Influence of Feeding Flammuling veluipes Media on Productivity and Egg Quality in Laying Hens (팽이버섯 배지 부산물의 급여가 산란계의 생산성 및 난질에 미치는 영향)

  • This study was conducted to evaluate the feeding value of the Flammulina veluipes media by-product(FMB) in laying hens(Hy-Line Brown). A total of three hundreds sixty were fed corn-soy based experimental diets containing 0(control), 5.0(T1), and $10\%(T2)$ FMB for 12 weeks. There were no significant differences among the treatments in egg production, egg weight, egg mass, feed conversion and viability during the experimental period. Feed intake was significantly(P<0.05) lowered in control(120.5g) than 72(123.9g). There were no significant differences among the treatments eggshell breaking strength, thickness and haugh unit, whereas the yolk color of T1 and T2 were significantly love. than control(p<0.05). In conclusion, the FMB can be used as resource of feed in laying hen feed at $5.0\%$ level without effect on performance and egg qualify. However, dietary pigments must be added when the FMB was used as a feed ingredients more than $10\%$ in diets.

Effect of Fermentation Periods on the Qualities and Physiological Functionalities of the Mushroom Fermentation Broth (발효기간이 버섯 발효액의 품질과 생리 기능성에 미치는 영향)

  • To establish the optimal fermentation periods in the manufacture of mushroom fermentation foods using sugar, changes of quality characteristics of the mushrooms fermentation broth were investigated with changes of enzyme activity and physiological functionality during fermentation. Viscosity, L value (lightness), a value (redness) and b value (yellowness) were significantly decreased after 3 months of fermentation and after that, increased. In sensory evaluation test, unique flavors and tastes of mushrooms in the fermented broth were decreased during fermentation, whereas the other tastes and flavors were gradually increased. Overall acceptability was the highest in the 3 months of fermentation broth. ${\alpha}-Amylase$ activities of the fermented broth were significantly increased to 1 month of fermentation, however invertase and cellulose activities were low or not detected in the fermented broth. Antioxidant activities were the highest in 4 months of fermentation and after that, decreased. Tyrosinase inhibitory activities were high in all samples and they were not changed during fermentation. SOD-like activity was high in the fermentation broth of Flammulina velutipes and it was also not changed during fermentation. In conclusion, optimal fermentation periods in the manufacture of mushroom fermentation foods using sugar was 3 or 4 months.

  • A gram-negative bacterium was isolated from spent substrate of Agaricus bisporus and showed marked antagonistic activity against Pseudomonas tolaasii. The bacterium was identified as Pseudomonas azotoformans by based on the cultural, biochemical and physiological characteristics, and 16S rRNA gene sequence. The isolated bacterium was saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for P. tolaasii cell was not sufficient for inhibition in vitro. Control efficacy of Pseudomonas azotoformans HC5 to brown blotch of P. tolaasii was 73, 78, and 71% on A. bisporus, Flammulina velutipes, and Pleurotus ostreatus, respectively. In the future, the suppressive bacterium may be useful for development of a biocontrol system.

Changes of saccharide content in fruitbody composition of Flammulina velutipes during storage (팽이버섯의 저장기간 및 품종별 자실체내의 당 함량 변화)

  • Winter mushroom was monitored to investigate the influence of storage temperature on its quality during the storage and distribution phase. In measuring its quality, the contents of saccharides were quantified with its fruiting bodies using HPLC. Although it has been known to be difficult to separate saccharide isomers, our results indicated that Grace Prevail carbohydrate ES $5{\mu}column$ was the best in the separation to analyze the saccharide out of six columns used in this study. In our results, xylose was the main component of saccharide in the fruiting body of winter mushroom(White line mushroom:47.68mg/g, brown line mushroom: 63.28mg/g). In long-term storage, the total amount of saccharide tended to increase, but trehalose content of the disaccharide decreased. In comparison with the paramount amount of lactose and myo-inositol contents in long-term storage at $4^{\circ}C$, lactose wasn't detected when stored at $-1^{\circ}C$.

