Anti-Aging Effects of the Hanwoo Leg Bone, Foot and Tail Infusions (HLI, HFI and HTI) on Skin Fibroblast

  • Many researchers revealed that collagen contribute to maintaining the skin’s elasticity and inhibit wrinkling of skin. Korean native cattle (Hanwoo) bone (leg bone, foot and tail) infusion contains the various inorganic materials, collagen and chondroitin sulfate. All of this, a large quantity of collagen is included in Hanwoo infusion. Therefore, this study emphasized on the effects of collagen in the Hanwoo bone infusion. For the first time, Hanwoo bone infusions were directly added to the media of Human Dermal Fibroblast (NHDF-c) to test anti-aging effects. First, it was identified that growth rate of skin fibroblast was increased. Furthermore, the Hanwoo bone infusion increased a 50% of fibroblast collagen synthesis. Also, suppression of skin fibroblast aging was confirmed by treatment Hanwoo bone infusion. In conclusion, this study demonstrates the effects of infusion made from Hanwoo leg bone, foot and tail on anti-aging, wrinkle inhibiting and skin fibroblast elasticity maintaining. Therefore, this study identified that traditional infusion has effects that are good for skin elasticity.

Effects of Italian ryegrass diet on the performance, carcass characteristics, and blood metabolites of Hanwoo steers

  • This study was conducted to develop a pasture feeding program for reducing the Hanwoo feeding cost. Italian ryegrass (IRG) is a major plant which is abundantly produced in the southern area of South Korea. Locally produced Italian ryegrass has a 30 - 50% reduced pasture cost compared to imported pasture grasses. We hypothesized that Italian ryegrass feed could reduce the feeding cost but not change the physiological characteristics of the Hanwoo steers in the feedlot. Four treatments (Control, short fed IRG silage, long fed IRG silage, and IRG hay) in a completely random design were used to feed 27 Hanwoo steers. Three or four steers were fed in the same pen, and 8 pens were used for the treatments. Blood was drawn from each steer on the first week of every month from 11 to 30 months. Overall, the average daily gain and feed efficiency were not different among the treatments (p < 0.05) except for the final fattening diets (p < 0.05). The long fed IRG silage group had a low ADG compared to the other groups (p < 0.05). The serum non-esterified fatty acid and glucose concentrations were reduced during the fattening periods (p < 0.05); however, the serum albumin, total proteins, triglycerides, and phosphorus were increased during the fattening periods (p < 0.05). The carcass weight, longissimus muscle area and back-fat thickness were low in the long fed IRG silage group (p < 0.05). The IRG diet induced saturated fatty acids in the longissimus dorsi (p < 0.05). These results show that the IRG diet increased the serum triglycerides but reduced the glucose level at the final fattening diet and decreased the meat yield score in Hanwoo steers.

Analysis of epididymal sperm from Korean native bull (Hanwoo) aged at 8 and 15 months before freezing and after thawing

  • The recovery of epididymal sperm in animals is considered as one of the important tools to preserve high value or endangered species. However, there are no appropriate castrating indicators such as months of age in bull, sperm morphology, and motility, particularly in young Korean native bull (Hanwoo). Therefore, this study aimed to investigate sperm number, morphology, and motility of sperm in the epididymis tail of young Hanwoo bulls at 8 and 15 months of age. After castration, epididymal tails were collected and minced with blades to recover sperm. In experiments 1 and 2, sperm number, morphology, and motility were examined. Total number of sperm and percentage of normal sperm from bulls at 8 months of age was lower than that of bulls at 15 months of age after collection (P<0.05). Percentage of abnormal head, tail, proximal cytoplasmic droplet, dead and damaged acrosome of sperm from bulls at 8 months of age were higher than those of bulls at 15 months of age (P<0.05). In experiment 3, sperm motility from bulls at 8 and 15 months of age were examined before freezing and after thawing. Frozen-thawed sperm at 8 months of age showed low total motility and motile sperm with ${\geq}25{\mu}m/sec$ compared to those at 15 months of age and commercially-used sperm (P<0.05). In conclusion, sperm derived from the epididymal tail of bulls at 8 months of age showed high abnormal morphology and poor motility, which are not adequate for AI and IVF. On the other hand, sperm derived from the epididymal tail of bulls at 15 months of age showed high normal morphology and motility.

