Molecular Identification of Asian Isolates of Medicinal Mushroom Hericium erinaceum by Phylogenetic Analysis of Nuclear ITS rDNA

  • A reliable molecular phylogenetic method to identify Hericium erinaceum, the most industrially valuable species in the Hericium genus, was established. Sequencing and phylogenetic analyses of the PCR-amplified ITS and 5.8S rDNA from Hericium fungi, including 6 species and 23 isolates, showed that variation in nucleotide sequences and size exists in both ITS1 and ITS2 regions, but not in the 5.8S region. These two ITS regions provided different levels of information on the relationship of H. erinaceum to other Hericium species. Based on the ITS1 sequence, both the parsimony and neighbor joining trees clearly distinguished Asian H. erinaceum isolates from other Hericium species and isolates. The intraspecific divergence of the ITS2 region was suitable to dissect the Asian H. erinaceum isolates into a few groups.

Quality Characteristics of Jook Prepared with Hericium erinaceum Powder (노루궁뎅이버섯(Hericium erinaceum) 분말을 첨가한 죽의 품질 특성)

  • The study was conducted to evaluate the effects of Hericium erinaceum Powder (HEP) on the quality characteristics of Jook. The proximate composition of HEP was as follows: moisture, 6.10%; crude protein, 32.69%; crude lipid, 4.63%; crude ash, 11.40%; and carbohydrates, 45.18%. Each sample contained 3, 6, and 9% Jook of Hericium erinaceum Powder (JHEP) compared to the control and their physicochemical properties assessed. As the level of HEP increased, there was a significant decrease in pH (p<0.05) and significant increase in total acidity (p<0.001). In addition, L and a values decreased, whereas b value increased with increasing addition of HEP. The Jook showed lower spreadability and higher viscosity values as its content increased. According to the results, addition of HEP positively affects the quality characteristics of Jook.

  • Recently proteasome inhibitors have been emerged as potential anticancer agents. In a continuous study on exploring proteasome inhibitors from natural products, fruiting body of Hericium erinaceum was investigated. The MeOH extracts of the fruiting body of Hericium erinaceum was fractionated with several solvents and the fractions were evaluated on the activity to screen the proteasome inhibitors. The n-Hexane and CHCl3 frs. showed potent activity, of which chemical investigations led to ergosterol peroxide (1), hericenones C (2) and D (3). Their structures were determined by spectroscopic methods such as $^1H$-NMR, $^{13}C$-NMR, and FABMS spectra.

  • Lion's Mane Mushroom, Hericium erinaceum, is a traditional edible mushroom widely used in culinary applications. It has been also used as a medicine in East Asian countries due to its various biological activities. Chemical investigation of fruiting bodies of this mushroom afforded many aromatic compounds, which were mostly isolated from polar fraction of its extracts. Herein we tried to investigate non-polar compounds from the extracts of this mushroom. $CHCl_3$-soluble fraction of the extracts was subjected to chemical investigation, which resulted in isolation of four compounds. Their chemical structures were elucidated as ircicerebroside (1), cortenuamide A (2), 1-D-arabinitol-monolinoleate (3), and cinnamic acid (4) on the basis of spectroscopic data. To the best of our knowledge, this is the first report of compounds 1, 2, and 4 from Hericium erinaceum.

Enhanced Activities of Alcohol Methabolism by Extracts from Hericium erinaceum Hypha Cultivated with Artermisia capillaris(HEAC) (인진쑥 배지에서 배양한 노루궁뎅이버섯 균사체 추출물(HEAC)의 알코올 대사촉진 활성)

  • Alcohol concentration in the blood was effectively decreased by extracts from Hericium erinaceum hypha cultivated with Artermisia capillaris medium(HEAC), Hericium erinaceum hypha and Artermisia capillaris after dirnking. Also, the activities of alcohol dehydrogenase and acetaldehyde dehydrogenase in the blood was studied. As a result of testing an alcohol concentration in the blood, the alcohol in the blood was not detected after 170 min, in case of HEAC and after 210 min, in case of Hericium erinaceum. Compared to control, each activities of alcohol dehydrogenase of HEAC and Hericium erinaceum hypha was showed up to 154% and 148% respectively. The activities of the acetaldehyde dehydrogenase of both extracts from HEAC and Hericium erinaceum was maintained in the range of 104 to 110% compared to control. In conclusion, such extracts represent significant effect to facilitate decomposition of alcohol.

Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System (Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up)

  • For the study of Hericium erinaceum as a useful functional foods and materials, liquid cultivation under two different bioreactors(air-lift fermenter and jar fermenter) which was not studied systematically until now, was conducted as a method of mass cultivation for H. erinaceum. A batch cultivation in an air-lift fermenter and a jar fermenter was examined for enhancing the productivity because of small amounts of mycelial weight and slow growth in case of a liquid culture for H. erinaceum. We found that air lift fermenter system was more effective than jar fermenter for mycelial production of H. erinaceum, and mycelial morphology was a critical factor of the growth. By scale-up and cultivation based on morphological analysis, the conditions for mass production with 30 L and 500 L jar fermenter was 200 and 150 rpm of agitation speed at 1 vvm of aeration rate, respectively, and mycelial dry weight under these conditions was enhanced to about $13{\sim}14g/L$.

