Effect of evaporation-induced osmotic changes in culture media in a dry-type incubator on clinical outcomes in in vitro fertilization-embryo transfer cycles

  • Objective: This study investigated whether adding outer-well medium to inhibit osmotic changes in culture media in a dry-type incubator improved the clinical outcomes of in vitro fertilization-embryo transfer (IVF-ET) cycles. Methods: In culture dishes, the osmotic changes in media (20 µL)-covered oil with or without outer-well medium (humid or dry culture conditions, respectively) were compared after 3 days of incubation in a dry-type incubator. One-step (Origio) and G1/G2 (Vitrolife) media were used. Results: The osmotic changes in the dry culture condition (308 mOsm) were higher than in the humid culture conditions (285-290 mOsm) after 3 days of incubation. In day 3 IVF-ET cycles, although the pregnancy rate did not significantly differ between the dry (46.2%) and humid culture (51.0%) groups, the rates of abortion and ongoing pregnancy were significantly better in the humid culture group (1.5% and 49.5%, respectively) than in the dry culture group (8.3% and 37.8%, respectively, p<0.05). In day 5 IVF-ET cycles, the abortion rate was significantly lower in the humid culture group (2.2%) than in the dry culture group (25.0%, p<0.01), but no statistically significant difference was observed in the rates of clinical and ongoing pregnancy between the dry (50.0% and 25.0%, respectively) and humid culture groups (59.5% and 57.3%, respectively) because of the small number of cycles. Conclusion: Hyperosmotic changes in media occurred in a dry-type incubator by evaporation, although the medium was covered with oil. These osmotic changes were efficiently inhibited by supplementation of outer-well medium, which resulted in improved pregnancy outcomes.

Depositional Characteristics of Atmospheric PCBs in Several Media Used Passive Air Samplers (대기 중 PCBs의 PAS에 이용되는 여러 매체의 PCBs 침착 특성)

  • Objective: This study was carried out to determine the depositional characteristics of pine needles, pine tree bark, moss, and soil, which are used as a passive air sampler (PAS) of atmospheric polychlorinated biphenyls (PCBs). Methods: All four media were sampled from the same site. PCB concentrations were analyzed by GC/MSD, and the lipid contents were measured using the gravimetric method. Results: The total PCB concentration (IUPAC No. 28, 52, 101, 138, 153, 180) was the highest in soil (227.97 pg/g dry), followed by pine needles (71.36 pg/g dry), pine tree bark (44.58 pg/g dry), then moss (21.91 pg/g dry). Pine needles contained the highest lipid contents (21.31 mg/g dry), whereas soil (10.01 mg/g dry), pine tree bark (4.85 mg/g dry), and moss (1.92 mg/g dry) contained less. The concentration of lower-chlorinated PCBs was relatively high in pine needles, pine tree bark and moss, but not in soil. The PCB concentrations were proportional to their lipid contents in pine needles, pine tree bark and moss, but a different trend was revealed in soil. Conclusions: The PCBs concentrations in the media were affected by various factors including atmospheric PCB concentrations, lipid contents, and depositional pathways. However, each of these factors had a varying influence depending on the type of medium.

Effect of Application Rate of Dolomite in Three Media Containing Polyacrylic Acid Sodium Salt on Growth and Nutrient Contents of Potted Chrysanthemum 'Lima Honey' (Polyacrylic Acid Sodium Salt를 혼합한 세 종류 상토에 고토석회의 시비 수준이 포트멈 'Lima Honey'의 생육 및 무기원소 흡수에 미치는 영향)

  • This research was conducted to determine the plant growth and nutrient contents of potted chrysanthemum 'Lima Honey' as influenced by application rate of dolomite in three root media, peatmoss+vermiculite (1:1, v/v), peatmoss+composted rice hall (1:1, v/v), and peatmoss+composted pine bark (1:1, v/v). All root media contained polyacrylic acid sodium salt (PASS) at a rate of $4.5g L^{-1}$. In crop growth at 40 and 80 days after transplanting, elevated application rates of dolomite up to $6.0g L^{-1}$ in three root media increased fresh and dry weights per plant. But the treatment of $9.0g L^{-1}$ had less fresh and dry weights than those of $6.0g L^{-1}$. Tissue phosphorus content decreased and those of Ca and Mg increased by the elevated application rates of dolomite. Elevated application rates of dolomite in three root media resulted in the increase of pH, EC, and the concentrations of K, Ca, and Mg at 43 days after transplanting. Those at 80 days after transplanting were also increased, but the differences among treatments were less significant as compared to those at 43 days after transplanting. From the results of dry weight in potted chrysanthemum 'Lima Honey', the proper application rate of dolomite to three root media containing PASS at a rate of $4.5g L^{-1}$ were $6.0g L^{-1}$.

