A Study of Anticoagulation Activity from Perillae Folium Extract (자소엽(紫蘇葉) 추출물의 항응혈(抗凝血) 활성에 관한 연구(硏究))

  • Objectives: This research was investigated to find out the effect of the anticoagulant Perillae folium extract. Methods: To examine an active effect of anticoagulation in Perillae folium extract, the study measured Prothrombin time(PT) and activated partial thromboplastin time(APTT) of human plasma in vitro and measured bleeding time and arterio-venous shunt model in rats in vivo. Results: Bleeding time of Perillae folium extract in vivo had a significant increase 1.6 times and thrombus weight of Perillae folium extract had a significant reduction of thrombus weight as 68%. Perillae folium extract had an effect of anticoagulation by operating on extrinsic pathway factor II, V, VII, X and intrinsic pathway factor VIII, IX, X, VI, VII in the coagulation system. Conclusions: Considering the above mentioned results, it is judged that a Perillae folium extract has a control effect of thrombus creation.

Studies on the Application for Cosmetics Natural Materials of Folium Perillae (소엽(蘇葉)의 화장품 천연소재 활용(活用)에 관한 연구(硏究))

  • Objectives : In this study, Folium Perillae were examined the possibility to apply as the cosmetics natural materials. Methods : Normal skin softener containing Folium Perillae extracts was manufactured and then its physiological activities function was experimented on. And emollient lotion containing Folium Perillae extracts was manufactured and then it was left under the condition of $-10^{\circ}C,\;0^{\circ}C$, room temperature and $37^{\circ}C$ for a month. Then its stability and safety were tested. Results : The physiological activities function of the normal skin softener was almost same with the electron donating ability, SOD like activity and xanthine oxidase inhibitory effect of Folium Perillae extracts. To find the changes of emollient lotion containing Folium Perillae extracts, the emollient lotion was left under the condition of $-10^{\circ}C,\;0^{\circ}C$, room temperature and $37^{\circ}C$ for a month. Then, when the emollient lotion was observed with the naked eye, pH, viscosity and particle diameter were measured, its changes were not nearly found. Futhermore, as a result of doing patch test to identify the safety of emollient lotion containing Folium Perillae extracts, there was no stimulus on skin. Conclusions : From the above results, it was expected that the physiological activities of Folium Perillae extracts can be maintained when cosmetics containing Folium Perillae extracts are manufactured. And it was proved that Folium Perillae extracts didn't affect the change of cosmetic when they were applied to cosmetic materials. And it was concluded that emollient lotion containing Folium Perillae extracts was safe for skin.

  • In this study, 15 compounds were elucidated from the hot-water extract of Perillae Folium. Fifteen isolates were determined to be protocatechuic acid (1), caffeic acid (2), (R)-rosmarinic acid (3), (S)-shisoflavanone A (4), luteolin-7-O-β-D-glucuronopyranoside (5), scutellarein-7-O-β-D-glucuronopyranoside (6), apigenin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (7), luteolin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (8), kelampayoside A (9), trans-N-feruloyloctopamine (10), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acrylamide (11), perilloside C (12), perilloside A (13), (6S,9R)-9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside (14) and (6S,9R)-roseoside (15) through spectroscopic evidences. The HPLC analysis revealed that hot-water extract of Perillae Folium contained caffeic acid, rosmarinic acid and glycosides of apigenin, luteolin and scutellarein as main constituents.

  • Effects of hydrogen peroxide$(H_2O_2)$ on polyphenol oxidase (PPO) in Perillae Folium were investigated. The inactivation of this enzyme was dependent on $H_2O_2$ concentration. and the initial lag period was not shown. Preincubation of Perillae Folium PPO with $H_2O_2$ in the absence of a substrate resulted in rapid loss of enzymatic activity. The inactivation of PPO by $H_2O_2$ dependents temperature and pH. OH radical scavengers such as mannitol and sodium formate did not protect the enzyme against inactivation by $H_2O_2$. Substrate analogue such as phenylalanine protected the enzyme against inactivation by $H_2O_2$. and copper chelator such as sodium azide also protected the enzyme.

  • The volatile components of Perillae folium were isolated by simultaneous steam distillation-extraction method, and analyzed by combined GC and GC-MS. Among seventeen components indentified 6 alcohols (3-octanol, 1-octen-3-ol, linalool, nerolidol, supathuleol and phytol), 2 ketones, 1 aldehyde, 1 phenol and 7 hydrocarbons were confirmed. The most abundant component was myristicin comprising about 53.4%.

