  • A biosurfactant producing bacteria strain, D2D2 was selected from diesel-contaminated soil, and identified as Pseudomonas aeroginosa. A glycolipid produced by P. aeroginosa D2D2 was purified by ethyl acetate extraction and adsorption chromatography. The biosurfactant was Identified as glycolipid which has two types of biosurfactants as a results of TLC analysis. The purified glycolipid biosurfactant reduced the surface tension of water to 27 dyne/cm. In time course studies of growth and rhamnolipid production in a minimal salts medium containing 1.5% glucose and 1.5% olive oil, a maximum rhamnolipid yield of $11.45gL^{-1}$ was obtained after 5 days.

  • Biosynthesis of glycolipid from glucosamine and $C_{13:0}$ fatty acid was attempted using an enzyme preparation which was extracted from Selenomonas ruminantium grown in lactic acid medium by means of ultrasonication, and with cofactors of ATP, Co A, $Mg^{++}$, and UTP. The results are summarized as follows: 1. The rate of synthesis of glycolipid from $^{14}C-glucosamine$ and tridecyl CoA by an enzyme from Selenomonas ruminantium was promoted by the presence of co-factors, ATP, Mg, and UTP. 2. The rate of incorporation of $^{14}C-glucosamine$ into glycolipid by the centrifugal fraction of bacterial preparation was the highest in the 105,000 g supernatant fraction, indicating about twice the enzyme activity as the 6,000 rpm supernatant fraction. 3. The biosynthesis of glycolipid from $^{14}C-glucosamine$ and tridecyl-CoA by the crude enzyme of the 105,000 g supernatant fraction of Selenomonas ruminantium cell preparation proceeded the most part of it in 30 minutes and completed in an hour.

  • For myelination, Schwann cells and neuron cells from dorsal root ganglion (DRG) of rat embryos (E16) were cultured in vitro system. The purified DRG cells with anti-mitotic agents and purified Schwann cells were cocultured and then accomplished myelination processing. Treatment of M. leprae-specific phenolic glycolipid-1 (PGL-1) into this coculture system was performed and then accomplished demyelination. Therefore, we identified demyelination processing using antibody of myelin basic protein (MBP).

A Study on the Changes of Fatty Acid Composition of Phospholipid and Glycolipid during Storage Period of Kaesojoo (저장기간에 따른 개소주의 Phospholipid 및 Glycolipid의 지방산 변화에 관한 연구)

  • This experiment was carried out to investigate the changes in phospholipid and glycolipid of Kaesojoo and Kaesojoo added medicinal herbs during storage(30 days) at 4$^{\circ}C$and -18$^{\circ}C$. Two dogs with 12kg live weight(♀, The Korean Jindo dog Hy-breed, 11∼12 month) were slaughtered to obtain samples. The result obtained were as follows: The saturated fatty acids found in phospholipid of Kaesojoo were palmitic acid, stearic acid, myristic acid, while the unsaturated fatty acid found in phospholipid of Kaesojoo with medicinal herbs were palmitic acid, stearic acid, arachidonic acid, while the unsaturated fatty acid were mostly oleic acid, increased during the period of storage. The saturated fatty acid found in glycolipid obtained from Kaesojoo were mostly oleic acid, linoleic acid and linolenic acid. The saturated fatty acid obtained from Kaesojoo with medicinal herbs were palmitic acid, stearic acid, while the unsaturated fatty acid were mostly oleic acid, linoleic acid and linolenic acid. The rates of changes in unsaturated and saturated fatty acid were higher at 4$^{\circ}C$ than that of -18$^{\circ}C$ during storage.

  • The contents of crude fat were remarkably different according to parts in porcine tissues. The most contents of total lipids in variety tissues were neutral lipid, and the nexts were phospholipid and glycolipid, where these of the most composition in liver were phospholipid and the next neutrallipid and glycolipid. The major component of the nonpolar lipids in fresh was triglyceried(58.09∼74.68%) and phospolipid in the polar lipid were phosphatidyl choline(20.14∼45.17%) and phosphatidyl ethanolamine(18.5∼30.14%) and the main component in glycolipid acylsterol glyceride(26.10∼45.15%), respectively.

Growth Inhibition of Microcystis aeruginosa by a Glycolipid-Type Compound from Bacillus subtilis C1

  • We attempted to identify the compound responsible for the growth inhibition of Microcystis aeruginosa occurring when a culture broth of Bacillus subtilis C1 was added to the medium. The active compound was purified from B. subtilis C1 culture broth by adsorption chromatography and HPLC, and was identified as a type of glycolipid based on $^1H$ NMR and MS analyses. The purified active compound completely inhibited the growth of M. aeruginosa at a concentration of 10 ${\mu}g/ml$. This is the first report of a glycolipid produced by a Bacillus strain that has potential as an agent for the selective control of bloom-forming M. aeruginosa.

Glycolipid Biosurfactants Produced by Pseudomonas aeruginosa D2D2 from Diesel-Contaminated Soil

  • A biosurfactant-producing bacterial strain was selected from diesel-contaminated soil by measuring the oil-film collapsing activity and identified as Pseudomonas aeruginosa D2D2. When glucose and olive oil were used as carbon sources, 11.46 g/1 of biosurfactant was obtained. Based on TLC analysis, the biosurfactant produced from P. aeruginosa D2D2 was identified as a glycolipid, consisting of two types of biosurfactants (Type I and Type II). The purified glycolipid reduced the surface tension of the culture from 72 dyne/cm to 27 dyne/cm. The hydrophilic and hydrophobic moiety of the biosurfactant were rhamnose and ${\beta}$-hydroxydecanoic acid, as determined by FAB-MS and NMR analyses, respectively.

  • This experiment was carried out to investigate comparison of total cholesterol, lipid and fatty acid composition of breast and thigh from female and male broiler meats. Total lipid and neutral lipid content of female and male broiler breast meats were lower, but phospholipid and glycolipid contents were highter than thigh meats. Unsaturated fatty acid composition of broiler thigh meats were higher than breast meats on neutral and phospholipid, but breast meat was higher than thigh meat on glycolipid. Glycolipid content in total lipid was lower in female than male broiler meat. Contents of palmitic acid in neutral lipid, palmitic stearic linolenic arachidic and arachidonic acid in phospholipid, palmitic and stearic acid in glycolipid were higher than male broiler meat. The highest content of total cholesterol in defatted tissue was thigh tissue of male and undefatted thigh tissue of female.

  • The change in fatty acid compositions of the glycolipid and phospholipid of sesame seed was analyzed during the storage for 4, 9, and 15 months at four different storage conditions, that is, storage under light with linen bag(LA), storage under light with polyethylene bag(LS), storage in dark with linen bag(DA), and storage in dark with polyethylene bag(DS). In the fatty acid composition of glycolipid, palmitic and stearic acid decreased during the storage but the linoleic acid increased significantly, particularly, between 4 to 9 months of storage. The content of palmitic and stearic acid in phospholipid decreased until 9 months, then increased until 15 months of storage, but reverse tendency was, observed in the content of oleic and linoleic acid. The ratio of unsaturated to saturated fatty acids in glycolipid increased during the storage of 15 months in all the storage conditions. The ratio of unsaturated to saturated fatty acids of phospholipid in DA and LA was slightly higher than that in DS and LS conditions during the storage for 15 months.
