The Effects of Some Oriental Herbs Which Have Been Used in the Treatment of Alcoholic Diseases on Alcoholic Metabolism and Alcoholic Liver Damages (주상에 활용되는 수종의 한약물이 알콜올대사 및 간장해에 미치는 영향)

  • Objectives : This experiment was conducted to verify the effects of some oriental herbs(Alny Cortex et Ramulus, Artemisiae Capillaris Herba, Aurantii Nobilis Pericarpium, Giseng Radix, Hoveniae Semen, Puerariae Flos, Puerariae Radix, and Xanthii Fructus) which have been used in the treatment of alcoholic diseases, on alcoholic metabolism, and on alcoholic liver damage. Methods : The effects of the herbs on the activities of alcohol dehydrogenase(ADH), aldehyde dehydrogenase(ALDH) were evaluated and their protective effects of liver function and cells from alcoholic damage were analysed. For the evaluation of the protective effects, the levels of glucose, triglyceride, BUN, AST, and ALT in serum of rats were measured. Results and Conclusions : It is concluded that Puerariae Radix interferes with the ADH activity directly, thereby reducing the toxicity of alcohol, resulting in enhancing alcohol-tolerance and protecting liver functions. Also Artemisiae Capillaris Herba interferes both ADH and ALDH activities. Isolation of the biologically active compounds from Puerariae Radix and its detailed characterization are matters for future research.

The protective effect of Red Giseng on the organotin compounds(TBTO) poison : Focusing on the Immunity and sex hormone (유기주석화합물의 독성에 대한 홍삼의 방어효과 - 면역 및 성 호르몬물질 중심으로 -)

  • This study was carried out effect of red ginseng extract(1.0g/kg) on organotin compounds (10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kindey, testis, ovary, serum immuinty and sex hormone activity of rats were examinned by gastric tubing for 3 weeks. The weight of each organ in TBTO treated group were significantly increased other organs which excepted kinedy in males and only liver in females.(p<0.05, p<0.01). In case of Immunity activity of each sex, IgM level was small change comparsion with that of control group in all sex. but IgG level was significantly decreased females rather than males comparsion with that of control group.(p<0.05, p<0.01) In case of sex hormone activity, the testosterone activity of males and the estradiol activity of females were significantly decreased rather than the control group. on the other hand, red singsong treated group was only significantly increased estradiol activity.( p<0.05, p<0.01)

Purification and Characterization of Invertase from Korean Ginseng Panax ginseng (고려인삼(Panax ginseng) Invertase의 정제와 그 특성)

  • In An invertase (EC 3.2.1.26) was extracted from Korean giseng (Panax ginseng C.A. Meyer) with distilled tvater The ginseng invertase was purified about 62.6 folds purified by procedures including ammonium sulfate fractionation , DEAE-cellulofine chromatography and gelfiltrations through Sephadex G-75 and the recovery of enzyme activity was 11.1%. The homogeneity of the purified enzyme was probed by polyacrylamide gel disc electrophoresis. The purifled enzyme was divided into two different subunits by treating with a mixture of SDS and 2-mercautoethanol, and the molecular weight of the large subunit was estimatedtobe 116,000 and that of the small one to be 14,000. The optimal VH and temperature of the enzyme were pH 6 and 45$^{\circ}C$, respectively. The enzyme hydrolyzed specifically the hydrolyzation of the -fructofuranosides such as sucrose, raffinose and inulin. The Km values of the enzyme for sucrose and raffinose were determined to be 0.85 and 0.6 mM, respectively.

The wound healing and anti-inflammatory effects of Panax ginseng C.A Meyer

  • Ginseng has been used as miraculous panacea since ancient times in oriental countries. In spite of voluminous work, ginseng still remains mysterious herb, but its value is becoming more recognized in the pharmaceutical and cosmetic fields. In this study, we investigated the effect of Panax ginseng on wound healing using two experimental methods. First, we studied the effect of ginseng on artificial wound of cultured human keratinocyte monolayer. Indivisual components from ginseng (ginsenoside Rb2, Rc, Re, Rg1, and panasenoside) and giseng extrats were examined. Of them, compared with control, ginsenoside Rb2 and Rg1 needed much shorter time to recover original appearance of momolayer. Second, we investigated the effect of ginseng on acute injury on dorsal skin of hairless mice. We here observed that ginseng has prominent effect than Madecasol(asiaticoside), a well known wound healing agent. These results were deduced that ginseng promoted wound healing in the wound region due to its stimulation of biosynthesis of various endogeneous materials that have relation to wound healing. Furthermore, we conformed that ginsenoside Rg1 exhibited anti-inflammatory activity on rat paw edema induced by carageenan. These results suggest that Panax ginseng C.A Meyer can be used in the cosmetics in that its wound healing and anti-inflammatory effects.

