The Effect of Herbal Formula KH-305 on Erectile Dysfunction in Hyperlipidemic Rat (고지혈증 흰쥐에서 발기부전에 대한 생약제제 KH-305의 효과)

  • This study was designed to investigate effects of KH-305 on erectile dysfunction in hyperlipidemic rat. KH-305 has been evaluated for antihyperlipidemic and antierectile dysfunction effects on experimental hyperlipidemic rats induced by high cholesterol-diet. After oral administration of KH-305 water extract (50, 100, 200, 300 mg/kg) to hyperlipidemic rats for 8 weeks, the variables including body weight, total cholesterol, HDL-, and LDL-cholesterol levels in serum, the expression of eNOS and nNOS in penis were measured. Also, erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve. Oral administration of KH-305 extracts significantly inhibited the increase of serum total cholesterol and LDL-cholesterol levels and the decrease of serum HDL-cholesterol level in hyperlipidemic rats induced by high fat diet. The penile expression level of the two enzymes (eNOS, nNOS) were increased significantly after oral administration of the KH-305 extracts. Erectile function after 10 volts stimulation was significantly decreased in the hyperlipidemic rat compared with the normal rat, but increased in KH-305 group compared with hyperlipidemic group. These results suggest that KH-305 is effective for erectile dysfunction in hyperipidemia.

Neuroprotective Effect Yanggyuksanhwa-tang on Cerebral Infarction Induced by MCAO in Hyperlipidemic Rats (양격산화탕(凉膈散火湯)이 고지혈증 흰쥐의 중대뇌동맥 폐쇄에 의한 뇌경색에 미치는 영향)

  • Objectives : Yanggyuksanhwa-tang is a prescription used for cerebral infarction clinically it is known that this formula reduces body fat, serum cholesterol and triglyceride in hyperglycemia and obesity patients. According to previous research data, controlling these types of lipid is considered to decrease the risk of cerebral infarction. Based on this fact, we investigated the relationship between hyperlipidemia and cerebral infarction, and the effect of Yanggyuksanhwa-tang on hyperlipidemic cerebral infarction. Methods : We induced cerebral infarction by middle cerebral artery occlusion (MCAO) in high-fat diet rats, and the rats were administered Yanggyuksanhwa-tang. Results : Infarct area and serum lipid were measured, and the level of elements such as c-Fos, Bax and caspase-3 in penumbra of infarct were expressed by immunohistochemical staining. Conclusions : Yanggyuksanhwa-tang showed neuroprotective effect through preventing neuronal cell apoptosis as well as reducing serum lipid level in hyperlipidemic condition.

Effects of Herbal-cheonggukjang on Serum Cholesterol Levels in Rats (생약초 청국장이 흰쥐의 혈중 콜레스테롤 함량에 미치는 영향)

  • Ojectives : Cheonggukjang(natto) is known to have anti-hyperlipidemic action. This study was designed to investigate whether Herbal-cheonggukjang (HC, Herbal-natto) is more effective than cheonggukjang only, Methods : We investigated anti-hyperlipidemic effects of HC in hyperlipidemic rats induced by high-fat diet in terms of observation on levels of total cholesterol, HDL-cholesterol and triglyceride in serum. HC was made by cheonggukjang added Codonopsis Lanceolata, Houttuynia cordata and Lentinus edodes in indicated concetrations. Results : In this experiment, hyperlipidemic rats showed elevated levels of total cholesterol and triglyceride, and also showed lowered level of HDL-cholesterol. Oral administration of HC decreased levels of total cholesterol and triglyceride, Cheonggukjang(natto) did not affect levels of total cholesterol, HDL cholesterol and triglyceride. Conclusions : In these results, oral administration of HC decreased total cholesterol and triglyceride effectively. These results demonstrate that HC is useful to modulate cholesterol and triglyceride in serum.

Effect of Lactobacillus rhamnosus hsryfm 1301 on the Gut Microbiota and Lipid Metabolism in Rats Fed a High-Fat Diet

  • Accumulating evidence indicates that lactic acid bacteria could improve host physiology and lipid metabolism. To investigate the effect of the gut microbiota on host lipid metabolism, a hyperlipidemic rat model was established by feeding rats a high-fat diet for 28 days, and the gut microbiota of the rats was analyzed using real-time PCR before and after administration of Lactobacillus rhamnosus hsryfm 1301 and its fermented milk for 28 days. The findings showed that the Lactobacillus spp., Bifidobacterium spp., Bacteroides spp., and Enterococcus spp. content in the hyperlipidemic rats gut was increased significantly (p < 0.05), while the Clostridium leptum and Enterobacter spp. content was decreased significantly after intervening with L. rhamnosus hrsyfm 1301 and its fermented milk for 28 days (p < 0.05). Furthermore, the lipid levels of the serum and the liver were decreased significantly (p < 0.05) and the fecal water content was increased significantly (p < 0.05) in the hyperlipidemic rats after the intervention, and hepatocyte fatty degeneration of liver tissues was also prevented. A positive correlation was observed between the Clostridium leptum content and the level of serum cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, and a negative correlation was observed between the Enterobacter spp. content and the Lactobacillus spp. and Bifidobacterium spp. content in the hyperlipidemic rats gut. These results suggest that the gut microbiota and lipid metabolism of hyperlipidemic rats could be improved by supplementation with L. rhamnosus hsryfm 1301 and its fermented milk.