한우 FASN 유전자변이와 한우 지방형질 및 지방산조성과의 연관성 분석 (Association of genetic polymorphism in fatty acid synthase with BodyFat and fatty acid composition in Hanwoo)

  • The aim of this study was to identify the polymorphism on fatty acid synthase (FASN) gene and its association with fat traits and fatty acid composition in Hanwoo. We genotyped a SNP (g.16024G${\rightarrow}$A) detected in Exon34 of FASN on 90 Hanwoo steers by PCR RFLP. A linear mixed model revealed an association of g.16024G${\rightarrow}$A with total body fat contents (P=0.006), while there is no significant effect between g.16024G${\rightarrow}$A and other traits (IMF, BF and fatty acid composition). Regression coefficient and $R^2$ of the SNP was -1.5 kg and 0.36 in this anslysis. Expecially, AA type of g.16024G${\rightarrow}$A bas a less amount of body fat (1.5 kg) than GG type of the SNP in Hanwoo. In conclusion, this study indicates an important role for FASN gene in determining body fat content in Hanwoo.

Biological effects of dietary probiotics on blood characteristics in Hanwoo heifers subjected to lipopolysaccharide (LPS) challenge

  • This study hypothesized that dietary feed additive containing probiotics alter either immune-related serum substances or serum metabolites in Hanwoo heifers. A probiotic treatment was given at 0.5% top-dressing of concentrate diet for 6 months. The change of immunological indicators in the blood was analyzed under LPS (Lipopolysaccharide) challenge. One day before administration of LPS, all heifers were fitted with an indwelling jugular vein catheter for serial blood collections. Both a serum tube and an EDTA-coated tube were collected at 30-min intervals from - 2 to 8 hours relative to the LPS challenge at time 0 ($1{\mu}g/kg$ of BW). Serum was used for analyzing albumin (ALB), glucose (GLU), total protein (TP), triglycerides (TG), phosphorus (IP), and non-esterified fatty acids (NEFA). Plasma was used for analyzing white blood cell (WBC), red blood cell (RBC), platelet (PLT) and inflammation-related factors (NE, LY, MO, EO, BA, Hb, HCT, MCV, MCH, MCHC, RDW, MPV). There were significant differences in ALB, GLU, TG, IP, and NEFA concentration with the passage of hours post challenge (p < 0.05). The level of ALB, GLU, TG, and IP showed significant difference (p < 0.05) between treatments. However, none of the data showed interaction between time and treatments (p > 0.05). The level of WBC, EO, LY, and MO were reduced after LPS challenge (p > 0.05). In conclusion, LPS challenge after dietary supplementation of probiotics changed the levels of both serum metabolites and inflammation-related factors. The increase of GLU and TG indicated a probiotics-positive response under LPS challenge (p < 0.05).

  • Hanwoo cattle are an important food source in Korea and their supply can have a major impact on meat availability for Korean consumers. The Hanwoo population was 1.8 million head in 2005 and gradually increased to 2.6 million in 2015. Per capita beef consumption has also increased, to 11.6 kg per year in 2015, and is expected to continue to increase. Because intramuscular fat percentage is a critical contributor to meat quality, Hanwoo cattle are fed a high-energy corn-based diet for long fattening periods. Long fed diet causes significant alterations in fat percentage in the loin muscle and other areas of the carcass. However, these long feeding periods increase feeding costs and beef prices. Recently, there has been increased Korean consumer demand for lean beef which has less fat, but is tender and priced more reasonably. These consumer demands on the Korean beef industry are driving differing beef production systems and also changes to the beef grading methodology. Korean government has made a significant investment to select bulls with favorable production traits using progeny testing. Progeny tested bull semen has been disseminated to all Hanwoo farmers. A beef traceability system has been employed for all cattle breeds in Korea since 2009. Hanwoo cattle are ear-marked with a 12-digit identification number from birth to slaughter. This number allows traceability of the management history of individual cattle, and also provides information to consumers. Traceability including management information such as herd, farm, year of birth, and carcass data can determine estimated breeding values of Hanwoo. For a sustainable Hanwoo industry, research scientists in Korea have attempted to develop feeds for efficient fattening periods and precision feeding systems based on genetic information for Hanwoo cattle. These initiatives aim to Korean consumer demands for beef and provide more precision management in beef production in Korea.

