Effect of Polymer Shielding on Elution of G3PDH Bound to Dye-ligand Adsorbent

  • Batch binding experiments were performed to assess the recovery performance of glyceraldehyde 3-phosphate dehydrogenase (G3PDH) bound to the unshielded and polymer (polyvinyl pyrrolidone. PVP)-shielded dye-ligand (Cibacron Blue 3GA) adsorbent. The adoption of a polymer-shielded, dye-ligand technique facilitated the elution efficiency of bound G3PDH. It was demonstrated that the recovery of G3PDH using polymer-shielded dye-ligand adsorption yielded higher elution efficiency, at 60.5% and a specific activity of 42.3 IU/mg, after a low ionic strength elution (0.15 M NaCl). The unshielded dye-ligand yielded lower elution efficiency. at 6.5% and a specific activity of 10.2 IU/mg.

The Adsorption and Elution Characteristics of Copper Ions in Electrochemical Ion Exchange Electrode Fabricated by the Compressed Diecasting (압착성형법으로 제작된 전기화학적 이온교환 전극에서 구리이온의 흡착과 용출특성)

  • Electrochemical Ion Exchang(EIX) electrode containing Amberlite IRP-64 as a cation ion exchange resin and Stylene-Buthylene-Rubber(SBR) as a binder was fabricated by the compressed diecasting method. The adsorption and elution characteristics in copper sulfate solution were investigated at the various electrode potentials and electrolyte pHs. In the adsorption process, it was found that the maximum adsorption rate of copper was obtained at -1800 mV and the ratio of adsorption was 92% during 90 min. In the elution process, the elution rate of copper was increased in proportion to anodic potential in the present experimental range and the ratio of elution was 88% during 50 mins at 3600 mV. The adsorption and elution processes were significantly affected by the variation of local pH in the vicinity of electrochemical ion exchange electrode. The higher performances of adsorption and elution were elution were obtained at basic and acidic eletrolytes.

  • This paper surveys the physical properties of sheath/core nylon/PET high hollow composites filaments and its fabrics according to the various elution conditions such as concentration of elution, eluted time and eluted temperature. For this purpose, sheath/core nylon/PET filament was texturized and four kinds of fabric specimens were woven with different warp and weft densities. These grey fabrics were eluted with two kinds of concentrations of NaOH (30g/l, 40g/l), three kinds of eluted temperatures $50^{\circ}C,\;60^{\circ}C,\;85^{\circ}C$) and two kinds of eluted times (60min, l20min). The elution characteristics of these specimens were investigated and discussed with different elution conditions. In addition, the mechanical properties such as extensibility, bending rigidity, shear modulus and compressional work of these specimens aceording to the elution conditions were analysed and summarized with cross-sectional shapes of eluted filaments measured by SEM.

The Elution effects by Sulfuric acid in the EAF dust (황산용액에 의한 제강분진 중 중금속의 용출효과)

  • Elution effect was studied by the reactions between Electric arc furnace(EAF)dust and conc. or dil. sulfuric acid for the purpose of recoverying of the useful metals afterwards. The methods of mixing with acids and EAF dust for the elutions were depend on the ratios of acid/EAF dust. Experimental results showed that increased the ratios of sulfuric acids, decreased the elution effects are on the Zn, Fe and Mn, and conc. sulfuric acid showed higher elution effects than dil. sulfuric acids for the all metals. But in case of Pb, dil. sulfuric acid showed higher elution effects than conc. sulfuric acids conversely. Furthermore, the reason of the extreamly low quantities of eluted Pb is caused by the low solubilities by sulfuric acids compared to the other metals, as the results of the insoluble Pb is removed by the process of filtering.

  • Adsorption isotherm is the most fundamental information in adsorption separation-process. Directly from the elution profile of a peak, the elution-curve method and frontal analysis(FA) were utilized to measure the adsorption isotherm in this work. Using RP-HPLC, sample and the buffer added in mobile phase were 5'-GMP and sodium phosphate, respectively. In this experimental condition, the retention time was decreased with increase in the injected mass of sample. And the front part of a peak was very stiff, so Langmuir adsorption isotherm might be applied. By the elution-curve method, the parameters used in the isotherm were obtained by optimization method, while by the FA, the concentrations of stationary phase were measured from the elution curve and the isotherm was determined by regression analysis. Compared to FA, the consumption of sample was less, and only one or two injections were needed by the elution-curve method. Finally, the effect of concentration of sodium phosphate in mobile phase on the parameters of the isotherm was investigated.