  • For the purpose of external and internal morphological study on the origin of Folium, writing on herbolagy have been thoroughly reviewed. The object herb medicine(9 kinds) is Lophatheri Folium(Herba), Isatidis Folium, Eoriobotryae Folium, Mori Folium, Perillae Folium, Artemisiae argi Folium, Phyllostachys Folium, Clerodendri Folium, Biotae Folium(Cacumen). After this study, additional experimental verification is required as a matter of fact. As comparison materials, this thesis will be utilized.

Antioxidant and Antiatopic Effects of Refined Chaff Liquid Smoke (정제 왕겨초액의 항산화 및 항아토피 활성)

  • The refined chaff liquid smoke purified by the second or third filtering, supplemented with Artemisia extract or Perillae folium extract was investigated by measuring antioxidant and antiatopic activities. In DPPH and FRAP assays, three kinds of chaff liquid smokes had potential in antioxidant activity. Additionally, we assessed for antiatopic activity by using C57BLl6 mice with stimulation of dinitrofluorobenzene and observed that their ear swelling was alleviated by various fractions. Overall, all of the chaff liquid smoke reduced the response of swelling on ear epidermis. Especially, refined chaff liquid smoke supplemented with Perillae folium extract was most active than any other fractions.

Effects of Folium Perillae on Cerebral Ischemia in Rats (소엽(蘇葉) 추출물이 백서(白鼠)의 뇌허혈에 미치는 효과)

  • Objectives : Folium perillae (FP) can relieve superficial pathogenic factors to dissipate cold and promote the circulation of qi and regulate the function of the stomach and is often used for interior qi-stagnation. We hypothesized that FP could rescue cerebral ischemia in rats. Methods : The present study was carried out to investigate the effects of FP on cerebral ischemia in terms of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. Finally, lactate dehydrogenase (LDH) release was investigated, too. Results : In this study, treatment with FP elevated rCBF and MABP levels in dose-dependent manner. Pre-treatment with indomethacin, an inhibitor of cyclooxygenase, inhibited rCBF increase induced by FP effectively. However, FP did not affect stability during cerebral reperfusion. Finally, FP significantly inhibited LD H activity in vitro Conclusions : These results suggest that FP is useful to treat patient with diseases related to cerebral ischemia, because FP can increase rCBF and MABP.

Purification and Properties of Polyphenol Oxidase from Perillae Folium (들깨잎중의 폴리페놀 산화효소의 정제 및 특성)

  • Polyphenol oxidase (PPO) was purified from an extract of Perillae Folium by ammonium sulfate fractionation and sephadex G-150 gel filtration, which molecular weight estimated 65,000$\pm$1,000 in SDS-gel electrophoresis, and pI value was 4.8. The pH and temperature optima were 6.0 and $30^{\circ}C$ respectively. $K_{m}$ values of the PPO for various phenolics derived from Lineweaver-Burk plots were 4.0$\times$10$^{-4}$, caffeic acid; 4.2$\times$10$^{-3}$M, 4-methylcatechol. The inhibition by 4-nitrocatechoi, potassium cyanide, cysteine, 2-mercaptoethanol was competitive with $K_{i}$ values of 7.6$\times$10$^{-5}$M, 7.2$\times$10$^{-5}$M, 3.6$\times$10$^{-5}$M, 2.2$\times$10$^{-5}$M, respectively. Among the divalent cations, Cu$^{2+}$ ion was strong activator on PPO and Zn$^{2+}$, Ni$^{2+}$ ions were little effect on PPO activity. In comparing the amino acid composition of Perillae Folium PPO with that of wheat isozyme, grape, spinach showed similarity. But the content of glycine phenylalanine was abundant relatively.

Depressor Responses to Intravenously Administered Perrillae Folium Juice (PFJ) in Cats (임엽(荏葉)(Perillae Folium)의 혈압강하 작용)

  • The present study was undertaken to investigate effects of Perillae Folium juice on the respiration and blood pressure in cats. Also studied was the mechanism of depressor action of PFJ. The results obtained are as follows; 1) Following administration of 0.2 ml/kg, 0.4 ml/kg and 0.6 ml/kg PFJ into cats the maximum depressor responses observed were 48.5+3.6 mmHg, 56.8+4.3 mmHg and 71.1+2.9 mmHg respectively. 2) Depressor responses to PFJ were blocked makedly by atropine and partially by propranolol. Therefore it is strongly suggested that depressor action of PFJ results mainly from cholinergic effect and partly from activity of ${\beta}-receptor$. 3) After administration of PFJ into cats tachypnea preceded by a short period of apnea was observed invariably.
