Breeding of Flammulina velutipes Strains Adaptable to Elevated-temperature

  • Winter mushroom, Flammulina velutipes, needs low temperature during its cultivation. To save on farm costs, especially during summer, a strain adaptable to a higher or elevated-temperature must be developed. At the start of breeding program, parental strains which could endure high temperature were obtained. Seuenty four dikaryotic strains were collected and divided into four groups according to the nature of temperature. They also had different fruiting temperature. Finally we selected three brown strains ASI 4048, 4057 and 4072, and collected their spores. These selected strains can germinate even at a high temperature of $32^{\circ}C$, which were dramatically higher than the other strains. Based on these results, the new white strain adapted to mid-temperature by backcross mating was developed. Molecular markers were applied to select white fruitbody producing strains without cultivation. They showed a specific band which co-segregated with brown fruitbody forming strains in $BC_1F_1$ progenies. Selected white strains were tested under several elevated temperature conditions.

Antioxidative Effects of Mushroom Flammulina velutipes Extract on Polyunsaturated Oils in Oil-in-water Emulsion

  • The antioxidative activities of the water solution of crude extract from edible mushroom enokitake Flammulina velutipes were compared with those of ascorbic acid and ascorbic acid 6-palmitate in oil-in-water (O/W) emulsions of cod liver oil. Oxidation of the emulsions was carried out at 40 and $50^{\circ}C$ in the dark. The antioxidant activities were measured by in vitro assay against oxygen uptake, 2-thiobarbituric acid value, hydroperoxide formation of the oils. Also, residual docosahexaenoic acid content was measured as indices of lipid oxidation. The cod liver oil in O/W emulsions with added enokitake crude extract (ECE) was significantly more stable against lipid oxidation than the control emulsions without the extract in terms of any oxidation indices used. Moreover, ECE provided remarkable antioxidative properties to eicosapentaenoic acid ethyl ester in emulsion system. These observations demonstrate that F. velutipes can be used as a natural antioxidant, which effectively prevents oxidation of polyunsaturated oils in emulsion system.

반응표면분석에 의한 팽이버섯의 microwave 추출조건 최적화 (Optimization of Microwave Extraction Conditions for Flammulina velutipes by Response Surface Methodology)

  • 팽이버섯 추출물의 수율 및 전자공여작용, 총폴리페놀함량, tyrosinase 저해작용, 갈색도, 아질산염 소거작용에 대하여 반응표면분석에 의해 추출조건을 최적화 하였다. 팽이버섯 추출물의 수율변화는 에탄올 농도에 가장 큰 영향을 받았으며, 갈변도는 microwave power 보다는 에탄올 농도에 크게 영향을 받는 것으로 나타났으며, 아질산염 소거작용의 경우 에탄올 농도가 증가할수록, microwave power가 감소할수록 증가하는 경향을 나타내었다. 전자공여작용의 경우 에탄올 농도가 증가할수록 증가폭이 컸고, microwave가 증가할수록 조금씩 증가하는 경향을 보였다. 총 폴리페놀 함량의 경우 에탄올 농도가 증가할수록, microwave power가 클수록 추출물의 생리활성이 우수한 것으로 나타나 에탄올 농도가 추출시 중요한 공정인자임을 확인 할 수 있었다. 한편, tyrosinase 저해 작용의 경우 에탄올 농도보다는 microwave power에 영향을 받았는데 power가 작을수록 그 값은 크게 측정되었다. 따라서 팽이버섯의 최적 추출조건 범위는 microwave power $47.21{\sim}76.05$ watt, ethanol 농도 $10.25{\sim}43.56%$, 추출시간 5.72분으로 예측되었다.

느타리, 새송이, 팽이버섯의 용매추출에 따른 항산화활성 연구 (Study on Antioxidant Activities of Pleurotus ostreatus (Jacq.) Que Pleurotus eryngii and Flammulina velutipes Extracts by Different Solvent)

