Stomach Contents of the Sea Urchins, Anthocidaris crassispina and Hemicentrotus pulcherrimus and Characterization of the Marine Algal Community along the Tongyeong Coast of Korea (통영연안에 서식하는 보라성게(Anthocidaris crassispina)와 말똥성게(Hemicentrotus pulcherrimus)의 위 내용물과 서식지 해조군락)

  • The seaweed community and stomach contents of the dominant herbivorous sea urchins Anthocidaris crassispina and Hemicentrotus pulcherrimus were examined from November 2008 to October 2009, in coastal areas of Tongyeong, Korea. Thirty-five seaweeds including two green, nine brown, and 24 red algae, were found over the study period. Seventeen seaweeds (two green, two brown, and 13 red) and two invertebrates were found in the stomachs of A. crassispina. In H. pulcherrimus, stomach contents were two green, one brown, 11 red seaweeds, and two invertebrates. The shell diameter of A. crassispina and H. pulcherrimus ranged from 22.3 to 62.3 mm and 15.3 to 40.1 mm, respectively. Total body weights ranged from 48.5 to 86.7 g for A. crassispina and from 7.7 to 25.9 g for H. pulcherrimus. The total weight of stomach contents were 5.1 to 25.8 g in A. crassispina and 1.7 to 11.8 g in H. pulcherrimus. The range of gonad weight was 3.6 to 17.0 g in A. crassispina and 0.8 to 4.0 g in H. pulcherrimus. The gonad index (GI) for A. crassispina peaked in July and reached a minimum in December, whereas the GI for H. pulcherrimus was highest in February, and lowest in May.

Effects of Salinity and Standard Toxic Metal(Cu, Cd) on Fertilization and Embryo Development Rates in the Sea Urchin(Hemicentrotus pulcherrimus) (염분과 표준 독성물질(Cu, Cd)이 말똥성게(Hemicentrotus pulcherrimus)의 수정 및 배 발생률에 미치는 영향)

  • Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Hemicentrotus pulcherrimus. Spawning was induced by injecting $1{\sim}2$ mL of 0.5 M KCl into the coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. The fertilization rate was below 30% when salinity was 20 psu and lower, but was almost above 90% when salinity was 25 psu and higher. The embryo development rate was below 60% when salinity was 25 psu and lower, but was above 80% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition(not including Cu and Cd) were greater than 90%, but decreased a high negative correlation with the increasing of Cu(r=-0.80, r=-0.78) and Cd(r=-0.90, r=-0.82) concentrations, respectively. The fertilization and embryo development rates were significantly inhibited in the addition of Cu($EC_{50}$=17.30 ppb, $EC_{50}$=10.32 ppb) and Cd($EC_{50}$=364.57 ppb, $EC_{50}$=244.04 ppb), respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of H. pulcherrimus are above 25 psu and 30 psu, respectively, and the biological assays of fertilization and embryo development rates using H. pulcherrimus are useful methods for the ecological toxicity test of marine pollution elements.

Effect of Bisphenol A on Early Embryonic Development and the Expression of Glutathione S-transferase (GST) in the Sea Urchin (Hemicentrotus pulcherrimus) (말똥성게(Hemicentrotus pulcherrimus)의 초기배아 발생과 glutathione S-transferase (GST)의 발현에 대한 bisphenol A의 영향)

  • In this study, gametotoxicity and embryotoxicity experiments using Hemicentrotus pulcherrimus were carried out to investigate the ecotoxicological effects of bisphenol A (BPA). We examined the effects of BPA on fertilization and normal embryogenesis at various concentrations (0, 300, 500, 800, 1000, and 1500 ppb). The results demonstrated that the fertilization rates were not changed. The normal embryogenesis rates were gradually decreased in a dose-dependent manner, and were significantly lowered following 800 ppb BPA treatment ($EC_{50}$=1056.1 ppb, 95% Cl=981.8~1163.9 ppb). The observed effective concentration and the lowest observed effective concentration of the normal embryogenesis rate were 500 ppb and 800 ppb, respectively. The embryos showed retarded development at each tested concentration, indicating the fact the embryonic development was delayed due to the increasing concentrations of BPA. Furthermore, we examined the expression of glutathione S-transferase (GST) mRNA at various concentrations of BPA in H. pulcherrimus. Interestingly, it was found that the expression level of GST mRNA was significantly increased in the experimental group exposed to BPA. Based on these results, we suggested that BPA at greater than 800 ppb has a toxic effect during the early embryonic stages of H. pulcherrimus, and GST mRNA may be used as a biomarker for risk assessment of BPA contamination.

Toxicity Assessment of Ocean Dumping Wastes Using Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus) (말똥성게 (Hemicentrotus pulcherrimus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가)

  • Toxicity assessment of ocean dumping wastes (dye waste, urban sewage, food waste) were examined in the fertilization and embryo development rates of the Sea Urchin, Hemicentrotus pulcherrimus. Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min the collection of both gametes. The fertilization and embryo development rates test were performed for 10 min and 64 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but suddenly decreased with increasing of ocean dumping waste sludge elutriate concentrations. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste ($EC_{50}$=4.37; $EC_{50}$=1.76), urban sewage ($EC_{50}$=5.79; $EC_{50}$=2.00) and food waste ($EC_{50}$=7.68; $EC_{50}$=2.16), respectively. The NOEC (<3.13) and LOEC (3.13) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest that biological assay using the fertilization and embryo development rates of H. pulcherrimus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

Toxicity Tests of Seawater on the Developmental Stages of Sea Urchin, Hemicentrotus pulcherrimus, in Ulsan Bay (말똥성게, Hemicentrotus pulcherrimus의 발생과정에 대한 울산만 해수의 독성조사)

  • Toxicity of surface seawater in Ulsan Bay on a sea urchin, Hemicentrotus pulcherrimus was assessed to investigate the correlation between the concentrations of five heavy metals (Fe, Co, Cu, Cd, and Ni) and the impairments during the developmental stage of the sea urchin eggs. The tests during the six developmental stages of the eggs, are fertilization rate, cell 4, cell 4, polyspermic cleavage, permanent blastula and exogastrula. Highly positive correlations were found between the concentrations of Cu, Co and Ni and the toxicities on the eggs. Such tests will be useful as an indicator of seawater quality.

