Anti-hyperlipidemic Effect of Vitex doniana Ethanol Extract in Poloxamer Induced Hyperlipidemia

  • To determine the effect of Vitex doniana (leaves stem and root bark) ethanolic extracts on lipid profiles of Poloxamer 407 (P407) induced hyperlipidemic and normal rats. Fifty four mixed sex rats weighing 100-200g were divided into nine groups comprising six animals per group. At the end of the 21 day, the animals were sacrificed and blood sample were collected for determination of serum levels of: Total cholesterol (TC), Triacylglycerides (TAG), High-density lipoprotein cholesterol (HDL-c) and Low-density lipoprotein cholesterol (LDL-c). The studies showed that all induced treated groups significantly (P<0.05) lower serum levels of TC, TAG, LDL-c and significantly (P<0.05) increased HDL-c when compared to the P407 induced hyperlipidemic control. The normal treated groups showed no significant (P>0.05) difference in the serum levels of TC, TAG, LDL and HDL when compared to the normal control group. Calculation of atherogenic risk predictor indices of the induced treated groups showed that all the extracts significantly (P<0.05) lowered the LDL-c/HDL-c, log (TAG/HDL-c) and significantly (P<0.05) increased HDL-c/TC ratio when compared to the P407 induced hyperlipidemic control group. The atherogenic risk predictor indices of normal treated groups showed no significant difference (P>0.05) in LDL-c/HDL-c, Log (TAG/HDL-c) and HDL-c/TC ratio when compared to the normal control group. The study demonstrates the phytotherapeutic effect of Vitex doniana (leaves, stem and root bark) ethanolic extract in poloxamer 407 induced hyperlipidemia.

Effects of Web-Based Nutrition Counseling on Dietary Behavior and Food Intake in Hyperlipidemic Patients (고지혈증 환자의 식행동 및 식품섭취에 웹기반 영양상담의 영향)

  • The purpose of this study was to investigate the effect of the web-based nutrition counseling on the dietary behavior and food intake of hyperlipidemic patients and to evaluate the web-based nutrition counseling program. Forty hyperlipidemic patients, twenty of them were hypercholesterolemia and the other twenty were hypertriglyceridemia, participated in the web-based nutrition counseling program. At the first nutrition counseling, the patients were interviewed and then follow-up nutrition counselings were accomplished four times during eight weeks through a web-based internet program. The web-based nutrition counseling program was consisted of diet management, nutrition education and nutritional assessment and the follow-up counseling program. The dietary behavior score increased significantly in both groups after the nutrition counseling program(p<0.01). The diet therapy knowledge score also increased significantly (p<0.01). For food frequency, vegetable (p<0.01), fruit (p<0.05) and seaweed (p<0.01) consumption increased significantly, while meat (p<0.01), egg (p<0.01), snack (p<0.01) and instant food (p<0.01) intake decreased significantly in both groups after the nutrition counseling program. The evaluation of the web-based nutrition counseling program by the patients showed above average in all domains, specially the evaluation scores of younger aged patients habituated to practice computer were better than those of older aged patients. Therefore, this study shows that the web-based nutrition counseling program is effective for improving dietary behavior and food consumption the patients. In addition, these results indicate that internet presents us with potential as a new medium for nutrition counseling in informationized society.

The Anti-hyperlipidemic Effect and Constituents of the 19${\alpha}$-Hydroxyursane-type Triterpenoid fraction Obtained from the Leaves of Rusus crataegifolius

  • To demonstrate anti-hyperlipidemic activity of the 19${\alpha}$-hydroxyursane-type triterpenoid (19${\alpha}$-HUT)-rich fraction, this fraction was prepared from the extract of Rubus crataegifolius leaves. This fraction was found to have anti-hyperlipidemic effect in a high fat diet-induced rat model from the observation of reduction of abdominal fat pad weights, atherogenic index and hypercholesterolemia at 30 and 60 mg/kg (p.o.) The 19${\alpha}$-HUT fraction was subjected to SiO$_2$, ODS, and/or Sephadex LH-20 column chromatography to yield a new triterpenoid (1) called pomolic acid ester along with nine known triterpenoids which are all 19${\alpha}$-HUTs: euscaphic acid (2), tormentic acid (3), 23-hydroxytormentic acid (4), kaji-ichigoside F$_1$ (5), rosamultin (6), niga-ichigosides F$_1$ (7) and F$_2$ (8), suavissimoside F$_1$ (9) and coreanoside F$_1$ (10). The structure of compound 1 was established as 28-O-formyl-3,19-dihydroxyurs-12-en-28-oic acid on the basis of 2D-NMR spectroscopic data and mass spectrum. Compound 1 was isolated for the first time from natural sources.

