Effect of Rot Water Soluble Extract from Eleutherococcus and Senticosus and Dietary Carnitine on the Lipid Metabolism and Antioxidant Defense System of Rats on Hypercholesterol Diet (가시오가피 열수추출물과 카르니틴 공급이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 항산화계에 미치는 영향)

  • This study investigated the synergy effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system of rats on hypercholesterol diets. Sprague-Dawley male rats were fed either a hypercholesterol diet with water or a hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine. The experimental groups consisted of the control group (CO), the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES), and the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). Eleutherococcus senticosus was extracted, made into 0.5% solution, and fed to the ES and ESC groups. A 3% carnitine diet was supplied to the ESC group. Hypercholesterol diets contained 18% beef tallow and 5% cholesterol. After 4 weeks of administering these diets, serum and liver were obtained and the level of serum lipid and the activities of GOT and GPT were measured. In addition, the level of liver lipid and TBARS and the activity of GSH-Px were measured. The results were as follows: 1) Weight gain and FER in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC) was low significantly(P<0.05). 2) In the groups supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and the group supplied with hot water soluble extract from Eleutherocorcus senticosus and dietarty carnitine(ESC), total serum cholesterol levels were both significantly low (P<0.05,P.0.01). 3) Serum GOT activity was significantly low(P<0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietary carnitine(ESC). There was no difference between serum GPT activity in the control group and the groups supplied with hot water soluble extract from Eleutherococcus senticosus or dietary carnitine. 4) Liver triglyceride was low (P.0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). No difference was observed in other lipid composition, TBARS level, and GSH-px activity in liver between the control group and the experimental groups. In summary, the effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system (such as weight gain, FER, total serum cholesterol and liver triglyceride) were low when Eleutherococcus senticosus and dietarty carnitine were supplied simultaneously.

Vegetation Structure and Ecological Properties of Eleutherococcus senticosus Population (가시오갈피(Eleutherococcus senticosus)개체군의 식생구조와 생태특성)

  • This study was conducted to analyze the correlation between vegetation, environmental factors, and soil characteristic of an endangered species, Eleutherococcus senticosus. Eighteen quadrats in population zones of Seoraksan, Odaesan, Chiaksna, Duckhangsan, Sobaeksan, Juwangsan, and, Jirisan were set up and ordination was analyzed. In the study sites, soil pH, available phosphorous, organic matter and nitrogen were ranged from 5.06~5.43, 24.63~57.77mg/kg, 14.01~18.37%, and 0.63~0.75%, respectively. Eleutherococcus senticosus is classified into Morus bombycis dominant population, Cornus controversa dominant population, and Eleutherococcus senticosus typical population. The importance value of Acer pictum subsp. mono, Morus bombycis, Magnolia sieboldii, Cornus controversa, Tilia taquetii, and Ulmus laciniata in Eleutherococcus senticosus population were 23.76%, 23.61%, 18.06%, 17.82, 15.31%, and 13.57%, respectively. The correlation between vegetation and environmental factors of Eleutherococcus senticosus vegetation distribution were shown to be affected by altitude, slope, and direction.

Effect of Dietary Supplementation of Eleutherococcus Senticosus, Taurine and Carnitine on Endurance Exercise Performance in Rats (가시오갈피, 타우린 및 카르니틴 보충식이가 흰쥐의 지구력운동 수행능력에 미치는 영향)

  • The effects of dietary supplementation of Eleutherococcus senticosus, taurine and carnitine on maximal endurance exercise performance along with other related parameters were evaluated in rats that underwent aerobic exercise training for 6 weeks. Thirty-two male rats (4 weeks old) were randomly divided into 4 groups, and fed experimental diets and/or aerobic exercise trained according to the protocol: SC (sedentary control group), EC (exercise-trained control group), EE (exercise-trained Eleutherococcus senticosus-supplemented group), and EETC (exercise-trained Eleutherococcus senticosus, taurine and carnitine-supplemented group). The food efficiency ratio of EC rats was significantly lower than the value for SC rats (p < 0.01). Exercise-trained control animals (92 $\pm$ 8.8 min) could run significantly longer until exhausted on the treadmill than sedentary control rats (11 $\pm$ 0.8 min) (p < 0.001). Animals fed an Eleutherococcus senticosus-supplemented diet, and an Eleuthherococcus sonticosus, taurine and carnitine- supplemented diet while undergoing aerobic exercise training for 6 weeks exhibited, respectively, 8 and 5 minutes longer running performance until exhausted than the rats fed the control diet. The gastrocnemius muscle glycogen concentration of the rats, measured at 48 hours post maximal exercise performance test, was 43% higher in EC rats than the value for SC rats (p < 0.05), but was not different among EC, EE, and EETC rats. The mitochondrial citrate synthase activity of the soleus muscle was significantly higher in EC rats compared to the value for SC rats (p < 0.01), and showed a tendency to increase, without statistical significance, in EE or EETC rats compared to the value for EC rats. These results indicate that aerobic exercise training for 6 weeks significantly improved maximal exercise performance, muscle glycogen content along with citrate synthase activity, which are important in the energy metabolism of muscle under aerobic exercise. Dietary supplementation of Eleutherococcus senticosus in rats while undergoing aerobic exercise training improved maximal endurance exercise performance without significantly affecting muscle glycogen content and enzyme activities involved in energy metabolism during exercise. Taurine and carnitine supplementation failed to show an additive effect on maximal endurance exercise performance when consumed along with Eleutherococcus senticosus.

Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.