Effects of Salviae miltiorrhizae Radix (SMR) on Serum Lipid Level in Hyperlipidemic Rats (단삼(丹蔘)이 고지혈증 흰쥐의 혈중 지질 변화에 미치는 영향)

  • Objective : Salviae miltiorrhizae Radix (SMR) has widely used to treat patients with cardiovascular diseases such as coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of SMR on changes in serum cholesterol and protective effects on liver tissue damage in Hyperlipidemic rats. Methods : The present author investigated changes in serum glucose, cholesterols, AST/ALT and histopathological changes of liver tissue by oral administration of SMR in Rats. Results : In this study, body weights of hyperlipidemic rats induced high fat diet did not changed, and treatment with SMR did not affect body weights in hyperlipidemic rats. For experimental period, Food and Water uptake in SMR administered group were the same as those in hyperlipidemic control group. In this experiment, treatment with SMR decreased total cholesterol and AST in serum which elevated by high fat diet respectively. In addition, SMR administration protected liver tissue from damage induced by induction of hyperlipidemia. Conclusions: These results suggest that SMR is useful to treat patients with disease related to cardiovascular diseases including hyperlipidemia, because SMR can decrease cholesterol and AST in serum and also have non-specific protective effect on tissues including liver.

  • The HWND_G02 (Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino) and HWND_G03 (Cinnamonum cassia Blume, Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino, Allium macrostemon Bunge) are new natural mixture composed with several oriental herbs. The aim of the present study was to investigate the effects of HWND extracts on high cholesterol diet (HCD)-induced hyperlipidemic rats. Male Sprague-Dawley rats were divided into five groups: control, HCD, atorvastatin (5 mg/kg, po), ethanolic extracts of HWND_G02 (1,000 mg/kg, po) and HWND_G03 (1,000 mg/kg, po) were administered to the HCD-induced hyperlipidemic rats for 4 weeks to evaluate their anti-hyperlipidemic activities. HWND extracts markedly decreased body and liver wight gain, and recovered serum lipid levels, such as total cholesterol (TC), triglycerides (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) in the HCD-induced hyperlipidemic rats. Furthermore, the lipid levels (TC and TG) and the lipid accumulation were significantly lowered in the liver tissue of HWND-administrated rats. After a HCD, each group had a lower atherogenic index (AI) compared to the HCD group. In conclusion, these data suggest that HWND extracts could be the candidate for the material to prevent hyperlipidemia.

Effect of Uncooked Food on the Hyperlipidemic Rats (생식이 고지혈증 흰쥐에 미치는 영향)

  • The hypolipidemic effect of the diet containing uncooked food was evaluated in hyperlipidemic rats. Body weightgain, food intake and food efficiency ratio were not significantly different among the diet groups examined. Significant decrease in plasma total cholesterol(8.8%), triglyceride(10.0%) and LCL-cholesterol(34.8%) were observed in uncooked food diet groups, and the liver total cholesterol and triglyceride level to the extent of 8.7% and 16.7%, respectively. Uncooked food groups substantially increased the plasma HDL to total cholesterol ratio(47.2%) and decreased the atherogenic index(37.6%) as well. Gut transit time for uncooked food was significantly higher, up to 104 minutes, compared to that of control diet group. These results suggest that uncooked food may have beneficial effects on blood lipid level and gut transit time in hyperlipidemc rats.

  • The objective of this study was to investigate the effects of the Crataegus pinnatifida BUNGE extract supplementation of non esterified fatty acid (NEFA), concentrations of serum protein and electrolyte in sera on the hyperlipidemic rats. Concentrations of NEFA and globulin were remarkably lower in the Crataegus pinnatifida BUNGE extract group (HW group) than in the hyperlipidemic group (HD group), but no difference between control group (CO group) and extract of Crataegus pinnatifida BUNGE supplement in control group (NW group). However, concentrations of electrolyte K and A/G were higher in the HW group than HD group. Concentrations of total protein, albumin, electrolyte of total Ca, Pi, Na and Cl were no difference between HW group than HD group. The results indicate that Crataegus pinnatifida BUNGE extract was in the improvement of hyperlipidemic rats.

  • Anti-hyperlipidemic effect of Sophorae Fructus in male Spague-Dawley rats induced by high-rich diet were studied. Oral administration of Sophorae Fructus extract significantly decreased serum total cholesterol (TC), LDL-cholesterol, triglyceride (TG), GOT and GPT levels, and incresed serum HDL-cholesterol level and also decreased the level of liver TC, TG, GOT and GPT. These results suggest that Sophorae Fructus extract is effective for the hyperlipidemia.

  • This study investigated the inhibitory effects of sea mustard on high-fat diet-induced obesity and hyperlipidemia in Sprague-Dawley rats. Sea mustard (Undaria pinnatifida) powder, sea mustard ethanol extract, and sea mustard ethanol-extracted residue were tested. The ethanol extracted residue had the most beneficial anti-hyperlipidemic activity. Alginate in the sea mustard was considered to be the key component. The ethanol-extracted residue of sea mustard also had antioxidant activity, which may be effective in preventing hyperlipidemia by increasing the enzymatic activity of superoxide dismutase, which can remove active oxygen from the bloodstream.