In vitro physical, chemical, and biological evaluation of commercially available metal orthodontic brackets

  • Objective: This in vitro study was undertaken to evaluate the physical, chemical, and biological properties of commercially available metal orthodontic brackets in South Korea, because national standards for these products are lacking. Methods: Four bracket brands were tested for dimensional accuracy, (manufacturing errors in angulation and torque), cytotoxicity, composition, elution, and corrosion: Archist (Daeseung Medical), Victory (3M Unitek), Kosaka (Tomy), and Confidence (Shinye Odontology Materials). Results: The tested rackets showed no significant differences in manufacturing errors in angulation, but Confidence brackets showed a significant difference in manufacturing errors in torque. None of the brackets were cytotoxic to mouse fibroblasts. The metal ion components did not show a regular increasing or decreasing trend of elution over time, but the volume of the total eluted metal ions increased: Archist brackets had the maximal Cr elution and Confidence brackets appeared to have the largest volume of total eluted metal ions because of excessive Ni elution. Confidence brackets showed the lowest corrosion resistance during potentiodynamic polarization. Conclusions: The results of this study could potentially be applied in establishing national standards for metal orthodontic brackets and in evaluating commercially available products.

Chromatographic Enrichment of Lithium Isotopes by Hydrous Manganese(IV) Oxide

  • Separation of lithium isotopes was investigated by chemical ion exchange with a hydrous manganese(IV) oxide ion exchanger using an elution chromatography. The capacity of manganese(IV) oxide ion exchanger was 0.5 meq/g. One molar CH3COO Na solution was used as an eluent. The heavier isotope of lithium was enriched in the solution phase, while the lighter isotope was enriched in the ion exchanger phase. The separation factor was calculated according to the method of Glueckauf from the elution curve and isotopic assays. The single stage separation factor of lithium isotope pair fractionation was 1.021.

  • Optimal operation in chromatography is needed to save operation time and the solvent used in multiple chromatographic runs. To this end, many simulation studies of chromatography process have been performed. The relationship between the distribution coefficient and the ionic strength is important in gradient elution ion chromatography. Experimental runs and computer simulations were carried out under linear gradient elution condition in order to compare the experiments and the simulation. Experiments were performed with formic acid under isocratic conditions to determine the simulation equation parameters. Computer simulation was based on three equations which related distribution with ionic strength as follows; K=${\alpha}$I(sup)-${\beta}$, K=A+BI+Cl$^2$and K=y(sub)0+A$_1$$.$e(sup)(-I/m$_1$). The effects of gradient slope on the chromatograms are discussed, and good agreement between the experimental and the simulated results is shown.

Development of a Virus Elution and Concentration Procedure for Detecting Norovirus in Cabbage and Lettuce

  • In this study, a rapid and efficient concentrating procedure that can be used for detecting viruses in vegetables was developed. The Sabin strain of poliovirus type 1 was used to evaluate the efficiency of virus recovery. The procedure included: (a) elution with 0.25 M threonine-0.3 M NaCl pH 9.5; (b) polyethylene glycol (PEG) 8000 precipitation; (c) chloroform extraction; (d) 2$^{nd}$ PEG precipitation; (f) RNA extraction; (g) reverse transcription-polymerase chain reaction (RT-PCR) combined with semi-nested PCR. The overall recoveries by elution/concentration were 29.0% from cabbage and 13.7% from lettuce. The whole procedure usually takes 18 hr. The overall detection sensitivity was 100 RT-PCR units of genogroup II norovirus (GII NoV)/25 g cabbage and 100 RT-PCR units of GII NoV/10 g lettuce. The virus detecting method developed in this study should facilitate the detection of low levels of NoV in cabbage and lettuce.

  • This paper describes the influence of nutrient salts eluted from the bottom of a closed water area where polluted sediment has been deposited by inflowing river water. The elution pattern was monitored at our experimental facility. Both the sediment pore water and water above the bottom were sampled using a dialyzer sampler (peeper). The pore water of the eutrophicated sediment contained a large amount of nutrient salts, and the effect of elution was confined to a limited area of the bottom surface. The nutrient concentration of the sediment pore water was closely related to both the water temperature and dissolved oxygen (DO) concentration. The eluted nutrients from the sediment provided a source for phytoplankton and algae growth. This experimental data indicated that the water quality of the surface was not directly connected to the eluted nutrient salts, while it was indirectly affected by the total ecosystem, including all the organisms within an area and their environment.