A Study on the Filter Media and Performance of Intake Air Filter for Vehicular Engine (자동차 흡기 에어필터의 여재 및 성능에 관한 연구)

  • Since the vehicle is regarded as the third living space, the comfortable conditions are required in the passenger compartment. For this reason, customers are concerned about the filtering performances and the importance of filter media has been greatly placed. Therefore the dust holding capacity, the efficiency of these filter media (dry paper, wet paper, non-woven) and the configuration of air filter for vehicle were measured in this study. The following results were obtained on the basis of air filter test. It shows that the thickness and poresize of filter media should be lower for the higher efficiency. The measurement result shows that the performance of round shape filters are higher than the square shape filters. The dust holding capacity of the wet paper and the non-woven paper is higher than the dry paper. As a result, this research can provide an important design parameter and product guidance of the intake air filter for vehicular engine.

Effect of Application Rate of a Slow-release Fertilizer in Three Media Containing Polyacrylic Acid Sodium Salt on Growth and Nutrient Contents of Potted Chrysanthemum 'Lima Honey' (Polyacrylic Acid Sodium Salt를 혼합한 세 종류 상토에 지효성 비료의 시비 수준이 포트-멈 'Lima Honey'의 생육과 무기원소 흡수에 미치는 영향)

  • Objective of this research was to determine the effect of application rate of a slow release fertilizer (SRF) in three root media, peatmoss+vermiculite (1:1, v/v; PV), peatmoss+composted rice hall (1:1, v/v; PR), and peatmoss+composted pine bark (1:1, v/v; PB), on growth and nutrient contents of potted chrysanthemum 'Lima Honey'. All media contained polyacrylic acid sodium salt at a rate of $4.5g L^{-1}$. The fresh and dry weights at 43 days after transplanting did not show statistical differences among treatments in each root media. Elevated application rate of SRF increased fresh and dry weights at 80 days after transplanting in PV and PB media, but not in PR medium. Elevated application rates of SRF resulted in the increase of tissue phosphorus content and decrease of tissue Ca, Na, and Zn contents at both 43 and 80 days after transplanting. Elevated application rates of SRF resulted in the decrease of pH and increase of EC and concentrations of ${NO_3}^-$ and ${P_2O_5}^{3-}$, K, Ca, and Mg in the soil solution of PV and PR media. The trends of those in PR media were also similar except ${NO_3}^-$. The differences among treatments in EC at 80 days after transplanting were less significant as compared to those at 43 days after transplanting in three media.

Depositional Characteristics of Atmospheric PBDEs on Pine Needles, Bark and Soil (대기 중 폴리브롬화디페닐에테르의 소나무 잎, 소나무 껍질 및 토양으로의 침착 특성)

  • Objective: This study was carried out in order to determine the depositional characteristics of pine needles, pine bark, and soil used as a passive air sampler (PAS) for atmospheric polybrominated diphenyl ethers (PBDEs). Methods: All three media were sampled from the same site. The PBDE concentrations were analyzed by HRGC/HRMS, and the lipid contents were measured using the gravimetric method by n-hexane extraction. Results: The total PBDE concentration was the highest in soil (22,274.57 pg/g dry), followed by pine bark (20,266.39 pg/g dry), and then pine needles (7,380.22 pg/g dry). Pine needles contained the highest lipid contents (21.31 mg/g dry), whereas soil (10.01 mg/g dry), and pine bark (4.85 mg/g dry) contained less. There were poor correlations between lipid content and total PBDE concentrations in the media ($R^2$=0.8216, p=0.2814). Congeners BDE 47, 99, 183, 196, 197, 206, 207 and 209 showed peak concentrations. Among these, BDE 206, 207, and 209 are highly brominated PBDEs that exist as particulates in ambient air. They accounted for 81.2% [69.2 (pine needles) - 89.0% (tree bark)] of the concentration and therefore are noted as the main congener of the total PBDEs. Conclusions: It can therefore be concluded that for reducing error by improper sampling, the same species of media should be recommended for use as a PAS for atmospheric PBDEs due to the differences in depositional characteristics.