Occurrences of Major Mushroom Diseases and Microbial Densities of Mushroom Cultivation Facilities (국내 주요 버섯류의 병해 발생과 재배사의 미생물 밀도 조사)

  • The occurrences of the major diseases and the densities of air-born microbes were surveyed in the cultivation facilities for oyster mushroom (Pleurotus ostreatus), king oyster mushroom (Pleurotus eryngii), and enoki mushroom (Flammulina velutipes) in different areas of Korea. Green mold disease was most often developed in oyster mushroom bed cultivation with the disease incidence rate of approximate 10% while the disease incidences from bottle and plastic envelop cultivation were less than 1~2%. In the bed cultivation, the major air-born microbes in the growth room were Aspergillus, Penicillium, Trichoderma, and Curvularia with the total fungal population density of 567~1,297 CFU/$m^3$ . However, only Trichoderma and Penicillium were detected in the growth rooms and innoculation rooms of bottle and plastic envelop cultivation with the densities of 350~700 CFU/$m^3$ and 160~260 CFU/$m^3$, respectively. The bacterial diseases become evident in the growth rooms of bottle and plastic envelop cultivation with the approximate incidence rate of 10%. The identified bacterial species were Brevibacillus levelkil, Rhizobium radiobacter, Brevundimonas vesicularis, Pseudomonas mosselii, Microbacterium testaceum. Sphingomonas panmi, Sphingomonas yabuuchiae, Paracocus dinitrificans, Curtobacterium flaccumfaciens pv. flaccumfaciens and some unidentified bacteria with the densities of 40~6,359 CFU/$m^3$ in the growth rooms and 9 CFU/$m^3$ in the inoculation room. This study indicated that the green mold disease by fungal strains was the major mushroom disease in the bed cultivation and suggested that the contamination of bacteria and fungi together in the growth media could result in severe production loss. The plastic envelope and bottle cultivation were evidenced to be less susceptible to such contaminations.

Monitoring of Artificial Radionuclides in Edible Mushrooms in Korea (식용 버섯류에서의 인공 방사능 농도 조사)

  • To ensure food-safety of mushrooms from radioactive contamination, edible mushroom samples distributed in Gyeonggi province in Korea were collected according to species and country of origin. A total of 284 mushrooms, belonging to 10 species (Lentinula edodes, Pleurotus ostreatus, Pleurotus eryngii, Agaricus bisporus, Flammulina velutipes, Phellinus linteus, Inonotus Obliquus (Chaga), Auricularia auricula-judae, Ganoderma lucidum and Tricholoma matsutake) were subjected to radioactivity testing. The concentration of artificial radionuclides, 131I, 134Cs, and 137Cs, was analyzed using gamma-ray spectrometry. 131I and 134Cs were not detected more than MDA value from all samples. Among 204 domestic mushrooms, however, 137Cs were detected in 0.21~2.58 Bq/kg from six cases (3 Lentinula edodes, 1 Ganoderma lucidum and 2 Tricholoma matsutake), whereas 137Cs were detected in 0.21~53.79 Bq/kg from 38 cases (22 Inonotus Obliquus(Chaga), 14 Phellinus linteus, 1 Lentinula edodes and 1 Tricholoma matsutake) among 80 imported mushrooms. In addition, average concentration of 137Cs in 10 Chaga mushroom-processed products was more than twice as much as dried Chaga mushroom, and maximum concentration was 123.79 Bq/kg. Results suggest that radioactivity monitoring system for imported mushrooms and mushroom-processed products should be continuously intensified to secure food-safety in Korea.

Selection of optimum conditions and distribution temperature of complete substrates of Pleurotus species for export to Southeast Asia (동남아 수출을 위한 느타리류 완성형배지의 최적 조건 및 운송 온도)

  • Pleurotus species are the most consumed and cultivated mushrooms in Korea. Although oyster mushrooms (P. ostreatus) can be cultivated automatically, their storability is slightly lower than that of king oyster mushrooms (P. eryngii) and winter mushrooms (Flammulina velutipes); therefore, the export proportion of oyster mushrooms is very low. Since Korean mushrooms are highly preferred across Southeast Asian, the export of oyster mushrooms in the form of complete substrates is expected to be more promising than that of fresh mushroom. Here, 1 and 2.5 kg complete substrates of P. ostreatus 'Soltari' and P. sajor-caju 'Sambok' were prepared and stored at different temperature from 0 to 15℃ for 10 days. Thereafter, the formation of fruiting bodies was induced. Since the 2.5 kg complete substrates required 70 days of incubation, their mycelia were at an advanced age and their fruiting bodies did not grown normally. When 70%-incubated complete substrates were stored at 5-10℃, the growth was faster and more uniform and stable fruiting bodies were formed. Export test of complete substrates to Vietnam using distribution containers set at 0℃ and 15℃ revealed that the growth period was shortened by 1-2 days when the distribution containers were set at 15℃ and the yield of 'Soltari' increased by approximately 10%. In addition, even though the yield of 'Sambok' was similar between treatments at 0℃ and 15℃, the quality of fruiting bodies from 15℃-distributed complete substrates was much better than that of those from 0℃-distributed substrates.

