Comparison of Resistance of Root Rot Caused by Fusarium solani in Ginseng Breeding Lines (인삼 육성계통의 Fusarium sozani에 의한 근부병 저항성 비교)

  • Root-rot of ginseng caused by Fusarium solani is one of the most obstacles to ginseng cultivation. We evaluated some inoculating techniques of ginseng with Fusarium solani, for selection of disease resistant breeding lines. The most effective inoculating techniques evaluated were inserting toothpicks colonized by F. solani into the seedling roots in laboratory test and dusting seedlings with vermiculite after dipping in conidial sllspension and then replanting method in field test. The resistance to diseased by F. solani was lines of 82022 and 82066 in laboratory test. 82920-1 and 78093 in field test.

  • To confirm the antifungal activity to plant pathogenic fungi, Rhizoctonia solani, Rhizoctonia solani AG2-2, Fusarium oxysporium, Phytophthora dreschler, Alternaria sp. were selected. Helicosporium has the antifungal activity to Rhizoctonia solani, Rhizoctonia solani AG2-2, while Fusarium oxysporium, Phytophthora dreschler did not affected even though Alternaria made a feeble response to that antifungal compounds. From $^1H-NMR$ spectrum of antifungal compound, this compound was guessed to be a structure corresponding to cholesterol.

  • Incidence of stem rot on carnation (Dianthus caryophyllus L.) ranged 11 to 29% in Namwon and Chongup area during the growing seasons from 1993 to 1994. Among 129 isolates from carnations in Namwon, 77 isolates were identified as Rhizoctonia solani, 38 isolates were Fusarium oxysporum and 14 isolates were not identified. Among 169 isolates in Chongup, 19 isolates were identified as R. solani, 106 isolates as F. oxysporum and 44 isolates were not identified. Among 77 isolates of R. solani isolated from the specimens of Namwon, 52 isolates were classified as anastomosis group AG 2-2 by anastomosis test, 14 isolates as AG 2-1 and 11 isolates as AG 4. Among 19 isolates from specimens of Chongup, 14 isolates were classified as anastomosis group AG 2-2 and 5 isolates as AG 4. Pathogenicity tests revealed that isolates of F. oxysporum and R.solani AG 2-2 were highly virulent and isolates of R.solani AG 2-1 and AG 4 were mildly virulent.

Interactions of Virulent and Avirulent Fusarium species on Clonal Asparagus Plantlets and Mechanisms Involved in Protection of Asparagus with Avirulent Fusarium Species Against Stem and Crown Rots (아스파라거스에서 병원성 및 비병원성 Fusarium균의 상호작용과 비병원성 Fusarium을 이용한 아스파라거스 줄기 및 뿌리썩음병 방제 기작 연구)

  • 병원성 Fusarium에 의한 아스파라거스 감염은 비병원성 Fusarium을 5일과 7일 전에 접종하였을 때 방제효과가 있었다. 비병원성 F. oxysporum은 F. moniliforme에 대하여 방제효과가 있었고, F. solani는 F. oxysproum에 대하여 방제효과가 있음이 밝혀졌다. 실험에 사용된 Fusarium 균들은 모두 주근과 측근의 말단 부위, 상처부위, 그리고 표피의 세포벽 사이를 통하여 감염하였다. 경우에 따라 감염하는 동안 appressorium과 유사한 구조를 형성하기도 하였고, 직접 감염하는 경우도 있었다. 병원성 그리고 비병원성 Fusarium 균 모두 공통적으로 생장점 부위를 통하여 감염하였다. 병원성이 강한 Fusarium 균의 경우 비병원성 균들보다 감염의 속도가 빨랐고 더욱 생장이 왕성하였다. F. solani는 생장속도나 기주 조직 침입속도가 매우 느렸다. 기주 감염의 결과 처음에는 cortical rot을 유발시켰고 나중에는 tracheary elements를 감염하고 결국은 조직의 괴사를 유발하는 것이 관찰되었다. 비병원성 F. oxysporum은 표피조직에 두터운 균사층을 형성하였고, 이는 병원성 Fusarium 균에 대한 방제효과를 나타내는 원인을 제공한 것으로 여겨진다. F. solani는 측근의 생성을 촉진시켜 표면적을 증대시킨 것으로 여겨진다. 결론적으로 AVFO와 F. solani를 이용하여 아스파라거스에 발생하는 병원성 Fusarium균의 침입을 저지할 수 있는 생물적 방제가 가능함이 밝혀졌다.

  • The 543 microbial strains were isolated from the rhizosphere in ginseng root rot-suppressive soil. From these, isolate KGM-100 was finally selected for the screening of powerful antagonistic bacterium for the soil-born Fusarium solani causing root rot of many important crops. The isolate KGM-100 was identified as Pseudomonas aeruginosa. Antibiotics produced by the P. aeruginosa KGM-100 were partially characterized as heat-stable and low-molecular weight antibiotics. The strain also produced siderophore, which was assumed to be pseudobactin. Pot test showed approximately 30-50% biological control effects when Fusarium solani-infected soil was treated with the P. aeruginosa KGM-100.

