Production of Bacteriorhodopsin by Halobacterium halobium in the Internal Membrane Bioreactor (내부 필터 발효기에서 Halobacterium halobium의 배양에 의한 박테리오로돕신의 생산)

  • Bacteriorhodopsin in the purple membrane (PM) of halobacteria has recently been attracting much attention to be used as a component of molecular electron device and optical computers. In order to increase the productivity of bacteriorhodopsin in high cell density cultures of Halobacterium halobium R1, an internal membrane cell-retention bioreactor system was employed. As a result, the production of cell mass at OD660 of 12 and of bacteriorhodopsin at 125-130 mg/L were obtained using the internal membrane bioreactor system at a dilution rate of 0.066 hr-1. The productivity achieved by the internal membrane system (0.7 mg/L$.$hr) was 3.5-fold higher than that obtained by the corresponding batch cultivations (0.2 mg/L$.$hr).

Conserved COG Pathways and Genes of 122 Species of Archaea (고세균 122종의 보존적 COG pathways와 유전자)

  • The purpose of this study was to identify conserved metabolic pathways and conserved genes in 122 archaeal species. Using the Clusters of Orthologous Groups of Proteins (COG) database of conserved genes, we analyzed whether 122 species had 63 COG metabolic pathways, the 822 COGs that compose them, and a total of 4,877 COGs. Archaeal ribosomal proteins were the most conserved in metabolic pathways. 46 COGs in seven COG pathways among 63 COG pathways and 20 COGs in others were conserved in 122 species. Some genes involved in cell wall and extracellular matrix synthesis, replication, transcription, translation, and protein metabolism were common to all 122 species. When the distance value of the phylogenetic tree was analyzed at the phylum level or class level, the average was the lowest at the class Halobacteria of the phylum Euryarchaeota. Standard deviation was high for the class Nitosospharia of the phylum Thaumarchaeota, the unclassified members of phylum Thaumarchaeota, the class Halobacteria of the phylum Euryarchaeota, the class Thermoprotei of the phylum Crenarchaeota, and other archaea. Furthermore, the phylogenetic tree analysis revealed six commonalities. The results of this study, along with data on conserved genes, could be used for drug development and gene selection for strain improvement.

  • The genes of Halohacteria. gvpD and gvpE. take part in formation of gas vesicle. These mRNA cause a lot of experimental prohlems due to its eharacteristic instahility in the analysis of transcripts. This study allowed easy cloning and sequencing of RNA hy substituting a stable complementary DNA for the mRNA of the genes for an analysis. The weak 111 RNA was reverse transcribed to DNA using reverse transcriptase. and was amplified using PCR method. The transcripts confirmed in this ~,tudy have not heen round in the northern hybridization covering almost all ranges of ORF of the gene. gvpD.

Production of Poly-3-Hydroxyalkanhoate by Haloarcular sp. EH-1 (Haloarcular sp. EH-1으로부터 생분해성 Poly-3-Hydroxyalkanoate의 생산)

  • The extremely halophilic archaebacterium Haloarcular sp. EH-1 was isolated from solar salts. Haloarcular sp. EH-1 accumulated poly(3-hydroxyalkanoate) (PHA) as intracellular granules. PHA production in batch culture have been studied. The PHA was identified as poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid) (PHB/HV) of 3-hydroxybutyric acid and 3-hydroxyvaleric acid by the analysis of GC, IR and NMR. The melting temperature of PHB/HV was 152.46$^{\circ}C$, viscosity was 1.25 ㎗/g, and molecular weight was $1.44 X 10^5.$