  • This study was conducted to find out the optimal solvent extraction method [Distilled water (DW), 70% ethanol, 99% ethanol] of mushrooms, including Pleurotus ostreatus (Jacq.) Que, Pleurotus eryngii and Flammulina velutipes and improve their usability as natural antioxidants. To analyze antioxidant activities in each mushroom, total polyphenol, flavonoid contents, 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) cation radical (ABTS+) and fluorescence recovery after photobleaching (FRAP) were measured. All mushrooms showed the highest total polyphenol contents in DW mushroom extract (p<0.001). Total flavonoid contents were the highest in P. eryngii and F. velutipes DW and 70% ethanol mushroom extracts (p<0.05). All mushrooms showed the highest activities using DPPH and FRAP assays in the DW extraction method (p<0.001). P. ostreatus (Jacq.) Que and P. eryngii showed the highest ABTS+ radical scavenging activity in the DW extraction method, and F. velutipes showed the highest activity in the 70% ethanol extraction method (p<0.001). As a result of comparing IC50 values of DPPH and ABTS+ radicals and FRAP EC50 values, the DW P. ostreatus (Jacq.) Que extract showed high antioxidant activities (p<0.001). Pearson's correlation between total polyphenol contents and antioxidant activities showed a positive correlation in all mushrooms (p<0.01). Therefore, extraction of the mushrooms with DW can enhance the extraction of effective bioactive substances and antioxidant activity.

  • The production of lac case by the funguson various media was studied. The characteristics of the enzyme were also studied regarding to the optimum pH, stability, Km value, and inactivation. The maximum activity of laccase reached the 40 days of incubation and the barley straw extract appeared to be a strong inducer for laccase. The enzyme showed stability at wide range of pH with optimum pH of 6.6. Temperature stability of the enzyme was high. Laccase was not inactivated by the organic solvents used for the precipitation. The enzyme, how­ever, was completely inactivated by trichloroacetic acid and sodium azide.

B16 흑색종에 대한 팽이버섯 추출물의 항암활성 비교 (Analysis of Anti-Tumor Activity of Flammulina velutipes Extract on B16 cells)

  • 버섯은 예전부터 단백질, 당질, 비타민, 무기질 등 일반 영양소들이 다량 함유되어 있을 뿐 아니라 다양한 약리작용을 가지고 있기 때문에 예로부터 민식용 및 민간요법으로도 많이 이용되어 왔다(17). 또한 현재에는 건강식으로 다양하게 이용되고 있다. 본 연구는 여러 가지 버섯 중, 팽이버섯의 추출물을 이용하여 이의 항암 활성을 실시하였다. 팽이버섯(Flammulina velutipes)의 항암활성을 검증하기 위하여 in vitro 실험으로 wound healing 활성 실험을 하였으며, in vivo 실험으로 마우스를 이용한 항암모델 실험을 실시하였다. Wound healing 활성 실험 결과 팽이버섯 추출물의 농도를 0-100 ug/mL로 처리함에 따라 B16 세포의 운동성이 감소하는 것을 알 수 있었다. 이를 바탕으로 마우스를 이용하여 항암 모델에 적용한 결과 B16 세포로 인해 형성되는 colony가 팽이버섯 추출물을 처리한 군에서 약 60% 내외로 현저히 감소하는 것을 알 수 있었다. 본 실험 결과는 팽이버섯 추출물의 항암 활성을 확인 한 것으로 향후 이의 추출물을 이용한 세포신호전달과정 등에 관한 연구와 건강기능성식품의 소재로의 제품 개발에 유용하게 사용될 수 있음을 나타내는 주요한 자료로 판단된다.

팽이 신품종 '백이' 육성 (Breeding of a new cultivar of Flammulina velutipes: 'Baeke')

  • 팽이의 신품종을 개발하기 위하여 모균주 ASI 4216(한솔)과 4217(백중)으로부터 단포자를 분리하여 단핵균주를 얻고 이들의 교잡을 통해서 꺽쇠연결체가 확인된 교잡주총 129점을 선발하였다. 교잡주를 대상으로 3회에 걸쳐 재배시험 결과, 고온에서 배양이 가능하며 수량성이 우수한 계통 'Fv14a51'을 최종선발되었으며, 생산력검정 시험을 통해 2017년 농작물 직무육성 품종 심의회에서 '백이'로 명명되었다. 주요 특성은 균사생장이 $30^{\circ}C$의 고온에서도 균활력을 유지할 정도로 강한 온도적응성 품종이고 대조품종에 비해 갓이 두껍고 대가 얇는 형태를 띠는 것 외에는 갓색깔이 순백색이며, 갓모영은 반구형, 수량은 $257.4{\pm}13.5g$으로 대조품종과 비교해서 대체적으로 유사하였다. RAPD primer을 통해 유전적 다형성을 검정한 결과, 대조구인 '오키노메구미', 2016년에 육성된 '백승'과 신품종이 유전적으로 구별되는 것을 확인하였다. 전반적으로 '백이'는 수량과 품질이 외국품종과 거의 유사하였으며, 버섯의 균일한 형태로 상업적 가치가 있었다. 신품종의 현장실증을 확대하여 국산품종이 국내에 확산되기를 기대한다.