  • Fusarium oxysporum was isolated most frequently, followed by F. moniliforme, and F. solani from infected asparagus plants grown in the field. In pathogenicity tests both with seedlings and plantlets, F. moniliforme showed higher virulence than Fusarium oxysporum did in general. Fusarium moniliforme showed more consistent virulence on both seedlings and plantlets than F. oxysporum did. Fusarium oxysporum showed higher virulence on plantlets than on seedlings. Fusarium solani showed very weak or no sign of virulence on seedlings and plantlets, respectively, in both tests. In protection tests with plantlets, most protection of asparagus against virulent fusarial infections occurred when challenge isolates were inoculated five or seven days after inoculation of protective fusarial species. Avirulent F. oxysporum was a more effective protective agent against infection of F. moniliforme than it was against F. oxysporum. Fusarium solani was more effective against infection of F. oxysporum than it was against F. moniliforme.

Root Rot of Moth Orchid Caused by Fusarium spp.

  • Moth orchid plants with yellowing blight and root rot symptoms were collected, and a total of 54 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 54 isolates of Fusarium spp., 42 isolates were identified as F. solani, 5 isolates as F. oxysporum, and 7 as F. proliferatum. Isolates of the three Fusarium spp. were tested for pathogenicity to moth orchid plants by artificial inoculation. All the Fusarium spp. induced root rot of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused yellowing blight. The symptoms induced on the plants by artificial inoculation with the Fusarium spp. isolates were similar to those observed in greenhouses. The present study reveals that F. oxysporum, F. proliferatum, and F. solani cause root rot of moth orchid, and that F. solani is the main pathogen of the disease.

  • Chromosome numbers were studied for three species of the genus Fusarium from observation of vegetative nuclear division in hyphae with aid of Giemsa-HCl techniques. It was confirmed that observation on the nuclear division could best be made at the growing hyphal tip and near the cells. The general shape of chromosome was dot-like form. The results confirmed that the chromsome number in n=8 in F. solani and F. moniliforme, and n=6 in F. cocophilum.

Characterization of Fusarium solani Causing Fusarium Root Rot of Lisianthus in Korea (꽃도라지 뿌리썩음병을 일으키는 Fusarium solani 의 특성)

  • Lisianthus (Eustoma grandiflorum) is a flowering ornamental plant used widely in Korea. In 2015, wilting, damping-off, stunting, and root rot symptoms were observed in lisianthus plants of Yeoju and Gimhae, Korea. Affected seedlings appeared yellow and showed poor development of root systems in the field and in nursery boxes. Furthermore, affected plants were yellow, stunted, and died at approximately 2-3 months after transplanting. Fusarium species were consistently isolated from the basal stems of diseased plants. Nine isolates were identified as Fusarium solani based on morphological characteristics. Macroconidia of isolates were relatively wide, straight-to-slightly curved, and microconidia formed in false heads on long monophialides. Abundant chlamydospores were produced at the middle or tips of hyphae. To confirm this identification, a molecular analysis of the translation elongation factor 1 alpha (TEF) and RNA polymerase II subunit (RPB2) genes was conducted. The sequences of TEF and RPB2 showed 99.2-99.9% and 98.0-98.1% similarity, respectively, to those of reference F. solani strains in NCBI GenBank. Pathogenicity was tested using root dipping inoculation of healthy lisianthus seedlings. Symptoms were observed within 7 days of inoculation only in inoculated plants. This is the first report of F. solani causing Fusarium root rot on lisianthus in Korea.

Growth Competition between Trichoderma harzianum and Fusarium solani on a Plant Residue in Non-Sterile Soil (토양 식물 잔사에서 Trichoderma harzianum에 의한 식물 병원균 Fusarium solani의 성장 저해)

  • Plant residues serve as substrates for the proliferation and overwintering of plant pathogenic fungi in soil. Effects of the biocontrol fungus Trichoderma harzianum on the colonization of wheat straw by the plant pathogenic fungus Fusarium solani were investigated under different soil moisture regimes (-50 vs. -500 kPa) in non-sterile soil. T. harzianum ThzID1-M3 and/or F. solani were added along with wheat straw to non-sterile soils. ThzID1-M3, other Trichoderma species, and F. solani were monitored for a 21-day period using quantitative PCR. ThzID1-M3 reduced the colonization of F. solani on wheat straw (p < 0.05) under both moisture regimes, and F. solani reduced the colonization by ThzID1-M3 and other Trichoderma species (p < 0.05), thus suggesting competitive inhibition between ThzID1-M3 and F. solani. Colonization by ThzID1-M3 and generic Trichoderma was improved in the wet soil (p < 0.05), but colonization by F. solani did not differ between the two moisture conditions. Thus, the inhibitory effect of ThzID1-M3 was greater in the wet soil (p < 0.05). The growth competition between ThzID1-M3 and F. solani to colonize plant debris suggests that the biocontrol fungus T. harzianum may reduce the potential of the plant pathogen, F. solani, to survive and proliferate on crops.