Transcript profiling of expressed sequence tags from intramuscular fat, longissimus dorsi muscle and liver in Korean cattle (Hanwoo)

  • A large data set of Hanwoo (Korean cattle) ESTs was analyzed to obtain differential gene expression results for the following three libraries: intramuscular fat, longissimus dorsi muscle and liver. To better understand the gene expression profiles, we identified differentially expressed genes (DEGs) via digital gene expression analysis. Hierarchical clustering of genes was performed according to their relative abundance within the six separate groups (Hanwoo fat versus non-Hanwoo fat, Hanwoo muscle versus non-Hanwoo muscle and Hanwoo liver versus non-Hanwoo liver), producing detailed patterns of gene expression. We determined the quantitative traits associated with the highly expressed genes. We also provide the first list of putative regulatory elements associated with differential tissue expression in Hanwoo cattle. In addition, we conducted evolutionary analysis that suggests a subset of genes accelerated in the bovine lineage are strongly correlated with their expression in Hanwoo muscle.

학교급식의 한우 이용실태 조사 및 저지방 한우 부위를 이용한 학교급식 메뉴 개발 (Survey on Hanwoo Usage and Menu Development using Lean Cut Hanwoo for School Foodservice)

  • The purpose of this study is to analyze Hanwoo usage in school foodservices and to develop menus of lean cut Hanwoo. As a result, satisfaction of students, parents and teachers all increased regardless of school type after using Hanwoo. The high price compared to other meats (3.98) was the biggest difficulty for using lean cut Hanwoo followed by reduced taste preferences (2.84), lack of parent awareness (2.67) and lack of appropriate cooking facilities (2.13). According to the survey on how to increase the usage of lean cut Hanwoo, improvement of meat tenderness (3.80), development of various sauces (3.74) and development of seasoning for existing menu (3.61) were identified as major necessities. This analysis of Hanwoo utilization in school foodservice was used to develop menus of lean cut Hanwoo. The menu was particularly developed to overcome the supply-demand imbalance of different cuts. For the menu development process, potential Hanwoo utilization in school foodservice, cooking method, and diverse taste were considered. In order to evaluate the applicability in school foodservice, JARS was used and the final eight menus were selected. For the selected menu, a nutrition analysis was conducted. Further, systematic recipes with a mixing ratio;product process description and a simplified product process were also developed.

Analysis of extended haplotype in Korean cattle (Hanwoo) population

  • Korean cattle (Hanwoo) are categorized into three breeds based on color: brown, brindle, and black. Among these breeds, brown Hanwoo has been subjected to intensive selection to improve meat traits. To identify genetic traces driven by recent selection in brown Hanwoo, we scanned the genomes of brown and brindle Hanwoo using a bovine SNP chip. We identified 17 candidate selection signatures in brown Hanwoo and sequenced four candidate regions from 10 individuals each of brown and brindle Hanwoo. In particular, non-synonymous SNPs in the ADSL gene (K88M, L189H, and R302Q) might have had mutational effects on protein structure as a result of altering the purine pathway during nucleotide breakdown. The ADSL gene was previously reported to affect meat quality and yield in livestock. Meat quality and yield are main breeding goals for brown Hanwoo, and our results support a potential causal influence of non-synonymous SNPs in the ADSL gene.

한우 종모우의 지방괴사증에 대한 Isoprothiolane 투여효과 (Effects of Isoprothiolane administration on fat necrosis of Hanwoo(Korean Cattle) sire)

  • The effect of isoprothiolane(di-isopropyl-1,3-dithiolan-2-ylidenemalonate) aganist fat necrosis in Hanwoo(Korean Cattle) sire was evaluated. The 10 heads of Hanwoo sire suffering from fat necrosis were given 50mg/kg body weight of isoprothiolane(0.2g/kg of Fujix, Japan) orally once a day for 8 weeks. In 30% of these, the size of the necrotic fat masses had decreased significantly 7 months after the adminstration. Isoprothiolane did not affect on live body weight and semen characteristics. However the sire affected with fat necrosis had higher MCHC(Mean corpuscular hemoglobin concentration) than normal sire in hematologic values 10 weeks after administration. Number of RBC(red blood cell) and PCV(packed cell volume) 10 weeks after administration had been increased than those before administration(p < 0.05). The serum concentrations of creatinine, triglyceride, and total cholesterol 10 weeks after administration were higher than those before administration while the concentration of glucose was vice versa. The isoprothiolane may reduce the oxidation of glucose, increase the glucose transfer to lipids, and increase blood supply to necrotic masses. These results indicate that isoprothiolane may be useful as the therapeutic agent aganist fat necrosis.
