Residual Solvents and Mineral Contents in Ginseng Extracts with Different Extracting Conditions (추출조건에 따른 인삼농축액 중의 잔류용매 및 무기물 함량)

  • This study was conducted to investigate the contents of residual solvents and mineral components(11 kinds) in ginseng extracts with different extracting conditions(5 types) and commercial ginseng extract products(domestic, imported). Fine root was extracted with solution having various ethanol concentration after hexane treatment. Among 5 type extracts, residual solvent(hexane) was detected ginseng extracts treated ethanol mixed with hexane. But extracts that dried after soaked in hexane wasn't detected hexane. Mineral components(Al, Mn, Fe, Cu and Zn) were detected in fine root and 5 types of extracts. The contents of mineral components between fine root and extracts with various extracting conditions were similar, however, extracts that dried after soaked in hexane showed the lower amount in Al, Fe, Pb than the others. In comparison with commercial ginseng ex-tract products(domestic, imported), the distribution pattern of mineral was similar but the contents were a little different.

Antimicrobial and antiviral activity of Saururus chinensis extract by n-Hexane (n-Hexane에 의한 삼백초 추출물의 항균 및 항바이러스 활성)

  • This study was conducted to investigate the antimicrobial, antivirus properties of Saururus chinensis extracts. The n-hexane extracts from Saururus chinensis showed the active antimicrobial activity against gram-positive bacteria. Minimum inhibitory concentration (MIC) of Saururus chiensis n-hexane extracts was 1.25 mg/ml against B. subtilis and 2.5 mg/ml against S. aureus. The cytotoxicity effects on MDBK (Madin-Darby bovine kidney) cell were observed at the various n-hexane extract concentrations. In $TCID_{50}$ assay, 0.6 mg/ml of n-hexane extracts decreased BVD (bovine viral diarrhea) virus by 1.4 log, whereas other extracts did not show antiviral activity. In this study, The results suggested that n-hexane extracts and fractions of Saururus chinensis can be a candidate materal of feed additive to chemical antibiotics and antivirus substances.

Effects of Godulbaegi Leaf Extracts on $CCl_4$-Induced Hepatotoxicity in Rats (고들빼기 잎추출물이 흰쥐의 사염화탄소에 의한 간손상에 미치는 영향)

  • This study was carried out to investigate the effect of godulbaegi leaf extracts on detoxication of liver injury in $CCl_4$-treated rats. We divided normal group(5) and $CCl_4$-treated group and then $CCl_4$- treated group was divided into 6 groups; only $CCl_4$-treated group(CS), aqueous extract pretreated group (CSA), n-butanol extract pretreated group(CSB), Tween 80 pretreated group(CT), n-hexane extract pretreated group(CTH) and ethyl acetate extract pretreated group(CTE). The ratio of liver weight per body weight and the activity of GPT in hexane extract group(CTH) were lowest, similar to the results of S. Godulbaegi n-hexane extract(IS) groups intoxicated by $CCl_4$ had lower values of MDA than CS and CT which are control groups. Histological finding of liver tissue revealed less of necrosis in IS extracts groups than in control groups(CS, CT). From these results, IS extracts could predominently prevent hepat-otoxicity of rats. Especially, hexane extract was effective on the detoxication of liver injury among the other extracts.

Activities of Natural Plant Extracts against HIV-1

  • Anti-HIV-1 activities for the extracts (buthanol, hexane, chloroform, and water) of medicinal plants widely used in the folk medicine were evaluated for screening of anti-AIDS agents. The activities of the extracts to inhibit HIV-1 replication were also analyzed. The 50% effective concentration (EC50) of inhibition activity of the p24 production for chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph showed 5.8, 29.9, and 37.3 2g/ml, respectively, as good activities. Hexane extract of Sophora flavescens, buthanol extract of Tulipa edulis, hexane extracts of Tulipa edulis, Herba ephedra, and Pachyma hoelen Rumph in the 50% cytotoxic concentration ($CC_{50}$) in inhibition activities of recombinant HIV-1 RT showed 12.9, 19.5, 11.6, 12.0, and 36.8 % at concentration of 200 ${\mu}g$/ml, respectively, as good activities. From these results, chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph were very effective against HIV-1 among all extracts tested. Therefore, we expect these plants will be a useful for anti- HIV-1 therapeutics in future.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

Antioxidant Properties of Ginseng (P. ginseng C.A. Meyer) Extracts by Organic Solvent Fractionation

  • The objective of this study was to investigate antioxidant activities of Panax ginseng extracted with various solvents including n-hexane, chloroform, EtOAC, n-butanol and water. Among the various ginseng extracts, ethyl acetate (EtOAC) extracts showed the most powerful scavenging activities against DPPH radicals. Among the other solvent extracts, the butanol extract seemed relatively more effective in scavenging activity, followed by chloroform, water and hexane extracts. Moreover, the highest reducing power and ferrous ion chelating activity were found in the EtOAC extract followed by other extracts of ginseng. EtOAC extracts, which exhibited the best antioxidant activities of all solvent extracts of ginseng, possessed higher concentrations of total phenolics (777.61 mg/100 g) than other extracts. These results suggest that EtOAC extracts of ginseng (P. ginseng C.A. Meyer) have the most effective antioxidant capacity compared to n-hexane, chloroform, n-butanol and water tested in this study, and has important applications for the pharmaceutical and food industries.

