Effects of Gastrodia rhizoma on Lipid Components of Serum in Hypercholesterolemic Rats (천마분말이 고콜레스테롤혈증 흰쥐 혈청의 지질성분에 미치는 영향)

  • This study was designed to investigate the optimum recommendation level on Gatrodia rhizoma and the effects on the improvement of the lipids in the dietary hypercholesterterolemic rats. Experimental diets mixed with 5% Dioscorea batatas(Group 2), 10% Dioscorea batatas(Group 3), 15% Dioscorea batatas (Group 4) 5% Gastrodia rhizoma (Group 5), 10% Gastrodia rhizoma (Group 6), and 15% Gastrodia rhizoma (Group 7), were administered to the male rats of the Sprague Dawley for 3 weeks. Concentration of total cholesterol in serum was lower in the Gastrodia rhizoma groups than in the other groups, especially total cholesterol concentration of 10% Gastrodia rhizoma(Group 6) was the lowest in the Gastrodia rhizoma groups. Concentration of HDL-cholesterol in serum was higher in the 10% Gastrodia rhizoma and 15% Gastrodia rhizoma than in the other groups. Concentrations of cholesteryl ester, LDL, LDL-cholesterol in serum were the lowest in the 10% Gastrodia rhizoma. Concentration of glucose and activity of GPT in serum were the lowest in the 10% Gastrodia rhizoma group. The activity of GOT in serum was lower in the 10% Gastrodia rhizoma group and 15% Gastrodia rhizoma group than in the other groups. Therefore, we consider that there are effects on the improvement of the lipids in the Gastrodia rhizoma and the optimum recommendation quantity of Gastrodia rhizoma is 10% to quantity of food composition.

  • The present study tested to recommend optimum level of Gastrodia rhizoma and effect on the compositional changes of the lipids in Sprague Bawley rats which were fed control diet adding 5, 10, 15% Dioscorea batatas powder and 5, 10, 15% Gastrodia rhizoma powder for 3 weeks, respectively The contents of moisture. ash, fat, protein and carbohydrate in dried Gastrodia rhizoma were 5.9, 2.7 2.1, 0.9 and 88.4%(w/w) respectively. Concentrations of total cholesterol and free cholesterol in liver were the lowest in the 10% Gastrodia rhizoma group. Concentrations of triglyceride and phospholipid in liver were lower in the 10 and 15% Gastrodia rhizoma groups than in the other groups. Concentrations of total cholesterol, free cholesterol, triglyceride and phospholipid in brain were significantly lower in the group 6, 7 compared to the other groups. Concentrations of triglyceride and phospholipid in kidney were slightly lower in the Gastrodia rhizoma groups than those in the Dioscorea batatas groups.

  • This study evaluated the effects of Dioscorea batatas and Gastrodia rhizoma on fatty acid composition of serum, liver and brain in rats which were fed control diet adding 5%, 10%, 15% Dioscorea batatas powder and 5%, 10%, 15% Gastrodia rhizoma powder for 3 weeks, respectively. In the other total fatty acid composition of serum lipids, the percentage of SFA was higher in the control group than in the other groups. The percentages 18:2 and 20:4 in serum lipids were significantly higher in the group 5, 6 and 7 than control group. In phospholipid fatty acid composition of serum lipids, the percentage of 18:2 was significantly higher in the group 5, 6 and 7 than in the other groups. 16:0, 18:0, 18:1, 18:2 and 20:4 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of liver lipids. 16:0, 18:0, 18:1, 18:2, 20:4 and 22:6 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of brain lipids.

Combination with Korean Red Ginseng and Gastrodia rhizoma Enhances Vascular Protective Effects in Hyperlipidemic Rats (고지혈증 동물모델에서 홍삼과 천마 혼합투여에 의한 혈관 염증 개선연구)

  • Objectives : This experimental study was designed to investigate the inhibitory effects of combination with Korean red ginseng and Gastrodia rhizoma on vascular dysfunction in high-fat/cholesterol diet-induced hyperlipidemia. Methods : Sprague-Dawley rats were fed with 7.5% cocoa butter and 1.25% cholesterol for 10 weeks, with Panax ginseng (PG), and mixtures of Panax ginseng and Gastrodia rhizoma (PGM), respectively. Results : Chronic treatment with PG and PGM significantly decreased body weight. The aortic expression of cell adhesion molecules such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin were markedly increased in hyperlipidemia rats. Interestingly, PGM significantly decreased cell adhesion molecules expression. However, there was no significant decrease in PG group. In addition, PG and PGM group inhibited high-fat/cholesterol diet-induced cytokine such as monocyte chemoattractant protein (MCP-1) mRNA expression. Furthermore, PG and PGM group significantly decreased c-reactive protein protein (CRP) level. Especially, PGM significantly accentuated the decrease of MCP-1 mRNA expression and CRP level. Conclusions : the present study provides an evidence that combination with Panax ginseng and Gastrodia rhizoma enhances anti-vascular protective effects through suppression of vascular inflammation in hyperlipidemic rats.

Volatile Flavor Constituents in the Rhizoma of Gastrodia elata (천마의 휘발성 향기성분)

  • Crude oils in the rhizoma of fresh and dried Gastrodia elata were obtained by a simultaneous steam distillation and extraction(SDE) method using n-pentane/diethyl ether (1 : 1) as solvent, and their volatile constituents were analyzed by gas chromatography-mass spectrometry(GC/MS) A total of 39 volatile flavor constituents (11 acids, 13 alcohols,6 hydrocarbons,7 carbonyls,2 esters) and 25 constituents (6 alcohols, 13 acids, 4 hydrocarbons, 1 carbonyl, ester) were identified in the fresh and dried Gastrodia elate respectively. The major volatile components of the fresh and dried sample were hexadecanoic acid(66.78%, 50.72%), 9-hexadecenoic acid(8.07%, 9.58%), heptadecanoic acid(2.01%, 0.13%), pentadecanoic acid(6.41%, 4.94%), p-cresol(1.43%,0.52%) and cyclododecene(1.83%, 6.00%).