Study of Methylglyoxal and Phosphorus Stress on Algae (조류의 Methylglyoxal과 인 Stress 연구)

  • Effects of phosphorous (P) and methylglyoxal (MG) on the cell number, dry weight, chlorophyll content, photosynthetic and respiratory rate, phosphate uptake and protein content of green algae (Scenedesrnus obliquus) were studied. The algal cell number from the medium treated with 0.5-1.0 mM of MG at 1/2 P or 1/4 P concentration was significantly lower than those of algae treated :with full strength of phosphrous in medium. The inhibitory effect of MG on algal cell division was enhenced at low concentration of phosphorous in medium. At the beginning of logrithmic phase of algal growth, the mean dry weight of algae from the medium without MG-treatment in 1/2 P media was significantly higher than that of algae treated with MG. After logrithmic phase of growth cycle, the mean dry weight of algae from the medium with 1.0 mM of MG-treatment in 1/4 P media was significantly lower than that of algae treated with or without MG. At logrithmic phase of algal growth, there were significant differences in the chlorophyll content among all groups of tested algae with various concentrations of P and MG. At 15 days after inoculation, the mean chlorophyll content per algal cell from the media without MG-treatment in 1/2P was significantly higher than that of other cells from MG-treated media. The adverse effect of MG at concentration of 0.5-1.0mM in 1/2 and 1/4 P media on photosynthetic rate was observed. The mean photosynthetic rate of algal cell without P and MG treatment at 15 days after inoculation was significantly higher than that of MGtreated algae. After logarithmic phase, the algal cell treated with 0.5mM of MG with full strength of phosphorous showed significantly high respiratory rate than that of other cell groups. There were significant differences in mean phosphate uptake rate among all groups of Scenedesmus obliquus at logarithmic phase. At 12 days after inoculation, phosphate uptake rate per each algal cell from the basic media without MG and P treatment was rapidly reduced which shows early introduction to stationary phase.

Plant-growth promoting traits of bacterial strains isolated from button mushroom (Agaricus bisporus) media

  • A diverse group of plant-growth promoting bacteria were isolated in button mushroom (Agaricus bisporus) media to investigate the plant-growth promoting traits of compounds including indole acetic acid (IAA), ammonia, 1-aminocyclopropane-1-carboxylic acid deaminase, siderophore, and hydrogen cyanide. Twenty-one bacterial strains showing positive effects for all the test traits were selected and classified to confirm bacterial diversity in the media habitat. Plant-growth promoting traits of the isolates were also assessed. All strains produced IAA ranging from 20 ㎍/mL to 250 ㎍/mL. Most of the isolates produced more than 80% siderophore. Four strains (Pantoea sp., PSB-08, Bacillus sp., PSB-13, Pseudomonas sp., PSB-17, and Enterobacter sp., PSB-21) showed outstanding performances for all the tested traits. In a bioassay of these four strains using mung bean plant, the best growth performances (23.16 cm, 22.98 cm, 2.27 g/plant, and 1.83 g/plant for shoot length, root length, shoot dry weight, and root dry weight, respectively) were obtained from the plants co-inoculated with Bacillus sp., PSB-13. The resultant data indicate that button mushroom media have got a diverse group of bacteria with plant growth promoting abilities. Thus, the media could be a good recycling resource for using to an effective bio-fertilizer.

  • In this paper describes the comparisons of the surface flashover characteristics according to the change of the insulation media by experimental GIS(Gas Insulated Switchgear) chamber in accordance with change of pressure(P) and electrode distance(d). The using insulation medias are $SF_6$, Dry-Air, I-Air(Imitation Air, $N_2$ : $O_2$ = 79[%] : 21[%]), $N_2:O_2$ mixture gas and pure $N_2$. In this study, in order to compare the properties $SF_6$ and order insulation gas, we investigated the properties of the various insulation media with a knife to knife electrode under ac high voltage application. The gas pressure was changed from 1 to 5[atm]. as a result, it was found that dielectric strength is $SF_6$ > I-Air > Dry-Air and the best environmental preservation gas is Dry-Air.

Influence of Thickness of Styrofoam-Panel on Tobacco Seedling Growth in Newly Developed Floating System (새로 개발된 부상형 육묘에 있어서 스티로포움 판의 두께가 연초 묘 생육에 미치는 영향)

  • The inluence of styrofoam-panel thickness of floating frame on the seedling growth of flue-cured tobacco(Nicotiana tabacum L.) in the newly developed direct-seed float system was investigated . Floating frame used instead of styrofoam-tray consisted of styrofoam-panel (106×108cm) for floating, eight holes(1.Sx2S.0 cm) to uniform the water, and nutrition supply far seedling and water and nutrition absorption cloth placed on the styrofoam-panel. Each floating-frame may lay eight plastic-trays on it. Eight grades of styrofoam-panel thickness from 26 mm to 46mm and two kinds of media were used in the greenhouse system. Dry cells were found from the styrofoam-panel thickness of 43mm in carbonized chaff, compost and original soil rate of L3-3(v/v) media, and of 37mm in peat and perlite rate of 8-2(v/v) media. The thinner styrofoam-panel produced more tender and succulent seedling with the more trunk and the less root weight. When considering the appearance of dry cell, seedling growth, producing healthy tobacco seedlings, and utilization of styrofoam-panel for two kinds of medias the ideal styrofoam-panel thickness were suggested to be around 34mm for flue-cured tobacco reeling production in the newly developed direct-seeding float system.