Dose Dependent Effects of Panax Ginseng on the Reproductive Functions in Mice (농도별(濃度別) 인삼(人蔘) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Purpose : This study was conducted to investigate the dose dependent effects of Panax Ginseng on the reproductive functions in mice. Methods : We used the 8-week-old mice, and administered 0.2ml extract solution of Panax Ginseng in the different concentration once a day for 60 days. The control group was administered 0.2ml normal saline in the same way and duration. We counted the total, motile and normal sperm number of the cauda epididymis and measured the activities of sperm hyaluronidase, peroxidase and catalase of the isolated testis tissues. And we observed histological changes of surgically isolated testis by histochemical methods. Results : All Panax Ginseng extract solution groups showed significantly dose dependent differences in the total number, the motility and normality of sperms compared with the control group, respectively. In the histological analysis of the testicular tissues, all Panax Ginseng extract solution groups showed the enlargement of testicular lobe diameter and apparent angiogenesis between seminiferous tubules. And the activity of typical sperm enzyme, hyaluronidase, was significantly increased in the Panax Ginseng extract solution groups compared to the control group. In the antioxidant activity analysis, the activities of peroxidase and catalase were significantly increased in the Panax Ginseng extract solution groups compared to the control group. Conclusion : This study shows that Panax Giseng has the beneficial effects on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and catalase. We can suggest that Panax Ginseng be useful for the treatment of male infertility.

Hypoglycemic Action of Red Ginseng Components (II). Investigation of the Effect of Fat Soluble Fraction from Red Ginseng on Enzymes Related to Glucose Metabolism in Cultured Rat Hapatocytes (홍삼 성분의 혈당강하작용 연구 (ll) :쥐의 배양 간세포의 당대사 관련 효소 활성에 미치는 홍삼 지용성 분획의 영향 조사)

  • In this study, rat hepatocytes known to have active glucose metabolism were obtained to investigate the hypoglycemic action of fat soluble fraction of red ginseng by using the liver perfusion technique and incubated in two different media-one containing insulin and glucagon (control group), and the other containing glucagon only The activities of main regulating enzymes, such as glucokinase, glucose 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenate, and glucose 6-phosphatase, related to metabolic pathways of glucose in these two kinds of hepatocytes were compared between these two groups and the effects of addition of fat soluble fraction ($10^1$~$10^4$%) from red ginseng to these two groups on these enzymes were also detected. The results were as follows. The specific activity of enzymes such as glucokinase, flucorse 6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase related to glucose-consuming pathways of insulin-deficient group was much less than control one. However, their decreased activity was recovered after the addition of fat-soluble fraction at all range of concentrations. The specific activity of these enzymes after the addition of ginseng components to the control group was also increased. On the other hand, the specific activity of glucose 6-phosphatase related to glucose-producing pathway of insulin-deficient group was much higher than control one, but their increased activity was decreased obviously after the addition of fat soluble fraction at all range of concentrations. The same results were observed after the addition of fat-soluble fraction to the control group. These results suggest that the red ginseng saponin components might be effective on diabetic hyperglycemia by regulating the activity of enzymes related to glucose metabolism directly and/or indirectly. The effects of fat-soluble fraction ($10^2$%) and ginsenosides (mixture, $Rb_1$ and $Rg_1$, $10^4$%) on hypoglycemic action were compared. As a result, they showed considerable effect on hyperglycemia, but the best eff ect on the activities of glucokinase and glucose 6-phosphate dehydrogenase was appeared by ginsenoside $Rb_1$ and that of 6-phosphogluconate dehydrogenase and glucose 6-phosphatase was by ginsenoside mixture.

Overexpression of Farnesyl Diphosphate Synthase by Introducing CaFPS Gene in Panax ginseng C. A. Mey. (인삼에서 Farnesyl Diphosphate Synthase 과발현이 진세노사이드 생합성에 미치는 영향)

  • FPS (farnesyl diphosphate synthase) plays an essential role in organ development in plants. However, FPS has not previously been identified as a key regulatory enzyme in triterpene biosynthesis. In order to investigate the effect of FPS on ginsenosides biosynthesis, we over-expressed FPS of Centella asiatica (CaFPS) in Panax giseng adventitious roots. PCR analysis showed the integrations of the CaFPS and hygromycin phosphotransferase genes and we ultimately selected three lines. The result of Southern blot analysis demonstrated the introduction of the CaFPS gene into genome of ginseng. In addition, the results of RT-PCR analysis revealed that CaFPS gene overexpression induced an accumulation of its transcription in the ginseng adventitious roots. To determine whether or not the overexpression of the CaFPS gene contributes to the downstream gene expression associated with triterpene biosynthesis, the level of mRNAs was analyzed by real-time PCR. The result showed that no differences were detected in any expression of all genes. To determine quantitatively the content of ginsenosides in transgenic ginseng adventitious roots, HPLC analysis was conducted. The content of total 7 ginsenosides was increased to 1.8, 1.4, and 1.7 times than that of the controls, respectively. This indicated that the overexpression of CaFPS in ginseng adventitious roots causes an increase in ginsenoside content, although down stream genes of FPS gene were suppressed by CaFPS overexpression.