Effect of Nelumbo nucifera Leaves on Hyperlipidemic and Atherosclerotic Bio F1B Hamster (고지혈증 및 동맥경화 감응 Bio F1B 햄스터에서 연잎(Nelumbo nucifera)의 지질저하 효과)

  • Nelumbo nucifera Gaertner leaf (NNL) has been known that is effective to lowering blood pressure and hyperlipidemic levels. But, its pharmacological actions have not been demonstrated. Therefore, the present research was performed to find the effect of NNL on hyperlipidemic actions in Bio F1B hamster as a model for hyperlipidemia and atherosclerosis. The hyperlipidemic hamster were induced by basal diet containing 10% coconut oil and 0.05% cholesterol with high fat atherogenic diet(HFAD). A control group and treated groups NNL100, 200, 400 mg/kg/day were fed a HFAD, but a normal group was fed a basal diet only. In conclusion, the serum total cholesterol levels were significantly declined on day 40 in F1B hamster given the NNL extracts (200, 400 mh/kg/day) with p<0.05, when compared with a control value of HFAD fed hamsters. The hepatic TC level was significantly lower in NNL 200 and 400 mg/kg/day group than of control hamster on day 40. But, the HDL levels were not changed between control and treated NNL groups. The TG levels of NNL treated groups showed a decrease tendency compare to the control value. In addition, accumulation of fatty level showed a slight decline to NNL treated groups in aortic arch.

Effects of Ephedra Herba ethyl-acetate fraction on high fat diet induced hyperlipidemic mice (마황 에틸아세테이트 분획물이 고지방 식이로 유발된 생쥐의 지질대사에 미치는 영향)

  • Objective : Ephedra Herba has been widely used for patients with common cold, asthma in eastern countries, especially china japan and korea. Recently it has been also used for obesity in clinic with high frequency. This study was designed to investigate the effects of Ephedra Herba ethyl-acetate fraction (EEAF) on hyperlipidemic mice. Method : Effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured in this experiment, and in addition, histopathological and gene expression changes in liver tissue was also observed. Results : In our study, EEAF did not affect weight gain in hyperlipidemic mice. Oral administration of EEAF lowered levels of total cholesterol which were elevated by induction of hyperlipidemia. And administration of EEAF lowered fasting blood glucose significantly. By carrying out ontological analysis, large numbers of genes were identified in up or down regulated genes. The expression of the genes that were altered in response to high-fat diet was restored to normal levels in EEAF treated mice, with a recovery rate of 49%. And it was considered that fatty acid metabolism was one of important key pathway of the recovery. Conclusion : Results in our study suggest that EEAF can prevent obese and through regulation of dyslipidemia and hyperglycaemia.

Effects of Kyeongok-go and Its Two Added Precriptions on Hyperlipidemic Rats Induced by High-fat Diet (경옥고(瓊玉膏) 및 그 가미방(加味方)이 고지방 식이로 유발된 흰쥐의 고지혈증 병태에 미치는 영향)

  • This study was designed to investigate the antihyperlipidemic activity of aqueous extracts of Kyeongokgo (KOG), Kyeongokgo Dansambang (KDB), and Kyeongokgo Gamibang (KGB) on the hyperlipidemic rats induced by high-fat diet. The animals were divided into 4 groups of the hyperlipidemic control group, the KOG-fed group, the KDB-fed group and the KGB-fed group, and received oral administration of each prescription with diet for a period of six weeks. The changes in the body weight, the amount of food intake, the liver weight, the epididymal fat weight, and the plasma levels of total cholesterol, triglyceride, HDL-cholesterol and LDL-cholesterol were measured. In all experimental groups, the weight of liver and epididymal fat decreased significantly. There was a significant decrease in the amount of food intake, the weight of liver and epididymal fat, the plasma level of total cholesterol, and HDL-cholesterol in the KDB-fed and the KGB-fed groups. In addition, KGB-fed group represented a significant decrease in the triglyceride. These results suggest that the aqueous extracts of Kyeongokgo Dansambang and Kyeongokgo Gamibang have a good antihyperlipidemic activity and a potential to treat hyperlipidemia.