1주기 수확을 끝낸 팽이버섯(Flammulina velutipes) 재래용 톱밥배지 분해의 현미경적 특징 (Microscopical Characteristics of Softwood Sawdusts Cultivated with Enokitake (Flammulina velutipes))

  • 팽이버섯 1주기를 수확한 폐배지 톱밥을 버섯재배 용으로 재활용 할 수 있는지를 알아보고자 팽이버섯을 재배한 침엽수재 의 톱밥을 현미경적 관찰을 통해 그 분해정도와 분해양태를 파악하였다. 아울러 팽이버섯 재배에 사용된 톱밥의 수종 식별도 실시하였다. 팽이버섯 재배용으로 사용된 톱밥의 수종은 대부분 소나무속 경송류로 식별되었고 극히 일부만 미송 (Douglas-fir)톱밥이 존재하였다. 팽이버섯 1주기를 수확한 폐배지 소나무 톱밥에는 균사에 노출된 부분과 방사조직에서 균사의 밀도가 높게 나타났다. 팽이버섯균의 공격을 받은 세 포벽은 박벽화현상과 침식현상을 보였으나, 이 같은 부후양태를 보인 세포벽은 극히 일부에 지나지 않았다. 이상의 현미경 관찰결과 팽이버섯을 1주기 수확한 소나무 톱밥은 버섯재배용으로 재활용하기에 충분한 세포벽 물질을 보유하고 있는 것으로 나타났다.

Characterization of β-Glucosidase Produced by the White Rot Fungus Flammulina velutipes

  • β-Glucosidase production by the white rot fungus Flammulina velutipes CFK 3111 was evaluated using different carbon and nitrogen sources under submerged fermentation. Maximal extracellular enzyme production was 1.6 U/ml, corresponding to a culture grown in sucrose 40 g/land asparagine 10 g/l. High production yield was also obtained with glucose 10 g/land asparagine 4 g/l medium (0.5 U/ml). Parameters affecting the enzyme activity were studied using p-nitrophenyl-β-D-glucopyranoside as the substrate. Optimal activity was found at 50℃ and pHs 5.0 to 6.0. Under these conditions, β-glucosidase retained 25% of its initial activity after 12 h of incubation and exhibited a half-life of 5 h. The addition of MgCl2, urea, and ethanol enhanced the β-glucosidase activity up to 47%, whereas FeCl2, CuSO4, Cd(NO3)2, and cetyltrimethylammonium bromide inflicted a strong inhibitory effect. Glucose and cellobiose also showed an inhibitory effect on the β-glucosidase activity in a concentration-dependent manner. The enzyme had an estimated molecular mass of 75 kDa. To the best of our knowledge, F. velutipes CFK 3111 β-glucosidase production is amongst the highest reported to date, in a basidiomycetous fungus.

Gene Expression Analyses of Mutant Flammulina velutipes (Enokitake Mushroom) with Clogging Phenomenon

  • Regulation of proper gene expression is important for cellular and organismal survival, maintenance, and growth. Abnormal gene expression, even for a single critical gene, can thwart cellular integrity and normal physiology to cause diseases, aging, and death. Therefore, gene expression profiling serves as a powerful tool to understand the pathology of diseases and to cure them. In this study, the difference in gene expression in Flammulina velutipes was compared between the wild type (WT) mushroom and the mutant one with clogging phenomenon. Differentially expressed transcripts were screened to identify the candidate genes responsible for the mutant phenotype using the DNA microarray analysis. A total of 88 genes including 60 upregulated and 28 downregulated genes were validated using the real-time quantitative PCR analysis. In addition, proteomic differences between the WT and mutant mushroom were analyzed using two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF). Interestingly, the genes identified by these genomic and proteomic analyses were involved in stress response, translation, and energy/sugar metabolism, including HSP70, elongation factor 2, and pyruvate kinase. Together, our data suggest that the aberrant expression of these genes attributes to the mutant clogging phenotype. We propose that these genes can be targeted to foster normal growth in F. velutipes.