Development of Functional Drink Using the $\textit{Hericium erinaceum}$ Cultivated on the $\textit{Angelica keiskei}$ (신선초를 이용한 $\textit{Hericium erinaceum}$ 음료 개발에 관한 연구)

  • 본 연구는 생물공학적인 방법을 도입하여 폐기되는 신선초박에 H. erinaceum의 균사체를 발효시킨 배양생성물을 이용하여 기능성 건강 음료 개발을 검토 하고자 하였다. 1. 종균제조공정개발 : 기본 배지의 선발에서 Hericium erinaceum의 균사 생육에 적합한 배지를 선발하기 위하여 10여종의 고체배지를 사용하여 균사 생육 및 밀도를 조사한 결과는 YMPG 배지에서 59.8mm/14days로 균사 생육이 가장 우수한 것으로 나타났고, 최적 온도는 20-$25^{\circ}C$ 범위에 가장 생육이 좋았으며, 배지의 pH를 조절하여 균사생육을 조사한 결과, pH는 5.5, 접종비는 전배양액 9%(v/v), 배양에 적합한 배지액량은 50mL, 최적교반속도는 120rpm이었다. 이러한 최적 조건 하에서 배양 경시 변화를 살펴 본 결과 당은 거의 일정한 속도로 감소하는 반면에 건조균체량은 배양 8일째까지 증가하다가 더 이상 변화가 없었다. 2. 발효공정개발: 수분함량이 200%(v/v)에서, pH5에서의 생육속도는 90mm/30 days, $25^{\circ}C$에서의 균사생육속도는 89mm/30days로 각각 H. erinaceum균사의 생육이 가장 우수한 결과를 얻었다 3.추출공정 및 시제품 제조: 녹즙을 생산한 후 폐기되는 신선초박에 액체 종균을 접종하여 ,40일 동안 배양시켜 생육 상태가 우수한 배양생성물만을 선별하여, 열수추출방법으로 10$0^{\circ}C$, 10시간 추출한 것을 음료제조의 원료로 하고, 음료의 기호성을 향상시키기 위해 유기산 및 한방추출물을 첨가하는 균사체음료의 조성비를 얻었다.

Optimization of Submerged Cultivation of Hericium erinaceum (Hericium erinaceum 액체배양의 최적화)

  • Recently, it has been known that Hericium erinaceum is a one of the very useful functional materials with great attention in mushroom processing industry. In present study, a liquid culture which was not studied systematically until now, was conducted as a method of cultivation for H. erinaceum, and also examined the characteristics of the liquid culture and conditions of process optimization. A good basal medium was selected through the cultivation of 16 species mushroom media and the optimum condition for medium and cultivation were chosen by response surface method. From these results, the optimum condition of medium for mushroom was 3% glucose, 0.2% yeast extract/peptone(1:1) and 0.1% $KH_2PO_4/MgSO_4$(1:1) and also the optimal culture condition was obtained at inoculum of 13.42%, temperature of $22.3^{\circ}C$ and pH of 5.7. The mycelial dry weight of 9 g/I was obtained under these conditions and this amount was about 1.7 times higher than that which were cultivated in basal medium for 8 days.

Mycelial Growth and Fruiting Body Formation of Hericium erinaceum in Sawdust and Agricultural By-product Substrates (톱밥 및 농업부산물 이용 배지상에서 노루궁뎅이버섯(Hericium erinaceum)의 균사생장 및 자실체형성)

  • This study was carried out to investigate the suitability of various agricultural by-products as basal substrates for the mycelial growth and fruiting body formation of Hericium erinaceum. For this aim, oak sawdust, cotton waste, sugarcane bagasse, Job's tears, rice hull, Chinese cabbage, and coconut waste were used as sole or mixed substrate(s). Corn waste and rice bran were used as nutrient supplements. The growth and density of mycelium, yield of fruiting body, and biological efficiency were compared among tested substrates colonized by Hericium erinaceum. The best measurement of mycelial growth and density, yield of fruiting body, and biological efficiency in a laboratory test was found in a spawn substrate composed with oak sawdust 80% and rice bran 20%. The suitability of this spawn substrate composition for Hericium fruiting body production was testified through practical tests in plastic bottles (850 ml) in a mushroom farm which had bottle cultivation facility. However, test in a mushroom farm which had plastic bag cultivation facility, best production of Hericium fruiting body (520 g per one bag) was observed in a spawn substrate composed of cotton waste 40%, saw dust 40%, corn waste 10%, and rice bran 10%.

Hypolipidaemic Effect of Hericium erinaceum Grown in Artemisia capillaris on Obese Rats

  • In this study, ethanolic extracts from Hericium erinaceum cultivated with Artemisia capillaris (HEAC) were assessed for their ability to lower the cholesterol levels of male Sprague-Dawley rats fed a high-fat diet. Rats were randomly subdivided into seven test groups. Each group contained eight rats fed a high-fat diet during a growth period lasting 4 wk. Supplementation with the extracts was performed once a day for 2 wk after the high-fat diet. The control group (rats fed a high-fat diet) showed a high efficiency ratio (feed efficiency ratio) value compared to the normal group. Biochemical parameters, including total cholesterol (TC), low-density lipoprotein-cholesterol (LDL-c), and triglyceride (TG) levels dramatically increased in the control group compared to the normal group. High-density lipoprotein-cholesterol (HDL-c) content in the control group was also significantly lower relative to the normal group. Two positive control groups, treated with simvastatin and atorvastatin, had lowered TC, LDL-c, and TG levels, and increased HDL-c content compared to the control group. Treatment with the tested extracts, including HEAC, ethanolic extracts from Hericium erinaceum, and ethanolic extracts from Artemisia capillaris reduced TC, LDL-c, and TG levels and elevated HDL-c content in the hyperlipidemia rats. The atherogenic index and cardiac risk factor values for the HEAC-treated group were 0.95 and 1.95, respectively. Simvastatin- and atorvastatin-treated groups showed atherogenic index values of 1.56 and 1.69, respectively, and cardiac risk factor values of 2.56 and 2.69, respectively. These results show HEAC possesses an ability to cure hyperlipidemia in rats and may serve as an effective natural medicine for treating hyperlipidemia in humans.
