  • The purpose of this study is to examine the ladylike style of Grace Kelly focusing on her costume design of 1950s movie and star image, and to understand how she became a fashion icon today. Her city elegant look was usually composed of A line or H line suits and dresses, pearl jewelry, gloves, handbags and her classic ladylike attitude. Kelly's luxury casual look epitomized a relaxed elegance based on the American sporting image including the Hermes "Kelly" bag. Her sexual elegant look showed the combination of freshness, ladylike virtue and underlying sex appeal. Her self-confident, ladylike style appeals to modern fashionable women who likes to be elegant but also sexy.

Identification and Characterization of a Ringspot Isolate of Odontoglossum ringspot virus from Cymbidium var.'Grace Kelly'

  • An isolate of Odontoglossum ringspot virus (ORSV) was identified from Cymbidium var. 'Grace Kelly' showing ringspot symptom on the floral and leaf parts, and was denoted as cymbidium ringspot isolate (ORSV-CR). In ultrathin sections of leaf tissue from diseased Cymbidium plants, clusters of virus particles were observed in the vacuole and cytoplasm. In the Western blot hybridization, the virus strongly reacted with ORSV-specific antiserum indistinguishable from ORSV, suggesting that the vims is serologically identical with ORSV. ORSV-CR sap was inoculated onto 20 species belonging to 12 genera. Systemic infection occurred in Cymbidium sp., Nicotiana benthamiana and N. clevelandii, the host of which was found to be different from that of ORSV-Cy, the Korean strain of ORSV. The analysis of coat protein (CP) gene showed that ORSV-CR was highly homologous to the known isolates of ORSV, with over 95.6% identity in amino acid level. Phylogenetic tree analysis of CP showed that ORSV-CR was clustered with the known ORSV isolates, suggesting that ORSV is a very stable tobamovirus.

Complementary DNA Cloning of Genomic RNA in Orchid Strain of Tobacco Mosaic Virus

  • Viral RNA was extracted from a purified orchid strain of tobacco mosaic virus (TMV-O) from Cymbidium "Grace Kelly". Polyadenylated viral RNAs were primed with Not I-oligo (dT) primer-adapter. First-strand cDNAs were reversely transcribed by Moloney murine leukaemia virus reverse transcriptase (RNAse H-), and then second-strand cDNAs were synthesized by RNase H and DNA polymerase I. The resulting double-stranded cDNAs were ligated into pSPORT1 vector and transformed into competent E. coli strain JM109 cells. The size of cDNAs within the recombinant plasmids was ranging from 0.9 to 3.9 kb. Among the selected clones, pTMO-0205 and -0210 covered the 3' half and the 5' half of the viral genomic RNA, respectively, which were covering more than 99% of the viral genemo size based on sequencing analysis. Two cDNA fragments which were 3.1 kb BamHI and NotI fragement released from pTMO-0.205 and 3.3 kb SalI and BamHI fragment released from pTMO-0210 were ligated with T4 DNA ligase. The clone was almost entire length, lacking only 31 nucleotides from the 5' terminus based on the sequencing result. This method was shown to be efficiently applicable to other plant viral gnomic RNA for the construction of cDNA.n of cDNA.
