The Chemokine SDF-1α Suppresses Fibronectin-mediated In Vitro Lymphocytes Adhesion

  • Stromal cell-derived factor (SDF-1) is a CXC chemokine that selectively activates the CXCR4 chemokine receptor. Fibronectin is an intracellular matrix component that binds integrin and mediates cell-matrix adhesion. Activation of the integrin receptor can occur in two ways: by ligand binding (outside-in signaling), and in response to intracellular events (inside-out signaling). In the current study we showed that SDF-$1{\alpha}$ inhibited adhesion of T lymphocyte Jurkat cells resulting from binding high concentrations of fibronectin as well as that of THP-1 monocytes. The effect of SDF-$1{\alpha}$ on fibronectin-mediated adhesion was partly reversed by the CXCR4 receptor antagonist T140. Our results suggest that an SDF-1/CXCR4 signal pathway modulates fibronectin-mediated lymphocytes adhesion.

Regulatory Role of CD29 $({\beta}1-integrins)$ in Monocytic Cell Functions (단핵구 기능 수행에서의 $CD29({\beta}1-integrins)$ 조절 역할)

  • CD29 $({\beta}1-integrins)$ is one of major adhesion molecules involved in regulating cell adhesion, migration and morphological changes. In this study, we investigated the regulatory role of CD29 in monocytic functions using monocytic cell line U937 cells. CD29 was found to be one of highly expressed membrane proteins in U937 cells, according to flow cytometric analysis. The activation of CD29 by agonistic antibody MEM101A and extracellular matrix protein (ECM) fibronectin strongly induced cell-cell and cell-fibronectin adhesions. However, blocking antibodies to CD98 and CD147 showed different inhibitory features in these two adhesion events. Furthermore, U0126, an ERK inhibitor, only blocked cell-cell adhesion but not cell-fibronectin adhesion, indicating that cell-cell or cell-fibronectin adhesion events may be regulated by different molecular mechanisms. Meanwhile, CD29 activation also enhanced ROS generation but not phagocytic ability, and similarly radical scavenger N-acetyl-L-cysteine strongly blocked CD29-mediated cell-cell adhesion, implying that ROS may play a critical role in up-regulating cell-cell adhesion. Therefore, our data suggest that the activation of CD29 may be critically involved in regulating monocytic cell-mediated cell-cell adhesion and ROS generation.

Decrease of Surface Fibronectin Availability Required for Myoblast Adhesion by Tunicamycin (Tunicamycin에 의한 근원세포 접착에 필요한 표면 Fibronectin 유용성의 감소)

  • 근세포 융합에 있어서 당단백질의 역할을 세포내 glycosylation의 저해제인 tunicamycin을 이용하여 검토하였다. 근세포가 읖합하기 전 여러 시기에 tunicamycin을 0.04091m숙 농도로 처리하면 세포내 glycosylation과 근세포 융합은 크게 감소되지만, 단백질 합성률과 creatine kinase 활성은 별로 변하지 않는 점으로 미루어 보아 근세포 표면의 당단백질은 세포간의 recognition과 adhesion에 관여하는 것으로 추정할 수있었다. 따라서, 표지된 Con A 염색법을 써서 근세포 원형질막의 당단백질의 변화를 검토해 본 결과 tunicamycin을 처리한 경우 원형질막 당단백질의 대부분이 감소됨을 볼 수 있었으며, 아울러 근세포내 단백질의 분해속도는 증가하고 fibronectin은 감소하는 현상을 관찰할 수 있었다. 한편, fibronectin(20 ug/ml) 을 tunicamycin과 같이 처리한 경우에는 융합이 억제되지 않았다. 이상의 결과들은 tunicamycin이 근세포가 adhesion하는데 필요한 세포막표면의 fibronectin의 유용성을 감소시킴으로써 근세포의 융합을 억제할 가능성을 제시하는 것이다.

