaris and application possibility as a cosmetic resource. D. crassirhizoma crude extract and hexane fraction was prepared and its anti-acne effect against Propionibacterium acnes was investigated with minimum inhibitory concentration (MIC) and paper disk diffusion method. The MIC of D. crassirhizoma crude extract and hexane fraction was 0.008 mg/mL and 0.001 mg/mL, respectively. This implies that D. crassirhizoma extract may be an efficient anti-acne ingredient for cosmetics, as a crude extract. The paper disk diffusion assay showed that its anti-acne effect was similar to that of triclosan. The cytotoxic effect of D. crassirhizoma extract was determined by a colorimetric MTT assay using HaCaT cell line and D. crassirhizoma extract exhibited lower cytotoxic effects. Finally, we examine the stability of D. crassirhizoma extract to temperature and pH. The D. crassirhizoma extract was very stable to high temperatures ($25{\sim}121^{\circ}C$) and to wide pH range ($pH\; 2{\sim}11$), suggesting its utilization for cosmetics.

As Sclerotinia sclerotiorum causing cucumber sclerotinia rot was the fastest in the mycelial growth at $25^{\circ}C$, its pathogenicity was strong at the same temperature among several temperatures. All the isolates of Sclerotinia sclerotiorum showed a strong pathogenicity against cucumber fruits, which was confirmed by a disk assay and a wound assay. A wound assay was superior to a disk assay to develop the assay system for assessing the fungicidal activity of several fungicides against Sclerotinia sclerotiorum. In a disk assay, it was very difficult to assess the fungicidal activity, because the pathogenicity of isolates used in the experiment was very strong. At 500 and $3.0{\mu}g/mL$, the activity of dichloflouanid and the mixture of carbendazim and diethofencarb against cucumber sclerotinia rot was 14.3 and 42.3%, respectively, by using a disk assay. However, at same concentration two fungicides showed the high controlling activity as 100 and 92.5%, through a wound assay in a laboratory. Also, the activity of two fungicides was good against cucumber sclerotinia rot in the greenhouse where cucumber plants were cultivated in the field, showing the control value as 91.1 and 82.9% at 100 and $825{\mu}g/mL$, respectively. All the isolates of Sclerotinia sclerotiorum from cucumber fruits sampled in the polyvinyl house were subjected to monitoring for the resistance to 7 fungicides. The $EC_{50}$ value of 7 fungicides was as follows: fenhexamid; $0.13{\mu}g/mL$, procymidon and iprodione; 0.18 and $0.24{\mu}g/mL$, carbendazim and the mixture of carbendazim and diethofencarb; 0.13과 $0.05{\mu}g/mL$, iminoctadine and dichlofluanid; 1.94 and $8.95{\mu}g/mL$. Ultimately it was not found that resistant isolates of Sclerotinia sclerotiorum were appeared in the field.
Transgenic herbicide-resistant sweet potato plants [Ipomoea batatas (L.) Lam.] produced through a biolistic transformation were used in this study. The objective of this research was to find out a rapid and reliable assay method for confirming glufosinate-ammonium resistance. The techniques tested include whole-plant bioassay, one leaf bioassay, and leaf disk bioassay. Parameters investigated in this study were leaf injury and ammonium accumulation at 1 and 5 days after treatment of glufosinate-ammonium. In the leaf disk bioassay, leaf injury of the transgenic line 7171 was 1.9-fold less affected by glufosinate-ammonium than the wild type. The leaf injury of 7171 in one leaf and whole-plant bioassays was 59- and 92-fold less affected by glufosinate-ammonium, respectively, compared with that of the wild type. Leaf disk, one leaf, and whole-plant bioassays showed that ammonium accumulation of the 7171 was 2 to 20-, 4 to 43-, and 6 to 115-fold less affected by 0.5-5 mM glufosinate-ammonium than that of the wild type. All three bioassays successfully distinguished the resistance from the transgenic lines, but one leaf bioassay is the simplest and quickest. Leaf injury and ammonium accumulation were the same in leaves 1, 3, 5, 7, and 10 of 3 mM glufosinate-ammonium treated plants or nontreated plants. The one leaf bioassay was chosen as the standard procedure for future confirmation of resistance in transgenic sweet potato because it is a rapid and reliable assay.

Agrimonia pilosa Ledeb. is a perennial plant, which naturally habitats in whole area of Korea, and where it is popularly used for the traditional remedies. In the present study, A. pilosa Ledeb. extract was prepared to determine the anti-acne effects and application possibility as a cosmetic resource. A pilosa Ledeb. was extracted with methanol and its anti-acne effect against Propionibacterium acnes was investigated via minimum inhibitory concentration (MIC) and paper disk diffusion method. The MIC of A. pilosa Ledeb. extract and triclosan was 0.05 mg/mL and 0.04 mg/mL, respectively. This implies that A. pilosa Ledeb. extract nay be an efficient anti-acne ingredient for cosmetics, considering that it is a crude extract. The paper disk diffusion assay showed that its anti-acne effect was similar to that of triclosan. Furthermore, A. pilosa Ledeb. extract effectively inhibited the growth of several aerobic microorganisms including Staphylococcus aureus. Finally, we examine the stability of the extract to temperature and pH. The extract was very stable to high temperatures ($70{\sim}121^{\circ}C$) and to pH ($pH 2{\sim}11$), suggesting its utilization for cosmetics.