Beneficial effect of Combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori on Cholesterol and Erectile Dysfunction in Hyperlipidemia rats (홍삼, 천마, 적하수오 병용투여에 의한 고지혈증 랫드에서의 콜레스테롤 및 발기부전 개선효과)

  • Objectives : This study was designed to investigate effects of the combination with Korean Red Ginseng (Panax ginseng C.A. Meyer), Gastrodia Rhizoma (Gastrodia elata Blume) and Polygoni Multiflori Radix (Polygonum multiflorum Thunberg) on metabolic disorders including cholesterol and erectile dysfunction in hyperlipidemia rats.Methods : Animals were divided into six groups; Control with normal diet, high fat/cholesterol-diet (HFCD), fluvastatin, Korean Red Ginseng treated (KRG), and the combination treated (Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori Radix; 1:1:1 for KGP1 and 2:1:1 for KGP2). The experimental groups initially received HFCD for 10 weeks and then treated orally with fluvastatin, KRG, KGP1 and KGP2 during the final 6 weeks. Erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve.Results : KGP2 decreased the level of total cholesterol and LDL cholesterol in the sera of HFCD rats without no changes of body weights. KRG, KGP1 and KGP2 decreased the level of C-reactive protein (CRP) levels except of fluvastatin, synthetic HMG-CoA reductase inhibitor. KRG, KGP1 and KGP2 significantly increased the ICP, ICP/MAP ratio, area under the curve (AUC) compared with those of normal rat. Morphometric analyses showed that KRG, KGP1 and KGP2 increased the volume of smooth muscle and the regular arrangement of collagen fibers in corpus cavernosum of HFCD rats. The penile expression of eNOS was increased by KRG, KGP1 and KGP2.Conclusions : Based on these results, we suggest that the combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori may improve hyperlipidemia through regulating the lipid profiles and erectile dysfunction in rats.

A Study on Functional components, antioxidant activity of Gastrodiae Rhizoma and Gastrodiae Elata floral axis (천마의 근경과 지상부의 성분 및 항산화 활성 연구)

  • Objectives : This study was carried out to know the necessity of freezing and boiling process of Gastrodiae Rhizoma. Also we need to evaluate Gastrodia elata floral axis as a product ingridients. Methods : Frozen Gastrodiae Rhizoma (GF1, GF2) and Gastrodia elata floral axis (GFA) were prepared. They were divided into samples (GF1 : frozen at the freezer, GF2; frozen and boiled for 10 hours, GFA; dried at 7 $0^{\circ}C$ for 120 hours) for experiment. They were extracted using water, freeze dried and powdered. And we analyzed proximate compositions, free sugars, gastrodin, p-hydroxybenzyl alcohol and p-hydroxybenzyl aldehyde content, phenolic and flavonoid, electron donating ability and nitrate scavenging activity and antioxidant activity. Results : In moisture, crude ash, fructose, glucose, sucrose, p-Hydroxybenzyl alcohol GF2 showed lower level than GF1. But GF2 showed higher content than GF1, in crude fat (0.8% > 0.19%), gastrodin ($8.84{\pm}0.58%$ > $4.18{\pm}0.73%$), and p-hydroxybenzyl aldehyde ($2.45{\pm}0.26%$ > $2.07{\pm}0.16%$) content, phenolic ($9.98{\pm}0.07%$ > $3.35{\pm}0.03%$) and flavonoid ($3.01{\pm}0.06%$ > $1.09{\pm}0.04%$) content, electro donating ability ($15.21{\pm}6.51%$ > $10.44{\pm}4.78%$), nitrate scavening activity ($20.43{\pm}5.30%$ > $13.62{\pm}5.78%$). GFA has a relatively lower key indicators component, but has a enough impact on antioxidant effect in phenolic ($11.85{\pm}0.08%$) and flavonoid content ($1.45{\pm}0.03%$), electron donating ability ($18.58{\pm}9.06%$) and nitrate scavenging activity ($19.41{\pm}9.90%$). Conclusions : In the view of proximate compositions, free sugars, functional component and antioxidant activity, the results indicated that boiling process is effective for the frozen Gastrodiae Rhizoma. And Gastrodia elata floral axis has a significantly functional components and antioxidant activity.

Discrimination between steam processed and unprocessed Tubers of Gastrodia elata Blume by HPLC

  • In this study, to evaluate the effectiveness and safety of oral therapy using Gastrodiae Rhizoma, a new HPLC-PDA analysis method was developed for the simultaneous quantitation of the three major components: (1) gastrodin, (2) gastrodigenin, and (3) p-hydroxybenzaldehyde, in steam processed and unprocessed tubers of Gastrodia elata Blume. The clear separation of the three components was achieved on a C18 column (250 × 4.6 mm, 5 ㎛) by gradient elution using water (including 0.1 % formic acid) and acetonitrile as the mobile phase. The flow rate was 1.0 mL/min, and the UV detector wavelength was set at 270 nm. The results demonstrate satisfactory linearity, recovery, precision, accuracy, stability, and robustness. The established HPLC-PDA method was applied to quantify three major compounds in 59 samples of G. elata Blume tubers. Finally, the steam processed and unprocessed tubers of G. elata Blume were successfully distinguished by pattern recognition analysis.