The Effects of Temperature on Biological Evaluation of Seawater with Seaurchins (성게를 이용한 해수의 생물학적평가에 있어 온도가 미치는 영향)

  • This study was done to research the effects of temperature on biological evaluation of seawater quality using gametes, embryos and early development system of seaurchin species, Hemicentrotus pulcherrimus, Anthocidaris crassispina and Scaphechinus brevis. As the result of performing effects of temperature on early embryo development, the conditions of appropriate temperature on formation of normal pluteus were appropriate at 5-16$^{\circ}C$ for H. pulcherrimus, 8-2$0^{\circ}C$ for A. crassispina, 12-2$0^{\circ}C$ for S. brevis. The conditions of optimum temperature on biological evaluation were 16$^{\circ}C$ for H. pulcherrimus and 2$0^{\circ}C$ for A. crassispina and S. brevis.

Effects of Lead and Zinc on the Fertilization and Embryo Development of the Sea Urchin (Hemicentrotus pulcherrimus) (납과 아연이 말똥성게(Hemicentrotus pulcherrimus)의 수정 및 배아 발생에 미치는 영향)

  • The individual toxicity of lead (Pb) and zinc (Zn) has been investigated by using the sea urchin (Hemicentrotus pulcherrimus) germ cell and pluteus-larvae. The gametotoxic and embryotoxic effects of Pb and Zn on H. pulcherrimus were each investigated at 31, 63, 125, 250, 500 ppb and 16, 31, 63, 125, 250 ppb, respectively. Spawning was induced by 0.5 M KCl solution and the fertilization and normal embryogenesis rates test were performed for 10 min and 64 h after fertilization, respectively. In exposure to Pb, the fertilization rate was not significantly changed compared with control but normal embryogenesis rate was significantly decreased with concentration dependent manner. Fertilization and normal embryogenesis rates showed a significant decreased with concentration dependent manner in exposed to Zn. The normal embryogenesis rates were significantly inhibited in exposed to Pb ($EC_{50}$=45.13 ppb, 95% Cl=40.12~50.05 ppb) and Zn ($EC_{50}$=19.82 ppb, 95% Cl=18.26~21.31 ppb). In exposure to Pb and Zn, the NOEC of normal embryogenesis rate was <31.25 and <15.63 ppb, respectively. The LOEC showed each 31.25 and 15.63 ppb in exposed to Pb and Zn. These results suggest that the early embryo development of H. pulcherrimus is highly sensitive to heavy metals such as Pb and Zn, H. pulcherrimus can be used as a test organism for risk assessment in marine ecosystems.

  • Tagged and untagged sea urchins, Hemicentrotus pulcherrimus (A. Agassiz) were reared for 84 days in the laboratory, Vinyl tape and trapall paper were used as tag materials. The natural mortality, mortality by predation and shedding rate of tags of the tagged groups were examined, and the results were compared with those of untagged control group. The results of Chi-square test of natural mortality between tagged and control groups were very significative, however, the comparison between the two tagged groups was insignificative. No effect of tag attachment seems to be present at the point of natural predation by starfish, and $x^2-test$ between two the tagged groups on the loss rate of tags showed also insignificance. The mortality shortly after tagging seems to be negligible, and the distinct decreasing effect on growth due to tag attachment was not revealed because of insufficient individuals and rearing periods.

Toxic Effects of Arsenic and Chromium on the Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus) (말똥성게(Hemicentrotus pulcherrimus)의 수정 및 배아 발생률에 대한 비소와 크롬의 독성영향)

  • Toxic effects of arsenic (As) and chromium (Cr) has been investigated using the sea urchin (Hemicentrotus pulcherrimus) germ cell and pluteus-larvae. The gametotoxic and embryotoxic effects of As and Cr on H. plucherrimus were each investigated at 6.25, 12.5, 25, 50, 100. Spawning was induced by 0.5 M KCl solution and the normal fertilization and embryogenesis rates were performed for 10 min and 64 hrs after fertilization, respectively. The normal fertilization and embryogenesis rates in the control condition (not including As and Cr) were greater than 94% and 93%, respectively. The fertilization rate was not significantly changed compared with control but embryogenesis rate was significantly decreased with concentration-dependent manner. As and Cr reduced normal embryogenesis rates and a significant reduction occurred at concentration greater than 6.25 ppb (P<0.01) and 25 ppb (P<0.05), respectively. The lowest-observedeffect- concentration (LOEC) of normal embryogenesis rate in As and Cr were each 6.25 and 25 ppb, respectively. From these results, normal embryogenesis rate of H. pulcherrimus have toxic effect at greater than the 6.25 ppb concentration of As and 25 ppb concentration of Cr in marine ecosystems. These results suggest that the normal embryogenesis rates of H. pulcherrimus are very useful test method for the toxicity assessment of heavy metal as As and Cr in marine ecosystems.