Effect of the MeOH Extract of Adventious Root Culture of Panax ginseng on Hyperlipidemic Rat Induced by High Fat-rich Diet (조직배양산삼 부정근 메탄올 추출물이 식이성 고지혈증에 미치는 영향)

  • We have investigated the effect of MeOH extract of the adventious root culture of Panax ginseng on hyperlipidemic rats induced by fat-rich diet. We also analyzed and compared the content ginsenoisdes of the cultured ginsengs by HPLC. After oral administration of the MeOH extract to hyperlipidemic rats for four weeks, the variables including body weight, cholesterol, HDL, LDL, and triglyceride levels in serum were measured. One of the cultured ginseng roots (CBN3) decreased cholesterol and LDL-cholesterol and increased HDL-cholesterol levels in serum. HPLC analysis demonstrated that CBN3 contains more $Rb_1$ and Rd ginsenosides than any other cultured ginsengs and cultivated ginseng. These results suggest that CBN3 with high contents of $Rb_1$ and Rd may be useful in lowering blood cholesterol.

Lactic Acid Bacteria Increases Hypolipidemic Effect of Crocin Isolated from Fructus of Gardenia jasminoides

  • The fructus of Gardenia jasminoides Ellis (GF) has been widely used as a natural colorant in Asian countries, and also as a Chinese traditional medicine for its homeostatic, antiphlogistic, analgesic, and antipyretic effects. In the present study, its main component, crocin, was fermented with lactic acid bacteria (LAB) and their antihyperlipidemic activity was measured. The GF extract, fermented GF (F-GF), crocin, and fermented crocin (F-crocin) significantly inhibited the increase of serum triglyceride (TG) level in corn oil feeding-induced triglyceridemic mice, as well as that of serum TG and total and LDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. These agents also showed hypolipidemic activity in hyperlipidemic mice induced by high fat diet for 5 weeks. F-GF and F-crocin exhibited more potent hyperlipidemic effects than GF and crocin. The results suggest that the hypolipidemic effect of GF and crocin can be synergistically activated by LAB, and that F-GF and F-crocin may improve hyperlipidemia in clinic, compared with GF and crocin.

Effect of Methanolic Extract of Pachymeniopsis elliptica on Lipids Component of Hyperlipidemic Rats (해조류 참도박의 메탄올 추출물이 고지방식이 흰쥐의 지질성분에 미치는 영향)

  • Hypolipidemic effect of methanolic extract of marine algae, Pachymeniopsis elliptica was examined in hyperlipidemic rats. Male $Sprague-Dawley(120\pm5g)$ rats were divided into five groups and fed high fat diets for four weeks. Each group was orally administered with methanolic extract of P. elliptica (PEM, 250, 500mg/kg), benzofibrate(BZF, 30mg/kg) and cholestyramine resin(CSR, 100mg/kg) daily for one week. Significant decreases in cholesterol, total lipid and triglyceride of both blood and liver were observed by the administration of the extract. But the levels of LDL- and HDL-cholesterol and the activity of lipase were not changed. Such results suggest that the extract may promote the fecal excretion of bile acid in hyperlipidemic rats.

Anti-hyperlipidemic and Anti-oxidant effects of Kgihwaetang-gamibang in High Cholesterol Diet-induced Mice Model (益氣化瘀湯加味方이 고지방 식이로 유발된 생쥐의 항고지혈 및 항산화에 미치는 효과)

  • This study was conducted to investigate the anti-hyperlipidemic and anti-oxidative activities of Ikgihwaetang-gamibang (IK). We observed the effect of IK on lipid factor, inflammation factor, body weight and organ weight, and liver after administering IK to hyperlipidemia mice triggered by high fat diet. Additionally, we used Raw 264.7 cells to investigate the changes in anti-oxidative activity and the effects of anti-inflammation. Administration of IK resulted in reduction of total cholesterol, LDL cholesterol, TG, and glucose in blood serum while a clear increase in HDL cholesterol. The production of IL-1β and IL-6, and ROS and NO also decreased. The results showed that IK has anti-hyperlipidemic and anti-oxidative effects, and suggested its potential use in prevention and treatment of hyperlipidemia and its associated diseases.