Alterations in the Level of Fibronectin and its Receptors during Chick Myoblast Differentiation (계배 근원세포 분화에 따른 Fibronection의 수준과 그 수용체의 변화)

  • Alterations in the amount of fibronectin during chick myogenesis were investigated. The amount of fibronectin as measured by immunoblotting was found to decrease during the process. As a first step in answering the precise mode of change in the level of ibronedin during myogensis, the interaction of 28,000 dalton(28 kDa) amino terminal fragment of fibronectin as well as 85,000 dalton (85 kDa) fragrxient with myoblasts was examined. The specific binding of 125 l-28 kDa fragment to myoblasts was time-dependent and reached a maximum within 60 min. Unlabelled 28 kDa fragment inhibited the binding of 125 I-28 kDa fragment, whereas 85 kDa fragment containing adhesion promoting activity did not inhibit it. This finding suggests that the 28 kDa fragment interacts with the matrix assembly receptors but not with the cell adhesion receptors. Accordingly, the decrease in the level of fibronectin is likely to correlate with the fall of fibronectin receptors on the myoblasts.

Effects of the Chestnut Inner Shell Extract on the Expression of Adhesion Molecules, Fibronectin and Vitronectin, of Skin Fibroblasts in Culture

  • The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.

  • 1. 연구 목적 Fibronectin은 세포외기질의 주요성분인 거대 당단백질로서, 조골세포의 부착과 증식 및 이동능에 중요한 역할을 담당한다고 알려져 있다. 이러한 fibronectin의 조골세포에 대한 영향을 실제 임상에 적용하기 위해서, 전체 fibronectin 단백질을 사용하는 것은 면역학적으로나 경제적으로 많은 단점을 안고 있어서, 유효한 반응단위만을 추출하여 활용하는 것이 바람직한 방법으로 알려져 있다. 이 연구의 목적은 세포부착에 주로 관여하는 fibronectin type III분절 중 10번 도메인이 조골양 세포에 미치는 영향을 전체 fibronectin단백질과 fibronectin type III 7-10 도메인 분절과 비교, 관찰하는 것이다. 2. 연구 방법 사람의 fibronectin을 기초로 한 적절한 primer로서, 유전자 재조합법을 이용하여 fibronectin type III 10 도메인과 fibronectin type III 7-10 도메인 분절을 얻었으며, 전체 fibronectin분자는 상용으로 준비하여 24-well 세포배양 용기에 도포하였다. 배양된 조골양세포(HOS cell)를 $1x10^5$ cells/well의 농도로 각 well에 분주하여 $37^{\circ}C$에서 1시간 배양을 하였다. Cell adhesion assay를 실시하기 위해 10% formaldehyde로 고정시키고 1% Crystal Violet으로 염색하여 광학현미경을 관찰 후 2% SDS를 처리하여 microplate reader기를 이용하여 570nm에서 혼탁정도를 측정하였다. 음성대조군으로는 RPMI 용액을 사용하였다. 동일한 방법을 이용하여 준비된 $35mm^2배양접시에 HOS cell을 $37^{\circ}C$에서 4일간 배양 후, MTS assay를 이용하여 세포 증식도에 미치는 영향을 관찰하였다. 6일째 405nm에서 활성화된 세포에서 분비된 p-nitrophenol을 이용한 alkaline phosphatase activity를 측정하였다. 3. 결과 및 고찰 Fibronectin type III 10 도메인은 HOS cell에 대한 생물학적인 효과면에서, 전체 fibronectin 분자 및 fibronectin type III 7-10 분절과 통계적으로 유사한 세포부착도를 보여주었으며, 세포증식도와 alkaline phosphatse 활성도면에서도 큰 차이가 나타나지 않았다. 이상의 연구결과로 볼 때, fibronectin type III 10 도메인이 조골세포의 증식을 목적으로 사용하는 생체재료의 표면개질 부착물질로 응용할 수 있는 가능성이 있다고 하겠다.