Mass Production of Eleutherococcus senticosus Plants through in vitro Cell Culture (세포 배양을 통한 가시오갈피(Eleutherococcus senticosus) 묘목의 대량 생산 시스템 개발)

  • Immature zygotic embryos of Eleutherococcus senticosus seeds matured rapidly within one month when the seeds comprising zygotic embryos were pieced to small size and cultured on 1/2 MS medium. Frequency of somatic embryos formation was declined rapidly when the zygotic embryos germinated and grew to plantlets. Embryogenic cells were induced by consecutive subculture of somatic embryos on MS medium with 1.0mg/L2,4-D. After heart-shaped somatic embryos were induced by suspension culture, these embryos were plated onto petri dish to support maturation of embryos. Germination of embryos occurred on medium with 5mg/L GA$_3$and transferred to culture bowl to stimulate the further growth. Frequency of soil survival of plantlets was influenced by soil mixture (perlite and peatmoss). The suitable combination of perlite and peatmoss was 1:5, and the soil survival rate was 78% after 4 months. The soil transferred plantlets were over-wintered in field condition after defoliation. New year sprouting of plants was achieved successfully and they grew to adult plants. These results indicate that the systematic procecure of plant production in E. senticosus for micro propagation.

Increasement of Eleutherosides and Antioxidant Activity in Eleutherococcus senticosus Adventitious Root by Jasmonic acid

  • This study was carried out to investigate the impacts of jasmonic acid (JA) on adventitious root culture of Eleutherococcus senticosus. Adventitious root of E. senticosus were treated with jasmonic acid (JA) and cultured for 30 days. JA inhibited the root growth but increased eleutherosides accumulation, total phenolic contents and antioxidant activity. Among various concentrations of JA, 1.0 mg/L JA increased the total phenolic contents in E. senticosus adventitious root to $39.81{\mu}g/g$, about 2.6 times higher than that of the control. Consequently, high accumulation of total phenolic contents led to increase the antioxidant activity to 82.41%. The antioxidant activity of control was 37.89% at $2500{\mu}g/mL$. A linear correlation ($R^2$ = 0.9937, 0.9648 and 0.9883) was also shown between antioxidant activity (at 1250, 1875, and $2500{\mu}g/mL$) and total phenolic contents of adventitious root of E. senticosus.

The Study for Optimal Layout of the Eleutherococcus Senticosus Sap Production Line Analyzed by Simulation Model (시뮬레이션 모델 구축과 분석을 통한 가시오가피 액즙 가공 라인의 최적 배치에 관한 연구)

  • The purpose of this study is basically for the use of simulations to enhance productivity. In this paper, the optimal number of allocation in a small and medium industry which produces eleutherococcus senticosus sap, is performed using simulations. The simulation model was developed under considerations of production layout, process & operation, process time, total work time, work in process (WIP), utilization, failure rate, and operation efficient as inputs, and was validated with careful comparisons between real behaviors and outputs of the production line. Therefore, we can evaluate effects and changes in productivity when some strategies and/or crucial factors are changed. Although too many workers and machines could decrease productivity, the eleutherococcus senticosus sap production line in this paper has been maintained many machines. To solve this problem, we determined the optimal number of workers and machines that could not cause any interrupt in productions using simulations. This simulation model considers diverse input variables which could influence productivity, and it is very useful not only for the production line of Eleutherococcus Senticosus Sap, but also for other production lines with various purposes, especially, in the small and medium industries.

Development and validation of SSR markers in Eleutherococcus senticosus

  • Eleutherococcus senticosus (Siberian ginseng) is an important medicinal tree found in Russian taiga, and northern regions of Korea, Japan, and China. In this study, we analyzed the genome-wide distribution of microsatellites in E. senticosus using developed SSR markers. A total of 711 clones from an SSR-enriched genomic DNA library were sequenced, of which 47 clones (6.6%) were redundant. Of the 664 independent clones, only 12 polymorphic SSR markers were obtained, which also revealed successful amplicons in E. senticosus accessions. Using the developed SSR markers, we estimated genetic diversity and population structure among 131 E. senticosus accessions in Korea and China. The number of alleles ranged from 2 to 11, with an average of 7.4 alleles. The mean values of observed heterozygosity ($H_O$) and expected heterozygosity ($H_E$) were 0.59 and 0.56, respectively. The average polymorphism information content (PIC) was 0.51 in all 131 E. senticosus accessions. E. senticosus accessions in Korea and China showed a close genetic similarity. Significantly low pairwise genetic divergence was observed between the two regions, suggesting a relatively narrow level of genetic basis among E. senticosus accessions. Our results not only provide molecular tools for genetic studies in E. senticosus but are also helpful for conservation and E. senticosus breeding programs.

Standardization of Eleutherococcus species and HPLC Method Validation for Quantitative Analysis (정량분석을 통한 Eleutherococcus species의 HPLC 분석법 검증과 표준화)

  • Objective : For the standardization and quality control of eleutheroside E in Eleutherococcus species, HPLC analysis was performed and eleutherosdie E content was compared in 23 kinds of Eleutherococcus species collected from Korea and China. Methods : The content of eleutheroside E in stem bark of Eleutherococcus species collected from Korea and China were analyzed by HPLC. 0.5% phosphoric acid and acetonitrile was used as mobile solvent. Validation of HPLC analysis method was confirmed by analyzing specificity, linearity, precision and accuracy following ICH guideline. Results : Content of eleutheroside E was determined to be 1.0-1.6% and 0.5-0.8% in Korean and Chinese E. senticosus, respectively. Content of eleutheroside E in E. sessiliflorus was 0.7-1.1% and 0.2-0.4% respectively in Korean and Chinese origin. All calibration curves showed good linear regression. The method showed good precision and accuracy with intra-day and inter-day variations of 0.880-3.442% (RSD) and 0.606-3.328% (RSD), respectively, and average recovery was of 0.141-1.363% (RSD), for the eleutheroside E analyzed. Conclusion : These results might be used to establish a criterion of eleutheroside E in Eleutherococcus species.