The Effect of Prunus Mume Extracts on the Growth of HepG2 and HeLa Cell Lines (간암 및 자궁암 세포주 증식에 미치는 오매 추출물의 영향)

  • This study was undertaken to investigate the inhibitory effect of prunus fume extracts on the growth of Hep G2 and HeLa cells. Prunus mums was extracted using the following solvents hexane, chloroform, ethylacetate, methanol, and hot water. The effect on the growth of each cancer cell line was examined by MTT (3-[4, 5-dimethylthiaeol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, cytotoxicity testing, and microscopic observation. The ethylacetate extracts of Prunus muse at the concentration of 250 $\mu\textrm{g}$/ml exhibited the greatest inhibitory effect on the growth of Hep G2 in the MW assay. In cytotoxicity testing, the treatment of the Hep G2 cells with ethylacetate extracts (1000 $\mu\textrm{g}$/ml for 72 hrs) destroyed 75% of the cells, and morphological changes were also observed. futhermore, the hexane extracts of Prunes muse at the concentration of 250 $\mu\textrm{g}$/ml exhibited the greatest inhibitory effect on the growth of HeLa cells in the MTT assay. The treatment of the HeLa cells with the hexane extracts (1000 $\mu\textrm{g}$/ml for 72 hrs) resulted in the destruction of 68% of the cells. Fibroblasts were not affected by either ethylacetate or hexane extracts of prunus muse.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

Cytotoxicity and Antimicrobial Effects of Extracts from Salvia miltiorrhiza (단삼 추출물의 세포독성과 항균효과)

  • This study was carried out to evaluate cytotoxic effects of Salvia miltiorrhiza extracts on NIH 3T3 fibroblasts and KB cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The comparison of $IC_{50}$ of values of Salvia miltiorrhiza extracts in KB cell lines showed that their susceptibility to these extracts decreased in the following order: hexane extract > chloroform extract > methanol extract> dichloromethane extract > ethyl acetate extract>ethanol extract by the MTT method. The dried roots of Salvia miltiorrhiza was extracted several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentrations (MIC's) of the extract against microorganisms were also examined. Amtimicrobial activity of ketoconazol as reference was compared to those of extracts of hexane, chloroform, dichloromethane, ethyl acetate, ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi. These results suggest that the hexane and chloroform soluble extracts of Salvia miltiorrhiza may be a valuable choice for the studies on the tumor cell lines and growth inhibition activity.

Antioxidant and Mixture Effects of Curry Spices Extracts Obtained by Solvent Extraction (용매에 따른 카레 향신료 추출물의 항산화 효과 및 혼합효과)

  • This study was performed to investigate the antioxidative effects of solvent extracts of eighteen curry spices. Yields of the curry spices extracted by methanol, ethylacetate and hexane were $6.4{\sim}42.9%$, $3.9{\sim}26.2%$ and $1.6{\sim}29.2%$, respectively. Methanol, ethylacetate and hexane extracts of spices were added up to 1,500 ppm in the rice bran oil and antioxidative effects of extracts were tested by Rancimat. Induction periods of spices extracted by methanol were in the order of rosemary(33.5hr)>sage(29.1hr)>ginger(28.6hr)>turmeric(26.9hr)>nutmeg(25.8hr)>oregano(25.6hr)>thyme(25.3hr)>BHT(24.5hr)>control(22.6hr). Those of ethylacetate extracts were as follows: rosemary(43.9hr)>sage(30.9hr)>oregano(29.0hr)>thyme(27.5hr)>ginger(27.1hr)>BHT(24.5hr)>control(22.6hr). Those of hexane extracts were in the order of rosemary(47.4hr)>sage(31.4hr)>oregano(27.3hr)>ginger(25.5hr)>thyme(25.1hr)>control(22.6hr). Solvent extracts of rosemary, sage, oregano, ginger and thyme showed significant antioxidative effects. Two thousands ppm hexane-extract of rosemary was the most effective. Addition of hexane-extracts of rosemary didn't show any synergism with the hexane extracts of sage, oregano, ginger, and thyme.