Regulatory Effect of Ginsenosides Rh1 on Monocytic U937 Cell Adhesion (홍삼유래 ginsenosides Rh1의 단핵구 U937 세포 유착조절 효과)

  • Cell-cell adhesion managed by various adhesion molecules is known to be one of pathophysiological phenomena found in numerous immunological diseases such as rheumatoid arthritis and allergic diseases. In this study, we examined the regulatory role of ginsenosides (G)- Rh1, reported to display anti-inflammatory and anti-allergic effects, on CD29-mediated cell adhesion. G-Rh1 significantly suppressed U937 cell-cell adhesion mediated by CD29 but not CD43. It also blocked U937 cell-fibronectin adhesion, mediated by activated CD29, up to 30%. In agreement, this compound also significantly decreased the surface level of CD29 but not CD43 as well as other costimulatory molecules such as CD69, CD80, and CD86. Therefore, these results suggest that G-Rh1 may have inhibitory function on CD29-mediated cell adhesion events, probably contributing to its anti-inflammatory and anti-allergic activities.

Modulatory Effect of Diethylstilbestrol on CD29-Mediated Cell-cell Adhesion in Monocytic U937 Cells (Diethylstilbestrol의 단핵구의 세포간 유착과정 조절효과)

  • Diethylstilbestrol (DESB) is a synthetic estrogen not only that routinely prescribed, but also that known to be a teratogen. In this study, we found a novel pharmacological feature that DESB is able to positively modulate CD29 $({\beta}1-integrin)$ function. Thus, DESB up-regulated homotypic cell-cell adhesion of monocytic U937 cells mediated by CD29. However, DESB did not increase the surface level of CD29 and its binding activity to ligand (fibronectin), according to flow cytometric analysis and cell-fibronectin adhesion assay. Instead, the DESB-mediated up-regulation of cell-cell adhesion was blocked by several signaling enzyme inhibitors. Treatment of U0126 [an extracellular signal-regulated kinase (ERK) inhibitor], SB20358 (a p38 inhibitor) or Rp-8-pCPT-cGMP (a protein kinase G inhibitor) clearly inhibited DESB-mediated up-regulation of cell-cell adhesion induced by CD29. However, estrogen receptor antagonist ICI 182,780 failed to abrogate DESB effect. Therefore, our data suggest that DESB may up-regulate CD29-mediated cell-cell adhesion via modulating intracellular signaling enzymes such as ERK, PKG, and p38, independent of estrogen receptor function.

Polymer Surfaces for Cell Adhesion II. Cell Culture on Surface-modified Polymers (세포적합성 고분자 표면에 관한 연구 II. 표면 개질된 고분자에의 세포 배양)

  • Chinese Hamster Ovary( CHO) cells were cultured on the surface-modified polymers described in the previous study( "Polymer Surfaces for Cell Adhesion. 1. Surface Modification of Polymers and ESCA Analysis, " J. of KOSOMBE, Vol. 10, No. 1, 43-51, 1989). Among the physicochemical treatment methods. the chloric acid treatment was found to be the best method of rendering the polymer surfaces adhesive for CHO cells probably due to the high density of hydroxyl groups on the surface. Among the biological methods, the fibronectin treatment was best for CHO cell-compatibility probably due to specific active sites existed on the tell-binding domains of the fibronectin structure. When we compare the cell-compatibility of the chloric acid - and the fibronectin -treated PET surfaces, the number of cells attached on the surfaces were increased by 460.5 % and 559.0 % and, respectively, after 32 hr CHO cell culture, compared to that of untreated PET.eated PET.

A Possible Role of Fibronectin on the Differentiation of Monocyte to Macrophase (단핵구 분화에 대한 Fibronectin 및 그 단편의 역할)

  • Monocyte interaction with fibronectin (FN) mediates specific cell surface receptors and results in cell attachment and differentiation. Several cell-mediated activities for various fragments of FN have been documented. To investigate the regulatory mechanisms of monocyte differentiation by cell binding domains of FN and their receptors, cell attachment-, cell migration-, and its respective inhibition assay were carried out. Monocyte recognizes 38-kDa domain distinctively from its recognition of 85-kDa domain, and the heparin-binding site of the 38-kDa fragment is not involved in monocyte adhesion. Based on these experimental results, it can be suggested that monocvte/macrophase interacts with at least two different sites in FN, which is critical step in cell adhesion and (or) migration.