Developed Vacuum Film Packaging Method Maintains Quality of Enoki Mushrooms (Flammulina velutipes) during Simulated Vessel Export to Vietnam (팽이버섯의 베트남 모의수출 중 진공포장방법 개선을 통한 품질 유지 효과)

  • Korean enoki mushrooms are exported to Southeast Asia and the United States, where there are complaints due to quality deterioration during the distribution process. In this study, we evaluated the efficacy of vacuum film packaging on quality maintenance in Korean enoki mushrooms during simulated vessel export to Vietnam using selected film, which was developed with vacuum packaging machine and oxygen absorber. We selected two MA film, one was 2-layerd 30 ㎛ CPP film (control) and the other was 3-layered 30 ㎛ CPP film (treatment) which is optimized film of higher gas and moisture permeability. The Korean enoki mushrooms were packaged with these two films using PAC-2000 or PAC-3000 vacuum packaging machine which was improved vacuuming of higher speed. Packed mushrooms were stored at 1℃ for 2 weeks and distributed at 20℃ for 2 days or 8℃ for 6 days. The efficiency of the film packaging was analyzed by vacuum maintaining index, and overall the quality characteristics such as off odor, color, cap cleavage, stem elongations and sensory evaluation were evaluated during storage and distribution. Results suggest that postharvest loss of fresh enoki mushroom could be reduced by packaging mushroom with 3-layered 30 ㎛ CPP films packaged using PAC-3000 machine during simulated vessel export to Vietnam due to vacuum maintaining. Oxygen absorber promoted off-odor at 20℃ distribution temperature, and did not affect storability at 1℃ storage compared to treated group without oxygen absorber treatment.

The characteristics of fungal-mycelium-based composite materials using spent mushroom substrates of Flammulina velutipes (팽이버섯 수확후배지를 이용한 곰팡이 균사체 기반 복합소재의 특성)

  • This study was conducted to develop a renewable and sustainable bio-material to replace polystyrene (EPS) in fungal-mycelium-based composite using agricultural by-products. Four mushrooms (Ganoderma lucidum, Fomitella fraxinea, Phellinus linteus, and Schizophyllum commune) were cultured in an oak sawdust plus rice bran substrate to select the mushroom with the best growth. The mycelia of G. lucidum showed the best growth. To investigate the optimal mixing ratio with spent mushroom substrate (SM) and oak sawdust (OS), samples were prepared by mixing SM and OS at ratios of 50%:50%, 60%:40%, and 80%:20% (w/w). Each substrate was then inoculated with G. lucidum. G. lucidum showed the best mycelial growth of 140.0 mm in the substrate with SM and OS mixed at a 60%:40% ratio. It was also found that the substrate with SM and OS mixed at a 60%:40% ratio had the best handling properties. The compressive strength of mycelial materials inoculated with G. lucidum was in the range of 300-302 kgf mm-1, and the materials were four times stronger than polystyrene materials. These results indicate that substrates comprising spent mushroom substrate mixed with oak sawdust can be successfully upcycled to mycelium-based composite materials using G. lucidum. This represents a sustainable approach.

Efficacy of Commercial Sanitizers for the Inactivation of Listeria monocytogenes on the Processing Equipment for Enoki Mushrooms (팽이버섯 재배 현장에서 Listeria monocytogenes의 성장을 억제하기 위한 살균 처리 기술 개발)

  • The consumption of enoki mushrooms has been associated with cases of listeriosis produced by Listeria monocytogenes, highlighting the significance of sanitizing food-contact surface, such as the velcro used in welding processing of enoki mushrooms, to ensure microbial safety. We investigated the inhibitory activity of nine chemical disinfectants at regular concentrations against L. monocytogenes isolated from a mushroom farm environment. The bacterial suspension was prepared in phosphate buffered saline and mushroom extract broth and inoculated onto the velcro surface. After inoculation, most disinfectants reduced the initial 8 log CFU/coupon concentration by less than 2 log CFU/coupon during a 5-min treatment. Slightly acidic hypochlorous water showed a reduction of approximately 4 log CFU/coupon when tested for more than 30 min at the maximum allowable concentration of 200 mg/L. Sodium hypochlorite solution showed a reduction of approximately 5 log CFU/coupon when used at 100 mg/L for 60 min. Peracetic acid, at the maximum allowable concentration of 300 mg/L, showed the most effective reduction of 5 log CFU/coupon or more when the surface was treated with 37.5 mg/L for 30 min. These results indicate that peracetic acid can be used as the disinfectant strategy to control cross-contamination of L. monocytogenes on the velcro surface of plastic wrappers used in the welding processing of enoki mushrooms.
